Single nucleotide polymorphisms (SNPs) are valuable DNA markers due to their abundance and potential for use in automated high-throughput genotyping. Numerous SNP genotyping assays have been developed. In this report, one of effective and high throughput SNP genotyping assays, which was named the template-directed dye-terminator incorporation with fluorescence polarization detection (FP-TDI) was described. Although the most of this assay succeed, the objective of this work was to determine the reasons for the failures, find ways to improve the assay and reduce the running cost. Ninety $F_2$-derived soybean, Glycine max (L.) Merr., RILs from a cross between 'Pureunkong' and 'Jinpumkong 2' were genotyped at four SNPs. FP measurement was done on $Victot^3$ microplate reader (perkinelmer Inc., Boston, MA, USA). Increasing the number of thermal cycles in the single-base extension step increased the separation of the FP values between the products corresponding to different genotypes. But in some assays, excess of heterozygous genotypes was observed with increase of PCR cycles. We discovered that the excess heterozygous was due to misincorporation of one of the dyeterminators during the primer extension reaction. After pyrophosphatase incubation and thermal cycle control, misincoporation can be effectively prevented. Using long amplicons instead of short amplicons for SNP genotyping and decreasing the amount of dye terminator and Acyclopol Taq polymerase to 1/2 or 1/3 decreased the cost of the assay. With these minor adjustments, the FP-TDI assay can be used more accurately and cost-effectively.
The design of a flat-field XUV spectrograph is optimized for a high throughput, aberration-corrected spectral image in the wavelength region of 50-300 $\AA$ The varied-line spacing (VLS) concave grating theory for an XUV spectrograph with a toroidal mirror in front of an entrance slit is derived. Since the derived theory includes the arbitrary shaped source, it is able to correct the limit of the simple optimization theory which considers only a point source at the center of the entrance slit. The reflection matrix at the toroidal mirror and the diffraction matrix at VLS grating are derived and compared with those of a holographic grating. The absolute energy efficiency of a flat-field spectrograph is calculated by considering the reflectivities of the toroidal mirror and the Au coated concave grating and the grating efficiency. The alignment sensitivity of the toroidal mirror and the concave grating is investigated, and the method to achieve the best imaging of XUV spectrum is discussed. The calculated resolving power of the flat-field XUV spectrograph is more than 4000 in the aberration-corrected wavelength range. The focused spot size at the dispersion plane is less than $20\mu \textrm m\times \mu \textrm m$at the wavelength 100$\AA$ It is shown that a high throughput characteristic can also be achieved through a careful adjustment of alignment parameters.
Nano imprint lithography(NIL) is a cost-efficient, high-throughput processing technique to transfer nano-scale patterns onto thin polymer films. Polymers used as the resist include UV cured resins as well as thermoplastics such as polymethyl-methacrylate(PMMA). In this study, an analytic investigation was performed for the NIL process of transferring nano scale patterns onto polymeric films. Process optimization calls for a thorough understanding of resist flow during the process. We carried out 2D and 3D numerical analyses of resist flow during NIL process. The simulation incorporated continuum-hypothesis and the effects of surface tension were taken into account. For a more effective prediction of free surface, fixed grid scheme with the volume of fluid (VOF) method were used. The simulation results were verified with experimental results qualitatively. And the parametric study was performed for various process conditions.
BCH code is one of the most widely used error correcting code for the detection and correction of random errors in the modern digital communication systems. The conventional BCH encoder that is operated in bit-serial manner cannot adequate with the recent high speed appliances. Therefore, parallel encoding algorithms are always a necessity. In this paper, we introduced a new systolic array type BCH parallel encoder. To study the area and speed, several parallel factors of the systolic array encoder is compared. Furthermore, to prove the efficiency of the proposed algorithm using tree-type structure, the throughput and the area overhead was compared with its counterparts also. The proposed BCH encoder has a great flexibility in parallelization and the speed was increased by 40% than the original one. The results were implemented on synthesis and simulation on FPGA using VHDL.

The use of high throughput screening (HTS) in drug development is principally for the selection new drug candidates or screening of chemical toxicants. This system minimizes the experimental environment and allows for the screening of candidates at the same time. Umbilical cord-derived stem cells have some of the characteristics of fetal stem cell and have several advantages such as the ease with which they can be obtained and lack of ethical issues. To establish a HTS system, optimized conditions that mimic typical cell culture conditions in a minimal space such as 96 well plates are needed for stem cell growth. We have thus established a novel HTS system using human umbilical cord derived-mesenchymal stem cells (hUC-MSCs). To determine the optimal cell number, hUC-MSCs were serially diluted and seeded at 750, 500, 200 and 100 cells per well on 96 well plates. The maintenance efficiencies of these dilutions were compared for 3, 7, 9, and 14 days. The fetal bovine serum (FBS) concentration (20, 10, 5 and 1%) and the cell numbers (750, 500 and 200 cells/well) were compared for 3, 5 and 7 days. In addition, we evaluated the optimal conditions for cell cycle block. These four independent optimization experiments were conducted using an MTT assay. In the results, the optimal conditions for a HTS system using hUC-MSCs were determined to be 300 cell/well cultured for 8 days with 1 or 5% FBS. In addition, we demonstrated that the optimal conditions for a cell cycle block in this culture system are 48 hours in the absence of FBS. In addition, we selected four types of novel small molecule candidates using our HTS system which demonstrates the feasibility if using hUC-MSCs for this type of screen. Moreover, the four candidate compounds can be tested for stem cell research application.

High data rates in long-term evolution (LTE) networks can affect the mobility of networks and their performance. The speed and motion of user equipment (UE) can compromise seamless connectivity. However, a proper handover (HO) decision can maintain quality of service (QoS) and increase system throughput. While this may lead to an increase in complexity and operational costs, self-optimization can enhance network performance by improving resource utilization and user experience and by reducing operational and capital expenditure. In this study, we propose the self-optimization of HO parameters based on fuzzy logic control (FLC) and multiple preparation (MP), which we name FuzAMP. Fuzzy logic control can be used to control self-optimized HO parameters, such as the HO margin and time-to-trigger (TTT) based on multiple criteria, viz HO ping pong (HOPP), HO failure (HOF) and UE speeds. A MP approach is adopted to overcome the hard HO (HHO) drawbacks, such as the large delay and unreliable procedures caused by the break-before-make process. The results of this study show that the proposed method significantly reduces HOF, HOPP, and packet loss ratio (PLR) at various UE speeds compared to the HHO and the enhanced weighted performance HO parameter optimization (EWPHPO) algorithms.

The design of cluster-based routing protocols is necessary for Wireless Sensor Networks (WSN). But, due to the lack of features, the traditional methods face issues, especially on unbalanced energy consumption of routing protocol. This work focuses on enhancing the security and energy efficiency of the system by proposing Energy Efficient Based Secure Routing Protocol (EESRP) which integrates trust management, optimization algorithm and key management. Initially, the locations of the deployed nodes are calculated along with their trust values. Here, packet transfer is maintained securely by compiling a Digital Signature Algorithm (DSA) and Elliptic Curve Cryptography (ECC) approach. Finally, trust, key, location and energy parameters are incorporated in Particle Swarm Optimization (PSO) and meta-heuristic based Harmony Search (HS) method to find the secure shortest path. Our results show that the energy consumption of the proposed approach is 1.06mJ during the transmission mode, and 8.69 mJ during the receive mode which is lower than the existing approaches. The average throughput and the average PDR for the attacks are also high with 72 and 62.5 respectively. The significance of the research is its ability to improve the performance metrics of existing work by combining the advantages of different approaches. After simulating the model, the results have been validated with conventional methods with respect to the number of live nodes, energy efficiency, network lifetime, packet loss rate, scalability, and energy consumption of routing protocol.

Compared to a terrestrial communication, the high BER(Bit Error Ratio) and low channel bandwidth are the major factor of throughput degradation due to characteristics of underwater channel. Therefore, a MAC protocol must be designed to solve this problem in UWASNs(Underwater Acoustic Sensor Networks). MAC protocols for UWASNs can be classified into two major types according to the contention scheme(Contention-free scheme and Contention-based scheme). In large scale of sensor networks, a Contention-based scheme is commonly used due to time-synchronize problem of Contention-free scheme. In the contention-based scheme, Each node contends with neighbor nodes to access network channel by using Back-off algorithm. But a Slot-Time of Back-off algorithm has long delay times which are cause of decrease network throughput. In this paper, we propose a new scheme to solve this problem. The proposed scheme uses variable Slot-Time instead of fixed Slot-Time. Each node measures propagation delay from neighbors which are used by Slot-time. Therefore, Slot-Times of each node are optimized by considering node deployment. Consequently, the wasted-time for Back-off is reduced and network throughput is improved. A new mac protocol performance in throughput and delay is assessed through NS3 and compared with existing MAC protocol(MACA-U). Finally, it was proved that the MAC protocol using the proposed scheme has better performance than existing MAC protocol as a result of comparison.

In this paper, image filter optimization method based on common sub-expression elimination is proposed for low-power image feature extraction hardware design. Low power and high performance object recognition hardware is essential for industrial robot which is used for factory automation. However, low area Gaussian gradient filter hardware design is required for object recognition hardware. For the hardware complexity reduction, we adopt the symmetric characteristic of the filter coefficients using the transposed form FIR filter hardware architecture. The proposed hardware architecture can be implemented without degradation of the edge detection data quality since the proposed hardware is implemented with original Gaussian gradient filtering algorithm. The expremental result shows the 50% of multiplier savings compared with previous work.

Wireless tactical network (WTN) is the most important present-day technology enabling modern network centric warfare. It inherits many features from WMNs, since the WTN is based on existing wireless mesh networks (WMNs). However, it also has distinctive characteristics, such as hierarchical structures and tight QoS (Quality-of-Service) requirements. Little research has been conducted on hierarchical protocols to support various QoS in WMN. We require new protocols specifically optimized for WTNs. Control packets are generally required to find paths and reserve resources for QoS requirements, so data throughput is not degraded due to overhead. The fundamental solution is to adopt topology aggregation, in which a low tier node aggregates and simplifies the topology information and delivers it to a high tier node. The overhead from control packet exchange can be reduced greatly due to decreased information size. Although topology aggregation is effective for low overhead, it also causes the inaccuracy of topology information; thus, incurring low QoS support capability. Therefore, we need a new topology aggregation algorithm to achieve high accuracy. In this paper, we propose a new aggregation algorithm based on star topology. Noting the hierarchical characteristics in military and hierarchical networks, star topology aggregation can be used effectively. Our algorithm uses a limited number of bypasses to increase the exactness of the star topology aggregation. It adjusts topology parameters whenever it adds a bypass. Consequently, the result is highly accurate and has low computational complexity.
