Effect of Nifedipine on Endocrine and Cardiovascular Responses to Angiotensin II in SHR Fed with Different Sodium Diets

  • Effects of a voltage dependent calcium channel antagonist, nifedipine, on the responses of blood pressure, and secretion of atrial natriuretic peptide (ANP) and aldosterone to angiotensin II (Ang II) were compared in male Wistar and spontaneously hypertensive rats (SHR). A low, control or high sodium diet (2, 10 or 25 mmol Na/100 g diet) was fed for 6 weeks from the age of 6 weeks. On the morning of the experiment catheters were inserted under ether anesthesia in the femoral artery for pressure recording and blood sampling, and in the femoral vein for drug infusion. Ang II was infused at a rate of 250 ng/kg/min for 20 min. Nifedipine mixed with Ang II was infused at a rate of $16{\mu}g/kg/min$ for 20 min. Arterial blood samples were collected before and after infusion of Ang II with or without nifedipine. The control plasma level of aldosterone was inversely related to the amount of salt intake, whereas the plasma ANP level was not different between the salt groups. SHR showed a higher basal plasma ANP but a lower aldosterone concentration than Wistar rats. Infusion of Ang II produced a significant increase in blood pressure and plasma levels of aldosterone and ANP: The % increase was not significantly different either between the salt groups or between SHR and Wistar rats. SHR showed a greater pressor response to Ang II but a remarkably smaller decrease in heart rate after Ang II infusion than Wistar rats, With increasing sodium intake, the effect of Ang II on aldosterone secretion was decreased, whereas that on ANP secretion or blood pressure was not changed. Nifedipine decreased the responses of blood pressure and heart rate to Ang II in all groups. Nifedipine caused almost a complete inhibition of Ang II induced ANP secretion, but only a partial inhibition of Ang II induced aldosterone secretion or vasoconstriction. These results indicate that calcium dependent processes were involved in Ang II induced vasoconstriction, and secretions of aldosterone and ANP. However, the calcium dependent process far ANP secretion was considerably different from that for aldosterone secretion or vasoconstriction evoked by ang II. The ang II induced increase in ANP secretion appeared to be caused primarily by activating voltage-dependent calcium channels, whereas Ang II induced aldosterone secretion and vasoconstriction was not.

Nickel Toxicity and Its Interaction with Zinc, Copper and Lead in Growing Chicks (초생추에서의 니켈의 독성과 아연·구리 및 납과의 상호작용)

  • Nickel toxicity and interactions of nickel with zinc, copper, and lead were studied in glowing chicks fed supplemented diet. Feed intake and growth rate of the chick were reduced by 250mg nickel as a sulfate salt per kg of feed. The toxicity of nickel was decreased by zinc or copper supplementation, but not lead. High nickel feed increased nickel level in kidney and decreased zinc levels in tibia and plasma. However, low zinc levels in tibia and in plasma were reversed by zinc supplementation. Hemoglobin, packed cell volume, and aortic elastin content were increased in chicks fed nickel. These results suggest that nickel toxicity is induced by interference with zinc metabolism.

The Effects of Dietary Chinese Cabbage Kimchi Juice on the Lipid Metabolism and Body Weight Gain in Rats Fed High-Calories-Diet (배추김치 즙 투여가 고 열량 섭취 흰쥐의 지질대사 및 체중변화에 미치는 영향)

  • Weanling male rats of Wistar strain were fed relatively high-calories-diet containing 1% (K$_1$ group), 5% (K$_{5}$ group) and 10% Chinese cabbage kimchi juice (K$_{10}$ group) for 4 weeks to study their effects on body weight gain rate, serum lipids levels and other blood biochemical parameters. In this study it was found out that significant decrease in body weight gain rate, diet efficiency ratio, serum triglyceride level, serum total cholesterol level, blood glucose level and LDL-cholesterol level was observed in $K_{10}$ group as compared to control group fed only high-calories-diet. Significant increase in HDL-cholesterol level, GPT activity, GOT activity and creatinine level was found in $K_{10}$ group as compared to control group. But creatinine level, GPT and GOT activity of $K_{10}$ group were within the normal ranges. The results suggest that long term intake of traditional Chinese cabbage kimchi in large dose have the lowering effect on blood lipid and blood glucose level. There is no possibility of adverse actions on renal and hepatic functions by ordinary intake of Chinese cabbage kimchi in spite of being rich in salt and hot spices such as garlic, onion, ginger and red pepper.r.r.

Effects of Lactobacillus plantarum CIB 001 on Lipid Metabolism of Hypercholesterolemic Rats (김치에서 분리한 Lactobacillus plantarum CIB 001 급여가 고콜레스테롤혈증 흰쥐의 지질대사에 미치는 영향)

  • The aim of the present study was to assess the anti-hypercholesterolemic effect of bile salt hydrolase-producing Lactobacillus plantarum CIB 001 (KCTC 11717 bp) in rats fed a high-cholesterol diet. Four treatment groups of rats (n=5) were fed experimental diets: a normal diet (ND), a ND plus L. plantarum CIB 001(NDL) at $5.0-7.5{\times}10^9$ colony forming unit (CFU)/day, a high-cholesterol diet (HCD), as well as a HCD plus L. plantarum CIB 001 (HCDL) at $5.0-7.5{\times}10^9$ CFU/day for 6 weeks. Compared with the HCD group, the HCDL group demonstrated a decrease in serum triglyceride (p<0.05), total cholesterol (p<0.05), and the corresponding HDL-cholesterol concentration increased at a rate of 40% (p<0.05). The HCDL group also induced a decrease in liver inflammation and steatosis. The present results suggest that supplementation of L. plantarum CIB 001 can have short-term (6 weeks) effects on blood lipids and liver injury, as well as on the atherogenic index and cardiac risk factors.

Comparison of Dietary Behaviors Related to Sodium Intake by Gender and Age (나트륨 섭취량과 관련된 식행동의 성별 및 연령별 비교)

  • This study was performed to understand recognition and behaviors related to sodium intake of Korean adults. The data were collected from subjects including 267 male and 285 female adults in nationwide and compared by gender and by age. We found that the male group showed significantly higher smoking, alcohol drinking, and exercise does the female group. The older group (40 to 59 years) revealed significantly higher exercise and lower alcohol drinking; however general disease and hypertension prevalence, diet therapy practice, and meditation for hypertension were higher. Recognizing sodium levels of foods containing high-sodium, and sodium-nutriture labels when purchasing foods, and knowing differences between salt and sodium of the male group or recognizing sodium levels of foods containing high-sodium of the older group was worse than the other. Among the 32 food behaviors, only 12 were identified as significantly correlated to sodium intake levels including behaviors of preferring Chinese and Japanese foods to Western foods, preferring kimchi to raw vegetables, completely consuming soup, stew, noodle liquid, liking of dried fish and salted mackerel, frequent eat-outs or delivered foods, and so forth. There were significant differences between gender or age groups in terms of sodium intake-increasing behaviors; the male group showed higher behaviors of preferring salty taste and eating all broths. And the older group revealed higher behavior of adding table salt as well as the previous two, however, the younger group showed more behaviors of eat-outs or delivered foods and not the liquid of kimchi.

Effects of Dietary Polyunsaturated Fat and Se on Levels of Serum Vitamin E and Creatine Phosphokinase and Morphology of Rat Hepatocytes (Polyunsaturated Fat 및 Se 식이가 흰쥐의 혈청 비타민 E 및 Creatine Phosphokinase 와 간세포의 형태에 미치는 영향)

  • Vitamin E status affected by dietary high PUFA and Se was examined by biochemical and morphological means. Rats were fed four different diets(I : 15% p/s=1 control diet, II : 15% perilla oil diet, III : 15% perilla oil, vitamin mix -vitamin E, IV : 15% perilla oil, vitamin mix-vitamin E and salt mix -Se ) for $4\frac{1}{2}$ weeks. Various dietary treatments had no significant effects on body weight gains of rats. Activities of serum creatine phosphokinase known as an indicator of vitamin E deficiency were significantly higher( P < 0.001) in groups fed diets high in PUFA, regardless of the addition or omission of vitamin E from the vitamin mixture than those in control group. Vitamin E concentrations of serum and liver were affected by experimental diets and serum levels were more affected than those in liver. Electron microscopic observations of the liver revealed 1) the presence of swollen and degenerated mitochondria and lysosome-like body(II), and 2) markedly swollen and degenerated mitochondria, numerous lysosomes and decreased in size and number of microvilli along the bile canaliculus ( III, and 3) a remarkable accumulation of lipid droplets, nuclear pyknosis, degenerated mitochondria and increased number of lysosomes scattered along the cell junction in the hepatocytes (IV).

The Effect of Insoluble Dietary Fiber Extracted from Chinese Cabbage Waste on Plasma Lipid Profiles in Rats Fed a High Fat Diet (배추 폐기물로부터 분리한 불용성 식이섬유가 고지방 식이를 급여한 쥐의 혈중 지질농도에 미치는 영향)

  • This study investigated the lipid profiles for the possible improving activity of insoluble dietary fiber extracted from cabbage waste discarded during kimchi manufacture and market distribution. Enzyme-treated Insoluble Dietary Fiber (EIDF) is the remnants of crude dietary fiber (CDF) treated by ${\alpha}$-amylase, protease, and amyloglucosidase after removal of soluble materials from cabbage waste. The insoluble dietary fiber of CDF and EIDF in cabbage waste was $65.33{\pm}0.33%$ and $73.57{\pm}0.09%$, respectively. To examine the effects of EIDF on plasma cholesterol concentration, two group of rats were fed either a high fat diet (HFD) or a HFD containing 0.1~1.0% EIDF for 4 weeks. The body weight of all groups was not significantly different (p<0.05) but the body weight of EIDF+HFD groups was less than that of the HFD group (p<0.1). Compared with the HFD group, EIDF also lowered serum levels of total triglycerides to 11.2~23.3% and cholesterol to 26.8~28.5%. In EIDF+HFD groups, the HDL-cholesterol level increased by 7.2~26.1%, while LDL-cholesterol especially decreased by 51.0~61.4% and VLDL-cholesterol by 16.9~26.4%. The atherogenic index of EIDF+HFD groups was also reduced twice that of the HFD group. From these results, EIDF from cabbage waste could be a potential effective food ingredient for improving lipid profiles.

Anti-Obesity Effects of Salted and Unsalted Doenjang Supplementation in C57BL/6J Mice Fed with High Fat Diet (식이유도 비만 Mice에서 된장 및 무염된장의 항비만 효과)

  • The aim of the present study is to elucidate the anti-obesity effects of Doenjang with and without salt in C57BL/6J mice. For the analysis, a total of forty mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HD), high-fat diet supplemented with 20% Doenjang group (DJ), high-fat diet supplemented with 20% unsalted Doenjang group (NS). During the study period, food intake and body weight were measured daily and weekly, respectively. The animals were sacrificed after 12 weeks. Body weight gain, epididymal fat pad weight and serum triglyceride levels of DJ group were found to be significantly lower than those of the HD and NS groups. Serum total-cholesterol levels of DJ and NS groups were significantly lower as compared to the HD group. There were significant decreases in plasma insulin and leptin levels in DJ group compared with the HD and NS groups. We did not observe any significant changes in the expression of hepatic lipogenic-related gene $PPAR{\gamma}$ among the HD, DJ and NS groups. However, ACC expression was found to be significantly decreased in DJ group. Lipolysis-related gene ($PPAR{\alpha}$ and CPT-1) expression was significantly higher in the DJ group as compared to HD and NS groups. In conclusion, the results of this study showed that Doenjang supplementation lowers body weight gain and improves obesity-related parameters.

Comparative Studies on the Fatty Acid Compositions of the Korean Salt Marsh Plants in the West Sea (한국서해에 서식하는 염생식물의 지방산 조성 비교)

  • Fatty acid is an important component of many biological processes. However, an imbalance in diet-especially, a n-3 polyunsaturated fatty acids (PUFAs) deficiency-causes several diseases such as diabetes, cardiovascular disease and cancer. In this study, we analyzed the fatty acid contents and compositions of 26 species of Korean salt marsh plants and found high fatty acid contents from S. herbacea ($148.75\;{\mu}g$/mg-dry wt.), S. komarvii ($119.05\;{\mu}g$/mg-dry wt.), C. heterocarpa ($79.23\;{\mu}g$/mg-dry wt.), A. capillaris ($71.65\;{\mu}g$/mg-dry wt.), and L. tetragonum ($67.02\;{\mu}g$/mg-dry wt.). In the case of saturated fatty acids (SFAs) composition, palmitic acid is richest in most salt marsh plants. On the other hand, oleic acid and linoleic acid are major components of monounsaturated fatty acid and n-6 PUFA, respectively. In addition, n-3 PUFAs such as LNA (linolenic acid), EPA (eicosapentaenoic acid), and DHA (docosahexaenoic acid) known as the main fatty acid components of fish oils and seaweeds, were also found in S. herbacea, S. komarvii, T. tetragonoides, A. capillaris and G. littoralis.

Effect of Age-related Changes in Taste Perception on Dietary Intake in Korean Elderly (노인의 영양상태에 미각변화가 미치는 영향)

  • This study was performed to investigate the change in taste perception during aging and its effects on dietary intake in Korean elderly. The subjects were female aged 65 through 90 in the Anyang area, and college women were included as a comparison group . Dietary intake of the elderly(n=155) and young subjects (n=38) was measured by a 3 day diet record. The taste threshold and 'just right' concentrations(JRC) for sweet and salty tastes were assessed by sensory evaluation . Sucrose solution (0.0.,0.4,0.6,0.8,1.0, 1.2%) and salt solution(0, 0.02, 0.03,0.06,0.09,0.12,0.15%) were used to establish thresholds. for JRC assessment, four suprathreshold sucrose concentrations of 5, 8, 11 and 14% in orange-pineapple flavored juice and salt concentrations of 0.20, 0.34, 0.50and 0.75% in beef stock were prepared. Mean intakes of energy, protein , vitamin A, thiamin , riboflavin, niacin ,calcium and iron of the elderly were below the Korean Recommended Dietary allowances. The elderly showed higher taste thresholds than young subjects of both sweet and salty tastes consumed less calories. Needs to bespecified. The older subjects having high threshold or JRCs for sweet and salty tastes consumed fewer calories from protein and fat. Pearson correlation coefficients was between JRC for sweet and salty tastes 0.54(p<0.01). The correlation coefficients between tastes threshold and nutrient intakes were very low for both age groups. Unlike the college women, in the elderly the JRC of sweet taste of the orange-pineapple juice were negatively correlated with intakes of energy, protein, fats, thiamin , riboflavin , niacin ,vitamin C , iron and consumption of meat and egg food groups (p<0.01). In summation age-related alterations in sweet and salty taste perception were observed in the elderly and dietary intakes of the elderly see to be influenced by these taste perception changes.
