In this study, hemicelluloses were pre-extracted from mixed hardwood chips using sodium hydroxide solution and then they were isolated from pre-extractives by using various solvents. Isolation was conducted by precipitating hemicelluloses with isopropanol, ethanol, 1,4-dioxane, dimethylsulfoxide (DMSO) and potassium hydroxide (KOH), respectively. The precipitate yield on alkali pre-extractives was the highest when hemicelluloses were isolated by DMSO and then precipitated with ethanol. Most precipitates were yellow colored. The efficiency of isolation was evaluated by analysing the characteristics of isolated hemicelluloses. Isolation using DMSO, KOH and 1,4-dioxane ($80^{\circ}C$) showed rather high efficiency. The highest total separation efficiency was about 77% when alkali pre-extractives were reacted with KOH and precipitated with ethanol. The quantity and purity of isolated hemicelluloses were affected by the solvent type.

Solid phase extraction (SPE) purification method is the efficient and well-known tool for automated [$^{11}C$]acetate synthesis. A fully automated homemade module adopting the SPE method and 'pinch' valves was developed very economically with a universal interface board, a relay card and an open source programmable logic controller. The radiochemical yield of the optimized [$^{11}C$]acetate synthesis by this system was $58.8{\pm}2.1%$ (n=10, decay-corrected) from $15.5{\pm}0.19GBq$ of $[^{11}C]CO_2$ as starting activity, and total synthetic time was 15 minutes. HPLC analysis showed its high radiochemical purity as $97.4{\pm}1.1%$ without possible by-products.

The extracts and pure saponins from the roots of Platycodon grandiflorum (PG) are reported to have a wide range of health benefits. Platycosides (saponins) from the roots of PG are characterized by a structure containing a triterpenoid aglycone and two sugar chains. Saponins are of commercial significance, and their applications are increasing with increasing evidence of their health benefits. The biological effects of saponins include cytotoxic effects against cancer cells, neuroprotective activity, antiviral activity, and cholesterol lowering effects. Saponins with commercial value range from crude plant extracts, which can be used for their foaming properties, to high purity saponins such as platycodin D, which can be used for its health applications (e.g., as a vaccine adjuvant). This review reveals that platycosides have many health benefits and have the potential to be used as a remedy against many of the major health hazards (e.g., cancer, obesity, alzheimer's) faced by populations around the world. Methods of platycoside purification and analysis are also covered in this review.

Seventy-six animals including cattle, sheep, horses, 6 species of zoo animals were employed for collection of fresh feces in order to detect verotoxigenic Esherichia coli (VTEC) by safe, quick and sensitive PCR-based molecular methods. Bacterial cell disruption with bead-beating followed by bacterial DNA purification with hydroxyapatide chromatography and gel filtration allowed DNA preparation from animal feces with high recovery and purity. A mountain goat was firstly shown by PCR and sequencing to shed verotoxin 2 gene (vt2) that was used to generate vt2 probe and second primer set for nested PCR to attempt more sensitive detection. Most sensitive nested PCR revealed that 45% of tested cattle and 47% of tested zoo animals were VTEC-positive, while least sensitive normal PCR detected VTEC from none of these animals except a mountain goat. Moderately sensitive detection by PCR in combination with hybridization suggested that the VTEC density varied between the VTEC-positive cattle.
Direct synthesis of cyclodextrin (CD) from extruded insoluble corn starch without liquefaction procedure using cyclodextrin glucanotransferase (CGTase) was carried out. Increased CD production rate and yield were achieved in heterogeneous enzyme reaction system containing extruded corn starch compared with those of conventional system employing liquefied or partially cyclized starch. At extruded starch concentration of 100 g/l the CD concentration and conversion yield were reached up to 54 g/l and 0.54, respectively. High purity of $\alpha \beta \gamma$-CDs without accumulation of undesirable malto-oligosaccharides was produced, furthermore, the residual extruded starch was easily separated by centrifugation from reaction mixture, whlch will facilitate the purification procedure. Granular structure of extruded starch was observed by SEM to investigate enzyme reaction mechanism. Supplemental addition of $\alpha$-amylase enhanced slightly the initial CD production rate, but it decomposed produced CD at the late stage. Various! extruded raw starches, such as, corn, rice, and barley were also suitable substrates for CD production.

We demonstrate a detail protocol for the radiosynthesis of an $^{125}I$-labeled MSTP prosthetic group and its application to the efficient radiolabeling of human serum albumin (HSA). Radioiodination of the precursor (2) was carried out by using $[^{125}I]$NaI and chloramine T as an oxidant at room temperature for 15 min. After HPLC purification of the crude product, the purified $^{125}I$-labeled MSTP ($[^{125}I]1$) was obtained with high radiochemical yield ($73{\pm}5%$, n = 3) and excellent radiochemical purity (>99%). Site-specific reaction between ($[^{125}I]1$) and HSA gave the $^{125}I$-labeled human serum albumin ($[^{125}I]3$) with more than 99% of radiochemical yield as determined by radio-thin-layer chromatography (radio-TLC). These results clearly demonstrate that the present radiolabeling method will be useful for the efficient and convenient radiolabeling of thiol-bearing biomolecules.
Efficient cellulolytic enzyme production is important for the development of lignocellulose-degrading enzyme mixtures. However, purification of cellulases from their native hosts is time- and labor-consuming. In this study, a constitutive expression system was developed in Penicillium oxalicum for the secreted production of proteins. Using a constitutive polyubiquitin gene promoter and cultivating with glucose as the sole carbon source, nine cellulolytic enzymes of different origins with relatively high purity were produced within 48 h. When supplemented to a commercial cellulase preparation, cellobiohydrolase I from P. funiculosum and cellobiohydrolase II from Talaromyces verruculosus showed remarkable enhancing effects on the hydrolysis of steam-exploded corn stover. Additionally, a synergistic effect was observed for these two cellobiohydrolases during the hydrolysis. Taken together, the constitutive expression system provides a convenient tool for the production of cellulolytic enzymes, which is expected to be useful in the development of highly efficient lignocellulose-degrading enzyme mixtures.
In this study, a tandem water and hexane washing process was developed to improve the recovery efficiency of paclitaxel derived from Taxus chinensis. The polar impurities contained in the sample were effectively removed by washing with water at a sample/water ratio of 1:40 (w/v) for 10 min. In addition, the non-polar impurities were effectively removed by washing with hexane at a sample/hexane ratio of 1:160 (w/v) for 20 min. A high purity of paclitaxel (>30.0%) was obtained in a short operating time (~30 min) by sequential washing with water and hexane.
An innovative process for high fructose corn syrup (HFCS) production coupled with direct saccharification of raw corn starch in the agitated bead enzyme reactor (bioattitor) was investigated. The required high concentration/purity of glucose solution suitable for isomerization was produced directly in a bioattritor. without condensation of hydrolyzate, 398 g glucose/$\ell$ and 98% glucose content from 400 g/$\ell$ (w/v) of raw corn starch after 24 hours. The unsaccharified residual starch could be separated easily upon centrifugation, and resaccharified. The obtained solution also possessed other desirable requirements as substrate for isomerization, such as. low concentrations of denatured protein and calcium ions, thereby, simplified the purification step. The obtained glucose solution was isomerized in an enzyme reactor paked with immobilized glucose isomerase to evaluate the suitability as a substrate. The proposed new HFCS process seems to have many advantages over the conventional process via liquefaction-saccharification steps. The follow-up investigations of the proposed process need to be conducted to evaluate the feasibility of industrial application.
