This study was carried out to investigate the effect of Lythrum salicaria L. ethanol extract on anti-obesity effects in rat fed a high fat diet for 8 weeks to induce obese rat model. Male SD rats were divided into normal group, control (high fat diet) group, positive control (Garcinia Cambogia extracts) group, high fat group supplemented with ethanol extracts of Lythrum salicaria L. (EELS). The body weight gain and control (high fat diet) were increased by a high fat diet, but decreased in the EELS. At the end of the experiment, the body weight in high fat diet groups was higher than that of normal diet group, while the body weights of EELS and positive control group were significantly reduced by 16.62%, as compared with that of high fat diet group (p < 0.05). The levels of serum triglyceride, total cholesterol in EELS group were significantly decreased as compared with high fat diet group (p < 0.05). The liver and mesenteric adipose tissue weights of control (high fat diet) increase than that for normal group, whereas EELS and positive control group were significantly decreased (p < 0.05). Levels of triglyceride in liver were significantly lower in EELS group than those in high fat diet group (p < 0.05). These results indicate that Lythrum salicaria L. extract may improve lipid metabolism and reduce fat accumulation and body weight.
신령버섯이 고지방식이를 섭취한 흰쥐의 지질대사에 미치는 영향을 조사하고자, 생후 21주링의 音쥐 에 표준식이를 급여한 정상군, 표준식이에 20% 돈지를 첨가한 식이를 급여한 고지방군, 고지방 식이에 신령버섯 자실체 분말을 3% 및 5%씩 첨가한 식이를 급여한 군(3% 및 5% 신령 버섯군) 등 4군으로 나누어 10주간 사육한 결과는 다음과 같다. 실험동물의 체중증가량 및 식이효율은 고지방군에 비해 3% 신령버섯군이 다소 감소되었으나, 5% 신령버섯군은 유의하게 감소되었다. 간과 신장의 무게는 고지방군과 신령버섯군들이 비슷하였으나, 부고환지방은 고지방군에 비해 신령버섯군들이 유의하게 감소되었다. 간의 콜레스데롤 농도는 고지방군과 신령버섯군들이 비슷한 수준으로 감소효과가 나타나지 않았다. 혈청 및 간의 중성지질 농도는 고지방군에 비해 5% 신령버섯군이 유의하게 감소되었다. 혈청의 총 콜레스테롤과 LDL-콜레스테롤 농도 및 동맥경화지수는 고지방군에 비해 3% 및 5% 신령버섯군이 유의하게 감소되었다. 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율은 고지방군에 비해 신령버섯군들이 유의하게 증가되었다. HDL-콜레스테롤 및 인지질 농도는 고지방군과 신령버섯군들이 비슷한 수준으로 신령버섯 섭취에 따른 영향은 나타나지 않았다. 이상의 결과로 보아 고지방식이를 급여한 흰쥐에 신령버섯을 5% 급여시 혈청과 간의 중성지질 농도, 혈청의 총 콜레스테롤, LDL-콜레스테롤 농도 및 동백경화지수를 낮추고, 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율을 증가시키는 것으로 나타났다.
Effects of newly developed functional rice grains Goami (high-amylose rice) and Nokwon (green-kerneled rice) on the glucose metabolism and antioxidative defense system in C57BL/6N mice under high fat diet condition were investigated. Animals were randomly divided into five groups (n = 8) and given experimental diets for eight weeks: normal control diet (NC), high fat diet (HF), and high fat diet supplemented with white rice (HF-WR), Goami rice (HF-GR), and Nokwon rice (HF-NR). At the end of the experimental period, the HF group exhibited markedly higher blood glucose level, insulin concentration, plasma lipid peroxidation and lower hepatic glycogen concentration than that exhibited by NC group. However, diet supplementation of Goami and Nokwon suppressed the high fat diet-induced hyperglycemia and oxidative stress through inhibition of the glucose-regulating enzymes and enhancement of the antioxidant enzymes activities. The results illustrate that the new functional rice Goami and Nokwon may be useful in the development of functional foods with preventive effect against high fat diet-induced hyperglycemia and oxidative stress.
We investigated the role of fatty acid availability on skeletal muscle AMPK activity and adenine nucleotide content. To investigate the chronic effects of elevated fatty acid in vivo Sprague-Dawley rats were fed a chow diet (15% fat) or a diet high in saturated (SAFA, 52% fat) or polyunsaturated (PUFA, 52% fat) fat for eight weeks. High fat diets increased (P < 0.05) plasma FFA levels by 25%. AMPK activity was increased in SAFA and PUFA rats and occurred in the absence of changes in ATP, AMP, phosphocreatine and glycogen content. These results suggest that increasing fatty acid availability increases AMPK activity independent of changes in the cellular energy charge, and implicate the regulation of AMPK by a covalent mechanism. These data also support the contention that increasing fatty acid availability can increase subsequent fatty acid oxidation by an AMPK-mediated process.
The effect of feeding various diets on plasma lipids, liver lipids, fecal excretion of lipids and triglyceride secretion rate was studied in Sprague-Dawley rats fed for 1 month diets containing high fat(40% of energy ; HF) high cholesterol(1% ; HC), low fat/high sucrose(70% ; HS) or control diet(CT). Weight gain in HS group but there were no differences in TG contents of HF or HC groups compared to CT group. Fecal excretion of lipid was higher in HC than in HF, HS and CT groups. Hepatic TG secretion was not significantly different in four dietary groups. Accordingly, hypertriglyceridemia shown in HS group does not seem to be resulted from the increased TG secretion rate.
[Purpose] The purpose of this study was to determine whether different types of carbohydrate diets with or without exercise changes energy metabolism at rest and during exercise. [Methods] To minimize differences in food and energy intake between experimental groups, mice were pairfed. After 1 week of adaptation, 40 male ICR mice (6 weeks old) were randomly divided into four groups: Sta. (high fat + high starch), Scu. (high fat + high sucrose), StaEX. (high fat + high starch + exercise), and SucEX. (high fat + high sucrose + exercise). StaEX. and ScuEX. groups underwent training by running on a treadmill five times a week. After 10 weeks of training, energy metabolism was measured for 24 h and during a 1 h exercise period. [Results] The final body weight showed no significant difference between the groups. However, the weight of abdominal tissues (epididymal, perirenal, and mesenteric adipose tissue) in training groups was markedly decreased following 10 weeks of training. Results of all energy metabolism (24 h at rest and during 1 h of exercise) showed no significant interactions between diet and exercise. A brief summary of the results of the energy metabolism is that the metabolism related indicators over 24 h were more affected by the dietary pattern than the exercise but during the 1 h of exercise, training had more effect on energy metabolism than diet. [Conclusion] Our findings confirm that: (a) the type of carbohydrates included in the diet influence the metabolic responses over 24 h, (b) training had more effect on energy metabolism than diet during 1 h of exercise, (c) both results; abdominal adipose tissue weight and fat oxidation during exercise are suggestive for a beneficial effect of moderate physical activity on weight maintenance.
The effects of water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower on serum lipid concentrations were evaluated in rats. Forty-eight male Sprague-Dawley rats weighing l00±l0 g were divided into six groups and fed high fat diets for six weeks. Experimental groups were administered with following diets; Control diet, animal, plant high fat diet and control and high fat diets with 2% water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower. Tissue weights of liver, lung, stomach, heart, kidney and spleen of high fat diet exposed rats were reduced by water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups. The concentrations of serum triglyceride in rats fed the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower were lower than those in other groups. The concentrations of total cholesterol in water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower group were lower than those in high fat diet groups. The concentrations of HDL-cholesterol in serum of the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups were significantly higher than those of other groups. The levels of LDL-cholesterol in serum of the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups were tended to be lower than those of other groups. GPT and GOT activities were decreased in water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups and than in the high fat group. LDH activity was lower in the water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups than in the high fat group. These results suggest that water extracts of green tea scented with lotus Nelumbo nucifera Gaertner flower groups may reduce elevated levels of serum lipid concentrations in rats fed high fat diets.
Objective : In this study, the anti-obesity effect of Fragaria orientalis (FO) on high-fat diet-induced obese mice was investigated. Drug treatment methods are widely used as obesity treatment methods, but research using various natural products is being conducted due to safety concerns. This study aims to evaluate the anti-obesity effect of FO extract, a natural product derived from Mongolia. Methods : C57BL/6 mice were used and divided into three groups, normal diet group, high-fat diet group, and high-fat diet with FO oral treatment group at a dose of 300 mg/kg. Extract was orally provided everyday for 6 weeks. Body Weight and food intake were measured every 2 days and blood lipid profiles and liver function in the sacrificed mice were evaluated. In addition, protein expression in hepatic tissue and histomorphological changes in liver and adipose tissue were observed. Results : Body weight, adipose tissue weight and FER were significantly lower in a high-fat diet with FO treatment than fed only high-fat diet. There was a significant difference between the high-fat diet and the FO-treated high-fat diet mice. As a result of analyzing lipid metabolism-related genes in hepatic tissue, all of p-AMPK, p-ACC, PPAR-α, CPT-1, and UCP-1 showed significant increases, and PPAR-γ also decreased significantly compared to the high-fat diet group. Conclusion : Overall, these results indicate that FO is effectual in improving obesity, suggesting that it can be used as a possible material for anti-obesity agents or functional supplements for weight control.
This study is to investigate the effects of capsaicin with high-fat diet on tissue glycogen contents in exercise-trained rats. Forty male Sprague-Dawley rats were offered a high-fat diet for 2 wks in individual cages and were exercise-trained by a animal treadmill running throughout the experimental period. After 2 wks of the prefeeding with high-fat diet, the rats were divided into two group: high-fat diet group(CON)and high-fat diet + capsaicin(0.014%) group(CAP). The rats were killed by decapitation at 10 hr(rest), 1 hr and 2 hr after treadmill running(27m/min, 6$^{\circ}$). Body weight and epididymal adipose tissure weight were significantly lower in CAP than in CON, but soleus muscle weight was not different between the two groups. Glycogen contents in liver, soleus and gastrocnemius white muscles were significantly lower in CAP than in CON at rest, 1 hr and 2 hr (p<0.05). However, glycogen content in gastrocnemius red muscle was significantly higher in CAP compared with CON at 2 hr after the exercise(p<0.05). These results indicate that capsaicin intake with high-fat diet would decrease glycogen contents in liver and muscle, however, this effect on glycogen metabolism could be changed by muscle type.
This study was performed to investigate effects of Chiarella vulgaris on lipid metabolism in rats fed high fat diet. Sixty 6-week-old male Wistar rats were divided into two groups; normal diet group and high fat diet group, then the rats in each group were further divided into three subgroups and fed 0%, 5% and 10% (w/w) chlorella-containing diets, respectively, and raised for 9 weeks, Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activity and total protein and albumin concentration were not different among groups. Serum total lipids and liver TG concentration were significantly lower in 5% and 10% chlorella groups than 0% chlorella group in high fat diet groups (p<0.05). Serum TG, serum total cholesterol, liver total lipid and liver total cholesterol concentrations were significantly lower in 10% chlorella groups than 0% chlorella group in high fat diet groups (p<0.05). Fecal total lipid, TG and total cholesterol excretions were significantly higher in 5% and 10% chlorella groups than 0% chlorella groups in normal diet and high fat diet groups, respectively (p<0.05). These results suggest that Chlorella vulgaris is effective for prevention of dyslipidemia which may be due to the modulation of lipid metabolism and increased fecal excretion of lipid.
