  • Background: Sepsis still has a high mortality rate despite adequate supportive care. Newer therapeutic modalities have been developed but they have generally ended in failure. Recently, insulin was reported to have an anti-inflammatory effect by inhibiting the $I{\kappa}B/NF-{\kappa}B$ pathway, and may have therapeutic potential in sepsis. However, the precise mechanism of the anti-inflammatory effect of insulin is unclear. This study examined the role of insulin in activating $I{\kappa}B/NF-{\kappa}B$ in macrophage. Methods: Raw 264.7 cells, a murine macrophage cell line, were used in this experiment. Western blotting using $I{\kappa}B$ Ab and phosphor-specific $I{\kappa}B$ Ab was performed to evaluate the degradation and phosphorylation of $I{\kappa}B$ cells. For the $I{\kappa}B$ Kinase (IKK) activity, an immune complex kinase assay was performed. The level of interleukin-6 (IL-6) was measured by ELISA to determine the level of proinflammatory cytokine. Results: $I{\kappa}B{\alpha}$ degradation began 30 min after lipopolysaccharide (LPS) treatment. However, an insulin pretreatment suppressed the $I{\kappa}B{\alpha}$ degradation caused by the LPS treatment. The phosphorylation of $I{\kappa}B{\alpha}$ and IKK activity was also inhibited by the insulin pretreatment. Finally, the insulin pretreatment showed a tendency to suppress the induction of IL-6 by LPS. Conclusion: Insulin might have an anti-inflammatory effect though partial inhibition of the $I{\kappa}B/NF{\kappa}B$ pathway in macrophage cell lines.

  • This study was carried out to produce high functional and biodegradable coating agents for pulp mold by evaluating various kinds of biodegradable polymers. Five kinds of biodegradable polymers were used. In addition, the mixture of the carboxymethylated starch and biodegradable polymers(${\kappa}$-carrageenan, chitosan) were used for mixed coating agents. Physical properties of coated paperboards were evaluated. Conclusions obtained were as follows. 4% ${\kappa}$-carrageenan and 5% chitosan showed higher water and oil resistance. 10% sodium alginate, 4% corn zein and 15% polycaprolactone showed high water resistance while no improvement was found on oil resistance. The optimum mixture ratios for the mixed coating agents were 90:10(carboxymethylated starch : ${\kappa}$-carrageenan) and 50:50(carboxymethylated starch : chitosan). Since these mixed coating agents have excellent biodegradability with higher water and oil resistance, these can be used for the environmental-friendly coating agents.

  • An investigation was conducted into the effects of $\kappa$-carbides on the cracking phenomenon, which often occurred in cold-rolled light-weight steel plates. Three kinds of steels were fabricated by varying the C content, and their microstructures and tensile properties were investigated. In the two steels that contained a high carbon content, the band structures of ferrites and $\kappa$-carbides that were severely elongated along the rolling direction were well developed, whereas continuous arrays of $\kappa$-carbides were formed in the steel that contained a low carbon content. Detailed microstructural analyses of the deformed region beneath the tensile fracture surface showed that the cracks initiated at arrays of $\kappa$-carbides or $\kappa$-carbides formed interfaces between the band structures, which initiated cleavage fractures in the ferrite bands, while the bands populated with a number of $\kappa$-carbides did not play an important role in propagating the cracks. Thus, the minimization of interfacial $\kappa$-carbides or $\kappa$-carbide arrays by increasing the carbon content was essential for preventing cracking from occurring during cold rolling.

  • This paper deals the effect of anthraquinone (AQ) on the contribution of sulphidity in kraft pulping of sapwood. The pulping conditions namely- active alkali concentration, pulpingtime, temperature and liquor ratio were varied in low ($15\%$) and high ($30\%$) sulphidity. $0.1\%$ AQ was added in the low and high sulphidity pulping with varying active alkali concentration and cooking time. At optimum conditions, low sulphidity kraft process produced about $44\%$ pulp yield with kappa number of about 23. But in high sulphidity kraft process kappa number was reduced to about 20 at the same yield. An addition of AQ reduced alkali requirement by $2\%$ on oven dried raw material and cooking time by 1 hour to produce pulp yield of about $44\%$ at kappa number 20. AQ is more effective in low sulphidity pulping than the high sulphidity pulping. The breaking length of kraft-AQ pulp was slightly higher than that of kraft pulp.

  • This study was conducted to analysis the distribution of the rectangular stress block for high-strength polymer concrete beam. C-shaped specimens were produced and tested to compute parameters of the rectangular stress block. They were $\kappa_{1}$ = 0.73, $\kappa_{3}$ = 0.94 and $\gamma$= 0.845, respectively. Experimental value of flexural strength of beam was same to be compared with theoretical value. But there is desirable to need many experimental data in order to exact design of polymer concrete structure.

  • The objectives of this study was to decrease pollutions of bleaching effluent and was to enhanced brightness of non-chlorine bleached pulps by xylanase treatments. Xylanase cloned Esherichacoli(E. coli) capable of each of endo, exo-xylanase and acetyl-esterase were obtained from Bacillus stearothermophillus. These xylanase was maintained high activity in alkali and high temperature. Especially endo-xylanase would be more active in $60^{\circ}C$ and pH 11. Xylanase pretreatment(X) of unbleached pulp increased brightness, and decreased the degree of delignification. The degree of increase in brightness of pulp due to xylanase pretreatment was similar to non-enzyme treated pulp, regardless of the amount of enzyme added. Therefore, the addition of xylanase of 2 unit was recommended when considering costs of enzyme. The pulp bleached XO sequence had higher brightness and lower Kappa no, than O bleached pulp, while pulp bleached XP sequence had similar brightness and Kappa no. with P bleached pulp. In XOC/D, XOZ and XOP bleaching sequences, brightness and degree of delignification were improved. The C/D and Z stage bleached pulp was good effect on rate of raise in brightness and Kappa no., but P stage bleached pulp had similar level in non-enzyme treated bleaching sequence.

  • Agmatine, an amine and organic cation, reduced $H_2O_2$ that was generated by hyperglycemia, and transcription factors such as NF-${\kappa}B$ and AP-1 activity in the mesangial cells that were exposed to high glucose. However, spermine which shares a strong nucleophilic structure with agmatine decreased the $H_2O_2$ levels and AP-1, but not the NF-${\kappa}B$ activity. Possible roles for agmatine and spermine in decreasing fibronectin are discussed, and the signaling pathway for agmatine-reduced fibronectin accumulation is presented.

  • Cholangiocarcinoma (CCA) is a tumor of biliary ducts, which has a high mortality rate and dismal prognosis. Constitutively activation of the transcription factor nuclear factor kappa-B (NF-${\kappa}B$) has been previously demonstrated in CCA. It is therefore a potential target for CCA treatment. Effects of diethyldithiocarbamate (DDTC) on NF-${\kappa}B$-dependent apoptosis induction in cancer have been reported; however, anti-metastasis has never been addressed. Therefore, here the focus was on DDTC effects on CCA migration and adhesiond. Anti-proliferation, anti-migration and anti-adhesion activities were determined in CCA cell lines, along with p65 protein levels and function. NF-${\kappa}B$ target gene expression was determined by quantitative RT-PCR. DDTC inhibited CCA cell proliferation. Suppression of migration and adhesion were observed prior to anti-CCA proliferation. These effects were related to decreased p65, reduction in NF-${\kappa}B$ DNA binding, and impaired activity. Moreover, suppression of ICAM-1 expression supported NF-${\kappa}B$-dependent anti-metastatic effects of DDTC. Taken together, DDTC suppression of CCA migration and adhesion through inhibition of NF-${\kappa}B$ signaling pathway is suggested from the current study. This might be a promising treatment choice against CCA metastasis.

  • This study investigated the physical properties of polymers and antimicrobial activities of organic acids on Listeria monocytogenes to develop hydrogels. ${\kappa}-carrageenan$ (1, 2, and 3%), carboxymethylcellulose (CMC; 1, 3, and 5%), and agar (1.5 and 3%) were mixed with cross-linkers ($Na^+$, $K^+$, $Ca^{2+}$, and $Al^{3+}$) or each other by stirring or heating to form cross-linkage, and their physical properties (hardness, elasticity, and swelling) were measured. The hydrogels formulated with organic acid (1, 3, and 5%) were analyzed by spot assay against L. monocytogenes. ${\kappa}-carrageenan$ formed hydrogels with high hardness without other cross-linkers, but they had low elasticity. The elasticity was improved by mixing with other cross-linkers such as $K^+$ or other polymer, especially in 3% ${\kappa}-carrageenan$. CMC hydrogel was formed by adding cross-linkers $Al^{3+}$, $Na^+$, or $Ca^{2+}$, especially in 5% CMC. Thus, stickiness and swelling for selected hydrogel formulations (two of ${\kappa}-carrageenan$ hydrogels and three of CMC hydrogels) were measured. Among the selected hydrogels, most of them showed appropriate hardness, but only 3% ${\kappa}-carrageenan-contained$ hydrogels maintained their shapes from swelling. Hence, 3% ${\kappa}-carrageenan+0.2%$ KCl and 3% ${\kappa}-carrageenan+1%$ alginate+0.2% KCl+0.2% $CaCl_2$ were selected to be formulated with lactic acid, and showed antilisterial activity. These results indicate that 3% ${\kappa}-carrageenan$ hydrogels formulated with lactic acid can be used to control L. monocytogenes on food surface.
