Fluctuation of Temperature Induces Pathogenicity of Streptococcus iniae and Changes of Immunology Related Genes of Korean Rockfish, Sebastes schlegeli

  • This study was designed to examine the immune response in Korean rockfish during water temperature fluctuation and to elucidate the factors contributing to streptococcal pathogenesis in cultured Korean rockfish, S. schlegeli. We investigated cumulative mortality against Streptococcus iniae (FP5228 strain) infection in the exposed Korean rockfish (39.7±5.8 g) to environmentally relevant temperature (Control, 23℃; High temperature, 28℃ and 23℃ and 28℃ with 12 hours interval exchange, 23↔28℃) for 48 hours. Also, the expression of the mRNA related to the immune response genes (heat shock protein 70, interleukin1β, lysozyme g-type and thioredoxin-like 1) were measured in spleen and head kidney by real-time PCR analysis in the exposed fish to thermal stress. In this study, the combined stress with bacterial challenge in fishes exposed to thermal stress lowered the survival rate than that of control (23℃). The cumulative mortality in the group of control, 28℃ and 23↔28℃ was 24%, 24% and 40% (P<0.05), respectively. Also, thermal stress modulated the mRNA level of immune related genes; heat shock protein 70, interleukin-1β, lysozyme g-type and thioredoxin-like 1 in Korean rockfish. The present study indicates that a high and sudden water temperature change affect immune responses and reduce the disease resistance in Korean rockfish.

A Study on High Temperature Creep and Stress Relaxation Properties of Zr-4 (Zr-4의 고온 크리프 및 응력이완 특성에 관한 연구)

  • Zr-4 used for a cladding and an end plug of reactor component has creep deformation under operation at high temperature. Creep is regarded as the time dependent deformation of a material under constant applied stress. Although the major source of the deformation of zirconium component in water-cooled reactors is irradiation creep, the thermal creep may give a rise to significant deformation in reactor component especially at relatively high temperatures and at various constant stresses, and therefore it must be predicted accurately. Stress relaxation is the time dependent change of stress at constant strain and it is a process related intimately to creep. In this paper, the creep behavior and stress relaxation of Zr-4 is examined at the temperature of 50$0^{\circ}C$ that is 40% of the absolute melting temperature of Zr-4 under the stress below yield stress and under the various constant strains. The results obtained are summarized as follows: 1) With an increase of stress, the steady state creep rate increases and the creep rupture time decreases. 2) The steady state creep rate $\varepsilon$(%/s) for the stress $\sigma$sub(c) (kgf/mm super(2)) of Zr-4 increases outstandingly. All the empirical equations computed for Zr-4 increases outstandingly. All the empirical equations computed for Zr-4 are in accord with Norton's model equation($\varepsilon$=K$\sigma$ sub(c) super (n)). The constants of materials computed are as follows: K=3.9881$\times$10 super(-5), n=1.9608 3) The rupture time T sub(r) (hr) decreases linearly with the increase of stress on the log-log scaled graph. The empirical equations computed for Zr-4 are in accord with Bailey's model equation (T sub(r)=K sub(1)$\sigma$sub(c) super(m)). The constants of materials computed are as follows: K sub(1)=1.2875$\times$10 super(16), m=-3.467 4) It seems clear that the strain could be quantitatively dependent on the high temperature creep properties such as creep stress, rupture time, steady state creep rate and total creep rate. It is found that these relationships are linear on the log-log graph. 5) In stress relaxation test, as the critical constant strain that can be allowed to the specimen is larger, stress relaxation becomes more rapid, and as the constant strain is smaller, the stress relaxation becomes slower.

Compression Behavior of Wood Stud in Light Framed Wall as Functions of Moisture, Stress and Temperature

  • There has been considerable research in recent times in light-timber med structures in fires. These structures have included horizontal (floor-like) panels in bending and walls under eccentric and approximately concentric vertical loading. It has been shown that compression properties are the most dominant mechanical properties in affecting structural response of these structures in fire. Compression properties have been obtained by various means as functions of one variable only, temperature. It has always been expected that compression properties would be significantly affected by moisture and stress, as well. However, these variables have been largely ignored to simplify the complex problem of predicting the response of light-timber framed structures in fire. Full-scale experiments on both the panels and walls have demonstrated the high level of significance of moisture and stress for a limited range of conditions. Described in this paper is an overview of these conditions and experiments undertaken to obtain compression properties as a functions of moisture, stress and temperature. The experiments limited temperatures to $20{\sim}100^{\circ}C$. At higher temperatures moisture vaporizes and moisture and stress are less significant. Described also is a creep model for wood at high temperatures.

  • In this paper, glass surface-mat reinforced epoxy(G.S.R.E.) is developed, It is assured that the material(G.S.R.E.) can be used as photoelastic model material and it satisfy with the required properties of photoelastic model material. Therefore, the material can be used as model material of transparent photoelastic experiment when we analyze the stress distributions of transversely isotropic material by photoelastic experiment. When we use G.S.R.E. as photoelastic experiment model material, we had better use the G.S.R.E. which fiber volume ratio is less than 0.7% in the high temperature(stress freezing method) and than 1.74% in the room temperature. Relationships between stress fringe value and elastic modulus in transversely isotropic material are developed in this paper, it is assured by experiment that they are established in the room temperature or in the high temperature. Therefore we can obtain stress fringe value or elastic modulus from the relationships between stress fringe value and elastic modulus.

  • We tested the hypothesis that alpine plants have special physiological and biochemical mechanisms in addition to their structural adaptation in order to survive under extreme conditions. The photosynthetic organs of Pinus pumila were used to examine the seasonal changes in sugar concentration, antioxidative enzyme, and lipid peroxidation. The concentrations of sucrose, glucose, fructose and reducing sugar were the highest in the leaves in April. But sugar contents in buds and inner barks did not respond sensitively on temperature change. Meanwhile superoxide dismutase (SOD) activity responded sensitively on the change of temperature and SOD in all tissues maintained high activity in April. Meanwhile anthocyanin content increased rapidly in June but the increase of anthocyanin content was not enough to prevent their tissues from the damage by the exposure of high temperature or other stress. In conclusion, under low temperature condition, P. pumila increased the concentration of soluble sugars and SOD activity in their tissues in order to overcome extreme environmental condition. But in summer, these stress defense system against high temperature might be disturbed slightly. This results in the increase of malondialdehyde (MDA) contents in three tissues by lipid peroxidation.

  • A new practical modeling of the Norton's power law creep is proposed and implemented to analyze the high temperature behaviors of Alloy 690 SG tube material. In the model, both the stress exponent n and the rate constant B are simply treated as the temperature dependent parameters. Based on the two-step optimization procedure, the temperature function of the rate constant B(T) was determined for the data set of each B value after fixing the stress exponent n value by using the prior optimized function at each temperature. This procedure could significantly reduce the numerical errors when using the power law creep equations. Based on the better description of the steady-state creep rates, the experimental rupture times could also be well predicted by using the Monkman-Grant relationship. Furthermore, the difference in tensile strengths at high temperatures could be very well estimated by assuming the imaginary creep stress related to the given strain rate after correcting the temperature effects on the elastic modulus.

  • Fracture behaviors of SCM435 high strength bolt have been studied including macroscopic and microscopic fracture observation, Energy Dispersive X-ray Spectroscopy, Vickers hardness test and applied stress evaluation. cracks (ratchet marks) were generated by the repetitive loads acting on the bolts, initial stress of bolt and the stress concentration. The applied stress was found to be slightly higher than the fatigue limit of the material. The initial stress of bolt must be removed, and the mold temperature during the process must be maintained by room temperature. Bolts are recommended to be peened to improve fatigue limit.

Prediction of High Temperature Plastic Deformation Variables on Al 6061 Alloy (Al 6061 합금의 고온 소성변형 조건의 예측)

  • The high temperature behavior of Al 6061 alloy was characterized by the hot torsion test in the temperature ranges of 400∼550℃ and the strain rate ranges of 0.05∼5/sec. To decide optimum deformation condition, three types of deformation maps were individually made from the critical strain (εc). deformation resistance(σp) and deformation efficiency (η). The critical strain(εc) for dynamic recrystallization (DRX) which was decided from the inflection point of strain hardening rate(θ) - effective stress (σ) curve was about 0.65 times of peak strain (εp). The relationship among deformation resistance (peak stress, σp), strain rate (ε), and temperature (T) could be expressed by ε=2.9×1013[sinh(0.0256σp]7.3exp (-216,000/RT). The deformation efficiency (η)which was calculated on the basis of the dynamic materials model (DMM) showed high values at the condition of 500∼550℃, 5/sec for 100% strain. The results from three deformation maps were compared with microstructures. The best condition of plastic deformation could be determined as 500℃ and 5/sec.

Thermo-mechanical stress analysis of feed-water valves in nuclear power plants

  • Feed-water valves (FWVs) are used to regulate the flow rate of water entering steam generators, which are very important devices in nuclear power plants. Due to the working environment of relatively high pressure and temperature, there is strength failure problem of valve body in some cases. Based on the thermo-fluid-solid coupling model, the valve body stress of the feed-water valve in the opening process is investigated. The flow field characteristics inside the valve and temperature change of the valve body with time are studied. The stress analysis of the valve body is carried out considering mechanical stress and thermal stress comprehensively. The results show that the area with relatively high-velocity area moves gradually from the bottom of the cross section to the top of the cross section with the increase of the opening degree. The whole valve body reaches the same temperature of 250 ℃ at the time of 1894 s. The maximum stress of the valve body meets the design requirements by stress assessment. This work can be referred for the design of FWVs and other similar valves.

High Temperature Deformation Behavior of Microalloyed Hot Forging Steels (열간 단조용 비조질강의 고온 변형 거동에 관한 연구)

  • The high temperature deformation behavior of microalloyed hot forging steels has been examined as a function of the temperature, the strain rate, and the alloying element by using high temperature compression test. The high temperature deformation mechanism, which was obtained by analyzing the flow stress-strain curve and microstructure, could be considered to dynamic recrystallization. The peak stress of Nb-V-Mo steel was more increased and the dynamic recrystallization of Nb-V-Mo steel was faster than those of Nb-V steel. The peak stress of 1.2Mn-0.09Nb steel was more increased and the dynamic recrystallization of 1.2Mn-0.09Nb was delayed a little bit than those of 1.0Mn-0.05Nb. The peak stress of C-Nb-V steel was more increased and the dynamic recrystallization of C-Nb-V steel was delayed than those of C-steel. The constitutive equation of high temperature deformation had a power law type. The grain size of dynamic recrystallization was refined as the Zener-Hollomon parameter was increased. The relation of the dynamic recrystallization grain size and Zener-Hollomon parameter could be quantified to power law.
