3T3-L1 지방전구세포와 고지방 식이로 유도된 비만 HR-1 마우스 피부에 도포한 한약 추출 복합물의 항비만 효과 및 안전성 평가 (Anti-obesity Effects and Safety of the Mixture of Herbal Extracts in 3T3-L1 Cells and HR-1 Mice Fed a High Fat Diet)

  • The aim of this study was to investigate whether a novel formulation of an herbal extracts has an inhibitory effect on obesity. To determine its anti-obesity effects, we performed anti-obesity-related experiments in vitro and in vivo. Thus, our present study was carried out to evaluate the anti-obesity effect of herbal extracts using a high fat diet (HFD)-induced obese mouse model and 3T3-L1 adipose cells. The effects of each herbal extracts on lipid accumulation in 3T3-L1 cells were examined using Oil Red O staining. Results showed that treatment with each herbal extracts at $10{\sim}100{\mu}g/ml$ had no effect on cell morphology and viability. Without evidence of toxicity, herbal extracts treatment decreased lipid accumulation compared with the untreated adipocytes controls as shown by the lower absorbance of Oil Red O stain. Futhermore, compared with control-differentiated mature adipocytes, each herbal extracts significantly inhibited lipid accumulation in mature 3T3-L1 adipocytes. In the HFD-fed obese mice, body weight, liver weight and white adipose tissue weights were significantly reduced by mixture of herbal extracts administration in mouse skin. Futhermore, we found that mixture of herbal extracts administration suppressed serum triglyceride (TG), and total cholesterol (TCHO) in HFD-induced obese mouse model. The mixture of herbal extracts of permeability was estimated by measuring the transepithelial electrical resistance (TEER) value in pig skin. The optimized formulations of herbal extracts (Test 3 formulation) showed skin permeation. However, test 1 formulation containing essential oil as enhancer showed maximum skin permeation. After confirming the enhanced skin permeability, in vivo studies were performed to assess whether skin irritation potential on the basis of a primary irritation index (PII) in rabbit skin. Reactions were scored for erythema/edema reactions at 24 h, 48 h and 72 h post-application. It was concluded that the test 1 formulation was not irritation (PII = 0). The present study suggests that the test 1 formulation might be of therapeutic interest with respect to the treatment of obesity.

Inhibition of endoplasmic reticulum stress in high-fat-diet-induced obese C57BL/6 mice: Efficacy of a novel extract from mulberry (Morus alba) leaves fermented with Cordyceps militaris

  • A few clues about correlation between endoplasmic reticulum (ER) stress and mulberry (Morus alba) leaves were investigated in only the experimental autoimmune myocarditis and streptozotocin-induced diabetes. To investigate whether a novel extract of mulberry leaves fermented with Cordyceps militaris (EMfC) could suppress ER in fatty liver, alterations in the key parameters for ER stress response were measured in high fat diet (HFD)-induced obese C57L/6 mice treated with EMfC for 12 weeks. The area of adipocytes in the liver section were significantly decreased in the HFD+EMfC treated group as compared to the HFD+Vehicle treated group, while their level was higher in HFD+Vehicle treated group than No treated group. The level of the eukaryotic initiation factor 2 alpha ($eIF2{\alpha}$) and inositol-requiring enzyme 1 beta ($IRE1{\alpha}$) phosphorylation and CCAAT-enhancer-binding protein homologous protein (CHOP) expression were remarkably enhanced in the HFD+Vehicle treated group. However, their levels were restored in the HFD+EMfC treated group, although some differences were detected in the decrease rate. Similar recovery was observed on the ER stress-induced apoptosis. The level of Caspase-3, Bcl-2 and Bax were decreased in the HFD+EMfC and HFD+orlistat (OT) treated group compared to the HFD+Vehicle treated group. The results of the present study therefore provide first evidence that EMfC with the anti-obesity effects can be suppressed ER stress and ER stress-induced apoptosis in the hepatic steatosis of HFD-induced obesity model.

고지방 사료 급여 마우스에서 인진쑥 추출물의 항비만 효과 (Anti-Obestic Effects of Artemisiae Capillaris Herba, Artemisia Capillaris Stem Aqueous Extracts on the High Fat Diet Supplied Mice)

  • Artemisia capillaries THUNB is a perennial herb that belongs to the family compositae spp. and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. The object of this study is to observe the dosage-dependent anti-obestic effects of an aqueous extracts of dried aqueous extracts of stems of Artemisia capillaris Thunberg. [Artemisiae capillaris Herba, In-Jin in Korean, INJ] on 45%/Kcal high fat diet (HFD) supplied mice. 45%/Kcal rodent HFD are supplied to ICR mice from 1 week before initiation of INJ administration throughout the 12 weeks, and after the end of 12 weeks of 62.5, 125 and 250 mg/kg/day of INJ administration, the efficacy was divided into five categories 1) hypoglycemic, 2) hepato-protective, 3) nephroprotective, 4) hypolipemic, and 5) anti- obesity effects. The effects were compared to those of simvastatin (for hypolipemic activity), silymarin (for hepatoprotective and free radical scavenger effects) and metformin (for hypoglycemic and related anti-obesity effects). 7 animals per group (8 groups; total 56adapted mice on HFD were selected base on the body weight at 6 days after initiation of HFD supply) were used in this experiment. INJ and all three different reference drugs were directly suspended or dissolved in distilled water, and administered at a volume of 10 mL/kg, once a day for 84 days from 1 week after HFD supply. As results of 91 days of continuous HFD supply, mice showed marked obese states, hyperglycemia, hyperlipemia, liver damages and kidney damages. These mean the obesity, diabetes, diabetic hepatopathies, nephropathies and hyperlipemia were induced by HFD supply. After end of 84 days of continuous treatment of three different dosages of INJ, all diabetes related complications were inhibited; relatively favorable anti-obesity, hypolipemic, hepatoprotective, hypoglycemic and nephroprotective effects. These favorable effects showed relatively good dose-relations between all three different dosages of INJ treated, and INJ 250 mg/kg showed enough favorable effects on diabetes and related four complications tested as compared with one of each three different references. Otherwise, the efficacy of 62.5 and 125 mg/kg of INJ was somewhat slighter than those of all three reference drugs. Therefore, the suitable effective dosage of INJ is considered as 250 mg/kg/day in the present study. The overall anti-obesity effects of INJ 250 mg/kg-treated group was similar or more favorable than those of metformin 250 mg/kg-treated group, and INJ 250 mg/kg showed slighter hypoglycemic effects with silymarin 100 mg/kg and metformin 250 mg/kg, similar hypolipemic effects with simvastatin 10 mg/kg, and similar hepatoprotective effects with silymarin 100 mg/kg, and similar nephroprotective effects with that of silymarin 100 mg/kg and metformin 250 mg/kg, respectively. Obese, hyperglycemia, hyperlipemia, steatohepatitis and related nephropathies induced by HFD supply were dramatically inhibited by 84 days of continuous treatment of all three different dosages of INJ. It is, therefore expected that INJ extracts will be a favorable alternative agent for diet-related diabetes and complications.

고지방식이 비만마우스 모델에서 강지환(降脂丸)과 강지환(降脂丸)+가미소체환(加味消滯丸)의 비알콜성 지방간질환 개선효과 비교 (Comparison of Gangjihwan and Combination of Gangjihwan and Gamisochehwan in the Improvement Effects of Nonalcoholic Fatty Liver Disease in a High Fat Diet-Fed NAFLD Mouse Model)

  • Objectives : This study investigated the improvement effects of Gangjihwan (DF) and combination of Gangjihwan and Gamisochehwan (GSH) on nonalcoholic fatty liver disease in a high fat diet-induced obese mouse model. Methods : Eight-week-old C57BL/6N mice were divided into five groups: a normal lean group given a standard diet, an obese control group given a high fat diet, and atorvastatin, DF, and DF+GSH groups given a high fat diet with atorvastatin (10 mg/kg), DF (40 mg/kg), and DF+GSH (80 mg/kg), respectively. After 8 weeks of treatment, body weight gain, blood lipid markers, ALT concentrations, liver weight and histology were examined. Results : 1. Body weight gain was significantly decreased in DF, DF+GSH and atorvastatin groups compared with control. The extent of decreases was eminent in DF+GSH group. 2. Circulating concentrations of total cholesterol, HDL-cholesterol and LDL-cholesterol were decreased in DF, DF+GSH and atorvastatin groups compared with control. The decreases were significant in DF+GSH and atorvastatin groups. 3. Liver weights were decreased in DF, DF+GSH and atorvastatin groups compared with control. In particular, liver weight was significantly reduced only in DF+GSH group. 4. Hepatic lipid accumulation was significantly decreased in DF, DF+GSH and atorvastatin groups compared with control, and the magnitude of which was more effective in DF+GSH group than in DF-only group. 5. Circulating ALT concentrations were decreased in DF, DF+GSH and atorvastatin compared with control, but ALS levels were significantly reduced only in DF+GSH group. Conclusions : In conclusion, these results suggest that DF decreases body weight gain, improves blood lipid metabolism, and reduces liver weight and hepatic lipid accumulation, contributing to the improvement of nonalcoholic fatty liver disease. In addition, these effects were more effective in DF+GSH combination group than in DF-only group.

Methods : Eight-week-old C57BL/6N mice were divided into five groups: a normal lean group given a standard diet, an obese control group given a high fat diet, and atorvastatin, DF-a, and DF-b groups given a high fat diet with atorvastatin (10 mg/kg), DF-a (80 mg/kg), and DF-b (80 mg/kg), respectively. In addition, these effects were similar between Pakistani and Chinese Ephedra herba-containing Gangjihwan.

절식이 마우스 장내미생물에 미치는 영향 (Comparison of mice gut microbiota before and after fasting for a day)

  • 본 연구에서는 절식이 일반사료 및 고지방사료를 섭취한 마우스의 장내미생물생태에 미치는 영향에 관하여 조사하였다. 일반 사료 및 고지방사료를 마우스에 16주 동안 무한급이 형태로 섭취하게 하여 실험종료 직전에 1일(24시간) 간 절식시켰으며, 절식 전 후의 분변 샘플을 채집하고 MiSeq을 이용하여 장내미생물생태 분석을 진행하였다. 본 연구의 결과는 고지방사료를 섭취한 마우스에서는 절식 전후 장내미생물생태 내의 종 풍부성과 균등성이 감소한 반면, 일반사료를 섭취한 마우스에서는 차이를 나타내지 않았다. 또한, 고지방사료를 섭취한 마우스의 장내미생물생태에서는 절식 후 변화하는 것을 확인하였으며, 일반 사료를 섭취한 마우스의 절식 전후 변화가 없었다. Difference abundance analysis는 일반사료를 섭취한 마우스에서 절식 이후 S24-7로 분류되는 OTU는 증가하고, Ruminococcaceae로 분류되는 OTU가 감소하는 것으로 확인되었다. 반면, 고지방사료를 섭취한 마우스에서는 절식 이후 10OTUs가 감소하고, 3OTUs가 증가하였는데, 대부분 Ruminococcaceae로 분류되는 것으로 확인되었다. 본 연구 결과는 마우스에서의 절식이 장내미생물생태 중 Ruminococcaceae의 abundance에 영향을 미치며, 일반사료를 섭취한 마우스에 비해 고지방사료를 섭취한 마우스에서 절식에 대한 효과가 더 명확하게 나타난다는 것을 시사한다.

간에서 지방산 분할에 대한 지방산결합 단백질 5의 역할 (The role of Fatty acid binding protein 5 (Fabp5) in fatty acid partitioning in the liver)

  • 간성 지방생성과 지방 대사에 있어서 FABP5의 역할에 대하여 알아보고자 하였다. 마우스에 바이러스 입자를 이용하여 간에서 FABP5를 과발현 그리고 침묵시켜 이용하였다. 마우스에 서양식 또는 일반 사료를 1주일간 섭취시키고 24시간 동안 금식한 후 희생하였다. 간을 기계적으로 분쇄하여 웨스턴 블?으로 단백질 농도를 측정하였고, mRANA 분석에는 RT-PCR을 이용하였다. 간과 혈청의 지질 분석에는 박층 크로마토그래피를 이용하였다. 서양식이나 고농도 포화 지방식을 급식한 쥐에서 FABP5 발현이 높은 증가를 보였으나, FABP5 mRNA 발현은 급식에 비해 단식조건에서 급격히 감소하였다. FABP5를 과발현 시킨 상태에서 급식과 금식을 실시한 결과 간성 TG가 중요하게 증가하였다. 간성 유리 콜레스테롤은 급식상태에서 현저히 감소하였다. FABP5 발현 억제 시 간성 TG가 상당히 감소하였다. 결과들에서 보듯이, FABP5가 간지질 생성에 중요한 역할을 하며, 정상 상태 및 탄수화물 유도 조건에서 간성 TG 저장형성에 관여할 것으로 사료된다. FABP5는 비알코올성 지방간 질환, 대사증후군, 비만 치료에 활용될 수 있을 것으로 사료된다. 더 나아가, FABP5 발현에 어떤 전사인자가 관여하는지에 대한 연구가 필요하다.

목이버섯의 항비만 효과 (Anti-obesity effect of Auricularia spp.)

  • 11 종의 목이버섯 자실체로부터 얻은 50% EtOH추출물을 사용하여 지방전구세포인 3T3-L1의 지방세포 분화억제 효과를 확인한 모든 strain에서 억제효과가 존재하였다. 털목이에서는 21001 균주가 가장 우수하였고 흑목이에서는 21002 균주가 억제효과가 우수하였다. 전반적으로 털목이가 흑목이에 비해 지방세포 분화억제 효과가 우수하였다. 또한 Adipocyte RNA 발현량과 protein 발현량 측정 실험에서도 털목이 21001 균주가 지방세포 분화억제 효과에 있어서 가장 우수하였다. 털목이 21001 버섯을 사용하여 동물실험을 진행한 결과 털목이 자실체 분말이나 50% EtOH 추출물을 급여한 실험에서 체중 감소효과가 나타났으며, 특히 EtOH 추출물 0.2%를 급여한 군에서 유의하게 감소함을 확인하였다(*P<0.05). 동물 희생 후 적출한 간 중량 및 부고환지방량과 혈청 내 중성지방 및 총 콜레스테롤, 혈당, ALT, AST 등도 털목이버섯 21001 EtOH 추출물 0.1% 이상 급여 시 고지방식이만을 급여한 군에 비해 유의차(*P<0.05 이상) 있게 감소함을 확인하였다.

고지방식이 동물모델에서 크릴오일의 아디포카인 조절을 통한 항비만 효과 (Anti-Obesity Effect of Krill Oil by Regulation of Adipokines in High Fat Diet-Induced Mouse Model)

  • 본 연구는 고지방식이로 비만이 유도된 동물모델에서 크릴오일 섭취를 통한 비만 개선 효능을 확인하고자 하였다. 실험동물은 16주간 고지방식이를 섭취시켜 비만 동물모델을 유도하였으며, 실험 종료 4주 전 크릴오일을 100, 200, 500 mg/kg/day의 농도로 4주간 투여하였다. 실험동물의 체중 변화, 장기 무게 변화, 식이 효율과 혈청 내 아디포카인 인 렙틴과 아디포넥틴 농도를 측정하여, 농도별 크릴오일의 비만 개선 효과를 확인하였다. 고지방식이를 유도한 대조군은 정상식이를 유도한 군에 비해 유의적으로 체중이 증가하여 비만이 유도됨을 확인하였다. 반면 크릴오일 200, 500 mg/kg/day 농도로 투여한 군에서 비만을 유도한 대조군에 비해 유의적으로 체중을 감소시키는 효과를 확인하였다. 또한 고지방식이를 유도한 대조군은 혈청 렙틴 증가 및 아디포아인 감소로 비정상적인 아디포카인 조절능을 나타낸 반면 크릴오일을 500 mg/kg/day 농도로 투여한 군에서는 대조군에 비해 유의적으로 혈청 렙틴 감소 및 아디포넥틴 증가를 나타내어, 아디포카인 조절을 통한 비만 개선 효과를 나타내었다. 따라서 크릴오일의 투여는 아디포카인 분비 조절을 통해 체중을 감소시켜 비만 개선 효능이 있음을 알 수 있었으며, 체지방 개선용 기능성 소재로서의 활용 가능성이 있을 것이다.

도인승기탕에 의한 당뇨병성 혈관장애 개선효과 (Therapeutic Effect of Doinseunggi-tang on Diabetic Vascular Dysfunction)

  • Objectives : Diabetes mellitus is the leading cause for vascular complications such as atherosclerosis. The present study is to investigate whether Doinseunggi-tang (DST) improves diabetic vascular dysfunction in type II diabetes. Methods : The db/db mice were treated with high fat/high cholesterol diet and DST (200 mg/kg/day) for 8 weeks. Results : DST significantly lowered blood glucose and systolic blood pressure. In addition, DST also markedly decreased total plasma cholesterol, triglyceride, and LDL-cholesterol, whereas increased the HDL-cholesterol. Vascular relaxation of aortic rings by acetylcholine or SNP was ameliorated by DST in a dose-dependent manner. Damage of vascular intima and hypertrophic of media was improved by DST. Immunohistological study revealed that DST attenuated the increase of ICAM-1, VCAM-1, and ET-1 expression in thoracic aorta. Conclusions : Taken together, DST suppressed hyperglycemia and diabetic vascular dysfunction in type II db/db mice. The present data suggests that Doinseunggi-tang may be prevent a development of diabetic atherosclerosis.
