열수 녹차추출물이 생쥐의 체중 및 부고환 지방축적 조절에 미치는 영향 (Aqueous Spray-dried Green Tea Extract Regulates Body Weight and Epididymal Fat Accumulation in Mice)

  • 본 연구에서는 기존 주정추출 방법에 의하여 인위적으로 카테킨 함량을 높인 녹차추출물의 효능이 아니라 음용시 사용되는 녹차가 지닌 일반적인 카테킨 함량에 의한 항비만 효능을 검토하기 위해, 열수 분무건조 방법을 적용하여 제조된 국내산 제주 열수 녹차추출물(aq-GTE)을 C57BL/6J 생쥐에 고지방 식이를 통한 비만을 유도시키며 동시에 농도를 달리하여 8주간 섭취시킨 뒤, 체중과 체내 지방함량 및 혈중 지질함량에 대한 분석을 실시하여 항비만 효능을 확인하였다. 그 결과, aq-GTE 첨가시 HFD 군간 식이 섭취량에는 차이가 없었으나 체중 증가량에 있어서는 aq-GTE에 농도 의존적으로 체중 증가량이 감소됨을 확인하였다. 또한 부고환 지방의 무게는 aq-GTE의 섭취 농도에 따라 유의하게 감소하였고, 체중 변화에 따른 비교에서도 통계적으로 유의하게 부고환 지방 무게의 감소가 확인되었다. 비만을 식이로 유도한 실험군에 나타나는 steatosis 증가 현상이 aq-GTE를 처리시 농도의존적으로 억제되어 감소하는 현상이 발생함을 관찰할 수 있었으며, 그 외에 혈액 분석을 통하여 확인할 수 있는 이상증상은 없는 것으로 확인되었다. 결론적으로 본 시험에서 사용한 aq-GTE는 C57BL/6J 생쥐에서 체중 증가와 체지방 축적 감소 및 지질대사 개선 효과를 나타내었으며, 이는 기존의 주정추출 방식으로 카테킨 함량을 높인 녹차 추출물뿐만 아니라 일반 녹차의 카테킨 함량도 충분히 그 효과를 볼 수 있다는 결과를 검증한 것이고, 향후 aq-GTE의 인체에 미치는 효과에 대한 연구가 수행되어야 할 것으로 사료된다.

고지방식사로 유도된 신장 산화스트레스를 개선하는 가압볶음 무말랭이 열수추출물 효과 (Effects of hot water extracts of roasted radish against renal oxidative stress induced by high-fat diet)

  • 고지방식사를 섭취한 쥐의 신장에서 가압볶음 무말랭이 추출물의 산화방지, 항염증 효과를 확인하였다. 실험군은 chow diet와 증류수를 경구 투여하는 NOR group, 고지방식사와 증류수를 경구 투여하는 CON group, 고지방식사와 237 mg/kg bw/day농도의 DR과 RDR를 각각 경구 투여하는 DR group, RDR group으로 나누어 12주간 사육하였다. 신장의 산화스트레스 지표인 ROS, $ONOO^-$, 그리고 지방질과산화물의 농도를 확인한 결과 CON group이 NOR group에 비해 유의적으로 증가하였다. 반면 DR group과 RDR group에서 유의적으로 감소하였다. 체내 산화방지 지표인 글루타싸이온의 농도와 산화방지단백질의 발현은 DR group과 RDR group 모두 CON group에 비해 증가하였다. NF-${\kappa}B$ 발현은 CON group이 NOR group에 비해 증가하였으나 DR group과 RDR group은 모두 감소하였다. RDR group은 DR group에 비해 $ONOO^-$ 및 지방질과산화물 농도가 감소하였고 글루타싸이온 농도와 산화방지 관련 단백질 발현 중 Nrf2, HO-1, 카탈레이스, 그리고 GPx가 증가하였고 염증반응 전사인자인 NF-${\kappa}B$의 발현이 낮아졌다. RDR group의 산화스트레스 개선효과는 유의적으로 높은 것으로 확인되었다. 따라서 가압볶음무말랭이를 차로 제조하여 섭취할 때 고지방식사로 유도된 신장의 산화스트레스를 억제할 수 있을 것으로 사료되며, 그 효과는 무말랭이보다 높은 것으로 확인되었다.

The Herbal Composition Gangjihwan from Ephedra intermedia, Lithospermum erythrorhizon and Rheum palmatum Ameliorates Hepatic Inflammation and Fibrosis in Obese C57BL/6J Mice and HepG2 Cells

  • It was demonstrated that Gangjihwan (DF), which is the herbal composition composed of Ephedra intermedia, Lithospermum erythrorhizon, and Rheum palmatum, inhibits obesity and hepatic steatosis in high fat diet (HFD)-fed obese mice. The aim of this study was to determine the effects of DF on visceral obesity, hepatic inflammation and fibrosis and the mechanism of actions involved in this process using in vivo and in vitro approaches. DF was extracted with water (DF-FW), 30% grain alcohol (DF-GA30), and 70% grain alcohol (DF-GA70). Administration of DF to HFD-fed control mice decreased visceral tissue mass and visceral adipocyte size without adverse effects. Visceral fat mass was decreased by DF-GA30 and DF-GA70, and visceral adipocyte size by all three DF extracts compared with obese control mice. Histological analysis revealed that three kinds of DF extracts reduced toluidine blue-stained mast cells and collagen accumulation in the liver, the extents of which were most eminent in DF-GA70-treated mice. DF-GA70 decreased the mRNA levels of the inflammation ($TNF{\alpha}$ and VCAM-1), fibrosis (${\alpha}-SMA$), and apoptosis (caspase 3) genes, but increasing the anti-apoptosis gene (Bcl-2) mRNA levels in the liver of obese control mice. Consistent with the in vivo data, GA-70 also altered the expression of inflammation genes ($TNF{\alpha}$ and MCP-1) in HepG2 cells. These results indicate that DF not only inhibits visceral obesity, but also ameliorates visceral obesity-induced hepatic inflammation and fibrosis and that this process may be mediated by regulating the hepatic expression of inflammatory and fibrogenic genes.

목이버섯 품종간 생리활성 비교 연구 (A Comparative Study on the Physiological Activities of Auricularia spp.)

  • 본 연구에서는 목이버섯 품종간의 생리활성을 실험하였다. ORAC assay로 항산화활성을 측정한 결과 털목이, 갈색목이, 흑목이 순으로 활성이 높게 나타났다. ${\alpha}$-Amyloglucosidase 저해활성을 통한 항당뇨 활성은 3품종 모두 10 mg/mL에서 60% 정도의 높은 저해활성을 나타냈으며 품종간 유의적인 차이는 나타나지 않았다. Raw 264.7 macrophage의 nitric oxide 생성 저해를 통한 항염활성은 1 mg/mL 처리시 털목이와 흑목이는 dexamethasone 1 ${\mu}g$/mL에 상응하는 활성을 나타냈다. 20% 지방을 첨가하여 고지방식이로 비만을 유도한 ICR 마우스의 목이버섯 급여에 따른 항비만 효과를 확인한 결과 모든 군에서 체중 및 증체량, 식이효율의 감소를 확인할 수 있었고 그 중 갈색목이버섯인 JNM21012군은 0.45 g/day로 7% 지방을 첨가한 일반식이(NC) 군과 동일 수준의 증체량을 보였다. 부고환 무게는 털목이 및 흑목이, 갈색목이 모두 고지방식이(HFC) 군에 비해서 낮은 수치를 보였으며, 갈색목이 군은 HFC군과 유의적인 차이를 보였다. 혈액성분 중 triglyceride의 경우 목이버섯 첨가 식이군 모두 HFC군에 비해 낮은 수치를 보였으며 갈색목이 군은 고지방식이 HFC군에 비해 42% 감소시키는 것으로 나타났다. 또한, total cholesterol과 LDL-cholesterol은 털목이와 갈색목이군이 HFC군 보다 낮은 수치를 나타냈으며, 갈색목이군은 HFC군에 비해 각각 81%와 66% 감소하여 유의적인 차이를 보였다. HDL-cholestrol의 경우 각 군간의 유의적인 차이를 보이지 않았지만 NC군에 비해 높은 수치를 나타냈다.

Aspergillus Oryzae로 발효시킨 비지메주의 항비만 및 지질개선 효과 (Effect of fermented soybean curd residue (FSCR; SCR-meju) by aspergillus oryzae on the anti-obesity and lipids improvement)

  • In this study, we designed to confirm the dietary effect of anti-obesity of fermented soybean curd residue (FSCR; SCR-Meju; Biji-meju) by A. oryzae, which is well known as a Korean traditional meju microbe. We observed that body weight gain, serum and hepatic lipid profile, as well as the activity of ROS generating enzyme and ROS scavenging enzyme in high-fat diet induced obese mice fed experimental diet (SCR and SCR-meju). Body weight gain and epididymal fat weight of HC (high-fat diet control) was markedly higher than that of NC (Normal control). Conversely, body weight gain and epididymal fat weight of the SCR (Biji) and SCR-meju (Biji-meju) group was significantly lower than that of HC; these of the SCR-meju group was lower than that of the SCR group. Furthermore, serum TG and total-cholesterol and LDL-cholesterol contents of SCR and SCR-meju groups were lower than that of HC, and HDL-cholesterol level of the SCR-meju group was significantly higher than that of HC. In conclusion, although precise mechanisms of the antiobese effects of SCR-meju in this study are unknown, the present study provides an experimental evidence that SCR-meju may prevent obesity and obesity related metabolic syndromes, such as hyperlipidemia, hypertension and diabetes, and liver disease by high-fat diet. Nevertheless, further study in this filed will be needed.

A Mixture of Ethanol Extracts of Persimmon Leaf and Citrus junos Sieb Improves Blood Coagulation Parameters and Ameliorates Lipid Metabolism Disturbances Caused by Diet-Induced Obesity in C57BL/6J Mice

  • This study investigated the effects of a flavonoid-rich ethanol extract of persimmon leaf (PL), an ethanol extract of Citrus junos Sieb (CJS), and a PL-CJS mixture (MPC) on mice fed a high-fat diet (HFD). We sought to elucidate the mechanisms of biological activity of these substances using measurements of blood coagulation indices and lipid metabolism parameters. C57BL/6J mice were fed a HFD with PL (0.5% (w/w)), CJS (0.1% (w/w)), or MPC (PL 0.5%, CJS 0.1% (w/w)) for 10 weeks. In comparison with data obtained for mice in the untreated HFD group, consumption of MPC remarkably prolonged the activated partial thromboplastin time (aPTT) and prothrombin time (PT), whereas exposure to PL prolonged aPTT only. Lower levels of plasma total cholesterol, hepatic cholesterol, and erythrocyte thiobarbituric acid-reactive substances, hepatic HMG-CoA reductase, and decreased SREBP-1c gene expression were observed in mice that received PL and MPC supplements compared with the respective values detected in the untreated HFD animals. Our results indicate that PL and MPC may have beneficial effects on blood circulation and lipid metabolism in obese mice.

Effects of natural raw meal (NRM) on high-fat diet and dextran sulfate sodium (DSS)-induced ulcerative colitis in C57BL/6J mice

  • BACKGROUND/OBJECTIVES: Colitis is a serious health problem, and chronic obesity is associated with the progression of colitis. The aim of this study was to determine the effects of natural raw meal (NRM) on high-fat diet (HFD, 45%) and dextran sulfate sodium (DSS, 2% w/v)-induced colitis in C57BL/6J mice. MATERIALS/METHODS: Body weight, colon length, and colon weight-to-length ratio, were measured directly. Serum levels of obesity-related biomarkers, triglyceride (TG), total cholesterol (TC), low density lipoprotein (LDL), high density lipoprotein (HDL), insulin, leptin, and adiponectin were determined using commercial kits. Serum levels of pro-inflammatory cytokines including tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, and IL-6 were detected using a commercial ELISA kit. Histological study was performed using a hematoxylin and eosin (H&E) staining assay. Colonic mRNA expressions of TNF-${\alpha}$, IL-$1{\beta}$, IL-6, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) were determined by RT-PCR assay. RESULTS: Body weight and obesity-related biomarkers (TG, TC, LDL, HDL, insulin, leptin, and adiponectin) were regulated and obesity was prevented in NRM treated mice. NRM significantly suppressed colon shortening and reduced colon weight-to-length ratio in HFD+DSS induced colitis in C57BL/6J mice (P < 0.05). Histological observations suggested that NRM reduced edema, mucosal damage, and the loss of crypts induced by HFD and DSS. In addition, NRM decreased the serum levels of pro-inflammatory cytokines, TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 and inhibited the mRNA expressions of these cytokines, and iNOS and COX-2 in colon mucosa (P < 0.05). CONCLUSION: The results suggest that NRM has an anti-inflammatory effect against HFD and DSS-induced colitis in mice, and that these effects are due to the amelioration of HFD and/or DSS-induced inflammatory reactions.

저식이섬유 및 고지방 사료 급여 마우스의 장내 미생물 생태 변화 (Comparison of gut microbiome between low fiber and high fat diet fed mice)

  • 경제발전으로 인해 한국인의 식습관이 점차 서구화됨에 따라 웰빙(Well-being)의 문제가 야기되고 있다. 웰빙은 장내 미생물 군집과 밀접하게 연관되어 있으며, 이는 섭취한 음식에 따라 가변적이다. 이에 본 연구에서는 장내 미생물의 16S rRNA 유전자를 기반으로 하여 MiSeq을 진행하였고, 고지방 식이(HFD) 및 저식이섬유 식이(LFD)로 인한 장내의 미생물 생태 비교 및 분석하고자 수행되었다. 일반 대조군(CTL) 그룹과 비교하여 각각 LFD 그룹과 HFD 그룹은 species richness가 유의적으로 감소하였고, species evenness에서는 차이가 나타나지 않았다. phylum 수준에서는 Proteobacteria는 두 처리군에서 유의적으로 증가하였고(p<0.05), 그 중 Sutterella genus가 유의적으로 가장 많이 증가하였다. Bacteroidetes는 HFD 그룹에서 유의적으로 감소하였고, S24-7 family가 가장 큰 비율로 감소하였다. 한편 Firmicutes는 HFD:LFD 그룹에서 차이를 보였고, LFD 그룹에서 Lachnospiraceae family가 유의적으로 낮은 비율로 나타난 것이 확인되었다(p<0.05). PICUSt 기반 신진대사 분석에서 LFD 그룹은 아미노산 대사 및 탄수화물 대사에 관여하는 미생물 수가 유의적으로 감소하는 양상을 보였고(p<0.05), 에너지 대사에서는 메탄 대사에 관여하는 미생물이 유의적으로 감소하였다(p<0.01). 한편 HFD 그룹에서는 아미노산 대사에 관여하는 미생물 수가 유의적으로 증가하였다(p<0.05). 글리칸 생합성 및 대사에 관여하는 미생물은 LFD 그룹과 HFD 그룹에서 유의적으로 증가하는 것으로 나타났다(p<0.01). 이상의 결과를 통해 지속적으로 불균형한 식단을 섭취하는 것은 장내 환경을 dysbiosis시켜, 대사성 질환 및 장 기능 저하를 유발할 것으로 예상된다.

Hepatoprotective Effect of Lactiplantibacillus plantarum DSR330 in Mice with High Fat Diet-Induced Nonalcoholic Fatty Liver Disease

  • Lactiplantibacillus plantarum DSR330 (DSR330) has been examined for its antimicrobials production and probiotics. In this study, the hepatoprotective effects of DSR330 were examined against nonalcoholic fatty liver disease (NAFLD) in a high-fat diet (HFD)-fed C57BL/6 mouse model. To induce the development of fatty liver, a HFD was administered for five weeks, and then silymarin (positive control) or DSR330 (108 or 109 CFU/day) was administered along with the HFD for seven weeks. DSR330 significantly decreased body weight and altered serum and hepatic lipid profiles, including a reduction in triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels compared to those in the HFD group. DSR330 significantly alleviated HFD-related hepatic injury by inducing morphological changes and reducing the levels of biomarkers, including AST, ALT, and ALP. Additionally, DSR330 alleviated the expression of SREBP-1c, ACC1, FAS, ACO, PPARα, and CPT-1 in liver cells. Insulin and leptin levels were decreased by DSR330 compared to those observed in the HFD group. However, adiponectin levels were increased, similar to those observed in the ND group. These results demonstrate that L. plantarum DSR330 inhibited HFD-induced hepatic steatosis in mice with NAFLD by modulating various signaling pathways. Hence, the use of probiotics can lead to hepatoprotective effects.

Hypotriglyceridemic effects of brown seaweed consumption via regulation of bile acid excretion and hepatic lipogenesis in high fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: The present study aimed to further investigate the potential health beneficial effects of long-term seaweed supplementation on lipid metabolism and hepatic functions in DIO mice. MATERIALS/METHODS: Four brown seaweeds (Undaria pinnatifida [UP], Laminaria japonica [LJ], Sargassum fulvellum [SF], or Hizikia fusiforme [HF]) were added to a high fat diet (HFD) at a 5% ratio and supplemented to C57BL/6N mice for 16 weeks. Triglycerides (TGs) and total cholesterol (TC) in the liver, feces, and plasma were measured. Fecal bile acid (BA) levels in feces were monitored. Hepatic insulin signaling- and lipogenesis-related proteins were evaluated by Western blot analysis. RESULTS: Fasting blood glucose levels were significantly reduced in the LJ, SF, and HF groups compared to the HFD group by the end of 16-week feeding period. Plasma TG levels and hepatic lipid accumulation were significantly reduced in all 4 seaweed supplemented groups, whereas plasma TC levels were only suppressed in the UP and HF groups compared to the HFD group. Fecal BA levels were significantly elevated by UP, LJ, and SF supplementation compared to HFD feeding only. Lastly, regarding hepatic insulin signaling-related proteins, phosphorylation of 5'-AMP-activated protein kinase was significantly up-regulated by all 4 types of seaweed, whereas phosphorylation of protein kinase B was up-regulated only in the SF and HF groups. Lipogenesis-related proteins in the liver were effectively down-regulated by HF supplementation in DIO mice. CONCLUSIONS: Brown seaweed consumption showed hypotriglyceridemic effects in the prolonged DIO mouse model. Specifically, combinatory regulation of BA excretion and lipogenesis-related proteins in the liver by seaweed supplementation contributed to the reduction of plasma and hepatic TG levels, which inhibited hyperglycemia in DIO mice. Thus, the discrepant and species-specific functions of brown seaweeds provide novel insights for the selection of future targets for therapeutic agents.
