Effect of Genistein and Soy Protein on Lipids Metabolism in Ovariectomized Rats (난소절제 랫드에서 제니스테인과 콩단백질이 지질 대사에 미치는 영향)

  • Postmenopausal women or ovariectomized rats are associated with increased cholesterol levels, which are risk factors of metabolic syndrome and cardiovascular diseases. Increased prevalence of metabolic syndrome after menopause might be associated with estradiol deficiency. Harmful effect of estradiol hampers the casual usage of hormone to prevent the metabolic syndrome. Soy protein has been reported to show several beneficial effects on health, however it is unclear which components of soy protein is responsible for anti-obesity and hypocholesterolemic effects. Soy isoflavones, gem-stein and daizein, are suggested to have anti-obesity and hypocholesterolemic effects but with inconsistency. The present study investigated the effect of supplementation of genistein (experiment I) and soy protein containing isoflavones (experiment II) to high fat diet on body weight gain, food intake, liver and fat tissue weight and the lipid levels in ovariectomized rats. Plasma and hepatic lipid contents and the mRNA levels of genes encoding lipid metabolism related proteins, such as CPT1 and HMGR were measured. Ovariectomy increased body weight, fat tissue weight and plasma and hepatic lipid levels which increase the risk of metabolic syndrome. Soy protein could improve plasma and hepatic lipids levels. Soy protein also increased hepatic CPT1 and HMGR mRNA levels. Plasma and hepatic lipids levels could not be decreased by dietary genistein alone. In contrast, lipids levels could be decreased by isoflavone-fortified soy protein, suggesting that the ingestion of soy protein enriched with isoflavone gives more benefit for protecting postmenopausal women from metabolic syndrome.

The Relationships of Plasma Leptin, Backfat Thickness and TDN Intake across Finishing Stage of Holstein Steers

  • Six 16 months old Holstein steers were offered ad libitum feed for 7 months, to determine the (1) relationships of backfat thickness (BFT) to plasma leptin, and insulin; and (2) associations of TDN intake/kg body weight (BW) to plasma leptin, BFT and insulin. Feed intake, body weight and BFT were measured on selected monthly ages from day 1 to 8, day 1 and 8, and day 8, respectively. Blood was sampled on day 8 and the plasma was analyzed for leptin, insulin, glucose, NEFA, total cholesterol and triglyceride. Body weight and BFT increased, while TDN intake per kg BW decreased from 16 to 23 months old. Plasma leptin increased and mimicked the level of insulin, resulting to significant correlation (r=0.54; p<0.002). TDN intake was negatively related to plasma leptin (r=0.49; p<0.004), insulin (r=0.41; p<0.02) and BFT at 12 to 13th rib (r=0.48; p<0.005). Backfat thickness at 12 to 13th rib was positively related to plasma leptin (r=0.45; p<0.01). Negative associations of TDN intake with plasma leptin and BFT during finishing period suggest long-term involvement of adipose tissues in the feed intake regulation of steers fed high concentrate diet.

Silymarin's Protective Effects and Possible Mechanisms on Alcoholic Fatty Liver for Rats

  • Silymarin has been introduced fairly recently as a hepatoprotective agent. But its mechanisms of action still have not been well established. The aim of this study was to make alcoholic fatty liver model of rats in a short time and investigate silymarin's protective effects and possible mechanisms on alcoholic fatty liver for rats. The model of rat's alcoholic fatty liver was induced by intragastric infusion of ethanol and high-fat diet for six weeks. Histopathological changes were assessed by hematoxylin and eosin staining (HE). The activities of alanine transarninase (ALT) and aspartate aminotransferase (AST), the levels of total bilirubin (TBIL), total cholesterol (TC) and triglyceride (TG) in serum were detected with routine laboratory methods using an autoanalyzer. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GPx) and the level of malondialdehyde (MDA) in liver homogenates were measured by spectrophotometry. The TG content in liver tissue was determined by spectrophotometry. The expression of nuclear factor-${\kappa}B$ (NF-${\kappa}B$), intercellular adhesion molecule-1 (ICAM-1) and interleukin-6 (IL-6) in the liver were analyzed by immunohistochemistry. Silymarin effectively protected liver from alcohol-induced injury as evidenced by improving histological damage situation, reducing ALT and AST activities and TBIL level in serum, increasing SOD and GPx activities and decreasing MDA content in liver homogenates and reducing TG content in liver tissue. Additionally, silymarin markedly downregulated the expression of NF-${\kappa}B$ p65, ICAM-1 and IL-6 in liver tissue. In conclusion, Silymarin could protect against the liver injury caused by ethanol administration. The effect may be related to alleviating lipid peroxidation and inhibiting the expression of NF-${\kappa}B$.

Effects of the Integrated Obesity Management Program, which Includes Exercise Therapy, Psychotherapy and Dietary Treatment, on the Body Composition and Blood Composition of Obese Patients Worked in the Industry (운동치료, 심리치료 및 식이요법이 포함된 통합비만관리프로그램이 산업체에 근무하는 비만 환자의 체성분 및 혈액성분에 미치는 영향)

  • Purpose : The purpose of this study was to find out how changes in body shape of obese patients could be affected by the combined management of diet and psychotherapy along with exercise therapy to control energy imbalance, the cause of obesity. Methods : In this study, the aerobic exercise program was conducted on 12 industrial workers of "D" company for a total of eight weeks for obese patients with a high body mass index (25 kg/㎡ or higher). The experimental group did not apply the program three times a week. The body type analysis was carried out using the body analyzer (Inbody 770, Inbody, Korea) and blood test and psychological test were performed after 8 weeks of exercise before and after exercise. Results : The results of the group showed a change in weight loss of 6.03 kg (p=.000) on average, which is nearly 10 % of the change in weight. It was also shown that the BMI (body mass index) decreased by 1.76 kg/㎡ (p=.000). Abdominal fat (AO) is also 0.14 % (p=.000), waist circumference (WC, 12.72 cm, p=.000), internal fat (VF, 25.12 %, p=.000), liver function levels AST (5.47 U/L, p=.04), ALT (13.64 U/L, p=.000), total cholesterol, pTC -14.22. Conclusion : Based on the results of this study, the obesity management program will be able to maximize the effects of obesity control if exercise programs and psychotherapy are combined.

A Study of the Dietary Intake Status and One Portion Size of Commonly Consumed Food and Dishes in Korean Elderly Women (노년기 여성의 식이 섭취실태와 주요 음식의 1일 1회 섭취분량 조사연구)

  • The use of semiquantitative food frequency questionnaires to estimate dietary intake has become widespread in nutritional epidemiologic studies. It had been suggested that knowledge about a person's usual portion size of each meal in addition to consumption frequency will improve the accuracy of this method. This study was performed to investigate the dietary intake status(nutrient, food, and dish intake) and one portion size of commonly consumed food and dishes to develop a semiquantitative food frequency questionnaire that will be used to expore the relationship between the incidence of cardiovascular disease and diet in Korean women after menopause. Intake of food or dish in 123 elderly women were measured by 3-day food records in March to May, 1996. The results obtained in this study were summarized as follow : 1) Calcium, vitamin A, and vitamin E intakes were less adequate than Recommended dietary allowances(RDA). 2) The subjects who consumed more than 20% of the energy from fat were 35% and the subjects consumed dietary cholesterol more than 300mg were 18$\%$. 3) The most frequently consumed foods or dishes by the subjects were Korean cabbage kimchi〉cooked rice, well-milled〉laver〉apple〉strawberry〉mixed rice〉soybean soup. 4) The major dish groups which contributed to most daily nutrient intakes were cooked rice, Korean cabbage kimchi, soybean soup because of large amount and high frequency of intake although they do not contain large amount of most nutrients. 5) One portion size of commomly consumed foods and dishes in elderly women was different from one portion size suggested in the Korean nutrition society. This study provides information for the design of data-based semiquantitative food frequency questionnaire. for Korean elderly women. Naturally Derived Probiotic Supplementation Effects on Physiological Properties and Manure Gas Emission of Broiler Chickens

  • To investigate the influence of multi-probiotic, fermented ginseng byproduct and fermented sulfone on the performance, intestinal microflora and immunity of broiler, a five weeks trial was conducted with 340, 1-d-old $Ross{\times}Ross$ broiler. All broilers were divided into five different groups having 68 birds in each treatment, and they were assigned as control, antibiotic avilamycin (AB), multi-probiotic (MP), fermented sulfone (FS) and fermented ginseng byproduct (FGB). Each artificial or naturally derived probiotic was inoculated 0.1% level with the basal diet, and all diets were provided to birds for five weeks. Weight gain and feed intake were measured weekly basis, and blood, spleen and feces were collectedand used for the physiological properties of broiler chickens. All performances and cholesterol profiles were not significantly differed but numerically lower level of neutral fat and LDL was found in multi-probiotics and FGB treatments respectively. The salmonella spp and E. coli numbers in the ileum were high in control in relation to those of other treatments and were significantly decreased in antibiotics treatments (p<0.05). In addition, Lactobacillus spp. showed significantly higher proliferation in MP as compared to that of others (p<0.05). Fecal ammonia and $CO_2$ gas emission was significantly decreased in MP, FGB and FS, respectively (p<0.05), but significantly increased proliferation of spleen was determined in MP group in comparison of other treatments (p<0.05). Therefore, the results indicates that multi-probiotics would be valuable feed additives to improve the salmonella, E. coli and Lactobacillus proliferation, and manure gas emission of broiler chickens, but further study related to the production of manure gas emission of MP is necessary.

Case Report of Impaired Fasting Glucose Improved with Korean Medicine Treatment and Dietetic Therapy (양격산화탕 투여와 식이요법을 병행하여 호전된 공복혈당장애 증례 보고)

  • Diabetes mellitus, commonly known as diabetes, comprises a group of metabolic disorders characterized by a high blood sugar level over a prolonged period of time. Diabetes is classified into type 1 diabetes and type 2 diabetes, and the incidence and prevalence of diabetes, mostly type 2, have increased remarkably in recent decades. A diagnosis of type 2 diabetes is greatly influenced by heredity, and it is important to prevent diabetes in people with a family history by improving lifestyle and environmental factors. Controlling overeating, obesity, lack of exercise, and stress is effective to prevent diabetes. The patient in this case report had impaired fasting glucose and mild hyperlipidemia. The patient experienced improvements in these sequelae after the administration of herbal medicine (Yangkyuksanwha-tang) for 12 weeks and the implementation of a plant-based diet. The complete blood count, XXXX, fasting blood sugar, HbA1c, insulin, and C-peptide levels were measured, and the Homeostatic Model Assessment for Insulin Resistance (HOMA-IR) and HOMA-β were calculated according to the FBS and fasting insulin levels. Total cholesterol, triglyceride, FBS, and HbA1c levels all decreased after 12 weeks compared with baseline measures. There was no change in the insulin secretory ability; the C-peptide level did not change as a result of β-cell function; and the HOMA-β level reflected an improved insulin secretory ability.

Lipid Improvement Effect of New manufacturing method produced Kyeongok-go on hyperlipidemia rats (제조방법을 달리한 경옥고의 고콜레스테롤혈증 흰쥐에 미치는 효능 비교 연구)

  • Objectives : Kyeongok-go has the effect of antioxidant, anti-cancer, improving fatigue, and hyperlipidemia, but it is not easy to use in clinical practice becausee of the complicated manufacturing process of heat control and moisture control. The aim of this study was to explore the lipid improvement effect of new manufacturing methods prodeced Kyeongok-go in hyperlipidemia rats. Methods : The SD rats were divided randomly into 8 groups of six animals each : Normal rats (Nor), hyperlipidemia rats (Con), hyperlipidemia rats treated with Spirulina 60 mg/kg(SP60), hyperlipidemia rats treated with Commercial Kyeongok-go 1 g/kg (K0), hyperlipidemia rats treated with Kyeongok-go manufactured by heating mantle 1 g/kg (K1), hyperlipidemia rats treated with ginseng fermentation Kyeongok-go 1 g/kg (KF), hyperlipidemia rats treated with Kyeongok-go with black ginseng 1 g/kg (BK), hyperlipidemia rats treated with black ginseng fermentation Kyeongok-go 1 g/kg (BKF). The experimental group induced hyperlipidemia by eating high cholesterol diet for 8 weeks. Also, we compared Kyeongok-go manufacturing method using the new method compared with traditional method. Results : As a result, the improvement effect of hyperlipemia using the new method showed the similar or better improvement effect compared with the conventional method, and it is considered that the Kyeongok-go manufacturing method using the new method can be substituted for the traditional Kyeongok-go manufacturing method. Conclusion : In conclusion, these results suggest that new manufacturing methods prodeced Kyeongok-go has the potential as a anti-hyperlipidemia material which possess lipid improvement activities.

  • Rabbit meat has high nutritional and dietetic characteristics, but its consumption rate is comparatively lower than other meat types. The nutritional profile of rabbit meat, by comparison with beef, pork, and poultry, is attributed to relatively higher proportions of n-3 fatty acids and low amounts of intramuscular fat, cholesterol, and sodium, indicating its consumption may provide health benefits to consumers. But, the quality attributes of rabbit meat can be originated from different factors such as genetics, environment, diet, rearing system, pre-, peri-, and post-slaughter conditions, and others. Different rabbit breeds and the anatomical location of muscles may also affect the nutritional profile and physicochemical properties of rabbit meat. However, adequate information about the effect of those two factors on rabbit meat is limited. Therefore, cumulative information on nutritional composition and carcass and meat quality attributes of rabbit meat in terms of different breeds and muscle types and associated factors is more important for the production and processing of rabbits. Moreover, some studies reported that rabbit meat proteins exhibited angiotensin-converting enzyme inhibitory characteristics and antioxidant properties. The aim of this review is to elucidate the determinants of rabbit meat quality of different breeds and its influencing factors. In addition, the proven biological activities of rabbit meat are introduced to ensure consumer satisfaction.

Cryptotanshinone promotes brown fat activity by AMPK activation to inhibit obesity

  • BACKGROUND/OBJECTIVES: Activating brown adipose tissue (BAT) and browning of white adipose tissue (WAT) can protect against obesity and obesity-related metabolic conditions. Cryptotanshinone (CT) regulates lipid metabolism and significantly ameliorates insulin resistance. Adenosine-5'-monophosphate (AMP)-activated protein kinase (AMPK), a receptor for cellular energy metabolism, is believed to regulate brown fat activity in humans. MATERIALS/METHODS: The in vivo study included high-fat-fed obese mice administered orally 200/400 mg/kg/d CT. They were evaluated through weight measurement, the intraperitoneal glucose tolerance test (IPGTT), intraperitoneal insulin tolerance test (IPITT), cold stimulation test, serum lipid (total cholesterol, triglycerides, and low-density lipoprotein) measurement, hematoxylin and eosin staining, and immunohistochemistry. Furthermore, the in vitro study investigated primary adipose mesenchymal stem cells (MSCs) with incubation of CT and AMPK agonists (acadesine)/inhibitor (Compound C). Cells were evaluated using Oil Red O staining, Alizarin red staining, flow cytometry, and immunofluorescence staining to identify and observe the osteogenic versus adipogenic differentiation. Quantitative real-time polymerase chain reaction and the Western blot were used to observe related gene expression. RESULTS: In the diet-induced obesity mouse model mice CT suppressed body weight, food intake, glucose levels in the IPGTT and IPTT, serum lipids, the volume of adipose tissue, and increased thermogenesis, uncoupling protein 1, and the AMPK pathway expression. In the in vitro study, CT prevented the formation of lipid droplets from MSCs while activating brown genes and the AMPK pathway. AMPK activator enhanced CT's effects, while the AMPK inhibitor reversed the effects of CT. CONCLUSION: CT promotes adipose tissue browning to increase body thermogenesis and reduce obesity by activating the AMPK pathway. This study provides an experimental foundation for the use of CT in obesity treatment.
