Identification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • A novel purification method was developed for producing homoharringtonine from Cephalotaxus koreana, to guarantee high purity and yield. Our simple, efficient procedure for isolating and purifying homoharringtonine from C. koreanabiomass consisted of solvent extraction, synthetic adsorbent treatment, low-pressure chromatography, followed by high performance liquid chromatography (HPLC). The use of active clay treatment and silica gel low-pressure chromatography in the pre-purification process allowed for the rapid, efficient separation of homoharringtonine from interfering compounds and dramatically increased the yield and purity of crude homoharringtonine for high-performance liquid chromatography (HPLC) purification steps compared with alternative processes. Homoharringtonine could be obtained simply with high yield and purity from biomass using this purification method, while minimizing solvent use and the scale and complexity of HPLC operations for homoharringtonine purification. Purified homoharringtonine was identified and characterized.

Analytical Methods for the Isolation of Dehydrotomatine and ${\alpha}$-Tomatine in Tomato Fruits by Use of Alumina Column Chromatography and High-Performance Liquid Chromatography (Alumina Column Chromatography와 HPLC에 의한 토마토의 Dehydrotomatine 및 ${\alpha}$-Tomatine 단리방법 연구)

  • Tomato fruits(Lycoperisicon esculentum) synthesize the glycoalkaloids dehydrotomatine and ${\alpha}$-tomatine, possibly as defense against bacteria, fungi and insects. We developed a new effective method to prepare and purify dehydrotomatine and ${\alpha}$-tomatine that exists in tomato fruits using alumina column chromatography and high performance liquid chromatography (HPLC). The tomato glycoalkaloids(TGA) in tomato was extracted with 2% acetic acid, and then precipitated with ammonium hydroxide(pH=10.5). The dry precipitate substance was applied on alumina column, and then fractionated with water saturated n-butylalcohol. The TGA(Fr. No. 26~36) were collected and dried under reduced pressure. The TGA was performed on a reverse phase HPLC(Inertsil ODS-2, $5\;{\mu}m$), eluted with acetonitrile/20mM $KH_2PO_4$(24:76, v/v) at 208 nm. Two peaks were detected on HPLC, and individual peak was collected by repeating HPLC. Furthermore, to confirm the identity dehydrotomatine and ${\alpha}$-tomatine, each peak isolated was hydrolyzed with 1N HCl into sugar and aglycone tomatidine. The sugars were converted to trimethylsilyl ester derivatives. The nature and molar ratios of sugars were identified by gas-liquid chromatography(GLC) and the aglycone by high-performance liquid chromatography(HPLC). The first peak (Rt=17.5 min) eluted from HPLC was identified as dehydrotomatine, and second peak(Rt=21.0 min) was as ${\alpha}$-tomatine. This technique has been used effectively to prepare and isolate dehydrotomatine and ${\alpha}$-tomatine from tomato fruits.

Simultaneous detection for synthetic antimicrobials in muscle by high performance liquid chromatography-mass selective detector (HPLC-MSD) (HPLC-MSD 를 이용한 식육 중 합성항균제의 동시분석)

  • This study was conducted to develop the analytical method about simultaneous determination for synthetic antimicrobials in muscle by high performance liquid chromatography - mass selective detector (HPLC- MSD). Solid phase extraction (SPE), matrix solid phase dispersion (MSPD) and liquid-liquid extraction (LLE) have been adapted as pretreatment procedures for HPLC- MSD. Among various solvent tested, methanol was chosen for extraction of synthetic antimicrobials in muscles. For the optimized response, the values of various MS parameters including fragment voltage, drying gas flow, nebulizer pressure, drying gas temperature were verified. The average recovery rates using MSPD and SPE for muscles of bovine and pork were 78.9-127.1% and 78.3-121.7%, respectively. This method was verified the satisfactory performance for fourteen synthetic antimicrobials excepting carbadox in muscle of pork as detection limit of $0.05{\mu}g/g$ on API/ES SIM mode.

  • Currently in Korea, standard operating procedure for the analyses of phenolic compounds in water is the spectrometric comparison of colors developed by 4-amino antipyrin with phenolic compounds. It is however that this method cannot identify individual compound and that some phenolic compounds do not react with 4-amino antipyrin. Spectrometric determinations of phenolic compounds were compared with chromatographic analyses of gas chromatography (GC) and high pressure liquid chromatography (HPLC) of various phenolic compounds. Individual phenolic compounds could be determined by both chromatographic methods but HPLC methods were more precise with lower detection levels in general.

  • A new isolation and purification method was developed aiming at increasing yield and purity for homoharringtonine. This method was a simple andefficient procedures, for the isolation and purification of homoharringtonine from Cephalotaxus koreana, consisting of solvent extraction, adsorbent treatment, low-pressure chromatography, and high performance liquid chromatography (HPLC). The crude homoharringtonine was efficiently pre-purified adequately to perform HPLC through a combination withadsorbent treatment and low-pressure chromatogaphy. The homoharringtonine can be simply obtained with high purity and yield from crude homoharringtonine by HPLC. Purified homoharringtonine was characterized.

  • The hydrolysis of penicillin G, carbenicillin and ampicillin in pure water at room temperature was studied by high pressure liquid chromatography electrospray ionization mass spectrometry. Hydrolysis of ampicillin did not occur under these conditions; however, penicillin G and carbenicillin were completely hydrolyzed after seven days. A short interpretation of this difference is proposed. The mass spectrometric behaviour, namely ESI response and fragmentation pathway, of hydrolyzed penicillin G and hydrolyzed carbenicillin have been also discussed.

  • A new application of high pressure liquid chromatography for the determination of fenitrothion and cis- and trans-neopynamin in insecticidal preparations was investigated. Optimum conditions for a good separation and determination were determined; solvent system: dichloromethane + n-hexane = 17 + 83; Bow rate: 0.5ml/min; column: u-porasil ($4mm{\times}3Ocm$); absorbance wavelength: 254nm; 0.05 AUFS and sample size: 30 ul. Recovery of fenicrothion, cis- and trans-neopynamin from mixed artificial preparations was 99.6%, 99.7% and 99.8% respectively. Also reproducibility tests showed that the coefficient of variation was 0.89% for fenitrothion, 0.74% cis-neopynamin and 1.1% for trans-neopynamin. There was no interference with insecticidal preparation containing DDVP, allethrin, S-421 and kerosene. HPLC method was rapid, accurate and it gave better reproducibility and higher sensitivity than any other analytical method. It was considered that HPLC could be greatly applied to the analysis of fenitrothion and neopynamin in insecticidal preparations.

  • A Liquid Chromatogaphic Method was applied to analyze the following nine preservatives used for foods simultaneously; propionic acid, sorbic acid, benzoic acid, dehydroacetic acid, ethyl p-hydroxy benzoate, propyl p-hydroxy benzoate, i-butyl p-hydroxy benzoate, butyl p-hydroxy benzoate, ethyl salycilate. A cosmosil $5C_{I8}$ was used and 1% phosphoric acid-acetonitrile (60 : 40) was used as the mobile phase. Pretreatment of the food sample with steam distillation improved the resolution of peaks at 220 nm.

Analysis of Carotenoids in 25 Indigenous Korean Coral Extracts

  • In this study, methanol extracts from 25 indigenous Korean corals were prepared and their carotenoid constituents were analyzed by high-performance liquid chromatography-atmospheric pressure chemical ionization-mass spectrometry (HPLC-APCI-MS). Among them, extracts from nine species showed detectable peaks in the HPLC chromatogram at 450 nm and the ultraviolet/visible spectra exhibiting carotenoid-specific characteristics were chosen. The mass data of carotenoid peaks revealed that only peridinin could be identified based on literature comparison and suggested the potential presence of novel carotenoid structures. This is the first reported investigation of indigenous Korean coral carotenoids and further work is needed to explore the carotenoids and their potential roles in the ecosystem of indigenous Korean corals.

  • Attempts were made to elucidate the process of biodegradation of furfural. Zoogloea sp. isolated from soil could convert furfural into 2-furoic acid by a certain enzyme (s) and also accumulated it extracellularly. This substance was extracted with diethyl ether and identified with U.V. spectrophotometry, high pressure liquid chromatography (HPLC) and IR spectrophotometry.
