무궁화 초콜릿의 품질특성 및 항산화활성 (Quality Characteristics and Antioxidant Activity of Chocolate with Mugunghwa (Hibiscus syriacus L.))

  • This study was conducted to determine the mixing conditions for Hibiscus syriacus L. chocolate depending on different addition ratios of Hibiscus syriacus L. and examine functionality in order to use Hibiscus syriacus L., which has excellent functionality and symbolizes Korea. To accomplish this, Hibiscus syriacus L. chocolate was manufactured by adding 0%, 2.5, 5, 7.5, and 10% of Hibiscus syriacus L. powder from the 'Samchulli' (Hibiscus syriacus 'Samchulli', which has reddish pink flower with red eye spot: SKK 14-2-72) variety to a coverture of white chocolate after which the antioxidant activity and quality characteristics were analyzed. As the amount of Hibiscus syriacus L. powder added to the chocolate increased, the chocolate color became darker, the pH decreased, and the sugar content and hardness increased. As a result of the sensory evaluation, the degree of preference increased as the amount of added Hibiscus syriacus L. powder increased to 7.5%. The degree of preference for color and flavor was the highest in the 5% added group, but the overall preference was 7.5% added chocolate. These findings demonstrate that adding Hibiscus syriacus L. powder to chocolate can increase chocolate functionality and preference and that there is a high possibility of Hibiscus syriacus L. chocolate development.

품종별 무궁화 분말의 항산화활성 및 품질특성 (Antioxidant Activity and Quality Characteristics of Hibiscus syriacus Cultivars Powders)

  • The aim of this study was to analyze the functional components of five cultivars of 'Mugunghwa (Hibiscus syriacus L.)' for the development of 'Mugunghwa'-added foods. There were five varieties, namely, Hibiscus syriacus 'Hwanhee' (HH), Hibiscus syriacus 'Samchulli' (SC), Hibiscus syriacus 'Harmony' (HM), Hibiscus syriacus 'Baekgu' (BG), and Hibiscus sabdariffa L (HB). Of the five varieties tested, HH extract showed the highest total phenolic content (131.85 GAE mg/g), DPPH free radical scavenging activity (95.18%), and anthocyanin and sugar contents. The sensory quality of powders from each Hibiscus variety was evaluated, and it was found that overall palatability decreased in the order: HH, SC, HB, HM, and BG. Thus, addition of Hibiscus variety, which has excellent functionality and palatability, to foods confers an extra element of Korean history and culture, which is expected to improve the marketability of various food products.

  • 무궁화의 종내(終內) 일대잡종(一代雜種)에서 화색(花色)과 화형(花型)의 분리현상(分離現象)을 조사(調査)할 목적(目的)으로 품종간(品種間) 교배(交配)를 하여 다음과 같은 결과(結果)를 얻었다. 1. 화판(花瓣)의 자주색(紫朱色)은 백색(白色)에 대(對)하여 dominance하고 세포질(細胞質) 유전(遺傳)에 의(依)한 것이라 추측(推測)된다. 2. 화판(花瓣)의 홑과 겹은 Ss, dd인자(因子)에 의(依)하여 발생(發生)한다. 3. 배수체(倍數体)무궁화와의 $F_1$의 화색(花色) 화형(花型)에는 큰 변화(變化)가 없었다. 4. Open-pollination되어 얻은 $F_1$에서 화색(花色)에 많은 변이(變異)가 있었다. 5. 유색(有色)견 화판(花瓣)무궁화와의 $F_1$에서는 화판(花瓣)에 적색(赤色) 반문(斑紋)이 있는 변이개체(變異個体)가 다수(多數) 발생(發生)되었다.

Variation in Flower Color among Hybrids of Jeoktanshim Hibiscus syriacus L

  • This study was performed to provide basic information of the development and breeding of new cultivars of Hibiscus syriacus L., which have more beautiful and diverse flowers. Morphological characteristics of the flowers and leaves, and genetic variation of the flowers color of two Jeoktanshim-line cultivars, Bulsae and Pyungsung, were crossed each other. The result of the cross between Bulsae and Pyungsung are as follows: Mean flower height and width were 5.35 cm and 7.69 cm, respectively. Mean length and width of petal were 5.43 cm and 3.80 cm respectively, and mean length and width of flower pistil were 4.67 cm and 0.44 cm, respectively. The flower color of all ten individuals was the color of the Jeoktanshim-line, and Pyungsung, and all flower type were I-c type.

  • Hibiscus syriacus L. (Malvaceae) is widely distributed over Korean, China, India and Siberia. The dried flower of Hibiscus syriacus is used as a folk medicine for curing of hematochezia, dysentery, obstruction due to wind-phlegm, regurgitation, and vomiting of food, and the dried root bark is used antipyretic, anthelmintic and antifungal agents. From a chloroform extract of root bark of this plant, compound I, II, and III were isolated and the structures were elucidated by various spectroscopic analyses. These compounds were identified as syringaresinol. E-N-feruloyltyramine, and Z-N-feruloyltyramine, respectively and were isolated from this plant for the first time. Compound II and III exhibited lipid peroxidation inhibitory activities with $IC_{50}$ of 15.5 and 28.6 ${\mu}g/ml$, respectively.

Immunostimulatory Activity of Hibiscus syriacus L. Leaves in Mouse Macrophages, RAW264.7 cells, and Immunosuppressed Mice

  • Under the COVID-19 pandemic, interest in immune enhancement is increasing. Although the immune-enhancing activity of plants of the genus Hibiscus has been reported, there is no study on the immune-enhancing activity of H. syriacus. Thus, in this study, we investigated the immune-enhancing activity of Hibiscus syriacus leaves (HSL) in mouse macrophages, RAW264.7 cells, and immunosuppressed mice. HSL increased the production of immunostimulatory factors such as nitric oxide (NO), inducible nitric oxide synthase (iNOS), interleukin-1β (IL-1β), and tumor necrosis factor-α (TNF-α) and activated the phagocytosis in RAW264.7 cells. The HSL-mediated production of immunostimulatory factors was dependent on toll-like receptor 4 (TLR4), p38, and c-Jun N-terminal kinase (JNK) in RAW264.7 cells. In the immunosuppressed mouse model, HSL increased the spleen index, the levels of the cytokines, and the numbers of lymphocytes, neutrophils, and monocytes. Taken together, HSL may be considered to have immune-enhancing activity and be expected to be used as a potential immune-enhancing agent.

Development of an efficient protocol for high-frequency regeneration system in Hibiscus syriacus L.

  • A rapid and efficient in vitro regeneration system was established for Hibiscus syriacus L. The successful regeneration protocol employs induction of shoot organogenesis on leaf, petiole, and root explants. Among the various plant growth regulators evaluated, thidiazuron (TDZ) was the most effective for inducing rapid shoot formation. Most efficient shoot regeneration frequency was obtained from Murashige and Skoog (MS) media containing 0.01 mg/L TDZ. Regeneration efficiency was highest in the roots, and lowest in the leaves. A combination of 0.01 mg/L TDZ with benzyladenine (BAP) markedly improved the frequency of shoot differentiation from the root (up to 98%) and petiole (up to 88%) explants. Furthermore, leaf and petiole explants showed the highest frequency of shoot induction in half-strength MS media containing 0.01 mg/L TDZ and 1.0 mg/L BAP, while root explants formed the greatest number of shoots when 0.01 mg/L TDZ and 0.1 mg/L BAP were added to half-strength MS media. Although the frequency of shoot differentiation from leaf explants was only 50%, the leaf is considered the most efficient plant organ for use in tissue culture because leaves are easier to obtain than roots and petioles. Our findings show that various organs of H. syriacus can be used for plant regeneration, and the protocol developed in this study may be applicable in the horticulture industry.

무궁화 품종내의 flavonoid 성분분포에 관한연구 (Studies on the flavonoids of the Hibiscus syriacus L. Complex)

  • 무궁화 품종 6계통, 총 68종류를 선발하여 flavonoid 성분을 비교하였다. 2차원으로 크로마토그래피를 전개 후 13개의 flavonoid가 관찰되었고 spot 5번의 flavonoid 함량이 전체의 50%를 차지하였으며 나머지는 미량으로 관찰되었다. 6개의 group별 spot출현 양상은 각각 차이를 보였으나 group 내에서는 거의 동일한 것으로 나타났다. 13개의 spot 중 1, 4, 6은 flavonol의 특성을 나타냈으며 나머지 10개의 spot들이 flavone의 특성을 나타내었다. 품종별 flavone함량 및 분포는 백색계통이나 자주 및 청색 계통에는 많은 양이 존재 하였고 pink와 적색 group에서는 매우 적은 양이 분포하였다. Purple spot들의 flavonoid aglycone은 4’, 5-OH의 aglycone 특성을 보였으며 이들 중 spot 5는 saponarin, 7은 vitexin, 9는 xylovitexin, 11은 rhamnosylvitexin으로 동정되었으며 13은 apigenin-7-O-diglycoside로 추정되었다. Saponarin은 백색무궁화에서 전체 flavone량의 50% 이상을 점유하였다.

왜성 변이 품종 '꼬마' 무궁화의 캘러스 유도 및 지상부 형성에 식물생장조절물질이 미치는 영향 (The Effect of Plant Growth Regulators on Callus Induction and Shoot Regeneration from the Dwarf Type Variety, Hibiscus syriacus L.var. Ggoma)

  • 'Ggoma' is a new Hibiscus dwarf type variety developed by gamma irradiation at the Korea Atomic Energy Research Institute (KAERI). This study was conducted to determine the best optimum cultural callus formation and shoot regeneration condition. Sterilized leaf tissues were cultured on MS (Marashige and skoog's) medium containing 3% sucrose, 0.8% agar with different concentration and combination of TDZ, 2, 4-D, KT, BA, and 2iP for 4 weeks in vitro culture. MS medium containing 2,4-D $0.1mg\;l^{-1}$ and BA $0.5mg\;l^{-1}$ were most effective on callus formation and growth. After 4 weeks, callus was transferred on BA (0.5, 1, $1.5mg\;l^{-1}$) and TDZ (0.1, 0.2, $0.3mg\;l^{-1}$) for shoot formation. The best condition for inducing the shoot from callus was BA $1.5mg\;l^{-1}$ and TDZ $0.3mg\;l^{-1}$. This result will be useful for the rapid multiplication of Hibiscus syriacus L.var. Ggoma.
