할미송이버섯으로부터 혈전용해효소의 정제 및 특성 연구 (Purification and Characterization of Fibrinolytic Enzymes from Tricholoma saponaceum)

  • 할미송이버섯으로부터 3단계를 거처 분자량이 각각 18 kDa(FE-1)과 18.2 kDa(FE-2)인 두 개의 혈전용해효소를 분리 정제하였다. 이 효소들은 지금까지 알려진 혈전용해 효소의 질량에 비해 비교적 작았고 두 효소(FE-1과 FE-2)의 15번째까지의 N-terminal amino acid서열은 A-L-Y-V-G-X-S-P-X-Q-Q-S-L-L-V로 같았다. FE-1은 ICP-MS의 결과 $Zn^{2+}$을 mole당 0.98 mole 포함하고 있는 metalloprotease로 pH 7.5와 $55^{\circ}C$에서 가장 큰 활성을 나타내고, 1,10-phenanthroline의 첨가 시 저하된 활성은 $Zn^{2+}$$Co^{2+}$의 첨가에 의해 재생되었다.

깍두기의 숙성(熟成)에 미치는 감압(減壓) 및 Polyethylene Film 포장처리(包裝處理) 효과(效果) (Effect of Subatomospheric Pressure and Polyethylene Film Package on the Kacdugi Fermentation)

  • Kacdugi fermented in the subatomospheric pressure(350mmHg) and polyethylene film package to improve the quality and to investigate the effect of fermentative control at $25^{\circ}C$. Brix degree, acidity, vitamin C content and number of total microbe and Lactobacilli was determined and also, the edible period of kacdugi was checked up by sensory assessment. The increasing rate of brix degree during kacdugi fermentation was high in the subatomospheric pressure, but decreased at the last period of fermentation as same tendency to the control, and it was preferably increased at the last period of fermentation in the polyethylene film package. However, the acidity was higher in the control than sbuatomospheric pressure but it was low in the polyethylene film package. Vitamin C content was high in the control at beginning and middle period of fermentation but high in sbuatomospheric pressure, and was low in the polyethylene film package at the last period of fermentation. The number of Lactobacilli was more in the subatomospheric pressure and polyethylene film package than the control, but it was suddenly incressed for the total microbe in the polyethylene film package at last period of fermentation. The edible periods of kacdugi by the sensory assessment of sour flavor, hardness and complex flavor was second days in the control. third days in the polyethylene film package, and fifth days in the subatomospheric pressure after soaking.

주의력결핍 과잉행동장애와 뚜렛증후군 아동의 모발 중금속 분석 (Analysis of Heavy Metals in the Hair of Children with Attention-Deficit Hyperactivity Disorder and Tourette's Syndrome)

  • Objectives : The purpose of this study was to examine the association of exposures to heavy metals with positive diagnosis for attention-deficit hyperactivity disorder (ADHD) and Tourette's syndrome (TS). Methods : Study participants included 27 children diagnosed with ADHD ($9.9{\pm}2.9$ years of age), 21 diagnosed with Tourette's disorder ($10.7{\pm}2.2$ years of age), and 45 normal control children ($9.6{\pm}0.5$ years of age). A Perkin-Elmer mass spectrometer was used to measure the concentrations of 5 heavy metals (Pb, Cd, U, Be, Hg) in hair samples obtained from each participant. Each heavy metal concentration was compared among the groups by use of a Kruskal-Wallis test. Results : The levels of lead (p=.006) and cadmium (p=.037) observed in the hair of children diagnosed with ADHD were significantly higher than those found in the control subjects. There were no significant differences observed for heavy metal levels when comparing TS and control subjects. Conclusion : We confirmed that lead exposure is a risk factor for ADHD. We also identified that cadmium may be a new candidate risk factor for manifestation of ADHD. We did not find an association between heavy metals and manifestation of TS.

톱밥으로부터 생산되는 개질 바이오오일 생산공장의 공정모사 및 경제성 분석 (Process Simulation and Economic Feasibility of Upgraded Biooil Production Plant from Sawdust)

  • 본 연구의 목표는 톱밥의 건조, 급속열분해, 바이오오일 응축, 바이오오일 수첨개질, 전기생산, 폐수처리를 포함하는 2개의 바이오오일 생산공정들에 대한 경제적 타당성을 평가하는 것이다. 첫번째 공정은 수소생산을 위한 steam-methane reforming (SMR)을 포함하는 바이오오일 생산공정이다(Case 1). 두번째 공정은 SMR을 포함하지 않고 수소를 외부로부터 공급받아 수첨개질을 수행하는 바이오오일 생산공정이다(Case 2). 상용공정모사기인 ASPEN Plus를 이용하여 이두 공정에 대한 공정흐름도를 구축하였고, 물질 및 에너지 수지식을 계산하였다. 원료로서 40%의 수분을 포함하는 톱밥 100 t/d, 30% 자기자본비율, 자기자본에 상응하는 자본지출, 수소 구입가 $1,050/ton, 완전건조된 원료대비 수송용 연료 수율 20% (Case 1) 및 25% (Case 2) 로 가정하고, 4단계 경제성 분석기법인 4-level EP를 사용하여 기술경제성 분석을 수행하였다. 총투자비(TCI), 총생산비(TPC), 연간판매금액(ASR), 그리고 개질 바이오오일의 최소판매가격(MFSP)은 Case 1에 대하여 $22.2 million, $3.98 million/yr, $4.64 million/yr, 그리고 $1.56/l 이고, Case 2에 대하여 $16.1 million, $5.20 million/yr, $5.55 million/yr, $1.18/l로 산출되었다. 수소를 직접 생산하는 Case 1과 수소를 외부로 부터 공급받는 Case 2의 투자회수율(ROI)와 투자회수기간(PBP)은 큰 차이를 보여주지 않았고, Case 1과 Case 2의 원료 공급량을 1,500 t/d로 증가할 경우, ROI는 15% 이상으로 향상될 것으로 예상되었다.

발 부분 온수 열자극시 인체 현상에 미치는 영향 (Effect of human biosignal according as foot was heating with hot water)

  • 족탕기(STYX ford202)를 이용하여 발 부분의 온열수 자극을 인가하여 생체 현상의 각종 변화를 계측하였다. 생체 현상의 변화 데이터는 발 부분의 온수 열 자극을 43$^{\circ}C$, 45$^{\circ}C$의 조건으로 10명의 남여 사람을 10일 동안 1회 측정시 5분, 10분, 15분, 20분, 경과 실험을 통해 9가지 생리적 요소를 측정하여 분석하였다. 생체신호요소의 측정의 분석 결과는 이마온도는 -0.69 $\pm$ 0.01 감소하였고, 다리온도는 1.51 $\pm$ 0.22 증가되었다. 혈류($m\ell/min$)는 1.18 $\pm$ 0.50 증가되며, 혈압(mmHg)는 -1.49$\pm$ 2.81, (min) -0.06$\pm$ 0.13 감소 현상을 보였다. 맥박수의 변화는 6.97 $\pm$ 0.72 증가하며, 혈당은 ($mg/d\ell$) : -2.41 $\pm$ 1.55 감소되었고, 피부산소포화도 (%): 1.34 $\pm$ 0.28 증가하였다. 또한 체지방(%) -1.75$\pm$ 0.15 및 체중은 (kr): -0.10 $\pm$ 0.04 감소 현상으로 나타났다.

고도 호염성 Halobacterium sp.가 생산하는 protease의 특성 (Characteristics of the protease from the extreme halophile, Halobacterium sp.)

  • Halobacterium sp.가 생산하는 protease를 ethanol 침전, S데hadexG-100 및 G-75로 겔 여과하여 비활성이 364units/mg protein 수율이 14%로 정제할 수 있었다. 정제호소의 casein에 대한 Km값은 $4.2{\times}10^{-4}M$이었다. 이 효소의 반응 최적온도 및 pH는 $35^{\circ}C$와 pH 8.0이었고, 온도 및 pH에 대한 안정성은 $50^{\circ}C$까지와 pH $5.0{\sim}11.0$범위이었다. $Ca^{2+}$$Mg^{2+}$ 이온은 효소의 활성을 증가시켰고 $Fe^{3+},\;Zn^{2+},\;Cu^{2+},\;Hg^{2+},\;Cd^{2+}$ 이온은 저해하였다. 정제호소는 기질용액중에 NaCl농도가 증가함에 따라 효소활성이 감소하고 2M NaCl 농도 이하에서 투석하면 활성이 저하되었으며 증류수로 2시간 투석으로 완전히 실활되었다. 그러나 0.1M $CaCl_2$용액으로는 6시간 투석하여도 활성은 그대로 유지되었다.

Theoretical gravity studies on roles of convection in crystal growth of $Hg_2Cl_2$-Xe by physical vapor transport under normal and high gravity environments

  • Particular interest in the role of convection in vapor crystal growth has arisen since some single crystals under high gravity acceleration of $10g_0$ appear considerably larger than those under normal gravity acceleration ($1g_0$). For both ${\Delta}T=60\;K$ and 90 K, the mass flux increases by a factor of 3 with increasing the gravity acceleration from $1g_0$ up to $10g_0$. On the other hand, for ${\Delta}T=30\;K$, the flux is increased by a factor of 1.36 for the range of $1g_0{\leq}g{\leq}10g_0$. The maximum growth rates for $1g_0$, $4g_0$, $10g_0$ appear approximately in the neighborhood of y = 0.5, and the growth rates shows asymmetrical patterns, which indicate the occurrence of either one single or more than one convective cell. The maximum growth rate for $10g_0$ is nearly greater than that for $1g_0$ by a factor of 2.0 at $P_B=20\;Torr$. For three different gravity levels of $1g_0$, $4g_0$ and $10g_0$, the maximum growth rates are greater than the minimum rates by a factor of nearly 3.0, based on $P_B=20\;Torr$. The mass flux increases with increasing the gravity acceleration, for $1g_0{\leq}g_y{\leq}10g_0$, and decreases with increasing the partial pressure of component B, xenon (Xe), $P_B$. The $|U|_{max}$ is directly proportional to the gravity acceleration for $20\;Torr{\leq}P_B{\leq}300\;Torr$. As the partial pressure of $P_B$ (Torr) decreases from 300 Torr to 20 Torr, the slopes of the $|U|_{max}s$ versus the gravity accelerations increase from 0.1 sec to 0.17 sec. The mass flux of $Hg_2Cl_2$ is exponentially decayed with increasing the partial pressure of component B, $P_B$ (Torr) from 20 Torr up to 300 Torr.

Erythropoietin 검사법(檢査法)에 관(關)한 연구(硏究) (Studies on Erythropoietin Bioassay Method)

  • It is the purpose of this paper to design the most preferable method of erythropoietin bioassay in Korea. Bioassay utilizing polycythemic mice are currently in general use for the indirect determination of erythropoietin. Assay animals are usually prepared either by transfusion or by exposure to reduced oxygen tension in specially constructed chamber. We prepared the polycythemic, mice by the specially constructed hypobaric chamber. We observed weights and hematocrits of the mice in the hypobaric chamber, then hematocrits and 72 hours $^{59}Fe$ red cell upatke ratio of the polycythemic mice induced by hypoxia after removal from the hypobaric chamber. We designed the method of erythropoietin bioassay according to the results obtained by above experiments. Then we measured the 72 hours $^{59}Fe$ red cell uptake ratio of the polycythemic mice with normal saline, normal plasma and anemic plasma according to the method we designed. The results are followed: 1. The hematocrits of the mice in hypobaric chamber increased to 74% in 14 days It is preferable to maintain the pressure of the chamber to 400mmHg for first 4 days then 300mmHg for last 10 days to reduce the death rate and time consuming in hypobaric chamber. 2. After removal from the hypobaric chamber, the 72 hours $^{59}Fe$ red cell uptake ratio decreased rapidly and maintained the lowest level from the fourth day to tenth day. 3. We design the method of erythropoietin bioassay according to the results of above experiment and to the half life of erythropoietin. 4. The Korean product S9Fe is mixture of $^{59}Fe\;and\;^{55}Fe$. And the $^{59}Fe$ red cell uptake ratio in normal mice was far less with Korean product $^{59}Fe$ than with pure $^{59}Fe$ of foreign product. So it is desirable to use pure $^{59}Fe$ in this method of erythropoietin bioassay. 5. Considering the cost, the technique, the time consuming and the sensitivity it is the most preferable method of erythropoietin bioassay in Korea using hypobaric chamber to induce the polycythemia.

Bacillus subtilis ${\beta}-Galactosidase$의 고정화에 관한 연구 (Studies on the Immobilization of ${\beta}-Galactosidase$ from Bacillus subtilis)

  • Bacillus subtilis HP-4의 ${\beta}-galactosidase$를 추출, 정제하여 효소의 고정화조건 및 고정화 효소의 특성을 조사하였다. B. subtilis를 배양하여 얻은 조효소액을 acetone분획 DEAE-cellulose에 의한 ion exchange chromatography, Sephadex G-100에 의한 gel filtration과정을 통하여 정제하였던 바 약 68% 정제되었으며, 이때의 회수율은 19.9%이었다. 본 효소의 최적 고정화조건은 2%(w/v)의 sodium alginate농도, 15%(v/v)의 효소농도, 그리고 2%,(w/v)의 $CaCl_2$ 농도에서 2시간 교반하였을 때 이었다. 고정화 효소의 최적온도와 pH값은 각각 $55^{\circ}C$와 6.5이었다. 고정화 효소의 활성은 Hg이온과 Cu이온에 의하여 저해되었으며, EDTA, 2-mercaptoethanol, KCN 등의 금속이온 그리고 보호제에 의한 영향은 없었다. 고정화 효소의 ONPG와 유당에 대한 $K_m$값과 $V_{max}$값은 각각 $1.82{\times}10^{-2}M$$2.94{\times}10^{-2}M$ 그리고 $3.57{\times}10^{-8}mole/min$$1.68{\times}10^{-7}mole/min$이었다. 고정화 효소를 $4^{\circ}C$에서 40일간 저장 후 잔존활성과 5회 재사용 후 잔존활성을 조사한 결과는 각각 95%와 81%이었으며, 탈지유(4.8% 유당)와 5% 유당용액에서 $50^{\circ}C$, 9시간 반응시켰을 때 유당분해율은 각각 51%와 43%이었다.

수도권 남녀대학신입생의 체질량지수에 따른 건강행동과 영양상태 (Nutritional Status and Health Habit of College Students by Body Mass Index)

  • It is important for young adults to establish good dietary behavior for healthy life. The prevalence of obesity in college students has increased gradually. The obesity is one of the most prevalent health problems in S. Korea. Moreover, obesity is closely related with the disease such as cardiovascular disease, diabetes mellitus. The purpose of this study was to evaluate health practice, serum components and nutrient intake of the college students according to Body Mass Index (BMI) level. 400 male and female freshmen students in 4 year university located near to Seoul metropolitan area were surveyed respectively through the health check-up procedure for college entrance in February. Survey was carried out for them by questionnaires, including a 24-hour dietary recall and anthropometric measurements. Blood samples were obtained while fasting. Nutrient intake was expressed by DDS(Dietary Diversity Score by 5 food groups) and DVS(Dietary Variety Score). Nutrition level was analyzed by Can-Pro (Korea Nutrition Association). And for the quantity intake, percentage ratio against daily requirement(by KDRIs) and MAR(Mean Adequacy Ratio) were calculated. Underweight group($BMI<18.5kg/m^2$), normal weight group($18.5) and overweight group($BMI<23.0 kg/m^2$) were 18.5%, 61.2% and 19.3% respectively. Nutrient intake levels and food intake status were not significantly different among three groups. The mean nutrient adequacy ratio (MAR) was not significantly different by BMI status. Nutrient intakes of calcium, iron, and riboflavin were especially low for all the students. Overweight group showed high blood pressure(120.9/79.3 mmHg) compared with the underweight group (116.4/ 75.8mmHg) but the difference was not significant. The serum triglyceride level in overweight group (94,0mg%) was higher than that in normal (70.9mg%) or underweight group(70.8mg%)(p<0.05). HDL-cholesterol level in overweight (43.8 mg%) group was not significantly different from that in underweight group (45.9mg%). BMI was positively correlated with blood pressure, hemoglobin, and the intake of total fat and cholesterol. But it was negatively correlated with HDL-cholesterol level. With these results overweight group is closely related with the cardiovascular disease risks, such as high blood pressure and high triglycerides and cholesterol. Thus, a health intervention program including weight control is required for young adults.
