Characterization of Hexane Biodegradation by Rhodococcus sp. EH741 (Rhodococcus sp. EH741에 의한 Hexane 생분해 특성)

  • As a strain EH741, having an excellent hexane degradability, was isolated from bacterial consortium using hexane as a sole carbon and energy source. EH741 was identified as a Rhodococcus sp. and the addition of a surfactant Pluronic F68(PF68), for increasing hexane solubility couldn't enhance the specific growth rate of the isolate EH741 n the mineral salt medium supplemented with hexane as a sole carbon source(hexane-BH medium). In the hexane-BH medium, the maximum specific growth rate(${\mu}_{max}$) of this strain was $0.04h^{-1}$, and the maximum hexane degradation rate($V_{max}$) and saturation constant($K_s$) were$161{\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$ and 10.5 mM, respectively. Rhodococcus sp. EH741 was one of excellent microorgamisms for hexane biodegradation processes.

Properties of a Hexane-Degrading Consortium (Hexane 분해 혼합균의 특성)

  • It was characterized the hexane biodegradation and mineralization using a hexane-degrading consortium, and analyzed its bacterial community structure by 16S rDNA PCR-DGGE (denaturing gradient gel electrophoresis). The specific growth rate (${\mu}_{max}$) of the hexane-degrading consortium was 0.2 $h^{-1}$ in mineral salt medium supplemented with hexane as a sole carbon source. The maximum degradation rate ($V_{max}$) and saturation constant ($K_{s}$) of hexane of the consortium are 460 ${\mu}mol{\cdot}g-DCW^{-1}{\cdot}h^{-1}$ and 25.87 mM, respectively. In addition, this consortium could mineralize $49.1{\%}$ of $^{14}C$-hexane to $^{14}CO_2$, and $43.6{\%}$ of $^{14}C$-hexane) was used for the growth of biomass. The clones isolated from the DGGE bands were closely related to the bacteria which were capable of degrading pollutants such as oil, biphenyl, PCE, and waste gases. The hexane-degrading consortium obtained in this study can be applied for the biological treatment of hexane.

The Measurement and Investigation of Combustible Properties for n-Hexane (노말헥산의 연소특성치의 측정 및 고찰)

  • For the safe handling of n-hexane, the explosion limit at $25^{\circ}C$, the temperature dependence of the explosion limits and the lower flash point were investigated. And AITs(auto-ignition temperatures) by ignition delay time for n-hexane were experimented. By using the literatures data, the lower and upper explosion limits of n-hexane recommended 1.0 Vol% and 8.0 Vol%, respectively. In this study, the lower flash points of n-hexane recommended $-23^{\circ}C$. This study measured relationship between the AITs and the ignition delay times by using ASTM E659-78 apparatus for n-hexane, and the experimental AIT of n-hexane was $240^{\circ}C$. The new equations for predicting the temperature dependence of the explosion limits of n-hexane is proposed. The values calculated by the proposed equations were a good agreement with the literature data.

Combinatorial effects of arginine and n-hexane extract from Korean red ginseng marc against Streptococcus mutans (Streptococcus mutans에 대한 홍삼박 n-hexane 추출물과 Arginine의 병용 효과)

  • In this study, the effect of the combined use of a lipid-soluble n-hexane extract of red ginseng marc (HERGM) and water-soluble arginine, which exhibits anticaries activity, on the growth of Streptococccus mutans was investigated. As a result of checkerboard assay, HERGM and arginine showed a synergistic effect in inhibiting the growth of S. mutans with a fractional inhibitory concentration index of 0.396. Combination treatments of HERGM and arginine resulted in leakage of nucleic acid components and decrease in the viable cell counts of S. mutans, all of which were proportional to the concentration of HERGM. In conclusion, the synergistic effect of HERGM and arginine on the growth inhibition of S. mutans is mainly attributed to HERGM.

Efficiency of Eluting Solvents for Solid-Phase Extraction During Multiresidue Analysis of 16 Pesticides in Cucumber (고상추출카트리지를 이용한 잔류농약 다중분석과정에서 정제용매에 따른 회수율 비교)

  • A study was carried out to optimize the cleanup step using a solid-phase extraction (SPE) cartridge, $SupelClean^{TM}$ Florisil, for multiresidue analysis of 16 pesticides in cucumber matrix. Eluting efficiencies of two solvent systems including acetone/n-hexane and ethyl acetate/n-hexane mixtures were critically evaluated by recoveries of target anaytes from the SPE Florisil column. Based on the recovery as a measure of eluting efficiency, an acetone/n-hexane (20/80,v/v) mixture provided more than 80% recovery for 15 pesticides except bifenthrin. In case of ethyl acetate/n-hexane, 14 pesticides showed recoveries higher than 75% while those far alachlor and bifenthrin were less than 30%.

Anti-microbial and Anticariogenic Activity of Yam and Prunella Extract against Oral Microbes (구강병인균에 대한 마와 꿀풀추출물의 항균.항우식효과)

  • Yam, Prunella was stepwise extracted with hexane, chloroform, ethyl acetate, butanol, and water. Anti-microbial activity of each extract was investigated. Hexane extract was tested for anti-microbial effect on Streptocaccus mutans, one of causative factor of dental caries. Methanol extracts of 7 plants were investigated to anti-microbial effects on S. mutans KCTC 5316, P. gingivalis KCTC 5352, S. aureus KCTC 1927 by means of agar diffusion method. Methanol extract of Yam and Prunella revealed anti-microbial activity against S. mutans, P. gingivalis, and S. aureus. Also, hexane fraction of Yam revealed anti-microbial activity against S. mutans. In sequence of hexane, chloroform, ethylacetate, butanol fraction by Prunelia acted as potent anti-microbial agent on P. gingivalis. The measured MIC of hexane fraction of Yam and Prunella on S. mutans KCTC 5316 strain was 0.25 mg/ml and 0.5 mg/ml and the MIC of hexane fraction of Prunella on S. aureus was 0.5 mg/ml. The hexane fraction of Yam and Prunella suppressed viable ceil counts(VCC) of S. mutans, especially after 24 hrs. The Prunella hexane fraction suppressed VCC of S. aureus, after 12 and 24 hrs. Tested concentrations were 0.1, 0.25 and 0.5 mg/ml. the results were compared with control (0 mg/ml). The pH of S. mutans media and GTase activity were determined to evaluate the anticariogenic activity of Yam, Prunella hexane fraction. The pH were increased from 5.6 to 7.0-7.2 in concentration of 2.0 mg/ml. Yam hexane extraction revealed 35% inhibition to GTase activity and Punella inhibited 25% of GTase. These results suggest that the hexane extracts of Yam and prunella have Antibacterial activities against S. mutans, P. gingivalis, S. aureus and have preventive effect on dental caries.

Effects of Ephedra Herba Hexane Fraction on Lipid Metabolism in Hyperlipidemic Mice (마황(麻黃) Hexane 분획물이 생쥐의 지질 대사에 미치는 영향)

  • ■ Objectives This study was designed to investigate the effects of Ephedra Herba hexane fraction on lipid levels in serum and lipid accumulation in liver tissue in Hyperlipidemic mice. ■ Methods Hyperlipidemia was induced by providing high fat diet for 4 weeks. Normal group was provided with normal diet. CTL groupwas provided with high fat diet. Ephedra Herba hexane fraction group was provieded with high fat diet and administered orally in the concentration of 1.5mg/kg body weight/day for 2 weeks. In this experiment, effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured. In addition histopathological changes in liver tissue were also observed. ■ Results Ephedra Herba hexane fraction did not affects weight gain, serum AST and ALT in hyperlipidemic mice. Oral administration of Ephedra Herba hexane fraction lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. In addition, Ephedra Herba hexane fraction group showed downward tendency of lipid accumulation compared with CTL group. Finally, administration of Ephedra Herba hexane fraction lowered fasting blood glucose significantly. And Ephedra Herba hexane fraction also ameliorates anti-oxidative stress systems in internal organs which play key role in disease prevention. ■ Conclusion These results suggest that Ephedra Herba hexane fraction can prevent lipid accumulation in liver tissue through regulation of dyslipidemia and hyperglycaemia.

A Study of Minimum Autoignition Temperature Behavior (MAITB) of Benzene and n-Hexane Mixture (벤젠과 노말헥산 혼합물의 최소자연발화온도 거동에 관한 연구)

  • The autoignition temperature (AIT) is important index for the safe handling of flammable liquids which constitute the solvent mixtures. This study measured the AITs and ignition delay time for Benzene and n-Hexane system by using ASTM E659 apparatus. The AITs of Benzene and n-Hexane which constituted binary system were $583^{\circ}C$ and $240^{\circ}C$, respectively. The experimental AITs of Benzene and n-Hexane system were a good agreement with the calculated AITs by the proposed equations with a few A.A.D. (average absolute deviation).

  • Adsorption experiments were carried out with several clay minerals and PCP hexane solution in order to clarify the status of adsorbed PCP on the clay surface. The amount of PCP adsorption on clay minerals was much greater in the clay-hexane system than in the clay water system. Among the clay minerals, allophane and imogolite ($SiO_2/Al_2O_3$ ratio of about 1) were the most efficient adsorbents of PCP. The PCP adsorption from hexane solution was greatly hindered by the presence of water, suggesting the occurrence of adsorption by a dipole-dipole interaction. PCP adsorption is dependent upon the nature of the clay surface and the exchangeable cations rather than the total surface area.

The Toxicity and Anti-cancer Activity of the Hexane Layer of Melia azedarach L. var. japonica Makino's Bark Extract

  • In this study, the 4-week oral toxicity and anti-cancer activity of the hexane layer of Melia azedarach L. var. japonica Makino's bark extract were investigated. We carried out a hollow fiber (HF) assay and 28-day repeated toxicity study to confirm the anti-cancer effect and safety of the hexane layer. The HF assay was carried out using an A549 human adenocarcinoma cell via intraperitoneal (IP) site with or without cisplatin. In the result, the 200 mg/kg b.w of hexane layer with 4 mg/kg b.w of cisplatin treated group, showed the highest cytotoxicity aginst A549 carcinoma cells. For the 28-day repeated toxicity study, 6 groups of 10 male and female mice were given by gavage 200, 100, or 50 mg/kg b.w hexane layer with or without 4 mg/kg b.w of cisplatin against body weight, and were then sacrificed for blood and tissue sampling. The subacute oral toxicity study in mice with doses of 200, 100, and 50 mg/kg b.w hexane layer showed no significant changes in body weight gain and general behavior. The cisplatin-treated group significantly decreased in body weight compared to the control group but regained weight with 100 and 200 mg/kg b.w of hexane layer. The biochemical analysis showed significant increase in several parameters (ALT, total billirubin, AST, creatinine, and BUN) in cisplatin-treated groups. However, in the group given a co-treatment of hexane layer (200 mg/kg b.w), levels of these parameters decreased. In hematological analysis, cisplatin induced the reduction of WBCs and neutrophils but co-treatment with hexane layer (100 and 200 mg/kg b.w) improved these toxicities caused by cisplatin. The histological profile of the livers showed eosinophilic cell foci in central vein and portal triad in cisplatin treated mice. These results show that hexane layer might have an anti-cancer activity and could improve the toxicity of cisplatin.
