The Selection of Heterokaryon by the Use of Different Buoyant Density of Protoplasts. (식물세포의 부유밀도를 이용한 융합원형질체의 선발)

  • This experiment was carried out to select of heterokaryon based on the different buoyant densities of protoplasts. Protoplats were isolated from cultured cells (calli) of Nicotiana tobacum(cv. BY4) and from mesophyll cells of N. glauca. The two types of protoplats were fractionated by centrifugation in an iso-osmotic (770 mOs/kg. H2O) density gradients condition. Major difference in the buoyant density exists between two types of protoplasts isolated from different cells. The mesophyll protoplasts were fractionated in the higher gradient interphases than that of callus protoplasts. The two types of fractionated protoplasts were fused with 40% polyethylene glycol (PEG), and the protoplasts treated with PEG were separated by centrifugation in the same density gradients condition. The heterokaryons were fractionated in the intermediate density gradients.

Transcription level of the ars-1 promoter of Neurospora crassa (Neurospora crassa ars-1 프로모터의 발현율 조사)

  • The ars gene of the Neurospora crassa encodes arylsulfatase and is expressed under sulfur limitation. An ars-1 promoter(Pars) translationally-fused to a lacZ gene was transformed into the N. crassa RLM 35-35, a his-3 inl strain and integrated into the his-S locus by a single crossover homologous recombination. $\beta$-galactosidase specific activity was measured from mycelia grown in sulfur-limited Vogel's medium. Enzyme activity reached its maximum at 14 hour after the shift to derepressing condition. When activity from homokaryon generated by microconidiation was measured, it was 17% a higher than that from heterokaryon.

  • Interspecific fusion products were obtained by protoplast fusion induced by polyethylene glycol, from auxotrophic mutants, Pleurotus cornucopiae and P. florida. The fusants were classified into allodiploidy, stable heterokaryon, and spontaneously segregated heterokaryons. Fruiting body of the clamped or clampless fusants was produced by light-dark cycle on the sawdust medium in glass bottles. Most of these clampless fusants produced mature basidiocarps. The pilei showed various mixed colors resembling the parents. All fruit bodies presented clamp connections except two fusants. When small tissues of stipe from basidiocarps were cultured on a complete medium, mycelial colonies grew more vigorously than that of the original clampless fusants. Five fusants in three crosses were analysed with the distribution of progenies and segregation of genetic characters by random spore analyses. The genetic markers were shown to segregate and recombine in the first generation of monospores isolated from basidiocarps. The analysis indicated the heter-okar-yosis and strong evidence for haploidy of vegetative nuclei, a sexual cycle consisting of nuclear fusion and meiosis. Genotypes of a large number of auxotiophic progenies were not detected. The aberration ratio of segregants indicated the gene interaction resulting from different genome structure between vegetatively incompatible species.

Cultural and Morphological Characteristics of a New White Button Mushroom Cultivar 'Saedo'

  • Since development of the first Korean button mushroom cultivar 'Sae-Ah' in 2010, mushroom farmers have demanded an improved cultivar. Although a new cultivar, 'Saejeong', was developed in 2011, this cultivar has thus far been difficult to cultivate in typical mushroom farms. To solve this problem, another new cultivar, 'Saedo', was developed in 2012. This new cultivar was selected by genetic analysis from crossing the heterokaryon line A175 and the homokaryon line ASI1346-15. The heterokaryon A175 was a crossed line between strains ASI1038-211 and ASI1346-20. The mycelium of 'Saedo' showed good growth on compost dextrose agar at $25^{\circ}C$. The morphological traits such as the pileus (cap) and stipe of the 'Saedo' cultivar were thicker than those of the reference control 'Saejeong', and the production yield was increased by 1.89-fold compared to that of the reference control. A field experiment of the 'Saedo' cultivar has been ongoing since 2013. The yield of the 'Saedo' cultivar was found to be substantially higher than that of the reference control during the $2^{nd}$ flush.

Characteristics of Fusion Products between Pleurotus ostreatus and Pleurotus florida Following Interspecific Protoplast Fusion (느타리버섯과 사철느타리버섯의 종간 원형질체융합 균주의 특성)

  • Interspecific heterokaryons were obtained between auxotrophic mutants of Pleurotus ostreatus and Pleurotus florida by polyethylene $glycol-Ca^{++}$ induced fusion of somatic protoplasts. The fusion products produced colonies of dense growing mycelium after 10-14 days culture on hypertonic Mushroom Minimal Medium. When they were transferred to Mushroom Minimal Medium plates, the sectors showed normal vegetative morphology but the colonies of irregular shape varied in growth rate. All of the colonies produced fruit body of normal pilei. Some heterokaryon colonies bearing none or only a small amounts of basidiospores were isolated. Fusion products, generally, gave higher basidiocarp yields than the parents.

Vegetable Compatibility Grouping of Fusarium oxysporum f. sp. lycopersici Isolated from Korea (국내에서 분리한 토마토 시들음병균(Fusarium oxysporum f. sp. lycopersici)의 체세포 화합성군)

  • Forty-six isolates of Fusarium oxysporum collected from infected tomato plants and soils in greenhouses in Sedo, Chungnam and Angang, Kyeongbuk and 8 isolates of F. oxysporum f. sp. lycopersici from Japan and USA were used to determine vegetative compatibility groups (VCGs). Vegetative comaptibility was assessed on the basis of heterokaryon formation among nitrate nonutilizing mutants. All Korean isolates of F. oxysporum f. sp. lycopersici used in this study belonged to the same type of VCG (003) regardless of their geographic origin, cultivar and race, but they were incompatible with the foreign isolates of VCG 0030, 0031, 0032 and 0033. Based on the results, the Korean isolates of F. oxysporum f. sp. lycopersici were classified as a new VCG 003.

  • Prion is a group of the proteins known for its infection mechanisms of Creutzfeldt-Jakob disease (CJD) and other diseases. Solved structures and proven biological roles of fungal prions add tremendous potential to conducting computational simulations. Our research focuses on the binding dynamics of HET-s(218-289), one of the heterokaryon fungal prion originated from Podospora anserina, by calculating the binding free energy using umbrella sampling at 300 K. The binding free energy calculated was $-54.5kcal\;mol^{-1}$, relatively similar to the binding energy of other amyloid fibrils. The simulation result suggests the thermodynamic properties of ${\beta}$-solenoid of HET-s prion and its similarity in dissociation pathways compared to amyloids.

Relationship Between Virulence and Vegetative Compatibility Group of Glomerella cingulata Isolates (Glomerella cingulata의 병원성과 체세포화합성군(Vegetative Compatibility Group) 과의 관계)

  • 국내의 고추, 사과, 포도, 스타티스(statice)에서 분리한 Glomerella cingulata 균주들의 병원성과 체세포화 합성군(vegetative compatibility group : VCG)과의 관계를 조사하였다. 사과, 포도, 스타티스에서 분리된 균주들은 전기주에 모드 병원성을 나타내었으나 고추에는 병원성이 없었다. 한편, 고추에서 분리된 균주들은 무상처접종시 병원성을 나타내지 못하였다. Nitrate nonutilizing(nit) mutant는 1.5%의 KClO\ulcorner을 첨가한 최소배지에서 선발하였다. nit mutnat들은 nit1, nit2, nit3, nitM의 4가지 표현형 중 하나에 속하였다. VCG는 표현형이 다른 nit mutan간의 대치배양을 통해 결정하였는데 사과, 포도, 스타티스에서 분리한 균주들간에는 같은 VCG를 나타낸 반면 고추에서 분리된 균주들은 다른 VCG로 나타났다. 따라서 G. cingulata 균주들의 체세포화합성은 병원성과 밀접하게 관련되어 있음을 알수 있었다. 또한 동일한 VCG에 속하며 기주가 다른 균주들의 대치배양시 heterokaryon을 형성했으나 자낭을 형성하지는 않았다.

Intergeneric Transfer of Isolated Nuclei from Lentinus edodes into Protoplasts of Pleurotus florida (사철느타리버섯 원형질체내에 표고 핵의 전이)

  • Transfer of the isolated nuclei from Lentinus edodes into protoplasts of Pleurotus florida was induced with polyethlene glycol (PEG) and $CaCl_2$. The intergeneric transfer products were classified into nuclear hybrid, heterokaryon or synkaryon, and reconstituted cell. These progenies except nuclear hybrids formed mature fruiting bodies on sawdust rice bran medium. Formation of fruit bodies was influenced by several factors such as light, temperature, nutrition and physic state of the culture media. Most of fruiting body characters were similar to those of P. florida in synkaryon and L. edodes in reconstituted cell, respectively. All these basidiocarps had clamp connections though initial heterokaryon colonies were lacking. Isozyme patterns of intergeneric progenies were quite different from those of parents. DNA polymorphisms of transfer products were also compared by random amplified polymorphic DNAs (RAPD) analysis based on polymerase chain reaction. The RAPD patterns were different from those of donor and recipient. DNA fingerprints ranged in size from 0.25 to 4.0 Kb. On the basis of RAPD, the transfer products were classified into five groups. Two synkaryon were analysed with distribution of progenies and segregation of genetic markers by random spore analyses. The genetic markers were segregated into wild type and riboflavine requiring auxotrophs.

Vegetative Compatibility Groups in Fusarium graminearum Isolates from Corn and Barley in Korea

  • Fifty-three isolates of Fusarium graminearum were obtained from corn and barley samples in several provinces of Korea. Gas chromatography-mass spectrometric analysis of trichothecenes produced by these isolates revealed that 37 and 16 isolates were nivalenol (NIV)- and deoxynivalenol (DON)-chemotypes, respectively. Two hundred and seventy-five nitrate-nonutilizing (nit) mutants were obtained from the isolates. Of these mutants, 187 were identified as nit1, nit3, or NitM, but 88 could not be identified as one of these classes. The highest frequency of nit mutant was nit1 (65%), followed by nit3 (20%) and NitM (15%). Higher frequency of NitM was observed in DON-chemotypes than in NIV-chemotypes. The mutants were used for vegetative compatibility group (VCG) analysis by examining heterokaryosis using complementary mutant pairs. No heterokaryon formation was observed among all 1,248 pairwise combinations, suggesting that all isolates tested belong to different VCGs. Higher frequency of self-incompatibility was observed in NIV-chemotypes than in DON-chemotypes. These results suggest that the like-lihood of asexual genetic recombination may be very low I F. graminearum under the field condition.
