식품의 조리.가공중 생성되는 발암성 이환방향족아민의 안전성 (The Safety of Carcinogenic Heterocyclic Aromatic Amines from the Cooked Foods)

  • 일상적으로 먹는 생선류, 육류 가공품들은 전형적인 방법으로 가열·조리하면 변이원성(mutagenic activity)이 나타난다. 식품의 제조 방법들은 변이원성의 생성에 뚜렷한 영향을 미치는데, 조리된 육류 제품들에서 발견되는 주요 식품 변이원들은 이환방향족아민(heterocyclic amines)들로 알려져 있다. 이중 몇 가지는 장기간의 동물실험 결과, 설치동물에서 발암성이 관찰되었으며, 사람에 있어서도 여러 암의 발병과 관련성이 높다는 증거들이 제시되고 있다. 모델 실험에서 몇가지 열(thermic) 변이원을 동정한 결과 creatine, creatinine, 아미노산 및 당 등 식품에 존재하는 성분들이 열 변이원 생성의 전구체인 것으로 나타났다. 건강상 관점에서 보면 이를 감소시키거나 식품 변이원의 생성을 막는 것이 바람직하므로 일반 가정에서 조리하는 동안 변이원이 형성되는 반응조건 및 전구체들에 대한 깊은 이해가 필요하다고 하겠다. 육류, 생선을 포함한 고단백 식품을 생산하는 식품업체에서도 가공설비의 최적조건 설정시 품질적인 측면뿐만 아니라 이환방향족아민의 안전성이 같이 고려되어야 할 것이다. 그러나 우리나라에서는 이환방향족아민에 대한 인식이 부족하고, 노출량 및 위해도 평가작업이 거의 이루어지고 있지 않아 국민보건향상 측면에서 이에 대한 연구가 시급하다고 본다.

  • Heterocyclic aromatic amines (HCAs) are established genotoxic human cancer risks, that display their activity also in a number of animal models and in vitro. They are formed during the trying or broiling of creatinine-containing foods, mainly fish or meat. We established that mixing soy protein with ground meat blocks the formation of HCAs. we also observed that coating the surface of meat with polyphenols green or from black tea gives a dose - related reduction of the formation of HCAs.(omitted)

Influence of Food Ingredients on the Formation of Heterocyclic Aromatic Amine in Cooked Pork Patties

  • The effects of cooking method, cooking time and various food ingredients on the formation/ inhibition of heterocyclic aromatic amines (HAAs) in pork products were investigated. Three HAAs, 2-amino-3,8-dimethylimidazo [4,5-f] quinoxaline ($MeIQ_x$), 2-amino-3,4,8-trimethylimidazo [4,5-f] quinoxaline ($DiMeIQ_x$) and 2-amino-1-methyl-6-phenylimidazo [4,5-b] pyridine (PhIP) were measured in pork products using solid-phase extraction and HPLC. Pork patties were boiled, oven-broiled and pan-fried to internal temperatures of 71, 77 and $88^{\circ}C$. Generally, HAA concentrations increased with increasing internal temperature, and HAA formation was greatest with pan-fried. Selected food ingredients (vitamin E, sodium nitrite, sodium tripolyphosphate, sodium ascorbate, Nanking cherry tissue and cherry tissue extract) inhibited HAA formation in pork patties fried at $225^{\circ}C$ for 10 min/side, with the greater inhibition provided by cherry tissue and its methanolic extract.

  • 2-Thiouracil-5-sulfonylchloride 1 reacted with a series of aromatic and heterocyclic amines to give 2a-j. The same compound 1 was reacted with a series of sulphonamides giving different sulphonamides of type 3a-e. On the other hand compound 1 was allowed to react with p-aminoacetophenone givining compound 4 which in turn was allowed to react with derivatives of alkyl thiosemicarbazides to give thiosemicarbazones of type 5a-e, also compound 4 was monobrominated to give compound 6 which in turn was reacted thiosemicarbazones of some aldehydes to give the corresponding thiazole derivatives 7a-f. In the same time compound 4 was reacted with a series of aromatic and heterocyclic aldehydes givining chalcones 8a-g (Claisen-Schemidt reaction). Also compound 4 was allowed to react with a series of aromatic and heterocyclic aldehydes, ethyl cyano acetate and/or malononitrile, and ammonium acetate giving pyridine derivatives 9a-d and 10a-e respectively. The biological effects of some of the new synthesized compounds was also investigated.

  • The objective of this study was to investigate the contribution of various smoke constituents to the toxicological activity of total particulate matter(TPM) or the gas/vapor phase(GVP). These components included phenol compounds, aromatic amines, polycyclic aromatic hydrocarbons, heterocyclic amines, and carbonyl compounds. The mutagenic and cytotoxic potencies were assessed using the Salmonella mutagenicity assay with S. typimurium TA98 strain and the neutral red uptake cytotoxicity assay(NRU) with BALB/c 3T3 fibroblast cells, respectively. The Salmonella mutagenicity test showed that heterocyclic amines exhibited significantly higher levels of toxicity compared to other smoke constituents. Among them, 2-amino-3,4-dimethylimidazo[4,5-f]quinoline(MeIQ) was shown the most mutagenic compound with a specific mutagenicity of $7.9{\times}10^5\;revertants/{\mu}g$. An analysis of the possible contribution revealed that MeIQ account for only 0.85% of the 2R4F-TPM mutagenicity in TA98. NRU data demonstrated that high cytotoxic activity was obtained for hydroquinone, formaldehyde, and acrolein. Based on the results of the present study, the contribution of acrolein to the cytotoxicity of the GVP fraction was calculated as 61%. Thus, a large proportion of the cytotoxic activity of this complex mixture, cigarette smoke gas phase, can be attributed to the acrolein.

A Novel Synthesis of Heterocyclic Compounds Containing Coumarin Moiety of Potential Antimicrobial Activity

  • The chemical behaviours of 4-methyl-2-oxo-2H-benzopyran-7-yl oxoacetyl hydrazine (2) towards some different reagents such as anhydride compounds, aromatic aldehydes, carb on disulphide, and nitrous acid yielded the corresponding pathalazine derivatives (3, 4, 5), hydrazone derivative (6), 1,3,4-oxadiazole derivative (7, 8, 9) and acid azide (10) respectively. Treatmen of 10 with absolute alcohols, amines and ethyl amino acid ester gave the corresponding carbamate derivative (11), substituted urea derivative (12) and ethyl substituted alkyl acetate (13) respectively. The biological activity of some synthesized compounds was evaluated.

  • Ethanolic tetra carbonylhydridoferrate solution combined with dialdehyde (no of carbon; 4,5,6) is very efficient for the selective transformation of amino group into N-heterocyclic compound. However, a large variety of both aliphatic and aromatic amines react with the ferrate-pimelaldehyde at room temperature under an atmospheric pressure of carbon monoxide to give the corresponding N-(cyclohexylmethyl)-N-alkyiamine derivatives in moderate yields instead of the corresponding N-substituted perhydroazocine derivatives.

Cancer Activation and Polymorphisms of Human Cytochrome P450 1B1

  • Human cytochrome P450 enzymes (P450s, CYPs) are major oxidative catalysts that metabolize various xenobiotic and endogenous compounds. Many carcinogens induce cancer only after metabolic activation and P450 enzymes play an important role in this phenomenon. P450 1B1 mediates bioactivation of many procarcinogenic chemicals and carcinogenic estrogen. It catalyzes the oxidation reaction of polycyclic aromatic carbons, heterocyclic and aromatic amines, and the 4-hydroxylation reaction of $17{\beta}$-estradiol. Enhanced expression of P450 1B1 promotes cancer cell proliferation and metastasis. There are at least 25 polymorphic variants of P450 1B1 and some of these have been reported to be associated with eye diseases. In addition, P450 1B1 polymorphisms can greatly affect the metabolic activation of many procarcinogenic compounds. It is necessary to understand the relationship between metabolic activation of such substances and P450 1B1 polymorphisms in order to develop rational strategies for the prevention of its toxic effect on human health.
