The tetracycline-controlled transactivator (tTA)-mediated gene activation system was examined in virus infected cells to determine its role in the control of gene expression. In the presence of tTA, the gene expression from the tetO-modified minimal promoter was efficiently activated in the uninfected cells, whereas essentially no activation was observed from the only minimal promoter without the seven direct repeats of 42 bp tetO sequences. However, essentially no activation was observed when only the minimal promoter was used, without the seven direct repetitions of the 42 bp tetO sequences. On the other hand, in the infected cells, a substantial background of $\beta$-glucuronidase expression was detected in the absence of tTA, even though tTA stimulated the gene expression by ~7-fold. This background expression indicates that the sequences within or nearby tetO are involved in the background stimulation of the gene expression by HCMV and HSV-1 . These results suggest that the application of the tTA-mediated gene activation system may not be extremely useful for studying the biological roles of HCMV and HSV genes In the viral replicative cycles, because of the basal activity of the gene expression.
Syntheses of new analogues of oxathiin carboxanilide (UC84) and their antiviral activities were described. The heterocyclic carboxylic acids including oxathiins (4), thiazines (9) and dithiins (13) in which the methyl was replaced either by lipophilic trifluoromethyl- or bulky phenylgroup were synthesized starting from $\beta$-keto esters (5). Reaction of 4, 9 and 13 with thionyl chloride followed by treatment of the substituted aniline 22 gave the corresponding carboxanilides (24a~24f). The carboxanilides were subjected to Laweson's reagent the corresponding thiocarboxanilides (24g~24k). The antiviral activities of the synthesized compounds against human immunodeficiency virus type 1 (HIV-1), poliovirus type 1 (PV-1 ), coxsackie B virus type 3 (CoxB-3), vesicular stomatitis virus (VSV) and herpes simplex virus type 1 (HSV-1) were presented. The antiviral activity against HIV-1 of dithiin carboxanilide (24e) was similar with that of UC84 (24a). The corresponding thiocarboxanilides (24g~24k) showed higher inhibitory activity against HIV-1 than the carboxanilides (24a, 24b, 24d, 24e). The compounds in which ether the lipophilic trifluorormethyl substituents (24d, 24f, 24i ,24k) or bulky phenyl substituent is present in the heterocyclic compounds showed lower inhibitory activity than that of the methyl substituents is present in the compounds against the HIV-1. But the trifluoromethylated dithiin (24f) showed higher inhibitory activity against PV-1 and CoxB-3 virus than commercial antiviral agents, ribavirin (RV).
연구배경 : HSVtk 유전자를 암세포에 이입하여 GCV에 대해 선택적으로 감수성을 증가시키는 HSVtk/GCV 유전자치료에서 bystander effect는 모든 암세포에 유전자를 이입하지 않고도 치료효과를 얻을 수 있도록 한다. 그러나 현존하는 viral vector는 유전자이입 효율이 낮아 임상적으로 치료효과를 기대하는데는 한계가 있다. 따라서 유전자의 이입효율이 높은 새로운 vector의 개발과 함께 bystander effect의 극대화를 통해 치료효과를 증가시킬 수 있는 방법 등이 요구된다. 최근 gap junction을 통한 세포간의 metablic cooperation이 bystander effect의 주요기전임이 밝혀졌고 retinoids는 gap junction을 통한 세포간의 communication을 증가시킨다고 보고되었다. 저자들은 HSVtk/GCV 유전자치료에서 bystander effect에 미치는 retinoids의 효과를 조사하였다. 방 법 : Adenovus와 retrovirus vector를 이용하여 connexin 43를 발현하는 악성중피종세포와 connexin 43를 발현하지 않는 SKHep-J 세포주에 HSVtk 유전자를 이입한 후 HSVtk 유전자가 이입된 세포와 HSVtk 유전자가 이입되지 않은 세포들을 여러가지 비율로 혼합한 mixing study를 시행하였으며 $10^{-10}M-10^{-6}M$ RA 처리 유무에 따른 bystander effect에 의한 살상효과를 비교하였다. 그리고 gap junction을 통한 세포간의 communication에 대한 retinoids의 영향을 조사하기 위해 retinoids 처리에 따른 세포간 communication을 FACS를 이용하여 double-dye transfer study로 측정하였다. 결 과 : Connexin 43를 발현하지 않는 SKHep-J 세포주에서는 RA 처리유무에 따른 bystander effect에 의한 살상효과의 차이가 없었다. 그러나 connexin 43를 발현하는 악성중피종 세포주에서는 $10^{-8}M-10^{-6}M$ RA처리시 세포간의 communication과 bystander effect가 RA를 처리하지 않은 대조군에 비해 유의하게 증가되었다. 결 론 : RA는 gap junction을 통한 세포간의 communication을 증가시켜 HSVtk/GCV 유전자치료에서 bystander effect에 의한 살상효과를 증가시켰다. 이러한 결과는 HSVtk/GCV 유전자치료의 효과를 증가시킬 수 있는 새로운 방법이 될 수 있을 것으로 생각된다.
연구배경 : 암 유전자치료에서 각광받고 있는 HSV-tk/GCV 전략의 항암효과에는 다음과 같은 장점들이 거론되고 있다 : 1) GCV 처리에 의한 암세포 직접살상효과 2) HSV-tk 이입된 세포에 의해서 HSV-tk 이입되지 않은 주변세포를 살상하는 bystander effect 3) 생체 내 bystander eff ect로 알려 진 anti-tumor immunity. Retrovirus와 adenovirus sequence를 이용할 경우 몇몇 세포주와 마우스에서 이들이 목적유전자의 발현을 억제할 수 있다는 것이 보고되고 있다. 본 연구에서는 retroviral나 adenoviral vector로 HSV-tk 유전자를 이입한 Lewis 폐암세포주와 폐암 마우스 모델을 통하여 HSV-tk/GCV 전략의 장점을 조사하였고 이 viral vector들 사이의 차이를 비교 조사하였다. 또한 Lewis 폐암세포주에서 butyrate를 처리한 후 HSV-tk 유전자의 발현증가를 관찰하였다. 방법 : Lewis retroviral vector와 adenoviral vector로 HSV-tk 유전자를 이입한 후 butyrate로 HSV-tk 유전자의 발현을 유도하고 Western blotting수행하여 분석하였다. 생체 외에서 HSV-tk/GCV에 의한 세포살상효과를 MTT 검사로 수행하였고 생체 내에서 LLC 나 HSV-tk 이입된 LLC 세포주를 이식하여 종양소멸 및 bystander effect를 조사 하였다. 결과 : 1. Butyrate로 HSV-tk adenovirus로 이입된 LLC에서 증가한 반면 retrovirus로 이입된 LLC에서는 증가하지 않았다. 2. 생체 외 그리고 생체 내에서 viral vector로 HSV-tk를 이입한 종양세포에 GCV 투여하는 것은 종양 세포의 살상에 효과적이었으며 LLC와 LLC-tk 세포주를 혼합한 실험에서 bystander effect도 종양세포의 성장을 억제하는 것으로 관찰되었다. 결론 : 향후 생체 외 그리고 생체 내 실험에서 adenoviral vector를 이용한 유전자 전달에 butyrate를 함께 사용하면 유전자발현을 증진시킬 것으로 사료되며 자살 유전자인 HSV-tk을 종양에 이입하여 GCV을 처리 하는 치료가 폐암유전자치료에 효과가 있을 것으로 생각된다.
목적: Hydrodynamic-based procedure는 손쉽고 간편한 비바이러스성 유전자 전달 방법으로 특히 간특이적으로 발현하는 특징을 가진다. 단순 헤르페스 바이러스 제 1 형 티미딘 키나제(herpes simplex virus type 1 thymidine kinase, HSV1-tk)와 다양한 기질을 이용한 비침습적 HSV1-tk 유전자 영상시스템이 널리 연구되어왔다. 본 연구에서는 HSV1-tk 유전자를 hydrodynamic-based procedure를 이용하여 전달한 후, HSV1-tk의 보고 기질로 알려진 5-(2-iodovinyl)-2'-deoxyuridine (IVDU)을 이용하여 간 특이적인 HSV1-tk 유전자 발현 영상을 획득하고자 하였다. 대상 및 방법: HSV1-tk 유전자와 녹색형광유전자를 가진 각 플라스미드 벡터를 마우스에 hyodynaminc injection을 통해 전달하고, 24 시간 뒤 유전자의 발현을 확인하기 위해 RT-PCR, 생체형광영상, 핵의학영상, 전신자가방사영상 그리고 생체분포를 시행하였다. 결과: 각 플라스미드 벡터를 전달한 간으로부터 추출한 전체 RNA를 이용하여 RT-PCR을 수행한 결과, 각각 HSV1-tk유전자와 녹색형광단백 유전자의 특이적인 밴드를 관찰할 수 있었다. 생체 분포 결과, pHSV1-tk 벡터를 전달한 마우스의 간에서 특이적인 [$^{123}I$]IVDU의 섭취를 보였다. 생체형광영상에서는pEGFP-N1 벡터를 전달한 마우스의 간에서는 유의한 형광신호를 나타내었다. 전신자가방사영상과 감마카메라 영상에서 pHSV1-tk 벡터를 전달한 마우스의 간에서 방사표지 IVDU가 국소적으로 집적되는 것을 확인하였다. 결론: 본 연구에서 hydrodynamic-based procedure는 간특이적으로 플라스미드 DNA를 전달하는데 효과적이며 전달된 유전자의 발현을 분자영상학적인 방법으로 확인하였다. 따라서 Hydrodynamic injection을 통해 HSV1-tk유전자와 목적 유전자의 공동발현은 방사표지 IVDU에 의해 목적 유전자의 발현을 정량평가하는데 유용할 것으로 기대된다.
Ganciclovir(GCV)와 Vidarabine(ara-A)을 단독으로 또는 동시에 HSV-1에 작용시켰을때 HSV-1의 복제, DNA합성 및 단백질 합성에 미치는 영향을 관찰할 목적으로 본 연구를 시행하였다. 본 실험에서는 4가지의 다른 HSV-1(Wild type KOS, $VCV^r$, $IUdR^r$, 및 $PAA^r5$)를 사용하였다. Virus복제에 미치는 항 virus약의 효과는 Vero 세포단층 배양에서 Yield reduction assay에 의해서 관찰하였다. 항 virus약의 virus DNA 합성에 미치는 영향은 NaI 밀도 구배초원심침전후 $H^3-$표지 virus DNA의 방사능에 의해서 관찰하였다. Virus 단백질의 합성에 미치는 항 virus약의 효과는 $^{35}S$를 표지한 후 polyacrylamide 겔 전기영동법, 자가방사기록법 그러고 virus bands의 음영농도주사법을 이용, 관찰하여 다음과 같은 결과를 얻었다. 1. GCV는 wild trype HSV-1 KOS와 $PAA^r5$의 복제를 강력하게 억제하였으나 $ACV^r$와 $IUdR^r$는 GCV에 대해서 중등도의 저항을 보였다. ara-A는 실험대상의 모든 HSV-(KOS, $ACV^r$, $IUdR^r$ 및 $PAA^r5$)의 복제에 대해서 거의 비슷하게 억제효과를 보였다. GCV와 ara-A 동시첨가는 KOS와 $PAA^r5$의 복제에 대해서 상승적인 억제효과를 보였고 $ACV^r$와 $IUdR^r5$의 복제에 대해서는 상가작용이하의 억제 효과를 보였다. 2. GCV또는 ara-A는 HSV-1감염 Vero세포에서 virus DNA의 합성을 유의하게 억제하였다. GCV와 ara-A의 동시첨가는 KOS 또는 $PAA^r5$ 감염세포에서 virus DNA 합성을 GCV 또는 ara-A를 단독 첨가하였을때 보다 현저하게 억제하였다. $ACV^r$ 또는 $IUdR^r$ 감염세포에서는 이런 현상을 관찰할 수 없었다. 3. Wild type HSV-1 감염세포에서 virus-단백질의 합성은 GCV, ara-A의 단독 첨가 또는 GCV와 ara-A의 동시첨가에 의해서 변경되지 않았다. Wild type HSV-1 감염말기에 GCV 또는 ara-A 단독 혹은 동시첨가에 의해서 virus 단백질의 합성은 경미하나마 유의성있게 증가하였다. Wild type HSV-1과 $PAA^r5$에 의한 단백질의 합성은 GCV 또는 ara-A 단독 첨가에 의해서 유의성있게 억제되었다. GCV또는 ara-A의 동시첨가는 GCV또는 ara-A를 단독으로 첨가했을 경우보다 단백질의 합성을 더욱 억제하였다. 이상의 실험결과로 보아 GCV와 ara-A의 동시사용은 HSV-1 혹은 ACV저항 DNA polymerase변이주인 $PAA^r5$에 대해서 상승적인 억제작용을 나타냈으며 이 효과는 virus DNA 합성 억제에 의한 것으로 생각된다. ACV저항 thymidine kinase 변이주인 $ACV^r$ 및 $IUdR^r$에 대해서는 ara-A가 유효하였다. 항 virus 약물에 의한 virus 단백질합성의 변화는 virus DNA 합성에 대한 억제효과에 인한 것으로 사려된다.

Mice immunized with equine herpesvirus type-1(EHV-1) L-particles skewed a significant increase (p<7.75) in serum antibody titers. Upon a booster dose four weeks lateral antibody titers increased significantly. Interestingly, immunization via intravenous or intramuscular route induced significantly higher (p<0.75) antibody titers. However, mice iummunized with UV-treated L-particles, visions or immunization via intranasal route induced lower antibody titers. Upon challenge inoculation with wildtype EHV-1, our data showed there was a poor correlation between antibody titers and protection against virus replication. Therefore, the role of cell-mediated immunity Inwards protection was investigated. As predicted, the strongest cell-mediated immunity, as measured by delayed-hypersensitivity test, was detected in mice immunized with live virus particles. The magnitude of cell-mediated immune response correlated with the efficacy of L-particles as immunizing agent. The highest efficacy, as indicated in mice immunized via intranasal routed was highly correlated with cell-mediated immunity. A similar phenomenon was also demonstrated in mice immunized intranasally with UV-treated L-particles. However, the degree of protection was reduced when mice immunized intravenously or intramuscularly with UV-treated L-particles. In conclusion, protection conferred in these animals was highly implicated by immune cells and the least by antibodies. The route of immunization and the nature of the antigen also contributed to the efficacy of L-particles as immunizing agent. In contrast to that of herpes simplex virus type 1, our data showed EHV-1 non-infectious L-particles are highly suitable for immunization of the host against EHV-1 disease.
The Herpes simplex virus type 1 (HSV-1) strain F glycoprotein H (gH) gene in the pHLB-4 plasmid was recombinated into a baculovirus expression vector (lacZ-HcNPV) to construct a recombinant virus GH-HcNPV expressing gH. The sequences of gH and its expression were analyzed. The gH gene was located in the 6.41 kb BglII fragment. The open reading frame (ORF) of the gH gene was 2,517 bp and codes 838 amino acid residues. Insect cells infected with this recombinant virus synthesized a high level of the matured and gX-gH fusion protein with approximately 112 kDa. The fusion gH protein was localized on the membrane of the insect cells as seen by using immunofluorescence assay and accumulated in the cultured media by the SDS-PAGE and immunoprecipitation assays. The amino acid sequence presents additional characteristics compatible with the structure of a viral glycoprotein: signal peptide, putative glycosylation sites and a long C-terminal transmembrane sequence. Antibodies raised in mice to this recombinant protein recognized viral gH and neutralized the infectivity of HSV-1 in vitro. These results demonstrate that it is possible to produce a mature protein by gene transfer in eukaryotic cells, and indicate the utility of the HcNPV-insect cell system for producing and characterizing eukaryotic proteins. Furthermore, the neutralizing antibodies would appear to protect mice against HSV; accordingly, this particular recombinant protein may be useful in the development of a subunit vaccine.

The murine leukemia virus-based semi-retroviral replicating vectors (MuLV-based sRRV) had been developed to improve safety and transgene capacity for cancer gene therapy. However, despite the apparent advantages of the sRRV, improvements in the in vivo transduction efficiency are still required to deliver therapeutic genes efficiently for clinical use. In this study, we established a gibbon ape leukemia virus (GaLV) envelope-pseudotyped semi-replication-competent retrovirus vector system (spRRV) which is composed of two transcomplementing replication-defective retroviral vectors termed MuLV-Gag-Pol and GaLV-Env. We found that the spRRV shows considerable improvement in efficiencies of gene transfer and spreading in both human glioblastoma cells and pre-established human glioblastoma mouse model compared with an sRRV system. When treated with ganciclovir after intratumoral injection of each vector system into pre-established U-87 MG glioblastomas, the group of mice injected with spRRV expressing the herpes simplex virus type 1-thymidine kinase (HSV1-tk) gene showed a survival rate of 100% for more than 150 days, but all control groups of mice (HSV1-tk/PBS-treated and GFP/GCV-treated groups) died within 45 days after tumor injection. In conclusion, these findings sug-gest that intratumoral delivery of the HSV1-tk gene by the spRRV system is worthy of development in clinical trials for the treatment of malignant solid tumors.
Purpose: The aims of this study was to compare and evaluate the clinical characteristics, laboratory data, and prognosis for infants under age 1 year with CMV hepatitis and those with viral hepatitis of unknown etiology. Methods: A retrospective study was conducted of infants under age 1 year who were admitted with acute hepatitis. The exclusion criteria consisted of: autoimmune, genetic, metabolic, toxic, HAV, HBV, HCV, toxoplasma, rubella, herpes simplex, and Epstein-Barr virus. The 30 patients included were divided into two groups based on markers for CMV (IgM anti-CMV, CMV PCR in urine, CMV culture in urine). Results: The median age of patients (n=15) was 2.8 months. No other organ involvement was detected in any patient. Peak serum total bilirubin levels (n=4) ranged from 2.6 to 6.7 mg/dL. Peak serum ALT levels ranged from 51 to 1,581 IU/L. The duration of ALT elevation ranged from 1.5 weeks to 26 weeks (median 9 weeks). All had recovered in full without ganciclovir; there were no cases of hearing loss. The median age of controls (n=15) was 2.5 months. Peak serum total bilirubin levels (n=4) ranged from 1.6 to 9.1 mg/dL. Peak serum ALT levels ranged from 26 to 1,794 IU/L. No significant differences were observed between both groups regarding the peak serum ALT levels, peak serum total bilirubin levels, duration of hyperbilirubinemia and ALT elevation. Conclusion: Although it was not possible to differentiate congenital infection with perinatal infection in this study, the prognosis of patients with CMV hepatitis without other organ involvement was good without ganciclovir treatment.

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
