This research was conducted to evaluate the safety of the herbicide-resistant rice, a genetically modified organism (GMO) developed by the Rural Development Administration by exposing it to 12 months old Sprague-Dawley rats for 8 weeks. The composition of herbicide-resistant brown rice with/without heating treatment was compared with those of conventional Ilpum brown rice with/without heating treatment to assess composition equivalence. Compositional analysis was performed to measure proximates, fiber, and minerals. The nutritional components of herbicide-resistant rice were similar to those of the nontransgenic control or were within the normal range of nontransgenic rice. Four groups of experimental male rats were fed one of the following diets for eight weeks: Ilpum brown rice (I) and its heated rice (IH) as non-GMO, and herbicide-resistant brown rice (G) and its heated rice (GH) as GMO- We checked clinical symptoms (anorexia, salivation, diarrhea, polyuria, anuria, fecal change), food intake, and water consumption every day, change of body weight once a week, and serum biochemistry and organ weights after 8 weeks of experimental feeding We did not find any significant differences in the above-mentioned items. These results suggested that genetically modified herbicide-resistant rice was compositionally equivalent to conventional Ilpum rice, and nutritional characteristics and safety of herbicide-resistant rice in old male rats treated for 8 weeks were not different from those of Ilpum rice, non-GMO.
This study was carried out to establish a new breeding scheme which is connected with conventional breeding method and anther culture method. To develop a herbicide resistant and direct seeding rice, $F_1$ plants were subjected to anther culture and regenerated plants from 5 crosses were studied to confirm the introduction of bar gene. After PCR analysis, we selected 227 plants which were carrying herbicide resistance gene (bar) out of 1,508 regenerated plants from anther culture. Among 169 $A_2$ lines carrying herbicide resistant gene from 5 crosses including YR23235 (Dongjin Ds3(Ba $r^{R}$)/ Milyang165), 42 lines that had superior agronomic characters were selected for further research. Among them, YR23235Acp79 which showed herbicide resistance, direct seeding adaptability and superiority in major agronomic characters was named Milyang 204. This breeding scheme proved that the anther culture of $F_1$ plants crossed between transformant and cultivar or transform ant alone could be utilized in breeding programs for a rapid progeny fixation and development of a variety.y.
Acetolactate synthase (ALS) is the first common enzyme in the biosynthesis of L-Ieucine, L-isoleucine, and L-valine. The sulfonylurea-resistant ALS gene from Nicotiana tabacum was cloned into the bacterial expression vector pGEX-2T. The resulting recombinant plasmid pGEX-ALS3 was used to transform Escherichia coli strain XL1-Blue, and the mutant tobacco ALS (mALS) was expressed in the bacteria as a protein fused with glutathione S-transferase (GST). The fusion product GST-mALS was purified in a single step on a glutathione-Sepharose column. ALS activities of 0.9-2.5 ${\mu}mol/min/mg$ protein were observed in the GST-mALS, and the Km values for pyruvate, FAD, and TPP were 10.8-24.1, $(1.9-8.9){\times}10^{-3}$, and 0.14-0.38 mM, respectively. The purified GST-mALS was resistant to both the sulfonylurea and the triazolopyrimidine herbicides, and lost its sensitivity to end products, L-valine and L-leucine. For comparision, the tobacco wild-type recombinant ALS fused with GST, GST-wALS, was also characterized with respect to its pyruvate and cofactor bindings. These results suggest that the purified mutant recombinant tobacco ALS was functionally active, that the mutations resulting in herbicide resistance has affected pyruvate and cofactor bindings," and that the two classes of herbicides interact at a common site on the plant ALS.

Development of herbicide resistant transgenic legume plants through Agrobacterium-mediated transformation has been worked in many previous studied. Plant regeneration after infection is the important step to obtain successful transgenic plants. Many attempts try to find the optimum media condition for plant regeneration after infection. However, the transformation efficiency of legume plants is still low. In this study, regeneration of some Korean legume species including two soybean cultivars (Dawon and Pungsan) and pea have been done with organogenesis which is used various kind of explants such as cotyledonary-nodes in soybean and bud-containing tissue in pea. We developed the optimum media condition for plant regeneration regulators under Agrobacterium-mediated transformation using different kind and various concentration of plant growth. As the results, B5 medium containing 2 mg/L of 6-benzylaminopurine was selected in this study for the optimum plant regeneration media. The segments were inoculated with Agrobacterium suspension harbored an IG2 vector containing bar gene which confers resistance to phosphinotricin (PPT) in 3, 5 and 7 days. The transformation efficiency was achieved in Dawon 3.03 % and pea 1.46 % with co-cultivation period of 7 days which is showed a high number of GUS positive expression period.

This study was to investigate the occurrence patterns of paddy weeds, their resistance levels to an ALS inhibiting herbicide, and to estimate the areas of resistance in these paddy fields. We used soil collected from 358 paddy fields of Jeonnam province in 2017. Based on their life cycles, weeds were 96% annuals and 4% perennial. Additionally, according to morphological classification, 59% were broad leaves, 28% were sedges and 13% were grasses. Different areas within Jeonnam province contained different numbers and occurrence rates of weed species. However, generally, we observed Lindernia dubia var. dubia, Monochoria vaginalis var. plantaginea, Ludwigia prostrata, L. procumbens, Cyperus difformis, Scirpus juncoides, Eleocharis Kuroguwai, Echinochloa oryzoides, and E. crus-galli var. echinata. We also observed seven weeds resistant to an ALS inhibiting herbicide. They were M. vaginalis, S. juncoides, C. difformis, L. dubia, Ludwigia prostrata, E. oryzoides, and E. crus-galli var. echinata. Although there were differences in the number and occurrence rate of resistant weed species to an ALS inhibiting herbicide among areas in Jeonnam province, the M. vaginalis, C. difformis, and S. juncoides occurred in 23 cities and counties in Jeonnam including Gwangju metropolitan city. Based on the rates (52%) of resistant occurrence to an ALS inhibiting herbicide in Jeonnam province, the area of weed resistant paddy fields was estimated to be 91,543 ha.
The expression of transgenic traits in genetically modified crops is sometimes associated with decreases in crop performance or fitness. These decreases in performance or fitness of transgenic plants in unfavourable conditions may provide valuable information about the ecological consequences of transgene escape. In a glasshouse trial, we tested the cost associated with resistance to herbicides by comparing the growth, yield, and competitive ability of transgenic rice with its parental non-transgenic line. This new line was developed for constitutive overexpression of protoporphyrinogen oxidase (PPO) to increase resistance to herbicides. We evaluated nine agronomic traits of transgenic and non-transgenic rice grown in a replacement series design over four densities. Competitive ability was also assessed between transgenic and non-transgenic plants by analyzing their relative yields based on biomass and seed weight data. Our results indicated that non-transgenic plants showed greater performance than did the transgenic plants when those genotypes were grown in mixtures. The non-transgenic rice plants exhibited superior competitive ability at certain combinations of planting densities and genotype proportions. These results suggest that PPO-herbicide resistance incurs some costs in plant performance and competitive ability.
An immunoassay method was developed to quantitatively detect phosphinothricin-N-acetyltransferase (PAT) encoded by the Bialaphos resistance (bar) gene in genetically modified (GM) pepper. The histidine-tagged PAT was overexpressed in Escherichia coli M15 (pQE3l-bar) and efficiently purified by $Ni^{2+}$ affinity chromatography. A developed sandwich enzyme-linked immunosorbent assay (S-ELISA) method (detection limit: $0.01{\mu}g/ml$) was 100-fold more sensitive than a competitive indirect ELISA (CI-ELISA) method or Western blot analysis in detecting the recombinant PAT. In real sample tests, PAT in genetically modified herbicide-tolerant (GMHT) peppers was successfully quantified [$4.9{\pm}0.4{\mu}g/g$ of sample (n=6)] by the S-ELISA method. The S-ELISA method developed here could be applied to other GMHT crops and vegetables producing PAT.

The nutritional composition of transgenic rice (Agb0101) with bar and modified cry1Ac1 genes grown with herbicides was compared with that of its non-transgenic counterpart. The analyzed components (proximates, amino acids, fatty acids, minerals, vitamins, trypsin inhibitors, and phytic acid) in the herbicide-treated Agb0101 brown rice were substantially equivalent to those of its non-transgenic counterpart.

Studies were carried out to provide basic information for establishing the weed control in Chungbuk, Korea. The present surveys targeting 260 farmers in Chungbuk province were conducted for the cultivation system, weeds occurrence and usage of herbicides. To estimate the occurring area of herbicide resistant weeds, soil samples from 400 paddy fields were collected twice on August, 2011 and April, 2012. In the results of survey, the 99.6% of farmers used the rice planting machine and the 78% of the farmers disseminated herbicides twice to control weeds before and after planting rice. The most commonly used herbicide were as follows; soil-applied herbicide : butachlor 46.6%, mid-term herbicide : mefenacet + pyrazosulfuron-ethyl 10.7%, foliar herbicides : bentazone 62%. The dominant paddy field weeds included Echinochloa crusgalli (16.2%), Scirpus juncoides (12.2%), Monochoria vaginalis (11.9%) and Sagittaria trifolia (9.5%). Occurrence area of sulfonylurea herbicide resistant weeds was 13,659 ha in 26.8% of the paddy area. Monochoria vaginalis showed the highest with 4,605 ha (36.4%) followed Scirpus juncoides (30.7%), and Lindernia dubia (10.6%) at 2011. Monochoria vaginalis and Scirpus juncoides occurred were evenly distributed and the most problematic weed in Chungbuk, Korea.
