Quantitative analysis of the marker compounds in the decoctions of Coptis chinensis-Scutellaria baicalensis at different proportion produced by 'Mixed decoction' and 'Single decoction mixture' (배합 비율에 따른 황련과 황금의 혼합 전탕액 및 개별 전탕 혼합액 내 성분 함량 분석)

  • Objective : The present study aimed to evaluate the change of the content of 7 active components in decoctions produced by various proportional pairs of Coptis chinensis Franch and Scutellaria baicalensis Georgi in 'Mixed decoction (MD)' and 'Single decoction mixture (SDM)'. Methods : The samples of MDs were prepared by decocting C. chinensis : S. baicalensis with the ratios of 10 g:10 g, 10 g:20 g, and 20 g:10 g. Those of SDMs were prepared by blending each single decoction from C. chinensis and S. Baicalensis with the ratios of 1:1, 1:2, and 2:1. The samples were evaluated by high-performance liquid chromatography with statistical analyses. Results : The analytical methods, which were optimized and validated, were reliably applied to present research. The content of all components in both MDs and SDMs at C. chinensis : S. baicalensis = 1:1 ratio were reduced compared with single herb decoction. The components from each compositional herb in MDs were proportionally increased with the ratio of original herb increased, but inversely proportional to paired herb. The contents of components in MDs were significantly lower than those in SDMs at all ratios, except for high content of baicalin at C. chinensis : S. baicalensis = 2:1. Conclusion : It was concluded that MDs and SDMs as well as the proportions of herbs could affect the contents of the components from original herbal medicines. These results provide the information for the quality control of herbal medicine combined C. chinensis with S. baicalensis.

Antimicrobial and cytotoxic properties of calcium-enriched mixture cement, Iranian propolis, and propolis with herbal extracts in primary dental pulp stem cells

  • Objectives: In this study, natural substances were introduced as primary dental pulp caps for use in pulp therapy, and the antimicrobial and cytotoxic properties of these substances were investigated. Materials and Methods: In this in vitro study, the antimicrobial properties of calcium-enriched mixture (CEM) cement, propolis, and propolis individually combined with the extracts of several medicinal plants were investigated against Enterococcus faecalis, Escherichia coli, Pseudomonas aeruginosa, and Staphylococcus aureus. Then, the cytotoxicity of each substance or mixture against pulp stem cells extracted from 30 primary healthy teeth was evaluated at 4 concentrations. Data were gathered via observation, and optical density values were obtained using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) test and recorded. SPSS software version 23 was used to analyze the data. Data were evaluated using 2-way analysis of variance and the Tukey test. Results: Regarding antimicrobial properties, thyme alone and thyme + propolis had the lowest minimum inhibitory concentrations (MICs) against the growth of S. aureus, E. coli, and P. aeruginosa bacteria. For E. faecalis, thyme + propolis had the lowest MIC, followed by thyme alone. At 24 and 72 hours, thyme + propolis, CEM cement, and propolis had the greatest bioviability in the primary dental pulp stem cells, and lavender + propolis had the lowest bioviability. Conclusions: Of the studied materials, thyme + propolis showed the best results in the measures of practical performance as a dental pulp cap.

Effects of Medicinal Herb Extracts of Artemisia iwayomogi Kitamura and Angelica gigas Nakai on Disease Resistance in Whiteleg Shrimp Litopenaeus vannamei (양식 흰다리새우(Litopenaeus vannamei)의 혼합생약재 투여에 따른 항병력 조사)

  • Although aquaculture shrimp production has increased, disease mortality has also increased. Acute hepatopancreatic necrosis disease (AHPND) is an emerging disease that significantly reduces production. In this study, we have investigated the potential of medicinal herbs in enhancing the immune system of shrimp. A mixture of medicinal herb extracts (Han In-jin, Artemisia iwayomogi kitamura and Cham Dang-gwi, Angelicae gigas Nakai) was absorbed into pellets and fed to whiteleg shrimp Litopenaeus vannamei for 12 weeks. The weight gain, immune activity, and disease resistance effects of the shirmp were examined. The growth performance of shrimp fed the mixture of medicinal herbal extracts was higher than that of the control group. The prophenoloxidase (proPO) activity was found to be significantly higher at 8 weeks in the 0.1% medicinal herb extract fed group. The cumulative mortality when artificially infected with AHPND pathogens commonly decreased in the 8 weeks fed group when compared with the control group. Furthermore, the RPS (relative percentage survival) was improved. This study confirmed that a mixture of medicinal herbal extracts has a positive effect on growth performance, innate immunity and disease resistance in shrimp. The optimum concentration of the medicinal herb extract for shrimp feed was 0.1% .

Effects of Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture on Osteoblast Differentiation and Osteoclast Formation (방사선 육종 차조기와 백출 복합물이 조골세포와 파골세포의 활성에 미치는 영향)

  • The effects of the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture on the activities of osteoblast differentiation and the restraint of osteoclast formation were investigated. the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture in the human osteoblast "MG-63" cell, was examined in relation to alkaline phosphatase (ALP) activity and alizarin red stains. In order to observe the effects of osteoclasts formation, we analyzed RAW 264.7 cell tartrate-resistant acid phosphatase (TRAP) activity and TRAP stains. In cytotoxicity testing, it was confirmed that apple extract is safe at a concentration of 50 ㎍/㎖ or less. The ALP activity and Bone calcification formation ability were the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture had a lower activity than that of control group. However the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture significantly reduced activity of TRAP in the RAW 264.7 osteoclastic cell generation and effectively Inhibited the TRAP(+) multinuclear cells. Thus, our results demonstrate that the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture enhances the inhibitory activity of bone-resorption in RAW 264.7 cells. In conclusion, the Perilla frutescens var. crispa and Atractylodes macrocephala Koidzumi mixture seem to be effective in the prevention and treatment of bone related disorders.

Enhancing effect of Multiherb extracts HT008-1 on Memory and Cognitive Function (한약복합물 HT008-1의 인지기능 및 기억력 향상효과)

  • Objectives : Investigation of the memory and cognitive enhancing effect of HT008-1 in scopolamine induced amnesia mice. Methods : At 60 min before acquisition trials, HT008-1 (30, 100, 300 mg/kg p.o.) was administered, and 30 min later, mice were injected with scopolamin (1.0 mg/kg, i.p.). In the passive avoidance test, acquisition trials were carried out 30 min after a single scopolamine treatment. Retention trials were carried out 24h after acquisition trials. Y-maze test was carried out 30 min after a single scopolamine treatment. Spontaneous alternation behavior during an 8-min session was recorded. Inhibitory effects of HT008-1 (0.01, 0.1, 1.0 mg/ml) on AChE activity was measured. Result : HT008-1 ameliorated scopolamine-induced learning impairments and spatial cognitive function in passive avoidance and Y-maze test, respectively. Moreover HT008-1 showed a significant inhibitory effect on AChE activity. Discussion: This study presented that eMultiherb mixture HT008-1 enhanced learning memory and spatial cognitive function in scopolamine-induced amnesia mice. These results suggest that the effect of HT008-1 may be dependent on the inhibition of AChE activity.

Antipruritic Effects of Puerariae Radix on 5-HT or AEW-Induced Itch Behavior in Mice (5-HT 및 AEW로 소양증(搔痒症)을 유발한 흰쥐에 대한 갈근(葛根)의 진양(鎭痒)효과)

  • Objectives : To find antipruritic herbal medicines, we screened the water extracts of four herbal medicines which have been frequently prescribed to treat dermatologic diseases in oriental medicine. Methods : Water extracts of Bupleuri Radix, Scutellariae Radix, Cimicifugae Rhizoma and Puerariae Radix were administered at a dose of 300 mg/kg to male ICR mice. 1 hour later, serotonin hydrochloride dissolved in physiologic saline at a concentration of 100 nmol / 50${\mu}{\ell}$ was administered intradermally to the nape of the neck at a dose of 50${\mu}{\ell}$. Then the scratching behavior was observed. Dry skin pruritus was induced with cutaneous application of acetone / ether (1:1) mixture and water (AEW) to the rostral back of male ICR mice, twice a day for 5 days. 14 hours after the last application, the most effective material in the first experiment was administered perorally at doses of 75, 150, 300, 600 and 1200 mg/kg. 1 hour later, the scratching behavior was observed. Results : Water extracts of Puerariae Radix significantly inhibited the serotonin-induced scratching behavior and the mean of scratching bouts was reduced by half compared with the control group. Bupleuri Radix group also showed a 29% decrease in the mean of scratching bouts, but there was no statistical significance. Water extracts of Puerariae Radix also inhibited the AEW-induced scratching behavior, in a dose-dependent manner. The dose of 150 mg/kg showed the highest and statistically significant antipruritic effect. Conclusions : These results suggest that Puerariae Radix and its constituents have antipruritic effect, and are new candidates as antipruritic agents.

Optical Characteristics of Two New Functional Films and Their Effect on Leaf Vegetables Growth and Yield (2종류의 기능성필름이 광학특성과 엽채류 생육과 수량에 미치는 영향)

  • Three leaf vegetables, namely green lettuce, red lettuce (Lactuca sativa) and red-veined chicory (Cichorium intybus) were grown in minigreenhouses covered with two new functional films and conventional polyethylene film (PE). Seedlings of leaf vegetables were transplanted in a plastic troughs filled with soil-perlite mixture. Two functional films were made from polyolefin (PO) material. Measurement of optical characteristics showed that polyolefin films have better transmittance for the photosynthetic active radiation (PAR, 400-700nm) and higher absorptance for the ultraviolet radiation (UV, 300-400nm) in comparison with the conventional PE film. After three months of utilization higher loss in PAR transmittance was observed for conventional PE film. Leaf vegetables growth was enhanced and yield was increased in greenhouses covered by new functional films.

Comparison of Biological Activities on Rehmanniae Radix and Fermented Rehmanniae Radix (지황(地黃)과 발효(醱酵) 지황(地黃)의 생리활성 비교 연구)

  • Herbal medicines are medicinal products containing a single or a mixture of two or more different herbal substances or herbal preparations as active principles. Recently, much attention has been paid to developing various kinds of fermented herbal extracts, a new type of traditional herbal medicine in the field of Korean traditional medicine. The fermentation of medicinal herbs is intended to exert a favorable influence on bioestability, bioavaliablilty and pharmacological activity of herbal extract in the gastrointestinal tract as well as intensifying the nutritional and pharmacological aspects of the medicinal herbs. The purpose of this study was to investigate biological activities of fermented Rehmanniae Radix by lactic acid bacteria at $30^{\circ}C$ for 3 days in comparison with those for Rehmanniae Radix The fermented Rehmanniae Radix exhibited different chemical profile to Rehmanniae Radix generated with HPLC, indicating production of new ingredients during fermentation. Rehmanniae Radix served as good nutritional sources for the growth of lactic acid bacteria showing increased number of bacteria during fermentation. Toxic effect of the fermented Rehmanniae Radix to cells were not seen judged by the MTT assay. The fermented Rehmanniae Radix exhibited better antioxidant effect than non-fermented Rehmanniae Radix analyzed by a SOD-likely assay. Both hypoglycemic and hypotensive effects of the fermented Rehmanniae Radix were also detected and better than those for Rehmanniae Radix in showing dose-dependent inhibitory effects on alpha-glucosidase and ACE, respectively. In conclusion, fermented Rehmanniae Radix appears to have more biological activities than non-feremented Rehmanniae Radix showing not only antioxidant effect but also cardiovascular protection.

Simultaneous Determination and Optimization Ultrasound-Assisted Extraction of Poncirin and Naringin in Poncirus trifoliata Rafinesqul (지실의 Poncirin, Naringin의 동시분석법 확립과 초음파 추출법 최적화)

  • The Ponciri fructus immaturus (Poncirus trifoliata Rafinesque) has been used in oriental medicine for uterine contraction, stomachache, abdominal distension and cardiovascular diseases. Two main compounds, poncirin and naringin were successfully analyzed by high performance liquid chromatography (HPLC) and carried out method validation according to ICH guideline. A successful resolution and retention times were obtained with a $C_{18}$ reversed phase column, at an $1m{\ell}min^{-1}$ flow rate, with a gradient elution of a mixture of methanol, water and acetonitrile. Poncirin and naringin showed good linearity ($R^2$ > 0.999) in relatively wide concentration ranged. The recovery of each compound was 95.81 ~ 101.48% with R.S.D. values less than 1.0%. The application of ultrasound-assisted extraction was shown to be more efficient in extracting poncirin and naringin from Ponciri fructus immaturus. The predicted optimal poncirin and naringin yield were poncirin 2.15%, naringin 1.65% under an extraction temperature of $40^{\circ}C$, an extraction time of 10 min in a solvent of 70% methanol.

The Effect of Anaerobic Fermentation Treatment of Wheat bran on the Root-Knot Nematodes and the Quality of Melons in Plastic Film House Soil (밀기울 토양 혐기발효 처리가 멜론의 뿌리혹선충 방제 및 품질에 미치는 영향)

  • This study was conducted to investigate the effect of anaerobic fermentation of wheat bran to prevent root-knot nematodes which are infected in plastic house due to over 10 years continuous cultivation of fruits and vegetables. Anaerobic fermentation treatment of wheat bran was done for 20 days by mixture of 2,000 kg fresh wheat bran per 10 are and soil with water in 30 cm soil depth. Chemical treatment of fosthiazate was done by mixture of 6 kg soil for 7 days. Both treatments show suppression of density of rootknot nematodes, especially in anaerobic fermentation treatment. Anaerobic fermentation treatment keeps the low level of root-knot nematode density until 90 days of cultivation and also showed good effect of melon growth. Related with death percentage of melon plant, anaerobic fermentation treatment shows only 3% and also large size of quality fruit but control 65%. Anaerobic fermentation treatment of wheat bran have proved to control the level of root-knot nematodes instead of synthetic chemicals for at least one cropping season and it showed good effect to fruit quality.
