Carthamus tinctorius seeds-Taraxacum coreanum combination attenuates scopolamine-induced memory deficit through regulation of inflammatory response and cholinergic function

  • BACKGROUND/OBJECTIVES: There is growing interest in herbal medicines for managing age-related diseases, such as Alzheimer's and Parkinson's. Safflower seeds (Carthamus tinctorius L. seeds, CTS) and dandelions (Taraxacum coreanum, TC) are widely used to treat bone- or inflammation-related diseases in Oriental countries. This study investigated the protective effect of the CTS-TC combination on scopolamine (Sco)-induced memory deficits through inflammatory response and cholinergic function. Moreover, marker components such as serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid in the CTS-TC combination were analyzed for their potential benefits on memory function. MATERIALS/METHODS: Water extracts of CTS, TC, and the CTS-TC combination at various ratios (4:1, 1:1, and 1:4) (100 mg/kg) were orally administered to mice for 14 days. Sco (1 mg/kg) was intraperitoneally injected into the mice before each behavioral test. T-maze and novel object recognition tests were conducted to monitor behavioral changes after the treatment. Western blotting was performed to detect protein expression. In addition, the presence of 5 biomarkers, serotonin, N-(p-coumaroyl) serotonin, N-feruloylserotonin, chlorogenic acid, and chicoric acid, was analyzed using high-performance liquid chromatography (HPLC). RESULTS: Behavioral tests showed that the CTS-TC combination enhanced memory function in Sco-injected mice. Inflammation-related proteins (inducible nitric oxide synthase, cyclooxygenase-2, and glial fibrillary acidic protein) were downregulated after treatment with the CTS-TC combination. The acetylcholinesterase protein expression was also downregulated. HPLC analysis revealed that N-feruloylserotonin and chicoric acid were the predominant components, followed by N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin. CONCLUSION: These findings suggest that the CTS-TC combination protects against Sco-induced memory deficits by inhibiting inflammatory responses and cholinergic dysfunction. N-feruloylserotonin and chicoric acid, along with N-(p-coumaroyl) serotonin, chlorogenic acid, and serotonin, might be biomarkers for the CTS-TC combination, and their effects on memory protection warrant further study.

Urtica Dioica and Lamium Album Decrease Glycogen Synthase Kinase-3 beta and Increase K-Ras in Diabetic Rats

  • Objectives: The aim of the present work is evaluating the special effects of Urtica Dioica and Lamium Album on the serum level of K-Ras and GSK-3 beta in diabetic rats. Methods: In the present experimental study, 32 male Wistar rats randomly divided into 4 groups (Group I: normal control rats; receiving daily PBS, Group 2: diabetic control rats; receiving single dose of streptozotocin (60 mg/kg) and daily PBS, Group 3: Diabetic rats treated with 100 mg/kg of hydroalcoholic extract of the U. dioica, Group 4: Diabetic rats treated with 100 mg/kg of hydroalcoholic extract of L. Album. Diabetes-induced by an intraperitoneal injection of streptozotocin (60 mg/ kg). On the 14 th day of treatment, the weight, fasting blood sugar (FBS) and on 28 th day blood glucose, K-Ras and GSK3 beta was measured. Results: In diabetic group blood GSK- 3 beta increase in comparison to control group (P < 0.05), also blood K-Ras decrease in the diabetic group (P < 0.05). Both extracts reduced GSK-3 beta level, however, this reduction was only statistically significant by U.dioica (P < 0.05). Compared to diabetic group, blood K-Ras level increased by both extract (P < 0.05). Also diabetes induction increase blood glucose levels and both extracts decrease its level significantly (P < 0.05).there is no significant differences among both extract effects on blood glucose, and K-Ras. Conclusion: For the first time shown that both extracts by regulating GSK-3 beta and K-Ras improve blood glucose level. More studies are needed to determine all the effects of these herbs.

Assessment of free-radical-scavenging and antibacterial activities, and brine shrimp toxicity of Scutellaria pinnatifida (Lamiaceae)

  • Scutellaria pinnatifida A. Hamilt. (Lamiaceae) is an endemic Turkish herb. This plant is also endemic to Iran, and grows abundantly in other central and western Asian countries. Several species of the Scutellaria are known for their traditional uses in the treatment of hypertension, arteriosclerosis, inflammatory diseases, hepatitis, allergy, cancer and diarrhoea. Free-radical-scavenging property, antibacterial activity and brine shrimp toxicity of the n-hexane, dichloromethane (DCM) and methanol (MeOH) extracts of S. pinnatifida were assessed using the 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assay, the resazurin microtitre plate based assay, and the brine shrimp lethality assay, respectively. The DCM and MeOH extracts exhibited free-radical-scavenging property, with the $RC_{50}$ values of 0.362 and 0.127 mg/ml, respectively. Among the solid-phase extraction fractions of the MeOH extract, the 50% aqueous-MeOH fraction showed the highest level of free-radicalscavenging activity ($RC_{50}$ = 0.039 mg/ml). While the DCM extract showed low level of antibacterial activity against Bacillus subtilis and ampicillin-resistant Escherichia coli, the MeOH extract was active against B. cereus, B. subtilis, E. coli and ampicillin-resistant E. coli. However, the minimum inhibitory concentrations (MIC) of the MeOH extract against these bacterial strains were >10 mg/ml. None of the extracts showed any significant toxicity towards brine shrimps ($LD_{50}$ = > 1.00 mg/ml).

  • Objectives : The histrionic personality disorder(HPD) is characterized by a pattern of excessive emotion and attention-seeking behavior, including a seductive behavior. It is known that HPD is closely related to a somatization disorder. The somatization disorder is characterized with various physical symptoms that have no pathologic manifestation. This report is on a case of 55 years old woman with HPD, who has been suffering from various physical symptoms that are coincided with diagnostic criteria for somatization disorder by DSM-IV. Methods : The patient was treated with oriental medical treatments (acupuncture, moxibustion, cupping therapy, and herb-medication), hypnotic therapy, and emotional freedom techniques. The effects of treatment were measured by VAS. Results : Chest pain was improved by hypnotic therapy and oriental medical treatments. But the patient's pantalgia and abdominal discomfort were controlled effectively by emotional freedom techniques. Conclusions : This result suggests that EFT might be effective for the defensive and dependent somatization disorder patient with HPD.

Houttuynia cordata Improves Cognitive Deficits in Cholinergic Dysfunction Alzheimer's Disease-Like Models

  • Cognitive impairment is a result of dementia of diverse causes, such as cholinergic dysfunction and Alzheimer's disease (AD). Houttuynia cordata Thunb. (Saururaceae) has long been used as a traditional herbal medicine. It has biological activities including protective effects against amyloid beta ($A{\beta}$) toxicity, via regulation of calcium homeostasis, in rat hippocampal cells. To extend previous reports, we investigated the effects of water extracts of H. cordata herb (HCW) on tauopathies, also involving calcium influx. We then confirmed the effects of HCW in improving memory impairment and neuronal damage in mice with Ab-induced neurotoxicity. We also investigated the effects of HCW against scopolamine-induced cholinergic dysfunction in mice. In primary neuronal cells, HCW inhibited the phosphorylation of tau by regulating p25/p35 expression in $A{\beta}$-induced neurotoxicity. In mice with $A{\beta}$-induced neurotoxicity, HCW improved cognitive impairment, as assessed with behavioral tasks, such as novel object recognition, Y-maze, and passive avoidance tasks. HCW also inhibited the degeneration of neurons in the CA3 region of the hippocampus in Ab-induced neurotoxicity. Moreover, HCW, which had an $IC_{50}$ value of $79.7{\mu}g/ml$ for acetylcholinesterase inhibition, ameliorated scopolamine-induced cognitive impairment significantly in Y-maze and passive avoidance tasks. These results indicate that HCW improved cognitive impairment, due to cholinergic dysfunction, with inhibitory effects against tauopathies and cholinergic antagonists, suggesting that HCW may be an interesting candidate to investigate for the treatment of AD.

데이터베이스를 이용한 식료찬요(食療纂要)의 통계적 연구 (A Statistical Study on Sikryo-chanryo by Applying Database)

  • 본 연구의 목적은 한국 고유의 전통지식에 기반하여 음식을 이용한 건강증진 방법에 대한 지식을 체계화하고 통계적으로 활용할 수 있는 방법에 대하여 연구하는 데 있다. 이를 위하여 한국 고유의 음식을 이용한 질병치료에 대하여 언급한 고문헌인 식료찬요(食療纂要)의 지식을 DB로 구축하였다. 데이터의 처리는 Relational Data Model을 이용하였고, 9개의 데이터 테이블을 사용하였다. 소프트웨어는 MS사의 Access 2014를 사용하였다. 그 결과, 데이터베이스 쿼리문장을 이용하여 식재료, 약재별, 증상, 병명, 효능, 변증 정보를 유기적으로 조건을 만들어 통계적 결과를 즉시 확인할 수 있었다. 따라서 이러한 Relational Data Model을 이용하여 기존 방식의 연구논문을 대체할 수 있었다.

탈모마우스모델에서의 송지추출물 및 그 성분인 아비에트산의 모발성장효과 (Hair Growth-promoting Effect of Resina Pini and Its Main Constituent, Abietic Acid, in Mouse Model of Alopecia)

  • 최근 남성성탈모증에 대한 관심이 증대되고 있으며, 이에 천연물 및 그의 활성성분을 활용한 새로운 약물 개발에 대한 연구가 증가하고 있다. 송지(Resina Pini, RP)는 Pinus sp. (Pinaceae)의 수지질로 전통의학적으로 감염, 우식증, 치주질환에 사용되어왔다. 본 연구진은 RP의 성분인 아비에트산(abietic acid, AA)이 남성성탈모기전에 중요한 효소인 $5{\alpha}$-reductase를 억제하는 효과를 세포 수준에서 입증한바 있으며, 이번 연구에서는 실제로 탈모억제 및 모발 성장에 대하여 실험동물 수준에서 입증하고자 한다. C3H/HeN 탈모마우스 모델에서 RP는 300 mg/kg에서 유의하게 탈모억제를 확인하였으며, 뿐만 아니라 AA는 30 mg/kg에서도 유의하게 탈모억제효과를 보였다. 이상의 결과로부터 RP는 그 활성성분인 AA가 $5{\alpha}$-reductase 억제하는 기전을 통해 남성성탈모억제효과를 보였다고 사료되며, 향후 탈모억제 보완치료법으로의 이용 가능성을 보였다.

보두산의 금속분해효소 활성 저해를 통한 사람 대동맥 평활근세포의 유주능 억제 효과 (Inhibitory Effect of Bodusan on $TNF-{\alpha}-induced$ Migration via Inhibition of Metalloproteinase Activity in Human Aortic Smooth Muscle Cells)

  • Objectives : This study was evaluated to elucidate the inhibitory potential of Bodusan (BDS) and its components, Strychnos ignatii semen (SIS) and Glycyrrhizae Radix (GR), on human aortic smooth muscle cells (HASMC) migration and production of matrix metalloproteinase (MMP)-2 and MMP-9 by $TNF-{\alpha}$ treatment. Methods : Cytotoxic activity of BDS and its components on HASMC was using 5-(3-caroboxymeth- oxyphenyl)-2H-tetra-zolium inner salt (MTS) assay. Effect of BDS, SIS and GR on $TNF-{\alpha}-induced$ HASMC migration underside of matrigel filter was stained with hematoxylin-eosin. And total number of cells that migrated to the underside of the filter was counted. MMP-2 and MMP-9 activity was evaluated by gelatin zymography assay. Results : The matrigel migration assay showed that BDS effectively inhibited the $TNF-{\alpha}-induced$ migration of HASMC. Moreover, BDS significantly inhibited MMP-9 activity. Our present study demonstrates that BDS and its components inhibits $TNF-{\alpha}-induced$ HASMC migration and MMP-9 activity. The inhibitory effect of BDS extract is more potent than that of its component herb extracts. Conclusions : These results provide evidence that BDS has multiple effects in the inhibition of HASMC migration and may offer a therapeutic approach to block HASMC migration.

인간 혈관 내피세포에서 NF-κB 억제를 통한 엉겅퀴 추출물의 VCAM-1 및 ICAM-1 발현 억제효과 (Cirsium japonicum var. Maackii Extract Suppress VCAM-1 and ICAM-1 Expression in TNF-α-treated Human Vascular Endothelial Cells by Blocking NF-κB Activation)

  • Cirsium japonicum var. maackii is a traditional Korean wild perennial herb used to treat blood circulation, high blood pressure, inflammation, diabetes, and kidney damage. However, it is not known whether C. japonicum var. maackii directly improves endothelial dysfunction. In this study, the effect of C. japonicum var. maackii (CJE) on tumor necrosis factor (TNF)-α-induced vascular inflammation was investigated in vitro using human umbilical vein endothelial cells (HUVEC). As a result, CJE inhibited the production of VCAM-1, ICAM-1 and ROS increased by TNF-α in HUVECs. In addition, treatment with CJE attenuated IκB phosphorylation and translocation of NF-κB to the nucleus. These results suggest that CJE can suppress TNFα-induced adhesion molecule expression by blocking NF-κB signaling and inhibiting ROS generation. The results of this study show that CJE has the potential to be used to treat and prevent inflammation associated with endothelial cell damage.

신장상피세포(腎臟上皮細胞)에서 호도약침액(胡桃藥鍼液)이 t-Butylhydroperoxide에 의한 세포막물질이동계(細胞膜物質移動系)의 장애(障碍)에 미치는 영향(影響) (Effect of Juglans sinensis Dode aquacupuncture(JS) on t-butylhydroperoxide-induced alterations in membrane transport function in renal epithelial cells)

  • Juglans sinensis Dode has been reported to have antioxidant activity. However, the effect of Juglans sinensis Dode aquacupuncture(JS) on reactive oxygen species(ROS)-induced alterations in membrane transport function in renal tubular cells. This study was performed to evaluate the effect of JS on the organic hydroperoxide t-butylhydroperoxide(tBHP)-induced inhibition of $Na^+$-dependent phosphate($Na^+$-Pi) uptake in opossum kidney (OK) cells, an established renal proximal epithelial cell line. tBHP inhibited $Na^+$-Pi uptake in a time-dependent manner. The inhibitory effect of tBHP was prevented by JS over concentration range of 0.05-1mg/100ml in a dose-dependent manner. Kinetic studies showed that tBHP caused an decrease in Vmax for $Na^+$-Pi uptake without any a significant change in Km. $Na^+$-dependent phosphonoformic acid binding, a irreversible inhibitor of renal $Na^+$-Pi uptake, was decreased by tBHP treatment. The reduction in Vmax and phosphonoformic acid binding by tBHP was prevented by JS. tBHP induced lipid peroxidation and its effect was completely inhibited by JS and antioxidant N,N'-diphenyl-p-phenylenediamine. These data suggest that the oxidant inhibits phosphate uptake by a reduction in the number of active carrier across the membrane. JS may prevent oxidant-induced inhibition of membrane transport function by a mechanism similar to antioxidants in renal epithelial cells. Although the precise constituents remain to be explored, JS may be employed as a useful candidate herb for drug development to prevent and treat oxidant-mediated renal failure.
