Effects of Dermal Cell Combination on the Formation of Basement membrane and Epidermis in Skin Equivalents (진피세포의 조성이 인공피부의 기저막과 표피형성에 미치는 영향)

  • European Union prohibited the marketing of cosmetic products containing constituents that have been examined through animal experiments. Thus, non-animal test models are needed to replace animal experiments. The reconstructed skin models are important as a test system for cosmetic, pharmaceutical, and medical device safety testing. In the present study, we tried to develop an optimal skin equivalent model containing basement membrane and epidermis. For this purpose, we used mesenchymal stem cells (MSCs) and/or preadipocytes as well as fibroblasts as the dermal matrix cells. The formation of basement membrane and epidermis was verified by immunohistochemical stains. Among various models, the epidermis was thickest when MSCs were used in the dermal matrix. Furthermore, PCNA and involucrin distribution showed that dermal matrix with MSCs resembled human skin. Therefore, skin equivalents with MSCs could be developed as a non-animal test model to replace animal experiments.

  • The purpose of this study is to find out effects of prescriptions according to the formula, manipulation of Rhei Rhizoma, configuration. The following results were reached through investigations on the prescriptions using Rhei Rhizoma as a main component. Objectives : Analysis of prescriptions According to the formula : Liquid Extract Prescriptions were used widely on three parts to treat fever and damp heat in the interior organs. Powder Prescriptions were taken with hot water, thin porridge, tea etc. to treat damp heat, congestion of phlegm, acute episodes. Liquid Mixed Pill treat congestion of QI, damp heat, phlegm. Honey Mixed Pill treat accumulated fever, distension, acute excessive fever. Paste Pill treat blood stagnation, excessive toxic-fever, epidemic diseases. External Application treat inflammation by injury, swelling due to severe fever by internal damage. Methods : Analysis of prescriptions According to the manipulation of Rhei Rhizoma : Prescriptions including Liquor processed Rhei Rhizoma treat excessive toxic-fever, congestion of phlegm, blocking orifices on the upper side. Steamed Rhei Rhizoma strengthen effects of making evacuate and cooling of heat. Processed Rhei Rhizoma with vinegar strengthen effects of removing blood stagnation by activating blood movement, releasing gathering. Results : Analysis of prescriptions According to the Composition of Rhei Rhizoma : 41% of the total prescriptions were on the area of less than 20%. In case of lower groups show increased frequency of combination with Pharbitidis Semen, Persicae Semen, Scutellariae Radix and manipulation of baking, steaming, roasting. In case of higher groups show increased frequency of treating excess syndrome, critical illness, acute severe illness, and using proccesed Rhei Rhizoma with vinegar. Treatment of damp heat on the liver and gallbladder, disorder of the spleen and stomach is done mostly by prescriptions on the area of less than 30%. Conclusions : Rhei Rhizoma-Coptidis Rhizoma pair treat damp heat, heat toxins in blood, and Constipation caused by excessive heat. Rhei Rhizoma-Glycyrrhizae Radix pair relieve effects of Rhei Rhizoma passing blocked feces, removing the poison, activating blood movement, releasing gathering with the effects of Glycyrrhizae Radix relaxing tension by harmonizing Middle. Rhei Rhizoma-Magnoliae Cortex pair are used to treat damp heat in middle area, excessive heat in the stomach and intestine. Rhei Rhizoma-Pharbitidis Semen pair act on both blood system and QI system treating edema, damp, stagnation, heat toxins, feces. Rhei Rhizoma-Persicae Semen pair treat blood stagnation with fever on blood system.

The Early External Medical Treatment of China described in the Mawangtew medical books, the Hwangjenaekyeong and Geumgweyoryak (마왕퇴의서, 황제내경, 금궤요약에 나타난 중국 고대 외치병법)

  • At present in many cases the methods of medical treatment in Oriental medicine Korea lays emphasis on internal use, but to improve the curative value and for the diversity of medical treatment it is desperately needed to study external medical treatment more deeply. From the viewpoint of medical science history, as various additives and new discoveries are developed this has made it easy to use and improved the curative value of it ; thus external medical treatment has been rapidly progressed both in quality and quantity. Therefore, in studying the external medical treatment, it is very important to know it's history. So, I intend to study the early external medical treatment first. The external medical treatments mentioned in the Mawangtew medical books (52Byeongbang, Yangsangbang, Japryobang, Taesanseo), Hwangjenaekyeong, and Geumgweyoryak which were written or presumed to be written in early times were examined to study the early external medical treatments. The parts of body or diseases in which external medical treatments were applied; the methods of external application and the administration of medicine; the number of prescriptions, and the herbs used are also examined. I came to the following conclusions as I compared the kinds of books with the itemized lists based on the results of examination. 1) Though Mawangtew medical books (52Byeongbang, Yangsangbang, Japryobang, Taesanseo) was written earlier than Hwangjenaekyeong and Geumgweyoryak, there are more incidents of the disease in which external medical treatments were applied, the methods of external application and the administration of medicine, the kinds of prescriptions , and the different herbs used in Mawangtew medical books than in Hwangjellaekyeong and Geumgweyoryak. 2) In the six medical books mentioned above there are more prescriptions for external diseases and dermatitis than for internal diseases. 3) From the viewpoint of the number of prescriptions, the most frequent use of a medicine type was a natural type of medicine. The second was ointment type, and the third was liquid type. A powder type was the fourth. 4) In the administration of medicine, combination types such as both pasting and plastering were used most frequently. 5) Vegetable, mineral and animal nature herbs were used diversely for the early external medical treatments. Things such as wine and vinegar were also used as a herb.

Effect of Jasmonic Acid and NaCl on the Growth of Spearmint(Mentha spicata L.) (Jasmonic Acid 및 NaCl 처리가 스피아민트의 생육에 미치는 영향)

  • This study investigated the effects of NaCl and jasmonic acid (JA) on the growth and physiological responses of spearmint (Mentha spicata L.). Spearmint was hydroponically grown for 3 weeks in modified Hoagland solution containing 0 (untreated control), JA ($20{\mu}M$ JA pretreatment), NaCl (50 mM NaCl treatment) and JA + NaCl ($20{\mu}M$ JA pretreatment + 50 mM NaCl treatment). Growth characteristics, chlorophyll, vitamin C, proline contents, DPPH scavenging activity and inorganic ion contents were evaluated. As a results, there were significant decreases in the plant height, leaf length, leaf width, and fresh weight of plants, treated with NaCl compared with control. On the other hand, the dry matters of shoot and root treated with JA + NaCl combination were better than control or NaCl treatment. Chlorophyll a and b contents in JA treatment was the highest. Vitamin C, antioxidant activity, and proline content in shoot were increased in NaCl treatment which showed low level of growth rate. The K/Na ratio, which is known to indirectly reflect the balance of ion uptake, was higher in a single treatment of JA than the control group, while lower in salt treatment (NaCl and JA + NaCl) because of high $Na^+$ absorption. In conclusion, these results showed that moderate stress treatment such as low level salt treatment and plant growth regulator jasmonic acid (JA) application would be potential strategies to improve the quality of spearmint by inducing the accumulation of secondary metabolites containing high antioxidant activity and essential oil.

Effects of Presown Cold Stratification, $GA_3,\;KNO_3$ and Acetone Treatment on Germination of Crotalaria sessiflora L. (저온, $GA_3,\;KNO_3$ 및 Acetone 처리가 활나물 종자발아에 미치는 영향)

  • Crotalaria sessiflora is one of a few medicinal herbs among the legumes used as antitumor herb medicine but has lower seed germination and afterward seedling emergence. This study was carried out to elucidate the effects of cold stratification as presown treatment as well as $GA_3$, $KNO_3$ and acetone on its seed germination and seedling emergence. Cold stratification, $GA_3$, $KNO_3$ and acetone treatments were performed with different concentrations and period levels. Cold treatment was done under 14 hour red light illumination a day or darkness and the two others under darkness. On the basis of the best germination rate of each treatment, $GA_3$ and cold stratification treatments showed the highest germination rates at 0.1 mM but 12 hours and 2 week periods under darkness, respectively. $KNO_3$ and acetone treatments came out at 400 mM and 200 mM lasted for, respectively. The best germination rates from $KNO_3$ and acetone were comparatively higher than those from the former $GA_3$ and cold treatments. In evaluation of seedling emergence using the last two treatments, $KNO_3$ treated seeds were better performed than acetone treated seeds showed somewhat higher germination rate, implying that presown seed treatments must be evaluated by the combination with indoor and field tests.

A Study of the Āyurveda Herbal theory in the 『Suśruta-saṃhitā·Sūtrasthāna』 (Āyurveda(아유르베다)의 약물 지식에 관한 연구 - 『Suśruta-saṃhitā·Sūtrasthāna (수슈르따-상히따·수뜨라스타나)』를 중심으로 -)

  • Through a simple translation and analysis of the "Su$\acute{s}$uta-saṃhit$\bar{a}$(The fascinating tales) S$\bar{u}$trasth$\bar{a}$na(The Introduction)", I have summarized the results of the research into the $\bar{A}$yurveda Herbal theory. 1. The title of chapter 40 is "dravya-rasa-guṇa-vip$\bar{a}$ka-vij$\tilde{n}$anīya(Medcation Taste Nature Effects Digestion)", and it states theoretical knowledge on materia medica. It says that 'matter' is the most important thing, and that it decides the curing effect. Although it does acknowledge the reason behind the doctors that emphasized things like 'taste', it mainly has a critical tone. The difference in philosophies and medical theory can be seen when we compare this to the "Caraka-saṃhit$\bar{a}$", 'theory on taste'. 2. The title of chapter 41 is "dravya-vi$\acute{s}$eṣa-vij$\tilde{n}$anīya(A Discussion on the Characteristics of Materia Medica)", and the contents of the chapter have a similar tone to the thoughts of the S$\bar{a}$mkhya(數論派). All 'matter' was created by the combination of the five elements of earth, water, fire, wind, and ether, and the predominant element makes it have an earth, water, fire, wind, or ether nature. Earth has an unmoving and down going nature, and makes the body strong. Water has an irrigating nature, and moistens the tissue and induces secretion and excretion. Fire has an upward-going nature, and improves sight and complexion. Wind relaxes and dries the body, and makes the mind and body agile. Ether gives flexibility, porosity, and candidness. The title of chapter 42 is "rasa-vi$\acute{s}$eṣa-vij$\tilde{n}\bar{a}$nīya(A Discussion on the Types of Rasa(taste))". "Rasa" is an important concept in $\bar{A}$yurveda. Sometimes it is translated as 'one of the seven types of bodily tissue' and seen as chyle, and sometimes it is seen as the tastes that can be felt with the tongue such as spicy, sweet, sour, bitter, salty, and astringent(澁). Volume 1, chapter 42 of the "Caraka-saṃhit$\bar{a}$" is dedicated to the "types of taste", and in chapter 26, it theorizes the concept of 'taste'.

Plant regeneration through the callus culture induced from bulb scales of an endangered species Lilium cernum Komarvo. (멸종위기종인 솔나리(Lilium cernum Komarvo.)의 인편 유래 캘러스 유도 및 기내 식물체 재분화)

  • Lilium cernum Komarvo. is an important endangered plant belonging to the family Liliaceae. A method was developed for the rapid micropropagation of L. cernum through plant regeneration from bulb scales explant-derived calli. The bulb scales segments were cultured on Murashige and Skoog (MS) medium supplemented with 0, 0.5, 1.0, $3.0mg{\cdot}L^{-1}$ kinetin and 0, 0.1, 0.5, $1.0mg{\cdot}L^{-1}$ NAA or 2,4-D for callus induction. In media with $0.5{\sim}3.0mg{\cdot}L^{-1}$ kinetin and $0.1{\sim}1.0mg{\cdot}L^{-1}$ NAA and 2,4-D, 95~100% of explants produced callus. They were then transferred to MS medium supplemented with various concentrations of NAA (0, 0.01, 0.05 and $1.0mg{\cdot}L^{-1}$) in combination with BA (0, 1.0 and $2.0mg{\cdot}L^{-1}$) for bulbet formation. Bulbet induction (78%), weight (468 mg) and size (15.5 mm) were obtained the highest on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. In vitro frequency of plant regeneration was not significantly treated in strength of MS and sucrose concentration. Chlorophyll contents in 1/2MS with $50g{\cdot}L^{-1}$ sucrose treatments were higher than those in control and another treatment. This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.

Antistress effect of Gagamsanzointang on rat chronic stress model (가감산조인탕(加減酸棗仁湯)이 만성 스트레스 모델 쥐에 대한 항스트레스 효과)

  • Objectives : To observe the effect of 4 kinds of Korean medicine and complex extracts on the anti-stress of chronic mild stress Rat model, so as to find out the machanism of its pharmacological effects. Methods : 64 healthy SD Rats were randomly divided into 8 groups. Model Design by chronic stress(CSM), and carried out the forced swimming test. We investigated the body weight, feed and water intake, feed conversion ratio and measured parameters to changes in swimming time, immobility time, 2 and 24 hours of feces, corticosterone hormone and blood glucose level. Results : In this experiments, body weight gain and feed intake were higher in all korean medicinal herb group. Swimming time were increased by Zizyphus jujuba and Gagamsanzointang(SZT) groups and immobility time was significantly lower by Zizyphus jujuba and Fluoxetine(FLX) groups. The result of corticosterone hormone level were significantly lower in Fluoxetine(FLX) and Gagamsanzointang(SZT) but blood glucose was not significantly difference. In 2 hours after fecal frequency result were significantly lower in Zizyphus jujuba group. but SZT group were increased. This result caused by acute stress but 24 hours of feces were restored to normal. Conclusions : 5 kind of Korean Medicinal Herbs extracts has good anti-stress effects, and also to clear the circulatory system and regulate blood glucose and improve bowel function. We suggest that use a single Korean medicine rather than the combination to relieve stress.

  • According to the Leukemia and Lymphoma Society, leukemia is a malignant disease (cancer) that originates in a cell in the marrow. It is characterized by the uncontrolled growth of developing marrow cells. There are two major classifications of leukemia: myelogenous or lymphocytic, which can each be acute or chronic. The terms myelogenous or lymphocytic denote the cell type involved. Thus, four major types of leukemia are: acute or chronic myelogenous leukemia and acute or chronic lymphocytic leukemia. Leukemia, lymphoma and myeloma are considered to be related cancers because they involve the uncontrolled growth of cells with similar functions and origins. The diseases result from an acquired (not inherited) genetic injury to the DNA of a single cell, which becomes abnormal (malignant) and multiplies continuously. In the United States, about 2,000 children and 27,000 adults are diagnosed each year with leukemia. Treatment for cancer may include one or more of the following: chemotherapy, radiation therapy, biological therapy, surgery and bone marrow transplantation. The most effective treatment for leukemia is chemotherapy, which may involve one or a combination of anticancer drugs that destroy cancer cells. Specific types of leukemia are sometimes treated with radiation therapy or biological therapy. Common side effects of most chemotherapy drugs include hair loss, nausea and vomiting, decreased blood counts and infections. Each type of leukemia is sensitive to different combinations of chemotherapy. Medications and length of treatment vary from person to person. Treatment time is usually from one to two years. During this time, your care is managed on an outpatient basis at M. D. Anderson Cancer Center or through your local doctor. Once your protocol is determined, you will receive more specific information about the drug(s) that Will be used to treat your leukemia. There are many factors that will determine the course of treatment, including age, general health, the specific type of leukemia, and also whether there has been previous treatment. there is considerable interest among basic and clinical researchers in novel drugs with activity against leukemia. the vast history of experience of traditional oriental medicine with medicinal plants may facilitate the identification of novel anti leukemic compounds. In the present investigation, we studied 31 kinds of anti leukemic medicinal plants, which its pharmacological action was already reported through many experimental articles and oriental medical book: 『pharmacological action and application of anticancer traditional chinese medicine』 In summary: Used leukemia cellline are HL60, HL-60, Jurkat, Molt-4 of human, and P388, L-1210, L615, L-210, EL-4 of mouse. 31 kinds of anti leukemic medicinal plants are Panax ginseng C.A Mey; Polygonum cuspidatum Sieb. et Zucc; Daphne genkwa Sieb. et Zucc; Aloe ferox Mill; Phorboc diester; Tripterygium wilfordii Hook .f.; Lycoris radiata (L Her)Herb; Atractylodes macrocephala Koidz; Lilium brownii F.E. Brown Var; Paeonia suffruticosa Andr.; Angelica sinensis (Oliv.) Diels; Asparagus cochinensis (Lour. )Merr; Isatis tinctoria L.; Leonurus heterophyllus Sweet; Phytolacca acinosa Roxb.; Trichosanthes kirilowii Maxim; Dioscorea opposita Thumb; Schisandra chinensis (Rurcz. )Baill.; Auium Sativum L; Isatis tinctoria, L; Ligustisum Chvanxiong Hort; Glycyrrhiza uralensis Fisch; Euphorbia Kansui Liou; Polygala tenuifolia Willd; Evodia rutaecarpa (Juss.) Anti leukemic compounds, which were isolated from medicinal plants are ginsenoside Ro, ginsenoside Rh2, Emodin, Yuanhuacine, Aleemodin, phorbocdiester, Triptolide, Homolycorine, Atractylol, Colchicnamile, Paeonol, Aspargus polysaccharide A.B.C.D, Indirubin, Leonunrine, Acinosohic acid, Trichosanthin, Ge 132, Schizandrin, allicin, Indirubin, cmdiumlactone chuanxiongol, 18A glycyrrhetic acid, Kansuiphorin A 13 oxyingenol Kansuiphorin B. These investigation suggest that it may be very useful for developing more effective anti leukemic new dregs from medicinal plants.

Plant regeneration and soil acclimatization through photoautotrophic culture from leaf explant of a rare species in Sedum tosaense Makino (희귀수종인 주걱비름(Sedum tosaense Makino)의 잎절편으로부터 기내 식물체 재분화 및 광독립배양을 통한 토양순화)

  • The aim of this study was to establish plant regeneration from leaf explants of Sedum tosaense Makino, which is globally rare and endangered species. The leaf explants of S. tosaense were cultured on the MS medium supplemented with different concentration of BA and NAA for callus induction. Callus induction was showed the highest (100%) on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. The highest number of shoots were regenerated when callus were cultured on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA for 5 weeks. The axillary bud were cultured on the MS media supplemented with combination of BA and NAA for in vitro propagation. The highest number of adventitious shoot (7.9 per explants) formed at $1.0mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ BA. For rooting, MS medium supplemented with or without $2.0g{\cdot}L^{-1}$ activated charcoal was tested. The optimal results were observed using MS medium supplemented with $2.0g{\cdot}L^{-1}$ activated charcoal, on which 85.7 (No. of root), 4.6 cm (length of root). 1,200 ppm $CO_2$ and 350 ppm $CO_2$ were supplied for make certain the effects of $CO_2$ on pre-acclimatization by photoautotrophic culture. 1,200 ppm $CO_2$ treatment was established higher than 350 ppm $CO_2$ treatment. Soil acclimatization of in vitro plantlets was the best in mixture soil consisted of peat moss and perlite with 100% survival rate and they showed the maximum growth.
