Effect of Cytokinins on the Number of Cotyledons of Sometic Embryos from Immature Zygotic Embryo Culture of Heracleum moellendorffii Hance (어수리(Heracleum moellendorffii Hance.)의 미숙배로부터 형성된 체세포배의 자엽수에 미치는 Cytokinin의 영향)

  • In order to investigate the effect of cytokinins on the cotyledonary variability of somatic embryos in Heracleum moellendorffii Hance., somatic embryos were induced from the immature zygotic embryo on MS medium containing 1.0mg/l 2, 4-D, and then transferred to 2,4-D-free and cytokinin-added medium for somatic embryogenesis after 4weeks culture. Polycotyledonary embryos were formed most abundantly(84.9%) on the 0.2mg/l BA medium as compared with the 0.2mg/l 2ip(42.6%) and kinetin(32.9%) media, and it was proportional to BA concentrations(0.01~0.1mg/l). The rate of polycotyledonary embryo formation increased proportional to the period of BA treatment and also increased more at the globular stage than at the heart, torpedo-shaped and cotyledonary stages.

Water Relations Parameters of Heracleum moellendorffii Hance Obtained from Pressure-Volume Curves (P-V 곡선법을 활용한 어수리의 수분특성 분석)

  • This study was carried out to establish a proper cultivation site and to diagnose the drought tolerance of Heracleum moellendorffii leaves by using pressure-volume curves. As a result of analysing data measured, the leaf of H. moellendorffii showed the osmotic pressure at full turgor (Ψosat) was -1.0MPa, and that at incipient plasmolysis (Ψotlp) -1.2MPa. Then, the value of maximum bulk modulus of elasticity Emax was 28MPa, showing the sightly strong drought tolerance of H. moellendorffii. Furthermore, the values of relative water contents RWCtlp and RWC* were above 88%, showing that the function of osmoregulation is somewhat better. Thus, responses to water relations such as Ψosat, Ψotlp, Emax, RWCtlp and RWC* of H. moellendorffii showed it's slightly high drought tolerance property.

  • A furocoumarin derivative, psoralen (7H-furo[3,2-g][1]benzopyran-7-one), was isolated from the n-hexane fraction of Heracleum moellendorffii Hance. We examined the effects of psor-alen on a human Kv1.5 potassium channel (hKv1.5) cloned from human heart and stably expressed in Uk- cells. We found that psoralen inhibited the hKv1.5 current in a concentration-, use- and voltage-dependent manner with an IC$_{50}$ value of 180 $\pm$ 21 nM at +60 mV. Psoralen accelerated the inactivation kinetics of the hKv1.5 channel, and it slowed the deactivation kinetics of the hKv1.5 current resulting in a tail crossover phenomenon. These results indicate that psoralen acts on the hKv1.5 channel as an open channel blocker. Furthermore, psoralen prolonged the action potential duration of rat atrial muscles in a dose-dependent manner. Taken together, the present results strongly suggest that psoralen may be an ideal antiarrhythmic drug for atrial fibrillation.

Comparison of Photosynthetic Responses in Heracleum moellendorffii and Aruncus dioicus var. Kamtschaticus in Relation to Atmosphere-Leaf Vapor Pressure Deficit (대기-엽 수증기압차(VPD)에 의한 어수리와 눈개승마의 광합성 반응 비교)

  • This study was conducted to investigate the effect of atmosphere-leaf vapor pressure deficit (VPD) in Heracleum moellendorffii and Aruncus dioicus var. kamtschaticus. The VPD was rapidly increased with increasing temperature and decreasing relative humidity. Taken as a whole, the stomatal transpiration reaction was slightly late with increasing of VPD. Maximum photosynthetic rate at high-VPD condition was 6.49 µmol CO2·m-2·s-1 in Heracleum moellendorffii Hance, which was a little lower than 5.57 µmol CO2·m-2·s-1 in Aruncus dioicus var. kamtschaticus, respectively. After 2 p.m, stomatal transpiration of Heracleum moellendorffii at the high VPD condition was rapidly decreased. The results indicated that physiological activities in Heracleum moellendorffii are more limited from high VPD conditions.

Anti-oxidative Activity and Chemical Composition of Various Heracleum moellendorffii Hance Extracts (추출 조건에 따른 어수리의 항산화 활성 및 성분 분석)

  • Anti-oxidative activities of Heracleum moellendorffii Hance extracts were measured after extraction with 50% (v/v) ethanol or water (at $80^{\circ}C$ or $20^{\circ}C$). The total polyphenol content was highest (64.73 mg GAE/g) in extract from 50% ethanol extraction at $80^{\circ}C$. In extracts obtained by water extraction at $80^{\circ}C$ and $20^{\circ}C$, the total polyphenol contents were 39.78 mg GAE/g and 23.17 mg GAE/g, respectively. The antioxidant activities of the ethanol extract at $80^{\circ}C$ were highest, as assessed by DPPH radical scavenging activity (69.14% at 50 ppm), reducing power (OD 0.93), and $ABTS^+$ radical-scavenging activity (85.62% at 1,000 ppm). At $80^{\circ}C$, the antioxidant activity of the 50% (v/v) ethanol extract was 8-20% higher than that of the water extract. Also, the chemical composition of extract from ethanol extraction at $80^{\circ}C$ was analyzed. The levels of moisture, crude protein, crude fat, and crude ash were 6.38%, 4.35%, 0.67%, and 1.96%, (all w/w), respectively. The Na, K, Mg, Zn, Fe, Ca, and P contents of the extract were 53.41, 398.26, 5.80, 0.40, 5.27, 3.56, and 47.17 mg/100 g, respectively. The total levels of polyphenolic compounds and flavonoids in the ethanol extract were 64.73 mg GAE/g and 49.54 mg RE/g.
