The methanol (MeOH) extract and ist fractions of Taraxacum mongolicum (Compositae) were examined for their scavenging effects on 1,1-diphenyl-2- phenylhydrazyl (DPPH) and superoxide radicals, and hepatoprotective effects on tacrine-induced cytotoxicity in human hepatoma cell line, Hep G2 cells. Both methylene chloride $(CH_2Cl_2)$ and butanol (n-BuOH) soluble fractions of the MeOH extract showed the free radicals scavenging and hepatoprotective effects. From these results, it is suggested that hepatoprotective effect of these fractions partly relies on their free radical scavenging activity.
The hepatoprotective effect of methanol extract of leaves of Caesalpinia bonducella was studied by means of paracetamol induced liver damage in rats. The degree of protection was measured by using biochemical parameters such as serum transaminase (SGPT and SGOT), alkaline phosphatase (ALP), bilirubin, and total protein. Further, the effects of the extract on lipid peroxidation (LPO), glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) were estimated. The methanol extract of C. bonducella (MECB) (50,100 and 200 mg/kg) produced significant (P<0.01) hepatoprotective effect by decreasing the activity of serum enzymes, bilirubin, and lipid peroxidation, while it significantly increased increased the levels of GSH, SOD, CAT, and protein in a dose dependent manner. The effects of MECB were comparable to that of standard drug Silymarin. However, at a lower dose (25 mg/kg) it could not restore the deleterious effect produced by paracetamol. The results indicate that Caesalpinia bonducella had antioxidant and hepatoprotective effects.
Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of subfractions has been evaluated by analyzing blood and hepatocyte biochemical analyses and biotransformation enzyme analyses. Treatment of BS-5, subfraction has significantly decreased the activities of alanine aminotransferase and aspartate aminotransferase. In addition, the levels of cholesterol and triglyceride in liver have been decreased as compared with that of CCl4 treated rats. The hepatoprotective effect of BS-5, subfraction on the CCl4-induced hepatotoxicity would be mediated of the attenuation of the level of cytochrome P450 and the enhancement of the activity of glutathion S-transferase.
Objective: To examine the in vitro antioxidant and hepatoprotective properties of Amomum xanthoides extract and its fractions. Methods: Methanol extract of Amomum xanthoides was fractionated into three fractions; petroleum ether fraction (PFAX), water fraction (WFAX) and ethyl acetate fraction (AFAX). The antioxidant and hepatoprotective effects of these fractions were compared by in vitro experiments. Results: The total phenols content and DPPH (1 1-Diphenyl-2-picrylhydrazyl) radical scavenging activity was found significant in AFAX when compared with other fractions (PFAX, WFAX). AFAX significantly inhibited NO production by RAW 264.7 cells in LPS-activated condition. In Hep $G_2$ cells, AFAX markedly prevented the leakage of LDH and release of AST against CCl4 challenge. Conclusion: Amomum xanthoides and especially ethyl acetate fraction have potent antioxidative and hepatoprotective properties.
Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.
The present study was undertaken to investigate whether or not the hepatoprotective activity of acetylbergenin was superior to bergenin in carbon tetrachloride ($CCl_4$)-intoxicated rat. Acetylbergenin was synthesized by acetylating bergenin, which was isolated from Mallotus japonicus. The hepatoprotective effects of acetylbergenin were examined against $CCl_4$-induced liver damage in rats by means of serum and liver biochemical Indices. Acetylbergenin was administered orally once daily for 7 successive days, then a 0.5 ${m/kg}$ mixture of $CCl_4$in olive oil (1:1) was intraperitoneally injected at 12 h and 36 h after the final administration of acetylbergenin. Pretreatment with acetylbergenin reduced the elevated serum enzymatic activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and $\gamma$-glutamyltransferase in a dose dependent fashion. Acetylbergenin also prevented the elevation of hepatic malondialdehyde formation and depletion of glutathione content dose dependently in $CCl_4$-intoxicates rats. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored to almost normal levels. The results of this study strongly suggest that acetylbergenin n has potent hepatoprotective activity against $CCl_4$-induced hepatic damage in rats by glutathione-mediated detoxification as well as having free radical scavenging activity. In addition, acetylbergenin doses of 50 ${mg/kg}$showed almost the same levels of hepatoprotection activity as 100 ${mg/kg}$ of bergenin, indicating that lipophilic acetylbergenin is more active against the antihepatotoxic effects of $CCl_4$ than those of the much less lipophilic bergenin.
Methanolic extract of Cassia tora leaves was evaluated for its hepatoprotective activity in rats by inducing hepatotoxicity with paracetamol (acute model). In this experiments, we evaluated pathophysiologically the effect of NYBT on swimming time in mouse and hepatoprotective activity in rats intoxicated with carbon-tetrachloride. NYBT was nontoxic in orally acute toxicity test ($LD_{50}$, 320 ml/60 kg): a nontoxic food in more four times of one-shoot dosage (80 ml) to human. Weight-loaded forced swimming test was carried out to measure the swimming time of mice with a 4% load of body weight in plastic cylinder (diameter $10{\;}cm{\;}{\times}{\;}height{\;}20{\;}cm$) on water bath at $25^{\circ}C$, and the anti-fatigue activity represented the ratio of swimming time of experimental group to that of control group. NYBT had dose-dependent anti-fatigue activity Mice administered NYBT at a dose of 320 ml/60 kg once daily for 5 days could swim about two times more than control. Hepatoprotective activities of NYBT were examined by the determination of malonedialdehyde (MDA) and pathological survey in liver and liver function test of rat intoxicated with $CCl_4$ at i.m. dose of 2 ml/kg once daily for 7days. NYBT decreased dose-dependently thiobarbituric acid reactive substance: Oral administration of NYBT at a dose of 20 ml/60 kg was $38.51{\;}{\pm}{\;}3.02$ nmol MDA/g of tissue, that of 80 ml/60 kg was $33.76{\;}{\pm}{\;} 1.84$ nmol MDA/g of tissue, and that of 320 ml/60 kg was $32.87{\;}{\pm}{\;}1.90$ nmol MDA/g of tissue as compared with control group ($43.61{\;}{\pm}{\;}2.85$ nmol MDA/g of tissue). All rats administered NYBT at a dose of 320 ml/60 kg were survival as compared with 40% survival of control animals, and GPT activity of rats administered NYBT at a dose of 80 ml/60 kg was decreased as compared with control. In histopathological survey, NYBT improved slightly the fatty changes of hepatocytes around centrilobular area. These results suggest that NYBT has anti-fatigue and hepatoprotective activity in rats and mice.
The ethanolic extract of Bacopa monnieri Linn. aerial parts were studied far its hepatoprotective effect on $CCl_4-induced$ hepatotoxic rats. The extract was found to decrease significantly $CCl_4-induced$ elevation of SGOT, SGPT, ALP, bilirubin and total cholesterol. But, it increased HDL-cholesterol level and liver weight with respect to $CCl_4$ toxic rats. The extract was also found to decrease significantly the $CCl_4-induced$ elevation of lipid peroxidation and increase the activity of antioxidant enzymes (SOD and CAT) and GSH level in the liver of extract treated rats when compared with $CCl_4$ induced rats. Histopatholosical profiles showed that the extract had significant protective effect against $CCl_4-induced$ liver injury, which corroborates the above findings. Hence it may be possible that the mechanism of hepatoprotection of the extract is due to its antioxidant effect.

The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl4. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.
