The aim of this study was to investigate the in vivo toxicity of di-2-ethylhexyl phthalate (DEHP), on the immune system of the rock bream, Oplegnathus fasciatus. Fish were injected twice intraperitoneally with DEHP (200, 400, and 800 mg/kg BW), and the cellularity and functional activity of phagocytes in the spleen and head kidney were measured. The number of head kidney leukocyte cells was significantly greater in fish treated with 800-mg DEHP/kg BW. Nonspecific immunity, as determined by the phagocytic index, was significantly decreased at 800-mg DEHP/kg BW in the head kidney. A significant reduction in phagocytic capacity was observed in the head kidney at ${\geq}$400-mg DEHP/kg BW. Furthermore, significantly increased levels of serum glutamic oxaloacetate transaminase and glutamic pyruvate transaminase indicated a marked hepatic dysfunction in immunosuppressed fish. Total serum protein was significantly reduced at ${\geq}$400-mg DEHP/kg BW; however, there were no significant changes in lysozyme activity. These results demonstrate that immune responses in the rock bream, Oplegnathus fasciatus can predict immunotoxicity at doses ${\geq}$400-mg DEHP/kg BW.
Nitrate poisoning was observed in Holstein calves raising for beef purpose in a dairy-farm of Gyeongbug province. The results observed were summarized as follows: 1. These calves had been fed mainly cornstalk, and a qualitative reaction by "dephenylamine blue test" for nitrate was positive in the serum of the affected calves and in the cornstalk provided. 2. In the clinical signs, the affected calves were suddenly recumbent without premonition and then dyspnea, followed by death. Death also occurred after inappetence, depression, dyspnea, cyanosis of mucous membrane, and terminal anoxic convulsions were observed. 3. In the macroscopical findings, the blood was dark brown (chocolate in color) and more or less poor coagulative. Petechiae of epicardium, severe cloudy swelling of liver, cloudy swelling and hemorrhage of kidney, congestion and hemorrhage of spleen, congestion and edema of lung, and congestion af abomasum and small intestine were observed in these cases. 4. Microscopical changes observed in these cases were peripheral congestion of lobules, centrilobular necrosis and sinusoidal dilatation in liver, hydropic degeneration of hepatic cells, hemorrhage and edema of kidney, necrosis of convoluted-tubular epithelium, and dilatation of Bowman's space and convoluted tubule. There were also congestion and hemorrhage of spleen, hemorrhage and edema of lung, cloudy swelling of myocardium, and congestion and hemorrhage of various organs.

An example of lymphoblastic lymphosarcoma was diagnosed in a 7-year-old male brown bear (Ursus arctos) that died after having a 7-month history of depression, anorexia and watery diarrhea. Grossly the mesenteric lymph nodes were enlarged to approximately 4 to 6 times their normal size and histologically diagnosed as lymphoblastic lymphosarcoma. The small intestinal mucosa was corrugated and had severe mural thickening due to infiltrated neoplastic cells. Hepatic metastasis was also noted. the mother of this bear which died at the age of 20 also had similar gross changes on necropsy, but histological examination was not performed at that time and therefore the exact type of neoplasm remained to be determined. Since the daughter also died of neoplasm, genetic factor could be suspected in this family. This is the first reported case of lymphosarcoma in Ursidae in Korea. As an incidental finding, endogenous lipid pneumonia was noted in the lung.
Hemophagocytic syndrome(HPS) has four subgroup, sporadic disorder, associated with acute infection, familial form seen in children, and associated with malignant disorders, immunodeficiencies, defective leukocyte function. Histologically, Hemopoietic cells are actively ingested by moncytes/macrophages in various organs, including lymph nodes, bone marrow, liver, and spleen. Epstein-Barr virus(EBV) is now thought to be one of the major causes for the virus-associated hemophagocytic syndrome(VAHS). Epstein-Barr(EB) virus infection is common, with up to 90% of individuals demonstrating positive titiers by age 20. Although elevated liver function tests commonly occur, severe hepatitis is rare. Only seven cases of ascites complicating Epstein-Barr infection are reported, but none clearly demonstrate the abscence of other causes of hepatic dysfunction. We are reporting a case of Epstein-Barr Virus-associated hemophagocytic syndrome with ascites.
To determine the chemopreventive potential of alyssin and iberin, the in vitro anticancer activities and molecular targets of isothiocyanates (ITCs) were measured and compared to sulforaphane in hepatocellular carcinoma cell HepG2. The SR-FTIR spectra observed a similar pattern vis-a-vis the biomolecular alteration amongst the ITCs-treated cells suggesting a similar mode of action. All of the ITCs in this study cause cancer cell death through both apoptosis and necrosis in concentration dependent manner ($20-80{\mu}M$). We found no interactions of any of the ITCs studied with DNA. Notwithstanding, all of the ITCs studied increased intracellular reactive oxygen species (ROS) and suppressed tubulin polymerization, which led to cell-cycle arrest in the S and $G_2/M$ phase. Alyssin possessed the most potent anticancer ability; possibly due to its ability to increase intracellular ROS rather than tubulin depolymerization. Nevertheless, the structural influence of alkyl chain length on anticancer capabilities of ITCs remains inconclusive. The results of this study indicate an optional, potent ITC (viz., alyssin) because of its underlying mechanisms against hepatic cancer. As a consequence, further selection and development of effective chemotherapeutic ITCs is recommended.
cAMP responsive element-binding protein (CREB) is one of the most intensively studied phosphorylation-dependent transcription factors that provide evolutionarily conserved mechanisms of differential gene expression in vertebrates and invertebrates. Many cellular protein kinases that function downstream of distinct cell surface receptors are responsible for the activation of CREB. Upon functional dimerization of the activated CREB to cis-acting cAMP responsive elements within the promoters of target genes, it facilitates signal-dependent gene expression. From the discovery of CREB, which is ubiquitously expressed, it has been proven to be involved in a variety of cellular processes that include cell proliferation, adaptation, survival, differentiation, and physiology, through the control of target gene expression. In this review, we highlight the essential roles of CREB proteins in the nervous system, the immune system, cancer development, hepatic physiology, and cardiovascular function and further discuss a wide range of CREB-associated diseases and molecular mechanisms underlying the pathogenesis of these diseases.

To evaluate physiological effect of Aralia elata, in vitro antioxidant activity and hepatic protective effects were investigated. Ethyl acetate fraction from Aralia elata (EFAE) had higher total phenolic content than other fractions (n-hexane, chloroform, and distilled water layers). EFAE also showed significantly greater radical scavenging activity against 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH), than other fractions. Moreover, EFAE showed dose-dependent inhibitory effect of malondialdehyde (MDA). Hepatoprotective effects of EFAE against ethanol- and $H_2O_2$-induced oxidative stress and cytotoxicity in H4IIE and HepG2 hepatic cells were examined using 2',7'-dichlorofluorescein diacetate (DCF-DA) and 3-[4,5-dimethythiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay. The results showed that EFAE reduced cellular oxidative stress, and increased hepatic cell viability. In addition, EFAE inhibited ethanol-induced lipid accumulation in HepG2 cells. Finally, physiological substances of EFAE were analyzed using high performance liquid chromatography (HPLC), and the major bioactive compounds identified were 3,5-dicaffeoylquinic acid and chlorogenic acid.
The purpose of this study was to demonstrate the effect of Momordica Charantia L. water extracts, one of the natural chelator, on the biochemical and enzyme activity changes in the rats liver and kidney caused by lead acetate. Rat approximately 250 g in weight were grouped into the control, lead acetate treated, and the Momordica Charantia L. boiling water extracts treated after lead acetate groups. Lead acetate (1,000 ppm) and Momordica Charantia L. water extracts (5%, 10%) were delivered drinking water. Serum AST, ALT and BUN were measured, histological alteration of liver and kidney were examined by light microscopy. Momordica Charantia L. extract group was decreased serum AST, ALT and BUN level induced by lead. Optical observations of liver tissue, lead group were observed necrosis of hepatic cells and infiltration of inflammatory cells, but Momordica Charantia L. extract group was observed only slight infiltration of inflammatory cells around the central vein. Optical observations of kidney tissue, lead acetate induced atrophy and necrosis of glomerulus and infiltration of inflammatory cell around renal tubule. For the group treated with Momordica charantia L. extract, the glomerulus was similar to the control, some around the renal tubule was observed infiltration of inflammatory cells. In conclusion, Momordica Charantia L. water extract may protect the lead-induced toxicity on liver and kidney.
Autosomal recessive polycystic kidney disease (ARPKD) is one of the important genetic disorders in pediatric practice. Mutation of the polycystic kidney and hepatic disease gene 1 (PKHD1) was identified as the cause of ARPKD. The gene encodes a 67-exon transcript for a large protein of 4074 amino acids termed fibrocystin, but its function remains unknown. The neoplastic-like in cystic epithelial proliferation and the epidermal growth factor/epidermal growth factor receptor (EGF/EGFR) axis overactivity are known as the most important characteristics of ARPKD. Since the misregulation of $Ca^{2+}$ signaling may lead to aberrant structure and function of the collecting ducts in kidney of rat with ARPKD, present study aimed to investigate the further mechanisms of abnormal proliferation of cystic cells by inhibition of PKHD1 expression. For this, a stable PKHD1-silenced HEK-293T cell line was established. Then cell proliferation rates, intracellular $Ca^{2+}$ concentration and extracellular signal-regulated kinase 1/2 (ERK1/2) activity were assessed after treatment with EGF, a calcium channel blocker and agonist, verapamil and Bay K8644. It was found that PKHD1-silenced HEK-293T cell lines were hyperproliferative to EGF stimulation. Also PKHD1-silencing lowered the intracellular $Ca^{2+}$ and caused EGF-induced ERK1/2 overactivation in the cells. An increase of intracellular $Ca^{2+}$ in PKHD1-silenced cells repressed the EGF-dependent ERK1/2 activation and the hyperproliferative response to EGF stimulation. Thus, inhibition of PKHD1 can cause EGF-induced excessive proliferation through decreasing intracellular $Ca^{2+}$ resulting in EGF-induced ERK1/2 activation. Our results suggest that the loss of fibrocystin may lead to abnormal proliferation in kidney epithelial cells and cyst formation in ARPKD by modulation of intracellular $Ca^{2+}$.

Protaetia Brevitarsis Seulensis(white grub) has been traditionally used as medicinal stuff to treat blood stasis, occlusion of menstruation, tetanus and liver cancer in Asian countries (Korea, Japan, China, Taiwan, India and Myanmar). Especially, Donguibogam, which is traditional korean medicinal book, described the white grub as traditional medicine to treat hepatic diseases and vascular disorders. The white grub has been considered as highly nutritional food. The major constituents of white grub are rich in protein, healthy fats, iron, calcium. Recent studies announced that white grub has hepatoprotective effect and anti-microbacterial effect. However, the immuno-enhancing effect of white grub extracts in RAW 264.7 macrophage cells has not been studied yet. In this study, the various concentrations of white grub extract were examined to find immuno-enhancing effects on RAW 264.7 cells. Cytotoxicity was determined by MTT assay and immuno-enhancing effect of white grub extract was investigated by measuring nitric oxide (NO) production compared with only lipopolysaccharide (LPS) treatment. White grub extracts (0.001 - 10 mg/ml) did not show cytotoxicity. Additionally, white grub extracts (0.001 - 1mg/ml) had Immuno-enhancing effect on RAW 264.7 cells compared with only LPS treated group. These results might be provided proof to develop beneficial immuno-enhancing material for human health.
