An assessment of responses to egg production and liver health of Japanese quails subjected to different levels of metabolizable energy

  • Objective: Current quail production is configured as an economic activity in scale. Advancements in quail nutrition have been limited to areas such as breeding and, automation of facilities and ambience. The objective of this study was to evaluate the performance responses, liver and oviduct morphometry, and liver histology of Japanese laying quails subjected to different levels of nitrogen-corrected apparent metabolizable energy (MEn). Methods: A completely random design was used that consisted of nine levels of MEn, six replicates, and five hens per cage with a total of 270 quails. The experimental period lasted for 10 weeks. The variables of performance were subjected to analysis of variance and then regression analysis using the broken-line model. The morphometric and histological variables were subjected to multivariate exploratory techniques. Results: The MEn levels influenced the responses to zootechnical performance. The broken-line model estimated the maximum responses for feed intake, egg production, egg weight, and egg mass as 3,040, 2,820, 1,802, and 2,960 kcal of MEn per kg of diet, respectively. Multivariate analysis revealed that the occurrence of hepatic steatosis and increased levels of Kupffer cells were not related to MEn levels. Conclusion: The level of 2,960 kcal/kg of MEn meets performance variable requirements without compromising hepatic physiology.

CD38 Inhibition Protects Fructose-Induced Toxicity in Primary Hepatocytes

  • A fructose-enriched diet is thought to contribute to hepatic injury in developing non-alcoholic steatohepatitis (NASH). However, the cellular mechanism of fructose-induced hepatic damage remains poorly understood. This study aimed to determine whether fructose induces cell death in primary hepatocytes, and if so, to establish the underlying cellular mechanisms. Our results revealed that treatment with high fructose concentrations for 48 h induced mitochondria-mediated apoptotic death in mouse primary hepatocytes (MPHs). Endoplasmic reticulum stress responses were involved in fructose-induced death as the levels of phosho-eIF2α, phospho-C-Jun-N-terminal kinase (JNK), and C/EBP homologous protein (CHOP) increased, and a chemical chaperone tauroursodeoxycholic acid (TUDCA) prevented cell death. The impaired oxidation metabolism of fatty acids was also possibly involved in the fructose-induced toxicity as treatment with an AMP-activated kinase (AMPK) activator and a PPAR-α agonist significantly protected against fructose-induced death, while carnitine palmitoyl transferase I inhibitor exacerbated the toxicity. However, uric acid-mediated toxicity was not involved in fructose-induced death as uric acid was not toxic to MPHs, and the inhibition of xanthine oxidase (a key enzyme in uric acid synthesis) did not affect cell death. On the other hand, treatment with inhibitors of the nicotinamide adenine dinucleotide (NAD)+-consuming enzyme CD38 or CD38 gene knockdown significantly protected against fructose-induced toxicity in MPHs, and fructose treatment increased CD38 levels. These data suggest that CD38 upregulation plays a role in hepatic injury in the fructose-enriched diet-mediated NASH. Thus, CD38 inhibition may be a promising therapeutic strategy to prevent fructose-enriched diet-mediated NASH.

  • 평활근종은 평활근세포에서 발생할 수 있는 흔한 양성종양으로, 주로 자궁에서 발생한다. 복막평활근종은 매우 드물며 대부분 복막성 평활근증으로 보고되었다. 저자들이 아는한 단일 복막평활근종의 영상의학적 소견은 영문으로 아직까지 보고된 바가 없다. 전산화단층촬영에서 비균질한 조영증강, 자기공명영상에서 점진적 조영증강 및 확산제한을 보여 영상의학적으로 간의 변성된 혈관종, 간선종, 혹은 염증성 근섬유아세포종 등을 감별하였으나 수술 후 병리학적으로 복막평활근종으로 진단된 34세 여성의 증례 및 영상의학적 소견을 보고하고자 한다.

Hepatoprotective Effect of Lactiplantibacillus plantarum DSR330 in Mice with High Fat Diet-Induced Nonalcoholic Fatty Liver Disease

  • Lactiplantibacillus plantarum DSR330 (DSR330) has been examined for its antimicrobials production and probiotics. In this study, the hepatoprotective effects of DSR330 were examined against nonalcoholic fatty liver disease (NAFLD) in a high-fat diet (HFD)-fed C57BL/6 mouse model. To induce the development of fatty liver, a HFD was administered for five weeks, and then silymarin (positive control) or DSR330 (108 or 109 CFU/day) was administered along with the HFD for seven weeks. DSR330 significantly decreased body weight and altered serum and hepatic lipid profiles, including a reduction in triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels compared to those in the HFD group. DSR330 significantly alleviated HFD-related hepatic injury by inducing morphological changes and reducing the levels of biomarkers, including AST, ALT, and ALP. Additionally, DSR330 alleviated the expression of SREBP-1c, ACC1, FAS, ACO, PPARα, and CPT-1 in liver cells. Insulin and leptin levels were decreased by DSR330 compared to those observed in the HFD group. However, adiponectin levels were increased, similar to those observed in the ND group. These results demonstrate that L. plantarum DSR330 inhibited HFD-induced hepatic steatosis in mice with NAFLD by modulating various signaling pathways. Hence, the use of probiotics can lead to hepatoprotective effects.

사람의 제대 및 양막유래 줄기세포의 간세포로의 분화 (Hepatogenic Potential of Umbilical Cord Derived-Stem Cells and Human Amnion Derived-Stem Cells)

  • 목 적: 사람의 제대유래 줄기세포 (HUC)와 양막유래 줄기세포 (HAM)를 간유사세포로 분화 시키고자 하였다. 연구방법: 산모의 동의아래 만삭 정상 산모로부터 양막 및 제대를 얻고 줄기세포를 분리, 배양하였다. 분리된 세포의 줄기세포로서의 특성을 규명하기 위하여 RT-PCR, 세포면역화학 분석 그리고 중배엽성세포로의 분화능 실험을 하였다. 또한 이들 줄기세포를 분화 배양액에서 간유사세포로 분화 유도 후, 간세포 특이 유전자 발현, 알부민 ELISA, 알부민에 대한 면역블로팅 및 세포면역화학염색 그리고 PAS 염색을 시행하였다. 결 과: 사람의 양막 및 제대로부터 세포를 분리하여 줄기세포로서의 특성을 조사하고, 이들 세포의 지방세포, 연골세포, 골아세포로의 분화능력을 확인하였다. 동일한 분화 배양액에서 HUC과 HAM을 간유사세포로 분화시킨 결과 HUC이 모든 조건에서 HAM보다 알부민과 요소를 더 많이 합성 분비하였다. 간유사세포로의 분화능력이 더 좋은 것으로 나타난 HUC을 가지고 최적의 분화를 유도한 결과 $1.2{\pm}0.8\;{\mu}g/mL$의 알부민을 합성 분비하였고, $8.9{\pm}1.2\;mg/dL$의 요소를 합성 분비하였다. 결 론: HUC과 HAM 모두 알부민을 분비하는 간유사세포로 분화할 수 있었으며, HUC이 더 잘 유도되는 것으로 관찰되었다. 이러한 결과로써 HUC과 HAM을 간질환 환자의 간세포 이식 치료에 필요한 세포치료제로써 이용할 수 있을 것으로 생각된다.

Anti-metastatic Potential of Ethanol Extract of Saussurea involucrata against Hepatic Cancer in vitro

  • The rates of morbidity and mortality of hepatocellular carcinoma (HCC) have not lessened because of difficulty in treating tumor metastasis. Mongolian Saussurea involucrata (SIE) possesses various anticancer activities, including apoptosis and cell cycle arrest. However, detailed effects and molecular mechanisms of SIE on metastasis are unclear. Thus, the present study was undertaken to investigate antimetastatic effects on HCC cells as well as possible mechanisms. Effects of SIE on the growth, adhesion, migration, aggregation and invasion of the SK-Hep1 human HCC cell line were investigated. SIE inhibited cell growth of metastatic cells in dose- and time-dependent manners. Incubation of SK-Hep1 cells with $200-400{\mu}g/mL$ of SIE significantly inhibited cell adhesion to gelatin-coated substrate. In the migration (wound healing) and aggregation assays, SIE treated cells showed lower levels than untreated cells. Invasion assays revealed that SIE treatment inhibited cell invasion capacity of HCC cells substantially. Quantitative real time PCR showed inhibitory effects of SIE on MMP-2/-9 and MT1-MMP mRNA levels, and stimulatory effects on TIMP-1, an inhibitor of MMPs. The present study not only demonstrated that invasion and motility of cancer cells were inhibited by SIE, but also indicated that such effects were likely associated with the decrease in MMP-2/-9 expression of SK-Hep1 cells. From these results, it was suggested that SIE could be used as potential anti-tumor agent.

간흡충에 감염된 흰쥐 담관 상피세포의 증식 양상 (Turnover of biliaiy epithelial cells in Clonorchis sinensis infected rats)

  • 간흡충에 감염된 담관은 확장되면서 상피세포의 수가 크게 증식하여 점막층이 두꺼워지고 과식증에 의해 점막의 단면이 유두상으로 관찰된다. Bromodeoxyuridine (BrdU)이 세포가 핵산을 복제할 때에 끼어 들어가는 성질을 이용하여, 이 연구에서는 담관 상피세포 중에서 합성기(Sphase)에 있는 세포의 분율을 파악하고자 하였다. 흰쥐에 간흡충의 피낭유충을 100 개씩 경구 감염시키고, 15주까지 간의 각 엽에 있는 담관의 조직절편을 만들어서 다세포군항체와 ABC complex를 이용한 염색을 실시하였다. 그 결과 충제가 들어 있는 담관에서는 감염 1주에 상피 세포의 2.9-10.2%가 염색에 양성이었고, 2주에 7.3-12.8%가, 5주에는 7.3-13.4%가, 15주에 8.4-14.8%가 BrdU를 함유하였다. 대조군 흰쥐나 실험군 동물의 감염되지 않은 담관의 경우에는 0-2.7%가 염색에 양성으로 나타났다. 염색된 상피세포는 주로 점막 상피세포층의 기저부에서 관찰되었다. 이 결과에 의하면 간흡충과 직접 접촉하는 담관의 상피세포가 뚜렷하게 증식하고 있어 충체의 어떠한 물리적인 또는 화학적인 자극이 있더라도 가까운 부위에서 주로 작용하고 있다고 판단하였다.

  • The study was designated to investigate the electron microscopic findings following diethylnitrosamine (DEN) treatment in rats. Forty four male (Srague Dawley) rats were continuously given water containing 0.01% DEN for 13 weeks and livers of five rats with more tumor lesions at 16 and 17 weeks after initial treatment were used as EM materials. In transmission electron microscopic findings, most small-sized hepatocytes were active cells containing large mount of organelles, but light (pale staining) hepatocytes among small-sized hepatocytes were injured cells containg disorganized organelles. Tumor cells among small-sized hepatocytes were irregularly arranged and have pleomorphic nuclei containing electron dense chromatin but the organelles in cytoplasm were swelled. Large-sized hepatocytes were active cells with condensed chromatin but the cytoplasm of these cells were pale due to be injured and dilated organelles. Dark hepatocytes were apoptotic cells with homogenous pyknotic nuclei and cytoplasm, and the cytoplasm of these cells contained dilated smooth endoplasmic reticulum (sER) but these sER were non-vesiculated. Cholangiocarninoma cells were crowded and were pale by far less number of organelles in cytoplasm and nuclei. In scanning electron microscopic findings, the lumens of portal veins, bile canaliculi, bile ductules, bile ducts and sinusoids were dilated and have irregular folded inner surface by protruded parenchyma.

사염화탄소의 장기(長期) 투여(投與)로 유발된 백서(白鼠) 간경변(肝硬變)에 대한 형방지황탕(荊防地黃湯)의 조직병리학적 영향 (The Histopathology of Hyeungbangjihwang-tang Extracts on the Liver Cirrhosis of Fisher 344 Rats Induced by Long Time Carbon Tetrachloride Injection.)

  • Objective : This study was carried out to investigate the effects of oral administration of Hyeungbangjihwang-tang (HBJHT) on the liver cirrhosis of rats induced by Carbon tetrachloride for 10 weeks. Method : The histopathological changes were observed. The HBJHT extracts were daily dosed at 50, 100 and 200mg/kg for 12 weeks. Results : Severe hepatocellular necrosis and ballooning, hyperplasia of connective tissue, subdued reduction of hepatic lobule, and hyperplasia of bile duct in portal triad were dramatically decreased in the HBJHT-treated group compared to that of the Carbon tetrachloride-treated control group in histopathological observation. The diameter of hepatic lobules was significantly enlarged in the HBJHT-treated group compared to that of the Carbon tetrachloride-treated control group, and the amounts of connective tissue, degenerative cells and bile ducts were significantly and dose-dependently decreased. Conclusion : It is concluded that HBJHT has a significant recovering effect on the liver of rats induced by carbon tetrachloride.

Acetaminophen에 의해 손상된 마우스 간세포에서 합마유의 간세포보호 효과 (Hepatoprotective Effect of Forest frog's oviduct oil on Acetaminophen-induced Liver Injury in Mice.)

  • Objectives : The purpose of this study is that the protective effects of habmayou on acetaminophen (AP)-induced hepatotoxicity were investigated in mice. Methods : Before administering AP mice supplied with only water were left alone for 18 hours. after concentration and dissolution in poly ethylglycol AP 400mg per 1kg of mouse weight, we injected AP titrated density with a physiological saline solution into the abdominal cavity of mouse to induce hepatotoxicity. we researched mortality rate and the shape of liver tissue of mouse. Results : Treatment with habmayou (250 mg/kg, p.o.) 0.5 h after AP administration significantly prevented an increase in plasma alanine aminotransferase and aspartate aminotransferase activities and AP-induced hepatic necrosis, and also reduced AP-induced mortality from 46% to 0%. In addition, oral treatment with habmayou significantly prevented AP-induced depletion of glutathione (GSH) contents. However, habmayou treatment, by itself, did not affect hepatic GSH contents. Conclusion : These results show that the hepatoprotective effects of habmayou against AP overdose may be due to its ability to block the bioactivation of AP by regeneration of hepatonecrotic cells.
