A study was conducted to investigate the effects of the combined stressor induced by high stocking density with feed restriction on immunological parameters such as leukocyte differential counts and cytokine expression in laying hens. A total of forty White Leghorn laying hens were randomly allotted into the control (12 kg of body weight/$m^2$) and the stress (44 kg of body weight/$m^2$) groups, and then birds of the stress group were given 75% of voluntary intake of the control birds for 12-d on a daily basis. There was a significant decrease in body weight without affecting the relative weights of the liver and spleen after 12-d of the combined stressor. In hematological values, no significant difference in leukocyte differential counts including heterophils (H), lymphocytes (L), monocytes and H:L ratio was observed between the two groups. In cytokines, hepatic lipopolysaccharide-induced tumor necrosis factor-${\alpha}$ (LITNF-${\alpha}$) and inducible nitric oxide synthase (iNOS) expression levels in the stress group were significantly (p<0.05) higher compared with those in the control group. However, the expression levels of interleukin (IL)-4 and IL-6 in the liver were not affected by the combined stressor. Splenic LITNF-${\alpha}$ expression in the combined stressor group was significantly (p<0.05) up-regulated compared with that in the control birds. However, the combined stressor did not affect splenic IL-4, IL-6 and iNOS expression. In conclusion, the combined stressor caused by high stocking density with feed restriction enhanced some pro-inflammatory cytokines including LITNF-${\alpha}$ and iNOS in lymphoid and non-lymphoid organs of birds, suggesting that altered cytokine expression to given stressors can be another parameter that can be used in assessing stress responses of birds.

The purpose of this study was to evaluate the effects of gobonyangjeonbang (GYB) on the endocrine function and the antioxidant efficacy of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced oxidative stress in rats. In 2017, to evaluate the efficacy of GYB on oxidative stress, 35 male SD rats were divided into five groups and tested. The normal control group was administered saline as a vehicle, while the TCDD-alone group was administered TCDD (2 ㎍/kg per week) intraperitoneally and with physiological saline, and the test group was administered GYB orally by dividing it into three concentrations (75, 150, and 300 mg/kg) for six weeks. Bodyweight decreased significantly after six weeks of TCDD exposure, when compared to rats in the NC group (p<0.001). However, weight loss from TCDD was significantly protected by administration of GYB at 300 mg/kg (p<0.01). The rat liver induced by TCDD showed cytoplasmic vacuole degeneration, and the hepatic sinusoid and weight increased. As a result of measuring MDA and SOD, both items tended to decrease under TCDD administration. On the other hand, there was no change due to GYB administration, and significance was observed in the GYB 300 mg/kg group compared to the NC group in the SOD result (p<0.05). These findings demonstrated that GYB may have a protective effect against TCDD-induced liver toxicity in rats.
This study was to compare the effects of dietary n-6 and n-3 fatty acids and fat unsaturation on plasma lipids and chemical composition of VLDL and LDL fraction and lipogenic enzymes activity in rat liver under the conditions providing 1) a similar amount of n-6, n-3 fatty acids(LA, ALA, EPA+DHA) in diets and 2) the various degree of fat unsaturation. Male Sprague-Dawley rats weighing 420g were treated for 6-n with six experimental diets providing 25% of energy as fat and which were different only in fatty acid composition. The fats used for a source of each fatty acid were beet tallow for saturated fatty acid corn oil for n-6 linoleic acid(LA) perilla oil for n-3 $\alpha$-linolenic acid(ALA) and fish oil n-3 eicosapentaenoic acid (EPA) and n-3 docosahexaenoic acid(DHA). Plasma cholesterol level was increased by corn oil to compare with beef tallow but was decreased by perilla oil or fish oil. Plasma TG level was significantly decreased by perilla oil or fish oil. Fish oil significantly reduced the level of HDL-Chol and the proportion of Chol in LDL fraction and that of TG in vVLDL fraction. Overall there was a singificant negative correlation between the level of each plasma lipid(Chol TG, VLDL-TG, LDL-C) and the degree of fat unsaturation. However this rerlationship is not always true when compared the hypolipidemic effect of each fatty acid at a similar level of fat unsaturation. There was a trend such taht glucose 6-P dehydrogenase 6-phosphogluconate dehydrogenase and malic enzyme activites were reduced by n-3 fatty acids. Perilla oil significantly increased the incorporation of c20:5 and c22:5 into liver tissue and fish oil suignificantly increased the incorporation of c20:5, c22:6 into liver tissue and the effect of long chain n-3 fatty acid incorporation was greater by fish oil. therefore the hypotriglyceridemic effect of n-3 fatty acid could be resulted from the interference of hepatic lipogenesis by long-chain n-3 fatty acids and the reduced proportion of TG in VLDL fraction and its effect was greater by n-3 EPA+DHA than n-3 ALA even though plasma Chol and TG levels were also influenced by the degree of dietary fat unsaturation.

The study was conducted to investigate the effect of Saengshik supplementation on lipid metabolism in rats fed high cholesterol diets. Male Spraque-Dawley rats were administrated 1% cholesterol to induce hypercholesterolemia and were fed on diet containing Saengshik (30%, w/w). Serum and liver lipid profiles and fecal bile acids excretion were examined after 7 weeks of experimental diet. The feeding of diet containing 30% Saengshik significantly decreased total cholesterol (TC) contents in liver and plasma. Since cholesterol was balanced by entero-hepatic circulation through bile acid synthesis and reabsorption, inhibition of bile acid re-uptake in intestine decreases total cholesterol in liver and blood. In this point, significant increament of fecal bile acid excretion in Shaengshik group decreased TC and improved hypercholesterolemia. Also plasma high density lipoprotein-cholesterol (HDL-C) decreasing risk for coronary heart disease in Saengshik supplemented group was significantly higher than control group, whereas low-density lipoprotein-cholesterol (LDL-C) accumulation in arterial vessel wall was significantly lower than control group. In this result, we supposed that supplementation of Saengshik, uncooked natural food, may improve hypercholesterolemia through increment of cholesterol excretion.

Soybean sauce fermented with soybean and wheat, has been a major condiment of Korean diets from centuries ago. Melanoidin, a brown pigment generally found in various food systems, is a final product produced in amino-carbonyl reaction during soybean sauce processing. Antioxidative activities of soybean sauce and melanoidin were investigated in vitro system using linoleic acid emulsion. Soybean sauce and glucose-lysine model melanoidin showed the stronger antioxidative effect than control by ferric thiocyanate and conjugated diene assays. In addition, DPPH radical scavenging effect of soybean sauce was higher than melanoidin, which was ascribed to soluble peptide and low molecular protein existing in soybean sauce. To ascertain antioxidative effect of dietary soybean sauce and melanoidin in vivo, the male Wister rats were fed 10% soybean sauce or 10% glucose-lysine model melanoidin with corn oil or fish oil for 5 weeks. Fatty acid compositions in liver and plasma were influenced by oil source. Therefore, EPA and DHA contents of fish oil group were higher than those of corn oil group. When the inhibitory effect of soybean sauce and melanoidin on lipid peroxidation using TBARS methods was measured, fish oil group (FC) showed higher malondialdehyde (MDA) content than corn oil group (CC). However, supplementation of soybean sauce and melanoidin to fish oil group attenuated MDA formation. In the levels of phosphatidyl choline hydroperoxide (PCOOH) in liver and plasma by CL (chemiluminescence)-HPLC method, PCOOH in FC group was significantly higher than that of CC group both in liver and plasma. Supplementation of soybean sauce to fish oil groups significantly inhibited the formation of PCOOH in plasma and liver, while melanoidin suppressed hepatic PCOOH formation. Based on these results, it can be suggested that soybean sauce possesses stronger antioxidative potential than melanoidin.

This study was designed to establish the effects of Zingiber mioga, Zingiber mioga root and Zingiber ofiicinale on the lipid concentrations in hyperlipidemic rats. A total of 24 male Sprague-Dawley rats weighing 60$\pm$5g were fed one of four experimental diets for 4 weeks: a control diet (hyperlipidemic diet), diets supplemented with 3% of Z. mioga, 3% Z. mioga roots or 3% Z. officinale. The concentration of total cholesterol in serum was significantly decreased by 31% in Z. mioga group compared to the control group. The HDL-cholesterol concentration was significantly higher in Z. mioga and Z. mioga root supplemented groups than in control group. Triglyceride concentration in serum and liver significantly decreased with 3% Zingiber supplemented groups compared with control group, but had no significant differences among three Zingiber supplemented groups. The contents of total cholesterol triglyceride free cholesterol and cholesteryl ester in liver were remarkably lower in Zingiber supplemented groups than in control group. However, hepatic phospholipid level was not showed significant effect.

The effects of cheonggukjang (traditional fermented soybean food, CK) and cheonggukjang added Phellinus linteus (CKP) on lipid metabolism were investigated in adult male rats. Twenty weeks old Sprague-Dawley rats were fed a purified hyperlipidemic diet (control diet: 0.5% cholesterol, 10% fat, 18% casein) for 4 weeks. The rats were randomly assigned to each treatment group control, two kinds of CK or CKP (powders of CK or CKP as dietary protein sources) After 5 weeks of CK or CKP diets consumption, the body weights, the hepatic and epididymal fat pad weights of the CK or CKP diets groups were significantly decreased than those of the control group. The liver cholesterol and triglyceride levels were significantly lower in both the CK and CKP diets groups than those in the control group. The concentrations in serum triglyceride, total cholesterol, LDL-cholesterol, and atherogenic index ratios were significantly decreased in the CK and CKP diets groups compared with those in the control group. The HDL-cholesterol/total-cholesterol ratios were significantly increased in the CK and CKP diets groups compared with those in the control group. Fecal cholesterol and triglyceride excretion of the CK and CKP diets groups were significantly increased than those of the control group. These results showed that both the feeding of cheonggukjang and cheonggukjang added Pheilinus linteus decreased the triglyceride, total cholesterol and LDL-cholesterol in serum as well as the triglyceride and cholesterol in liver, and increased the HDL-cholesterol/total-cholesterol ratio in serum of the rats.

This study was designed to evaluate lipid component of serum and organ in matured rats fed oil extract from ducks. Male Sprague-Dawley rats weighing 595g(52-week-old) were divided into 3 groups and fed experimental diets for 8 weeks. Rats were fed soybean oil(control group) diet, duck oil(DO group) diet and duck oil supplemented with 1% cholesterol(DOC group) diet. Serum triglyceride concentration was significantly lower in DO and DOC groups than that in the control group. Serum total cholesterol concentration was significantly higher in DOC group than those in the other groups, while serum HDL-cholesterol concentration was significantly lower in DOC group than those in the other groups. Hepatic total lipid, triglyceride and total cholesterol concentrations in DOC group were significantly higher than those in the other groups. These studies indicate that the duck oil decreases the concentrations of triglyceride and shows almost same concentrations of total cholesterol compared to the soybean oil supplemented group in serum and liver. But hypercholesterolemic effect of duck oil was not improved.

This study was carried out to investigate the antidiabetic, alcohol metabolism, anti-inflammatory, and hepatoprotective effects of Acer tegmentosum extracts (ATE). A. tegmentosum has been traditionally used as a folk medicine to treat hepatic disorders. The antioxidative activities of ATE were measured by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide (SOD) assay. DPPH radical scavenging and SOD activities of ATE were about 89% and 82.9% at $0.5{\mu}g/mL$, respectively. Alcohol dehydrogenase and acetaldehyde dehydrogenase activities were 118.0% and 177% at 2 mg/mL, respectively. ${\alpha}-Glucosidase$ inhibitory activity of ATE was 75% higher at $50{\mu}g/mL$ and remarkably increased in a dose-dependent manner. Nitric oxide productions in macrophage RAW 264.7 cells stimulated by lipopolysaccharide was reduced to 16.7% by addition of ATE at 1 mg/mL. ATE showed significant protective effects against tacrine-induced cytotoxicity in Hep G2 cells at $100{\mu}g/mL$. Based on our results, we conclude that ATE may be used as a major pharmacological agent and anti-diabetic, anti-hepatitis, and anti-inflammatory remedy.

The study aimed to investigate the anti-obesity effects of 1% Rhus verniciflua vinegar (RV) supplementation in high-fat-diet (60% fat)-induced obese rats. A total of 50 4-wk-old male Sprague-Dawley rats were fed normal chow diet or maintained on high-fat diet (HFD) for 12 weeks to induce obesity and were then randomized into five groups as follows: normal diet+ultra-pure water (CON), HFD+ultra-pure water (OB-DW), HFD+1% acetic acid (OBAA), HFD+1% RV (OB-RV), and HFD+0.1% caffeine (OB-CF). AA was used as a control for RV, and caffeine was used as a positive control due to its weight reducing effect. After 2 months, body weight, organ and adipose tissue weights, serum lipids, hepatic lipids, adipocyte size, and cell number per spot level were analyzed. As a result, food efficiency ratio, abdominal adipose tissue weight, serum levels of total cholesterol, triacylglycerol, free fatty acids, coronary artery index, and fecal lipid were significantly reduced in the RV treatment group. In this study, we found that dietary RV improved obesity by increasing lipid excretion and reducing lipogenesis. These results suggest that RV has potential as a functional anti-obesity food.
