  • The present study was done to determine the effect of trolox C. a hydrophilic analogue of vitamin E, on hepatic injury, especially alteration in vasoregulatory gene expression during ischemia and reperfusion. Rats were subjected to 60 min of hepatic ischemia in vivo. Rats were treated intravenously with trolox C (2.5 mg/kg) or vehicle (PBS, pH 7.4), 5 min before reperfusion. (omitted)

  • The activation of the hepatic stellate cell (HSC) is a key step in liver fibrogenesis. We investigated the changes of global gene expression during activation in hepatic stellate cells using a rat cDNA microarray with 5, 000 sequence-verified clones. We identified osteopontin (OPN), a secreted matrix protein, as one of the upregulated factors. Northern analysis showed OPN mRNA was increasingly expressed during progressive activation of cultured rat HSCs and in models of experimental liver fibrosis. (omitted)

  • Objective : This study was performed to investigate the activity of Whagan-Jeon (huaganjian) in protection against acetaminophen (AAP)-induced hepatotoxicity and the possible mechanisms in vivo. Methods : The following were performed : Serum ALT, depletion of hepatic glutathione (GSH) levels, the microsomal p. nitrophenol hydroxylation activity, microsomal aniline hydroxylation activity, genomic DNA fragmentation and its reversal, hepatic glutathione-S-transferase (GST) activity, and hepatic NAD(P)H:quinone oxidoreductase (QR) activity Results : Whagan-Jeon (huaganjian) protected against AAP-inducedhepatotoxicity by the increase of GSH levels, inhibition of P450 2E1-specific metabolic activities, attenuation of hepatic DNA damage, and induction of GST and QR activities in vivo. Conclusions : In conclusion, Whagan-Jeon (huaganjian) was effective in protection against AAP-induced hepatoxicity.

Primary culture of adult rat hepatocytes and assay of hepatic functions (쥐 간세포의 일차배양과 분화기능 측정)

  • Rat hepatocytes were isolated by collagenase perfusion method and cultured on the collagen coated dish or on the floating collagen membrane. Using the primary cu1tured hepatocytes, the efficiency of cell attachment and the hepatic functions such as gluconeogenesis, ureogenesis and albumin synthesis were studied. The cell viability was kept above 50% until 5 days and the hepatic functions of ammonia metabolism and albumin synthesis were maintained until 7 days. Floating collagen membrane was found to be more efficient than the collagen coated dish for the maintenance of hepatic function in-vitro.

  • Clinical signs, serum chemical values and histological findings of hepatic tissue after partial and complete obstruction of common bile duct in Korean goats were investigated. Abnormal clinical signs were not observed in partial obstruction of common bile duct, but in complete obstruction clinical signs such as jaundice, urine color change, were observed. Serum total bilirubin, total cholesterol, aspartate aminotransferase, sorbitol dehydrogenase, gamma glutamyltranspeptidase, and total protein values increased on the 1-4th day and then gradually decreased to normal level in partial obstruction. However, they tend to increase persistently by the 24th day in complete obstruction of common bileduct. Histologic features of hepatic tissue in partial obstruction were not changed as compared with normal hepatic tissue. On the other hand, in complete obstruction of common bite duct there were moderate bile duct proliferation in a portal area, rupture of bile canaliculi, phagocytosis of bile pigment by Kupffer cells, periportal fibrosis, intrahepatic bile stasis and hepatic cell necrosis.

The Effect of Vitamin C on Hypoxia/reoxygenation Induced Hepatic Injury in Isolated Perfused Rat Liver (흰쥐 관류간 모델에서 저산소 및 산소재도입시 vitamin C가 간장기능에 미치는 영향)

  • This study was done to investigate the effect of vitamin C on hypoxia/reoxygenation-induced hepatic injury ul isolated perfused rat liver. Isolated livers from rats fasted 18 hours were subjected to 45 min of hypoxia followed by reoxygenation for 45 min. The perfusion medium used was Krebs-Henseleit bicarbonate buffer (pH 7.4) and 0.5 mmol/L of vitamin C was added to the perfusate. Alanine aminotransferase (ALI) and lactate dehydrogenase (LDH) levels were significantly increased by hypoxia/reoxygenation. These increases were augmented by vitamin C. Glucose output and bile flow were markedly decreased by hypoxia/reoxygenation. Vitamin C aggavated the decrease of glucose output but had little effect on bile flow. Our findings suggest that hypoxia/reoxygenation diminishes hepatic metabolic and secretory functions, and vitamin C significantly aggravates these changes.

  • A castrated male, 4-year old, Dachshund weighing 5.3 kg with an acute history of anorexia, vomiting, severe depression, and hypothermia for 1 day was referred. Severe leukopenia, mild increased hepatic enzymes, and mild imbalanced electrolyte were found on laboratory tests. There are no remarkable findings on abdominal radiographs. Ultrasonography showed multifocal heterogeneous and hypoechoic round regions in the liver. Ultrasound guided fine needle aspiration and drainage were performed and hepatic abscess was diagnosed. The patient was recovered normally without any complications through aggressive medical treatments from the result of culture and sensitivity test.

  • Synchronous primary cancers in the liver and gallbladder have been rarely reported. We report a case of synchronous cancers of hepatic angiosarcoma and gallbladder adenocarcinoma, mimicking gallbladder cancer with hepatic invasion. Additionally, the clinical implications, the radiologic features, and the diagnostic difficulties are further discussed.

Effect of famotidine on propranolol elimination in the isolated perfused rat liver (Famotidine이 propranolol 대사에 미치는 작용)

  • The Ha-antagonist, cimetidine, has been shown to retard the hepatic elimination of low and high clearance drugs, and this has been attributed to inhibition of microsomal cytochrome P-450. This study was done to determine the effects of low (50$\mu\textrm{g}$) and high (1mg) dose of famotidine, another histamine H$_2$-receptor antagonist, on hepatic elimination of propranolol compared with cimetidine in the isolated perfused rat liver. Both low and high dose of cimetidine not only inhibited the elimination of propranolol but also increased the area under the perfusate propranolol concentration time curve (AUC). In contrast, low and high dose of famotidine did not affect hepatic elimination of propranolol. Our findings suggest that famotidine has not a propensity for hepatic microsomal inhibition.

  • The objective of the present study was to develop an equivalent circuit model of glucose kinetics including the hepatic glucose balance functions which were neglected in the previous compartmental models. Using this circuit model, the insulin resistivity parameter and hepatic glucose sensitivity parameter were estimated in optimal fitting of the model based data of glucose and insulin concentration to the reported clinical intravenous glucose tolerance test(IVGTT) data in normal and diabetic subjects. The addition of the hepatic function in the model has improved the overall performance of the simulation. Also, the computed tissue insulin resistivity and the hepatic glucose sensitivity are shown to be significant in distinguishin four clinical groups of normal and diabetic groups.
