Aerial Parts and Roots of Pulsatilla koreana Affect the Viability of HSC-T6 Hepatic Stellate Cells

  • During liver fibrosis, hepatic stellate cells (HSCs) undergo a complex activation process characterized by increased proliferation and extracellular matrix deposition, which is the major pathological feature of hepatic cirrhosis. Therefore, suppression of HSCs activation has been proposed as therapeutic strategies for hepatic fibrosis. We tried to screen the antifibrotic activity of natural products employing HSC-T6, hepatic stellate cell lines as an in vitro assay system. In the present study, we investigated the antiproliferative activity of aerial parts and roots of Pulsatilla koreana Nakai (Ranunculaceae). Our present study shows that roots of P. koreana exerted more strong inhibitory activity compared to its aerial parts. In addition, among the fractions of the aqueous methanolic extract of P. koreana roots, both n-hexane and $CHCl_3$ fraction showed the strong inhibitory activity on HSC proliferation. Further study also demonstrated that the n-hexane and $CHCl_3$ fraction of P. koreana roots significantly inhibited the HSC proliferation in time- and concentration-related manners.

  • This study was done to determine that vitamins I and C are synergistic in protecting liver cells during hepatic ischemia and repefusion. Rats treated with vitamins I and C were subjected to 60 min of hepatic ischemia and to 1 and 5 hr of reperfusion thereafter. Serum aminotransferase level and microsomal lipid peroxidation were markedly increased by ischemia/reperfusion. These increases were significantly attenuated by vitamins E, C or its combination. Hepatic wet weight-to-dry weight ratio was increased in ischemic group, but this increase was prevented by combination of vitamin I and C. Bile flow and cholate output were markedly decreased by ischemia/reperfusion and vitamin C alone and combination of vitamin I and C restored their secretion. Cytochrome P-450 content and aminopyrine N-demethylase activity were decreased by ischemia/ reperfusion and restored by vitamin C and combination of vitamin I and C to the level of sham-operated rat. Aniline p-hydroxylase activity was increased by ischemia/reperfusion and this increase was prevented by vitamin E. Our findings suggest that ischemia/reperfusion diminishes hepatic secretory and microsomal functions by increasing lipid peroxidation and vitamins I and C synergistically ameliorates these changes.

  • Hepatic hemangioma is the most common benign liver tumor and must be considered in the differential diagnosis of other space occupying hepatic masses. A 54-year-old man was referred to evaluate bone metastases of lung adenocarcinoma. In our case, we thought that a focal hepatic uptake in the bore scan was a metastatic lesion, because of underlying lung adenocarcinoma. However, the findings of abdominal CT and Tc-99m RBC scan results were deemed to be characteristic of hepatic hemangioma. The biopsy of the lesion was not performed.

  • The response of hepatic mixed function oxygenase (MFO) system was investigated in olive flounder exposed to formalin. Hepatic microsome of olive flounder incubated in vitro with formalin demonstrated the induction of cytochrome P450 (CYP), ethoxyresorufin deethylase (EROD), cytochrome P450 reductase (P450R) and cytochrome b5 reductase (b5R) activity. In addition, olive flounder was exposed to 100, 300 and 500 ppm of formalin for 1 h and then transferred to a flow-through type of 1000 L aquarium. Hepatic MFO enzyme activity was determined for 72 h. As the result, hepatic CYP, P450R and EROD activities increased following exposure of formalin, but b5R and GST showed no significant change. These results imply that CYP and P450R can be considered as main hepatic enzymes involving in detoxification of formalin.

  • Majority of hepatocellular carcinoma is evolved from a well differentiated cancerous condition such as hyperplastic lesions eg. adenomatous hyperplasia in cirrhotic liver or de no vo carcinogenesis and prolifenation along with dedifferentiation. Adenomatous hyperplasia is may be seen in severe acute hepatic injury, like submassive hepatic necrosis, or in chronic liver diseases, particularly liver cirrhosis and it has recently attracted much interest from both clinicians and pathologists because it is regarded as a precursor lesion of hepatocellular carcinoma. Hepatic adenomatous hyperplasia resembling focal nodular hyperplasia might have developed from localized vascular changes associated with chronic liver disease, pre-existing arterial malformation and early stage of angiogenesis in hepatocarcinogenesis. We present a patient who developed hepatocellular carcinoma after hepatic artery ligation.

Diagnostic importance of Ultrasound-Guided Fine Needle Aspiration in Diagnosing Hepatic Lesions among Sudanese Patients 2015

  • Background: Liver cytology is indicated and requested for evaluating hepatic masses in symptomatic or serendipitous cryptic discovered lesions. Objective: To determine the cytomorphological patterns of hepatic lesions identified among a group of Sudanese patients. Materials and Methods: This is an analytical descriptive hospital-based study included 165 patients undergoing ultrasound-guided fine needle is an aspiration cytology (FNAC)for hepatic lesions, at Al-Amal Military Hospital & Khartoum Teaching Hospital in Khartoum, Sudan. Clinical data were reviewed. Air dried Diff Quick stained smears were grouped into unsatisfactory samples, benign lesions, and malignant neoplasms. Results: Our population were consisted of 35 (21.2%) females and 130 (78.8%) males, with a male to female ratio 3.7:1 and an age ranged between 47 to 80, and a mean age $57{\pm}7$. Of 165 cases, 57 (34.5%) were benign, no atypia were noticed, 101 (61.2%) were malignant. Most investigated patients were found to have metastatic lesions. Conclusion: FNAC is a useful tool for investigating hepatic lesions.

  • Acute betaine treatment induces time-dependent changes in the hepatic glutathione (GSH), cysteine and S-adenosylmethionine (SAM) levels. Our previous study demonstrated that betaine administered $1{\sim}4$ hours prior to sacrifice decreased hepatic GSH levels, but these levels were increased when measured 24 hours following the treatment. The present study was aimed to determine dose-dependent effects of betaine on hepatic metabolism of sulfur amino acid in mice. Mice were sacrificed 2.5 or 24 hours after intraperitoneal treatment with betaine at different dose levels ranging from 50 to 1000 mg/kg. The concentrations of methionine and SAM were increased by a betaine dose of 100 mg/kg, and the concentrations of GSH and cysteine were decreased by a betaine dose of 200 mg/kg at 2.5 hours. These changes were augmented with increasing doses of betaine. At 24 hours following betaine treatment, increased GSH and decreased taurine levels were observed from dose levels of 400 mg/kg. Changes in hepatic activities of cystathionine beta-synthase, gammaglutamylcysteine ligase and cysteine dioxygenase were observed from dose levels of $200{\sim}400$ mg/kg of betaine administered 24 hours prior to sacrifice.

  • Objectives : This experiment was conducted to evaluate inhibitory effects against hepatic metastasis and promotion of immunocytes by cultivated wild ginseng Herbal Acupuncture. Methods : Colon26-L5 carcinoma cells were injected through hepatic portal vein to induce hepatic metastatic cancer. Changes in weight, morphology of the cancer, histological impressions were evaluated and cytokine level was analyzed to yield immunological changes. Mice treated with cultivated wild ginseng Herbal Acupuncture reduced metastatic size compared to the control group. 2. No distinctive differences were witnessed between the cancer cells of control and experimental group in histological observation, but experimental group was closer to the normal tissue condition. 3. Observing immunocytes from the spleen of experimental group, T-lymphocytes were significantly increased. 4. Measuring the level of cytokine IL-4 which stimulates Th 2 were significantly increased. These findings strong1y indicate cultivated wild ginseng Herbal Acupuncture enhances immunity to inhibit the growth of cancer and metastasis.

Inhibitory Effect of Rutaecarpine on Thioacetamide (TAA)-induced Hepatic Fibrosis

  • Rutaecarpine is one of the major alkaloids present in the fruits of Evodia rutaecarpa. In this study, rutaecarpine was evaluated, both in vitro and in vivo, for its hepatoprotective properties against thioacetamide (TAA)-induced hepatic fibrosis. The results showed that rutaecarpine inhibited TAA-induced cytotoxicity, reduced the expression of the fibrogenic cytokine transforming growth factor ${\beta}1$ ($TGF-{\beta}1$), and induced the expression of bcl-2. To evaluate its in vivo effects, animal models with TAA-induced hepatic fibrosis were utilized. Levels of liver tissue injury-associated enzymes, including alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were monitored. $TGF-{\beta}1$ and the ${\alpha}$-smooth muscle actin (${\alpha}$-SMA) were measured as markers of the protective effects on hepatic fibrosis. The AST and ALT levels in blood were greatly enhanced by TAA and completely blunted by rutaecarpine. Rutaecarpine led to the down-regulation of $TGF-{\beta}$ and Bax mRNA expression, as well as the up-regulation of Bcl-2 and $Bcl-X_L$ mRNA levels. In conclusion, rutaecarpine inhibited TAA-induced hepatic fibrosis and apoptosis by inducing the expression of Bcl-2 while blocking $TGF-{\beta}1$ in our TAA-intoxicated model.

Computed tomography diagnosis of secondary hepatic lipidosis in an awake cat with alimentary lymphoma

  • An 8-year-old castrated male Korean short hair cat was referred with chronic vomiting, diarrhea, and anorexia. The cat was diagnosed with alimentary lymphoma via fine needle aspiration. Three weeks later, he was presented again with anorexia. Hepatomegaly and hyperechoic change in the liver parenchyma were observed. Computed tomography while awake was performed and revealed hypoattenuation of the hepatic parenchyma. The cat was tentatively diagnosed with feline hepatic lipidosis secondary to alimentary lymphoma. In the feline patient with lymphoma, hepatomegaly is important to differentiate hepatic lipidosis from hepatic infiltration of lymphoma on liver. In the present case, fatty infiltration of the liver was identified via non-invasive, rapid, and convenient computed tomography examination in an awake cat with a lymphoma.
