Analysis of Transcriptional Activity and Estrogen Responsiveness of Regulatory Elements in Chicken Ovalbumin Promoter (닭 오브알부민 프로모터의 길이에 따른 유전자 발현 활성 및 에스트로겐 반응성 분석)

  • Chickens have been considered as well-defined animal bioreactor. The optimized ovalbumin promoter is essential for recombinant protein production in transgenic chicken. Here we try to compare the activity and identify the effect of estrogen on ovalbumin promoter according to each promoter length with estrogen response element (ERE) existence. We cloned two (2.8 and 5.5 kb) ovalbumin promoters that the 5.5 kb contained the ERE but the 2.8 kb did not, and these two promoters were cloned to pGL4.11 vector. Additionally, we constructed another pGL4.11 vector containing of the 4.4 kb (with ERE) ovalbumin promoter deleted with 1 kb between ERE region and the 2.8 kb promoter. For reporter assay, HeLa, MES-SA, LMH/2A, and cEF cells were transfected with all the pGL4.11 vectors. The comparative analysis showed that the mutated 4.4 kb promoter has more potent activity than the 2.8 and 5.5 kb promoters in HeLa, MES-SA, and LMH/2A cells. However, there is no significant difference in cEFs. Also, these cells transfected with the mutated 4.4 kb promoter were treated with the $17{\beta}$-estradiol (0~3,000 nM) and HeLa, MES-SA, and LMH/2A cells showed estrogen responsibilities, but cEFs did not. Besides, the mutated 4.4 kb promoter has still higher activity than the 2.8 and 5.5 kb promoter, and there is no transcriptional induction effect in 2.8 kb promoter at 500 nM estrogen that is blood concentration of laying hens. Hence our study strongly suggested that the mutated 4.4 kb promoter is considered as one of the most efficient length for generating transgenic chicken.

Production Performance of 12 Korean Domestic Chicken Varieties Preserved as National Genetic Resources (국가 보존 유전자원 한국토종닭 12종의 생산능력 고찰)

  • In this study, viability, growth performance and egg production performance were investigated to determine the productive characteristics of 12 Korean domestic chicken varieties which have been collected and conserved for over 25 years in National Institute of Animal Science, Rural Development Administration, Korea. The 1,134 hens were tested and their production traits including survival rate, body weight, age at first egg laying, hen-day and hen-housed egg production, egg weight, and egg quality were measured. Survival rate was the highest in Korean Rhode-D and Korean Native Chicken (KNC) White and the lowest in Korean Cornish Brown (92.2% and 54.3%, respectively). Body weights from 4 to 50 wks were consistently high in the order of Korean Cornish, Korean Rhode, KNC, Korean Ogye, and Korean Leghorn breeds. There was no significant difference in weight between varieties within a breed. Age at first egg laying was the shortest in Korean Leghorn, while it was the longest in Korean Cornish. The hen-day egg production from $1^{st}$ egg to 57 wks was the highest in Korean Leghorn-K, and the lowest in Korean Cornish Brown. Egg weight was the heaviest in Korean Leghorn-F and the lightest in KNC White. The Haugh unit was the highest in Korean Rhode-C and the lowest in Korean Ogye. Taken together, these results suggest that it is desirable that the Korean Cornish variety is improved as the Korean meat breed because of its excellent growth ability, the Korean Leghorn variety is improved as the Korean layer breed because of its good laying performance, and the Korean Rhode and KNC varieties are improved as strong viable breeds because of their good survival rate.

Comparison of Particulate Matter and Ammonia Emission in Different Types of Laying Hen Poultry Houses during Spring (봄철 산란계사 사육형태별 미세먼지 및 암모니아 농도 비교)

  • This study was conducted to determine the concentrations of particulate matter (PM) and NH3 emissions from different types of laying hens poultry houses during spring. The concentrations of PM and NH3 were measured three times (2-week intervals; March to May) in Floor-pen-, Aviary-, and Cage-type poultry houses. Overall, PM10 and PM2.5 concentrations were found to be low from 22:00 to 04:00. The PM10 and PM2.5 concentrations in Floor-pen and Cage houses were similar with no significant daily deviation. NH3 concentrations measured over 24 h at the center and end of Floor-pen house were relatively constant. Irrespective of measurement location, NH3 concentrations were the lowest in Floor-pen house. Moreover, NH3 concentrations were higher at the end of Floor-pen and Aviary houses than that at the center; however, lower concentrations of NH3 were detected at the end of Cage house. The concentrations of PM10 and PM2.5 around the poultry houses were 57.5 and 34.0 ㎍/m3, respectively, with the daily average PM10 and PM2.5 concentrations (4,730 and 447.7 ㎍/m3, respective) being the highest in Aviary house. The concentrations of NH3 at the center and end of Cage house were the highest at 12.0 and 9.31 ppm, respectively. Furthermore, in Cage house, the emission factor of NH3 was the lowest, whereas there was no significant difference on that of NH3. In conclusion, among the three types of poultry houses assessed, PM (PM10, PM2.5) concentrations were higher in Aviary house, whereas NH3 concentrations were higher in Cage house.

The Effect of Dietary Metabolic Energy Level of 'Woorimatdag' Breeder on Performance, Egg Quality, Fertility and Hatchability, and Chick's Weight (사료 내 ME 수준이 '우리맛닭' 종계의 생산성, 종란품질, 수정률 및 부화율, 병아리 체중에 미치는 영향)

  • This study was conducted to investigate the effects of the metabolic energy levels of Korean native chicken breeder diets on laying performance, egg quality, fertility and hatchability. On the basis of dietary metabolic energy values (2,800, 2,700, and 2,650 kcal/kg), 168 twenty-week-old 'Woorimatdag' breeders (maternal line) were divided into three treatment groups (four replications per treatment with each replicate containing 14 birds) and reared until 32 weeks of age. We found that the body weight of hens increased with age, although there were no significant differences among treatments with respect to body weight or egg weight. Egg production ratio at 20-24 and 24-28 weeks of age tended to be low in birds fed the 2,700 and 2,650 kcal/kg diets, although the differences among treatments were none significant. Feed intakes was significantly decreased as the metabolic energy level reduced (P<0.05). There were no significant differences among treatments with respect to feed conversion ratio or the strength and thickness of eggshell. Similarly, we detected no significant differences among the three metabolic energy levels with regards to fertility, hatchability, or chick weight. In conclusion, our findings indicate that during the initial phase of laying, the metabolic energy level of 'Woorimatdag' breeder diets could be lowered to 2,700 kcal/kg without significantly influencing the laying performance of breeders or egg quality, fertility and hatchability, and chick weight.

Comparison of the Growth Performance of 12 Crossbred Korean Native Chickens and Commercial Layer from Hatch to 16 Weeks (12개의 토종닭 교배조합과 실용 산란계의 육성기 성장능력 비교)

  • The current study was conducted to compare the effect of crossbred on the body weight and laying performance of Korean native chicken from hatch to week 40. A total of 873 one-day-old chicks were divided into twelve crossbreds (i.e., CFCK, CFYC, CFYD, CKCF, CKYC, CKYD, YCYD, YCCF, YCCK, YDCF, YDCK, and YDYC) and commercial layer (Hy-Line Brown) were obtained as a counterpart in the study. All the birds are raised in battery cages (76 × 61 × 46 cm3) and then raised until 14 weeks and subsequently moved to layer battery cages (60 × 25 × 45 cm3) and raised until 16 weeks. The body weight and viability were measured biweekly from hatching to week 16. The week of 16, body weight range was about 1,010.24 to 1,411.77 g. The body weight of specific crossbreeds (i.e., CKCF, YCYD, and YDYC) was found to be comparable to that of Hy-Line Brown). The viability hatch to week 14 range was about 55 to 100% and however week 14 to 16 range was 80 to 100%. The crossbred (i.e., CKCF) recorded superior (P<0.05) viability throughout the whole experiment period, even compared with Hy-Line Brown (100% vs 96%). Our results are indicating that crossbreds Korean native chicken including CKCF, and YDYC has the potential to enhance key features of laying hens during the growing phase like optimal body weight and higher viability.
