  • The influence of fluorescence, scattering, and absorbance in turbid material by light scattering was interpreted by the scattered fluorescence intensity and wavelength. The effect of optical property in scattering media was investigated. It is very important to study the charge coupled device(CCD) in spectrometry because we can use the molecular energy level, molecular structure, absorption or emission, intermolecular reaction, weakly bound molecular energy, photochemistry, fluorescence and photodynamic therapy. CCD is very essential to study the molecular structure and medical engineering combined laser spectroscopy in the modem physical and chemistry. Accordingly, this study has designed and manufactured the electromagnetic spectrometry with CCD, and has analyzed the hematoporphyrin derivative.

Toxicity of Hematoporphyrin-Coated Magnetic Ferrofluid in Rats

  • The purpose of this study was to investigate the toxicity of hematoporphyrin-coated magnetic ferrofluid (HP-MF) through intravenous administration in Sprague-Dawley rats. Each group was treated with either saline, or the HP-MF at 0.5, 1, 1.5, 2 and 4 ml/kg body weight (b.w.) for the observation of survival rate, clinical symptoms, laboratory values and histopathological findings. In this study, HP-MF was evaluated for the survival rates, symptoms, laboratory values and histopathological examination after treatments. The result revealed that the animals in the group of HP-MF at 2 and 4 ml/kg b.w. showed some lethality. In serum biochemistry, the levels of AST, ALT and ALP were increased in the MF and HP-MF treated groups. However, histopathological examination for the suspected organs showed no evidence of hepatotoxicity and nephrotoxicity of typical iron poisoning. Though the toxicity of HP-MF was higher than that of HP, long retention of hematoporphyrin via HP-MF provides additional benefit over conventional hematoporphyrin. HP-MF could be utilized as a potential photodynamic agent in cancer therapy. It is suggested to develop an efficient external magnetic device to attract hematoporphyrin in the target site, thereby enabling to administering a small amount of HP-MF.

  • The picosecond time resolved fluorescence spectra of Hematoporphyrin Derivative (HPD) in both solutions and cancer cell are measured by a time correlated single photon counting system with a synchronously mode locked dye laser. Two exoponential decay components in the fluorescence spectra were observed. The slow decay(6.3 ㎱)and the fast one(350 ㎰)are attributed to be originated from monomers and dimers, respectively. The absorption and fluorescence measurements in steady state also showed the presence of a monomeric and dimeric forms of HPD molecules. The monomer lifetime in the cancer cell was measured to be longer than that in solution, which was expected from the blue shift and narrowing of the absorption spectra for HPD-treated in vitro. The relative amplitude of the fast component was found to be enhanced in cancer cell, strongly indicating the higher affinity of the dimer for the cancer cell.

  • Occurrence of an electron (or H atom equivalent to one electron plus H+) transfer from $\alpha$- tocopherol $\alpha$(TOH) to a number of photosensitizers in theri triplet states were investigated by monitoring the ESR signal of $\alpha$-chromaoxyl radical ($\alpha$(TO.) in ethanolic solutions of $\alpha$TOH and the sensitizers under continuous illumination. Every sensitizer molecule examined, such as protocholorophyllide (Pchl), hematoporphyrin and rose bengal which are generally regarded as efficient type II photosensitizers and thus have long-lived triplet states, was found to actively participate in an electro transfer reaction with $\alpha$TOH even under air-saturation conditions, generating $\alpha$TOH complex as an intermediate in a fashion of Michaelis-Menten type of reaction. For the reaction of $\alpha$TOH with triplet Pchl, the rate law was derived by applying the steady approximation for the binary complex, triplet Pchl-$\alpha$TOH , which turned out to be well consistent with the kinetic data.

  • Mononitrotoluenes were reduced to aminotoluenes using porphyrin as a catalyst in the presence of several types of reductants including hydrogen sulfide and 1, 4-dithiothreitol(DTT). Intermediates and final products of porphyrin-catalyzed reduction of mononitrotoluenes were identified and a pathway for the reduction of the nitro group to the corresponding amino group was proposed. The optimum pH for the reduction was determined. The catalytic activity of the porphyrin was confirmed by UV/VIS absorption spectra and basic kinetics of porphyrin-catalyzed reduction were studied. Of several types of reductants tested, DTT sodium hydrosulfite, and hydrogen sulfide were seen to give significant reduction of nitrobodies. When hydrogen sulfide was used as a reductant hydrogen sulfide and nitrotoluenes were removed simultaneously.

  • PDT-mediated cyototoxicity basically depends on the penetrated light-dose into the tumor tissue. This limits the efficiency of PDT to the superficial tumor region typically less than 1 cm. The localized photochemical generation of reactive oxygen species, including singlet oxygen is known to increase expression of assortment of early response genes including heat shock protein. In order to increase PDT cytotoxicity in the treatment of solid tumor, it is desirable to combine PDT with other therapeutic effects. In this preliminary study we evaluated enhanced cytotoxicity from the PDT-mediated expression of thymidine kinase in a transfected tumor cell line. Two types of photo sensitizers, a hematoporphyrin derivative(Photogem, Russia) and aluminium sulphonated phthalocyanine(Photosense, Russia) were used to evaluate the overexpression of hsp-70 in PDT-treated cell. Transient increase of hsp-70 was observed at 6-8 hrs later following irradiation in the photosense-treated cell whereas it was not observed in Photogem-treated cell. In the presence of ganciclovia, transfected cell showed a 17% increase in the cytotoxicity compared to the PDT only cell.

  • To minimize the use of animals in toxicity testing, and to reduce the cost in vivo test, more rational test method was described which determines, in the same animal, photoxic and photoallergic potential of a substance, and is daptable to routine testing. The other purpose of this study was to investigate the usefulness of in vivo alternatives ; photostability and spectrophotometric carbonyl assay. In this modified photosensitization model, animal numbers and resting periods, the number and method of topical application were simplified. Two positive photoreactive agents, Benzocaine and 6-methyl coumarine, showed a similar photoallergic potential to that of Ichikawa's method. Two sunscreens, Octyl methoxy cinnamate, Butyl methoxyl dibenzoyl methane, hardly showed photoallergic potentials. The photostability test could be used in the step of prescreening of photosensitization potential because most of the photoreactive agents represented the reduction of more than 20% in the absorbance. And photoreactive agents have a high potential of photosensitization in the sddessment of spectrophotometric carbonyl level although two sunscreens have a low possibility of photosensitization. Therefore this method was assumed as a valuable in vivo alternatives in the respect even in the very low concentrations which phototoxicity test using almonella showed no phototoxic potential.

Preparation of Photosensitizer-Coated Ferrofluids and Fabrication of a Device for Photodynamic Therapy (광감제가 코팅된 자성유체의 제조와 광역학 치료용 장치의 구성)

  • For the purpose of annihilating tumor in body, hematoporphyrin as a photosensitizer was coated onto magnetic particles of $Fe_3O_4$ prepared by coprecipitation which could be concentrated around the tumor by magnetic field. The photosensitizer was applied differently before, during and after adsorbing the 1st surfactant on the particles. Its added amount was $5{\times}10^{-4}/mol$, and the coating reaction proceeded at temperatures of 60, 70 and 8$0^{\circ}C$. The amounts of photosensitizer coated on the magnetic particles were obtained by calculating an optical density with the maximum UV spectrum. As a result of the UV analysis, the coating amount of photosensitizer increased with higher reaction temperatures. When applied at 8$0^{\circ}C$ after adsorbing the 1st surfactant, the photosensitizer was coated with a maximum value of $3.8{\times}10^{-3}/mo1/$\ell$$. The TGA analysis revealed that the ferrofluids included the particles of 30.115 g/$\ell$. It was suggested that the magnetite particles was coated with photosensitizer of $1.26{\times}10^{-4}/mo1/g$. A small-sized device for magnetic field and light emission was designed, in which LED sheets coverts the permanent magnet of Nd-Fe-B. The LED sheet was connected in series circuit and also protected with a silicon tube. The power was supplied with rechargable battery of 9V and 100-120mA.
