Immunomodulatory effects of silymarin after subacute exposure to mice: A tiered approach immunotoxicity screening

  • Silymarin is a flavonoid complex extracted from the Silybum marianum plant with a wide range of pharmacological and biochemical effects. In the present study, the immunomodulatory effects of silymarin were investigated in BALB/c mice. Silymarin was administered daily by intraperitoneal injection at doses of 50, 100 and 150 mg/kg for 14 consecutive days. Following the exposure, host hematological parameters, spleen cellularity and histopathological examination, as well as delayed-type hypersensitivity (DTH) responses, hemagglutination titers (HA), splenocyte cytokine production and lymphocyte proliferation assay were studied in all of the test groups of animals. The results showed that the low dose of silymarin (50 mg/kg) could stimulate both cellular and humoral immune functions in the treated hosts. In addition, silymarin at 100 mg/kg appeared to impact on DTH responses and lymphoproliferation. Based on the finding here, it would seem that silymarin has efficient immunostimulant properties. As a recommendation, the application of silymarin along with acupuncture technique (herbal acupuncture) can be thought as a good plan to modulate and enhance the immune system for the management of several immunodeficiency disorders. However, further studies are required to demonstrate this hypothesis.

Study of the immunosuppressive activity of methanolic extract of Madhuca longifolia (Koenig)

  • The immunosuppressive activity of the Methanol extract of bark of Madhuca longifolia (Koenig) consisting of a mixture of saponins, flavonoids, tannins, steroids, phenol and glycosides was studied on the immune responses in mice. Methanol extract of Madhuca longifolia (MLL) was administered orally at doses of 50, 100 and 150 mg/kg/day to healthy mice divided into four groups consisting of six animals each. The assessment of immunomodulatory activity was carried out by testing the humoral (antibody titre) and cellular (foot pad swelling) immune responses to the antigenic challenge by sheep RBCs. Furthermore, the effect on hematological parameters as well as relative organ weight was determined. On oral administration MML showed a significant decrease delayed type hypersensitivity (DTH) response whereas the humoral response to sheep RBCs was unaffected. Thus MLL significantly suppressed the cellular immunity by decreasing the footpad thickness response to sheep RBCs in sensitized mice. With a dose of 100 and 150 mg/kg/day the DTH response was $7.66{\pm}2.75$ and $6.41{\pm}1.21$ respectively in comparison to corresponding value of $14.50{\pm}2.38$ for untreated control group. These differences in DTH response were statistically significant (P < 0.05). The study demonstrates that MLL shows preferential suppression of the components of cell-mediated immunity and shows no effect on the humoral immunity.

Vanadium Yeast의 독성저감 효과 (Toxic Reduction Effect of Vanadium Yeast)

  • Vanadium has been known as environmental polluants resulted from the burning of fossil fuels in nature. It led to toxic responses by prooxidant activity, inducing free radicals and the accumulation in the tissues. Recently, there has been growing interest in an essential nutritional requirement of vandium and especially the treatment of diabetes. But because of its strong toxicity, thease chemicals have narrow safety margin. In order to reduce metal toxicity, and increase absorption and biological activities, metal ions such as selenium and chromium were uptaken in yeast cells. In this study, Vanadium yeast was prepared by uptaking vanadate in yeast cells. Vanadate induced hematological and biochemical changes in the experimental rat blood were inhibited by the treatments of vanadium yeast. Lipid peroxidation and catalase activity were significantly increased in kidney and liver after a single intraperitoneal injection of vanadate to rats. However, these observations were apparently reduced in the vanadium yeast treated group. Vanadium amount in blood, kidney and liver after a single intraperitoneal injection of vanadium yeast was significantly reduced than that of vanadate treated group. In conclusion, vanadium yeast uptaken vanadate in yeast cells could reduce toxic effects of vanadate.

기니픽그에서의 실험적 trichothecene (T-2) 독소중독증 (Experimental Trichothecene (T-2) Toxicosis in Guinea pigs)

  • Guinea pigs were administrated with T-2 toxin at a rate of 1 and 0.6mg/kg body weight per day for 21 days to study the immunological and pathological effects of T-2 toxin in guinea pigs. Blood was collected before T-2 treatment and on days 7, 14 and 21 of the trial for hematological and biological examinations and for the mitogen assay using lymphocytes. Myeloid: erythroid ratios were examined from the fernur bone marrow samples taken a day before T-2 toxin treatment began, on day 12 and at death. Guinea pigs received with 1mg/kg body weight of T-2 toxin daily showed leukopenic, lymphopenic and anemic signs on day 7 and 14. The mitogenic responses to the T-cell mitogen, Concanavalin A and B-cell mitogens, lipopolysaccharide were significantly depressed on day 7. Histologically, marked cellular damages including karyorrhexis and depletion of lymphocytes were observed in the actively dividing cells of the gastrointestinal tract, lymph node, spleen and bone marrow of guinea pigs.

Multi-Target Cytotoxic Actions of Flavonoids in Blood Cancer Cells

  • To date, cytotoxic effects of flavonoids in various cancer cells are well accepted. However, the intracellular signaling cascades triggered by these natural compounds remain largely unknown and elusive. In this mini-review, the multiplicity of molecular targets of flavonoids in blood cancer cells is discussed by demonstrating the involvement of various signaling pathways in induction of apoptotic responses. Although these data reveal a great potential of flavonoids for the development of novel agents against different types of hematological malignancies, the pleiotropic nature of these compounds in modulation of cellular processes and their interactions certainly need unraveling and further investigation.

Unleashing the Therapeutic Potential of CAR-T Cell Therapy Using Gene-Editing Technologies

  • Chimeric antigen receptor (CAR) T-cell therapy, an emerging immunotherapy, has demonstrated promising clinical results in hematological malignancies including B-cell malignancies. However, accessibility to this transformative medicine is highly limited due to the complex process of manufacturing, limited options for target antigens, and insufficient anti-tumor responses against solid tumors. Advances in gene-editing technologies, such as the development of Zinc Finger Nucleases (ZFNs), Transcription Activator-Like Effector Nucleases (TALENs), and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR/Cas9), have provided novel engineering strategies to address these limitations. Development of next-generation CAR-T cells using gene-editing technologies would enhance the therapeutic potential of CAR-T cell treatment for both hematologic and solid tumors. Here we summarize the unmet medical needs of current CAR-T cell therapies and gene-editing strategies to resolve these challenges as well as safety concerns of gene-edited CAR-T therapies.

수온충격에 따른 금붕어(Carassius auratus)의 혈액 성상 및 생리적 반응 (Physiological Response and Hematological Characteristics of Goldfish (Carassius auratus) to Water Temperature Shock)

  • 금붕어(Carassius auratus)를 사용하여 수온변화에 따른 스트레스 반응에 대한 기초자료를 얻고자, 수온상승과 하강에 따른 혈액 성상 및 생리적 반응을 조사하였다. 수온은 $15^{\circ}C$로부터 급격하게 하강시켜 1시간이내에 $10^{\circ}C$로 사육수를 조절하였으며, 이후 자연적으로 수온이 상승하도록 조절하였다. 12시간째에 수온은 $15^{\circ}C$로 회복되어 24시간째까지 유지하였다. 이러한 수온자극 스트레스를 3일 연속하여 주었다. 혈장 cortisol, glucose 및 lactic acid에서 72시간까지 대조구보다 스트레스 실험구에서 유의적으로 높은 값을 보였다. 혈장 $Na^+,\;Cl^-$ 및 aspartate aminotransferase (AST)는 두 그룹에서 유의적 차이를 보이지 않았다. 본 연구결과 혈장 cortisol, glucose, lactic acid, hematocrit, red blood cell 및 hemoglobin에서 일반적인 스트레스 반응을 보였다.

Effect of Arsenic on Immunity, Oxidative Enzyme and Various Hematological Parameters in Cross Bred Calves

  • An experiment was conducted on crossbred male calves to study the effect of arsenic (As) on immunity status and certain hematological parameters. Ten crossbred male calves of 3-4 months of age were distributed into two equal groups. Group I was kept as control, whereas, group II was supplemented daily with 50 ppm As (as $As_sO_3$) up to 90 days, in the diet. Calves of both groups were fed as per ICAR standards and their requirements were fulfilled by feeding concentrate mixture and green oats. All calves were kept under similar managemental conditions. Blood samples were collected at fortnightly intervals to estimate various haematological parameters and superoxide dismutase (SOD) enzyme activity. Serum Ig and serum glutamic pyruvate transaminase (SGPT) were also measured. Cell-mediated immune responses of the calves were monitored at 0, 45 and 90 of experimental feeding, through lymphocyte proliferation. No change in blood total leukocyte counts (TLC), differential leukocyte counts (DLC), packed cell volume (PCV), haemoglobin (Hb) and SGPT was observed with As supplementation. A decrease in SOD activity was noticed in group II calves. Stimulation index (SI) for lymphocyte proliferation decreased from 1.14 to 0.79 in group II calves during 90 days experimental feeding, whereas, there was no change in SI values in group I indicating significant decrease in immune response of As supplemented calves. Blood As concentration increased in group II calves with the decrease in immune response. Short term supplementation of As to growing calves suggested suppressive effects on cell-mediated immunity. However, long term experiments are required to demonstrate clearly the efects of this toxic metal in calves.

Clinical Characteristics of Trauma-Related Chronic Osteomyelitis in 3 Wild Raccoon Dogs (Nyctereutes procyonoides)

  • Osteomyelitis typically occurs because of the direct inoculation of bacteria or fungi after penetrating trauma or surgical contamination or, by extension, from soft tissue infection. Osteomyelitis is rarely reported in wildlife animals, though severe chronic osteomyelitis cases do exist in wildlife owing to the scarcity of medical support in the wild environment. This report describes three cases of chronic osteomyelitis in wild raccoon dogs related to trauma. The typical symptoms of three reported cases were ataxia, stiffness, muscle atrophy, and lethargy. All three cases were relevant to traumatic or severe external injury, and skin infestation caused by ectoparasites was apparent on an ocular inspection. In the radiographic examination, diffuse sites of osteolytic lesions and remarkable periosteal responses were demonstrated around the injured limb in all three cases. Apparent neutrophilia with a left shift, lymphocytosis, and monocytosis in hematological examinations generally indicated chronic infection as shown in case 1 and 3. Treatment was attempted with broad-spectrum antibiotics and non-steroidal anti-inflammatory drugs, such as amoxicillin/clavulanic acid, enrofloxacin, clindamycin, and meloxicam. These treatment options helped improve the overall prognosis of chronic osteomyelitis, but the outcomes did not meet the treatment goal entirely. Osteomyelitis can be extremely challenging to treat, particularly in wild animals, because of their distinctive traits, such as masking phenomenon and uncontrolled exposure to ectoparasites. Earlier diagnosis with a radiographic examination, hematological examinations, and careful patient monitoring, followed by prolonged antibiotic therapy and restricted exercise, are the key factors leading to a better prognosis.

넙치(Paralichthys olivaceus)용 고효율 곤충배합사료 양식현장적용 평가 (Effects of Worm-based Extruded Pellets on Growth Performance of Olive Flounder Paralichthys olivaceus in Commercial Aquafarms)

  • We evaluated a worm-based extruded pellet diet with black soldier fly Hermetia illucens larvae (BSF) meal and BSF oil for olive flounder Paralichthys olivaceus through field feeding experiments at a commercial aquafarm. We prepared two experimental diets by replacing fish meal and fish oil with BSF meal and BSF oil (BEP-1, BSF meal 7%, BSF oil 1%) and (BEP-2, BSF oil 2%), respectively. We prepared raw-fish based moist pellets (MP) for comparison between the two experimental diets. We distributed the olive flounder (220±6.29 g) in square (10 m×10 m×1 m) concrete, 100 ton tanks at a density of 1,600 fish per tank (n=3) in triplicate for each dietary treatment. We fed the diets to the fish to apparent satiation for 7 months. At the end of the feeding trial, we found no substantial differences between the groups in terms of growth performance, survival, or feed utilization. None of the diet groups showed any changes in either hematological or non-specific immune responses. The histological observation of the intestine showed that the goblet cell number and cholecystokinin-producing cell activity increased in the fish fed the BEP diet compared with the those of the fish fed the MP diet. These results indicated that dietary BSF meal and oil can be used for olive flounder without compromising growth or, hematological and histological parameters.
