Effect of Inorganic Mercury on Hematological and Antioxidant Parameters on Olive Flounder Paralichthys olivaceus

  • The effects of inorganic mercury on hematological parameters and hepatic oxidative stress enzyme activity were studied in olive flounder Paralichthys olivaceus. Fish were injected twice intraperitoneally with mercuric chloride (2, 4, or 8 mg Hg/kg BW). The major hematological findings were significant decreases in the red blood cell count, hematocrit value, and hemoglobin level in olive flounder exposed to 8 mg Hg/kg BW. Remarkably low levels of calcium and chloride, and reduced osmolality, were also observed at 8 mg Hg/kg BW. In hepatic tissue, significant increases in glutathione peroxidase and catalase activity were observed above 4 mg Hg/kg BW Inorganic mercury also increased glutathione S-transferase and glutathione reductase activity at 8 mg Hg/kg BW in hepatic tissue. The present findings suggest that exposure to a low concentration (${\geq}4$ mg Hg/kg BW) of inorganic mercury can cause significant changes in hematological and antioxidant parameters.

  • Abnormal hematological values are associated with various disorders including cancer and cardiovascular, metabolic, infectious, and immune diseases. We report the copy number variations (CNVs) in clinically relevant hematological parameters, including hemoglobin level, red and white blood cell counts, platelet counts, and red blood cell (RBC) volume. We describe CNVs in several loci associated with these hematological parameters in 8,842 samples from Korean population-based studies. The data that we evaluated included four RBC parameters, one platelet parameter, and one associated with total white blood cell (WBC) count, exceeding the genome-wide significance. We show that CNVs in hematological parameters are associated with some loci, different from previously associated loci reported in single nucleotide polymorphism (SNP) association studies.

  • Changes in selected blood and serum components and electrocardiography (ECG) were investigated in 20 adults (13 females and 7 males) of water buffaloes suffering from severe theileriosis. The age of all animals used in this study ranged 1.5-5yr. Theileriosis was diagnosed by observation of parasites in the peripheral blood and the presence of schizonts in lymphocytes that were provided from swollen lymph nodes. Statistically significant decreases were observed in the means of RBC, WBC, and packed cell volume (PCV) in blood of infected animals. The means levels of sodium, calcium, phosphorus, and potassium of infected animals were lower than healthy animals, but only the decrease of potassium was significant. The mean serum activities of aspartate transferase and alanine aminotransfrase were significantly higher than in un infected animals. Three cases had atrial premature beat, 2 cases had sinus tachycardia, 2 had sinus arrhythmia, and 1 had first degree of atrioventricular block in ECG. The present study showed that T. annulata infection in cattle is associated with hematological and biochemical, and ECG changes.

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

  • This feeding experiment was conducted to investigate the effects of dietary inclusion of some additives on growth performance, hematological parameter and fatty acid composition of growing flounder. Triplicate groups of fish (average weight 120 g) were fed one of five diets containing 5% kelp meal (Ke), 10% krill meal (Kr), 1% garlic powder (Ga), 1% citrus meal (Ci) or control diet (Con) without supplementation for 15 weeks. After the feeding experiment, survival was not significantly different among the groups fed the different diets. Weight gain of fish fed the Ci diet was significantly higher than that of fish fed the Kr diet, but not significantly different from Con, Ke and Ga treatments. Feed efficiency and protein efficiency ratio of fish fed the Ga diet were significantly higher than those of fish fed the other diets. Total protein, glucose, GOT, GPT and total cholesterol contents in the plasma were not affected by the dietary additives. Composition of C20:4n-6 in the dorsal muscle of fish fed the Con diet was significantly higher than that of fish fed the other diets. The results of this study suggest that the dietary inclusion of garlic meal at 1% may improve feed utilization of growing flounder.

Hematological and Serum Biochemical Analyses in Experimental Caprine Besnoitiosis

  • This study was undertaken to investigate the hematological and biochemical changes in experimentally infected goats with Besnoitia caprae from the time of infection till 360 days post-infection (PI). Six male goats were inoculated subcutaneously with $13{\times}10^7$ bradyzoites of B. caprae, and blood samples were collected from the jugular vein. The total erythrocyte and total leukocyte counts, hematocrit value, and differential leukocyte counts were determined. Serum biochemical analysis, including the total protein, albumin, total globulin, cholesterol, triglyceride, chloride, testosterone, calcium ($Ca^{2+}$), inorganic phosphorus, sodium ($Na^+$), potassium ($K^+$), iron ($Fe^{2+}$), glucose, serum amyloid A (SAA), haptoglobin (Hp), fibrinogen, ceruloplasmin, aspartate aminotransferase, alanine aminotransferase, creatine kinase, lactate dehydrogenase, and alkaline phosphatase, was undertaken. Skin biopsy from the limbs were collected at weekly intervals and histologically examined for Besnoitia cysts. Cysts were present in the skin biopsies of the leg of the infected goats from day 28 PI. There were variations in hematological analyses, but no significant difference was seen. From day 30 to 360 PI, results showed that SAA, Hp, fibrinogen, and ceruloplasmin concentrations increased, whereas testosterone concentrations decreased. Infected goats exhibited decrease of albumin and increase of serum total protein and globulin concentrations. By contrast, there were no significant differences in the remained analyses concentrations.

Mice를 이용한 혈액 및 생화학적 특성에 대한 Nitrogen Dioxide의 위해성 평가 (Risk Assessment of Nitrogen Dioxide for Hematological and Biochemical Parameter in Mice)

  • 본 연구에서는 실험동물을 이용하여, $NO_2$의 반복적으로 폭로($0.025mg/m^3$, 8h/day /4 weeks)하여, 실험군 및 대조군에서의 혈액학 및 생화학적 요인들의 변화와 상관성을 평가하였 였으며, 그 결과를 요약하면 다음과 같다. $NO_2$의 흡입 폭로로 인한 혈액학적 요인의 변화는 백혈구의 경우 동물실험에서의 폭로군에서 통계적으로 유의하게 증가하였다(p<0.05). hematocrit의 경우 폭로군에서 통계적으로 유의하게 감소하는 경향을 나타내었다. 또한 hemoglobin과 hematocrit는 전혈 중에서 적혈구의 비율을 나타내는 것이므로 3가지 종류 모두 유사한 경향을 나타내었다. methemoglobin의 경우 동물실험에서 폭로군에서 유의하게 증가하였다(p<0.05). 생화학적 요인에 대한 $NO_2$의 영향은 동물실험에서 total protein과 triglyceride이 동물 실험에서의 폭로군에서 감소하였으나 통계적으로 유의하지는 않았다. albumin이 폭로군에서 감소하였고, blood urea nitrogen과 creatinine의 경우 폭로군에서 높은 경향으로 나타났다. uric acid 및 혈청 중 lactate dehydrogenase의 경우 폭로군이 대조군 보다 높은 농도를 나타내어 $NO_2$의 폭로에 의해 호흡기 조직세포 내 이상을 일으킨 것으로 판단된다. 따라서 실내오염물질 중 산화가스의 하나인 $NO_2$에 대한 위해성 평가는 저농도에서의 폭로연구가 장기적이고 지속적으로 이루어져야 할 것으로 사료된다.

백수오(白首烏) 엑스의 마우스 급성독성(急性毒性) 및 흰쥐 아급성독성(亞急性毒性)에 미치는 영향(影響) (Effect of Cynanchi wilfordii Radix Extract on the Acute Toxicity in Mice and Subacute Toxicity in Rats)

  • This study was attempted to investigate the acute toxicity in mice, the subacute toxicity in rats of Cynanchi wilfordii Radix extract, the effect on transaminase activities, hematological parameters, leukocyte parameters in serum of subacute-toxicated rats. In acute toxicity test, the death rate was not observed in 50, 100, 200, 300 mg/kg(i.p.), one tenths to two tenths in 300, 500, 1000, 1500 mg/kg(p.o.) for two weeks. In subacute test, rats were all died in 300 mg/kg(p.o.) during 4 weeks, in 500, 1000 mg/kg(p.o.) during three weeks. The cause of death believed to be stomach ulcer. The activities of S-GOT and S-GPT were significantly increased in all sample-treated groups, when compared with the normal groups. A number of WBC and neutrophil belong to hematological parameter were significantly increased, lymphocyte was decreased in all sample-treated group, when compared with normal group. The hemolytic action on water extract, saponin and alcohol extract showed very low activities.

Effect of Parathion on Hematological Parameters in the Serum of a Male Bagrid Catfish (Pseudobagrus fulvidraco )

  • To evaluate the impact of parathion on aquatic organisms, a freshwater bagrid catfish (Pseudobagrus fulvidraco) was exposed to sublethal concentrations $(63,\;95,\;190\;and\;380\;{\mu}g/L)$ of parathion, organophosphorus pesticide for 30 days. Glucose level in the serum of the bagrid catfish was significantly increased than that of control groups in the $190\;{\mu}g/L$ concentration at 30 days and in the $380\;{\mu}g/L$ concentration after 10 days. Bilirubin level was significantly increased in the $190\;{\mu}g/L$ concentration at the end of the experiment. After 10 days, a significant differences of lactate dehydrogenase (LDH) and alkaline phosphatase (ALP) activity increased in the ${\geq}190\;{\mu}g/L$ and $380\;{\mu}g/L$ groups. Though cholesterol concentration was stable, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) activities in serum were significantly reduced in the $190\;{\mu}g/L$ at 10 days and in the $380\;{\mu}g/L$ concentration at the end of the experiment. In conclusion, if P. fulvidraco was chronically exposed to ${\geq}190\;{\mu}g/L$ concentration of parathion, the hematological changes may be induced due to the stress response.

  • Olive flounder Paralichthys olivaceus (total weight 216.4±14.6 g, total length 28.4±1.7 cm) were exposed to different pH levels (3, 4, 5, 6, 7, 8, and 9) for 96 h. At pH 4, hemoglobin decreased significantly, while plasma calcium, glucose, cholesterol, and ALP increased significantly. Exposure to pH 4 also induced stress responses, as evidenced by a significant decrease in heat shock protein 70 (HSP 70) and a significant increase in cortisol. The results of this study indicate that acute exposure to acidic or alkaline pH (pH 3 or 9) induced significant mortality, while exposure to pH 4significantly affected hematological parameters and stress responses in P. olivaceus.
