A Novel Helicosporium Isolate and Its Antimicrobial and Cytotoxic Pigment

  • One Helicosporium strain, isolated from a wilted chestnut tree, evidenced in vitro antimicrobial activity against various types of bacteria and fungi, and generated a diffusible pigment. The antimicrobial compounds and the diffusible pigment of the Helicosporium sp. isolate were purified via solvent fractionation, column chromatography, and recycling preparative chromatography. Both the major antimicrobial compound and the diffusible pigment were identified as 2-methylresorcinol via nuclear magnetic resonance spectroscopy. Therefore, 2-methylresorcinol, a diffusible pigment generated by Helicosporium sp., appears to be an active antimicrobial principle. This pigment also exhibited considerable cytotoxicity against mammalian cells.

Antibiotic Properties of Helicosporium sp. KCTC 0635BP to Rhizoctonia solani AG2-2 IV (Rhizoctonia solani AG2-2 IV에 대한 Helicosporium sp. KCTC 0635BP의 항균활성)

  • Biocontrol potential of an isolate of Helicosporium spp. against Rhizoctonia solani, Fusarium oxysporium and Phytophthora drechsleri was evaluated in vitro and in vivo. A selected biocontrol agent designated as Helicosporium 0635BP strongly inhibited growth and lysed mycelium of Rhizoctonia solani and Fusarium oxysporium on PDA. Autoclaved culture filtrate of the agent also completely inhibited growth of the turfgrass large patch pathogen, R. solani AG2-2 IV at the concentration of 50 ml $L^{-1}$. The pathogen was killed when dipped under the 20% filtrate for four hours or 50% for one hr. In a field trial, plots applied with the crude or times diluted culture filtrate showed 100% control efficacy of the turfgrass large patch as a chemical applied for a comparison. Results indicated that Helicosporium 0635BP is a promising biocontrol agent on control of the turfgrass large patch disease and its culture filtrate contained unknown heat suitable antifungal substance (s). Further studies on mass production, purification and identification of the unknown compound (s) are in progress for practical use.

  • To confirm the antifungal activity to plant pathogenic fungi, Rhizoctonia solani, Rhizoctonia solani AG2-2, Fusarium oxysporium, Phytophthora dreschler, Alternaria sp. were selected. Helicosporium has the antifungal activity to Rhizoctonia solani, Rhizoctonia solani AG2-2, while Fusarium oxysporium, Phytophthora dreschler did not affected even though Alternaria made a feeble response to that antifungal compounds. From $^1H-NMR$ spectrum of antifungal compound, this compound was guessed to be a structure corresponding to cholesterol.

Isolation of antifungal compounds from Helicosporium nizamabadense KCTC 0635BP

  • One strain was isolated from wilted chestnut tree and identified as Helicosporium nizamabadense. Also, the strain was evaluated for biocontrol potentials against phytopathogens. Autoclaved fraction of the isolate also showed complete growth inhibition of the turfgrass large patch pathogen, Rhizoctonia. solani AG2-2 and plant pathogen, Botrytis cinerea. Culture extracts of Helicosporium nizamabadense were purified by Thin Layer Chromatography. The several fractions were analyzed by GC-MS, and NMR etc. Antifungal activities were presumably showed more than 5 fractions against the pathogens. Among them, the most effective antifungal fraction was identified as 2-Methylresocinol using GC-MS, and NMR. These results suggest that the 2-Methylresocinol may be useful for biochemicals.
