돼지감자 유래 1-sst와 1-fft 유전자의 형질전환 발현에 의한 벼의 fructan 생합성 증진 (Increment of fructan biosynthesis in rice by transformation of 1-sst and 1-fft genes isolated from jerusalem artichoke (Helianthus tuberosus L.))

  • Fructan은 식물이 저온에 노출 되었을 때 다양한 조직에 축적됨으로써 여러 스트레스에 저항을 나타내는 것으로 알려져 있다. 본 연구에서는 fructan 생합성 경로에 관여하는 효소인 1-sst와 1-fft 유전자를 돼지감자 구근으로 부터 분리하였다. 분리한 1-sst와 1-fft 유전자는 Ti-plasmid vector인 KJG V-B2 vector에 35S promoter에 의해 발현할 수 있도록 형질전환용 벡터를 구축하였다. Agrobacterium tumefaciens법에 의해 1-sst와 1-fft 유전자의 형질전환 벼를 육성하였고, 1-sst, 1-fft 및 HPT 유전자 특이적인 primer를 사용하여 PCR 분석한 결과 유전자가 벼의 callus 게놈내에 안정적으로 삽입되었음을 확인하였다. 또한 Southern 및 RT-PCR 분석에서도 같은 결과를 얻었다. 형질전환 벼의 후대에서도 안정적으로 유전자가 발현되는 homo 계통을 선발하였고 이를 이용해 1-sst와 1-fft 유전자의 삽입이 확인된 형질전환 벼에서 유전자의 발현양상을 알아보기 위해 RT-PCR 및 Real-Time PCR를 수행한 결과 형질전환 벼에서 1-sst와 1-fft 유전자 모두 안정적으로 발현되고 있음을 확인하였다. 또한 1-sst와 1-fft 유전자가 삽입된 형질전환 벼를 이용한 기능 분석 연구를 통해 식물체가 저온에 노출되었을 때 1-sst와 1-fft의 작용에 의해 fructan 생합성량이 증가됨을 알 수 있었다. 따라서 본 연구를 통해 얻어진 fructan 생합성 관련 유전자가 삽입된 형질전환 벼는 탄수화물대사 및 저온, 건조 등의 환경 stress에 대한 내성에 대해 좋은 육종 소재로 이용 가능할 것으로 사료된다.

돼지감자 수집클론의 우리나라 환경 적응성 (Performance of Several Jerusalem Artichoke Clones ( Helianthus tuberosus L. ) Screened for Adaptibility in Korea)

  • Nineteen clones of Jerusalem Artichoke (JA) from several countries were collected through the series of experiments about JA started in 1979. Collected clones were screened for adaptibility in Korea and showed introduction path way. The results about an ecological response of collected clones including flowering, tuberization, biomass production, sugar contents and grouping of collected clones for use of genetic material were as follows; 1. Nineteen clones collected were ffom Korea(2), Japan(l), USA(Z), Canada(2), France(4), Germany(7), and USSR(1). 2. Through the characteristics of top collected clones were divided to the types of branch-non branch, short and long plant height, and early and late maturity. Tuber characteristics were mainly grouped to the types of white skin color-violet skin color, clusters-single unit, round-elongate, and knotty-smooth. 3. Total sugar yields 6-om top at flowering time were 490 - 630kgl10a and 6-om the tuber were 420 -490 kg/ IOa through the high yielding clones. The top-high yielding clones were Mammoth French White, Fuseau 60, Nahodka, and JA3. The higher tuber yields were got from the clones of D- 19, Colombia, Bianka and Mammoth French White. 4. Collected clones were grouped to three and first group was characterized to early maturity and short plant height and second group to medium and finally, third group to late maturity and high plant height. 5. High yielding of top was 6-om the I group of early maturity and short plant height and high yielding of tuber h m III group of late maturity and high plant height.

부산광역시 장산 일대의 관속식물상 (Vascular Plants of Mt. Jangsan in Busan Metropolitan City)

  • This study was to carried out to survey and analyse the vascular plants of Mt. Jangsan, Busan Metropolitan City, South Korea. The vascular plants were surveyed from June to October, 2019. The numbers of vascular plants in this site were 497 taxa and comprised of 106 families, 306 genera, 454 species, 7 subspecies, 29 varieties, 5 forms and 2 hybrids. The planted species were 16 taxa including Ginkgo biloba, Cedrus deodara, Zea mays and so on. In this study, 309 taxa were identified, including Selaginella rossii, Cyrtomium falcatum, Cephalanthera falcata and so on. The rare plants were 4 taxa including Hololeion maximowiczii(EN), Ottelia alismoides(LC), Iris ensata Thunb. var. spontanea(LC) and Sagittaria trifolia(DD). All rare plants were found in Jangsan Wetland. The Korean endemic plants were 9 taxa including Populus × tomentiglandulosa, Hemerocallis hakuunensis, Hosta minor and so on. The floristic target species were totally 53 taxa, which were 2 taxa of grade IV, 11 taxa of grade III, 6 taxa of grade II and 34 taxa of grade I. The alien plants were 58 taxa including Rumex crispus, Ipomoea purpurea, Helianthus tuberosus, Vulpia myuros and so on.

돼지감자 Polyphenol Oxidase의 분리와 특성 (Isolation and Characteristic of Polyphenol Oxidase from Jerusalem Artichoke Tuber)

  • 돼지감자의 가공과정 중 갈변에 관여하는 polyphenol oxidase를 부분 정제하고 이의 특성을 조사하였다. 인산완충액으로 추출된 조효소가 ammonium sulfate에 의한 침전과 Sephadex G-100에 의한 gel filtration으로 48배 정제되었다 이 효소는 catechol을 기실로 하였을 때 최적 활성 pH가 6.5이었으며 $K_{m}$값은 3mM, 열불활성 화는 1차반응을 보였고, 이때 Z값은 $7.6^{\circ}C$, 활성화에너지($E_{a}$)는 72.6kcal/mole이었다. 이 효소를 인산완충액에 저장하였을 때 $4^{\circ}C$에서는 일주일 이상 안정하였으나 $25^{\circ}C$에서는 활성이 급격히 소실되었다. 이 효소는 monophenol에는 반응하지 않고 diphenol의 기질에만 친화력을 나타내었으며, 그 중 chlorogenic acid와 가장 잘 반응하였다. 그리고 1 mM의 L-cystein(HCl)와 L-hydrosulfite, L-ascorbic acid에 의해서 완전히 저해되었다.

경남 김해 화포천습지에 분포하는 관속식물상 (Vascular Plants Distributed in Hwapocheon Wetland, Gimhae, Gyeongnam)

  • The purpose of this study was to offer the basic data for conservation of wetland ecosystem by surveying and analysing the vascular plants distributed in Hwapocheon wetland, Gimhae, Gyeongnam, Korea. The numbers of vascular plants were summarized as 292 taxa including 72 families, 192 genera, 262 species, 1 subspecies, 26 varieties and 3 forms. There were 34 taxa of hydrophytes, 23 taxa of emergent plants, 4 taxa of floating-leaved plants, 5 taxa of free-floating plants and 5 taxa of submerged plants. The rare plants were 7 taxa including Aristolochia contorta, Penthorum chinense, Prunus yedoensis(planting), Ixeris tamagawaensis, Hydrocharis dubia, Iris ensata var. spontanea(planting), Acorus calamus and so forth. The Korean endemic plants were 2 taxa including Salix koriyanagi and Salix pseudolasiogyne. The specific plants by floristic region were 19 taxa including 2 taxa of grade V, 1 taxa of grade IV, 5 taxa of grade III, 2 taxa of grade II and 9 taxa of grade I. The naturalized plants were 62 taxa including Chenopodium album, Astragalus sinicus, Helianthus tuberosus, Panicum dichotomiflorum and so forth. The plants that were expected to spread nationwide were 14 taxa including Cerastium glomeratum, Bidens frondosa, Tagetes minuta, Festuca arundinacea and so forth. The invasive alien plants were 6 taxa including Rumex acetosella, Sicyos angulatus, Ambrosia artemisiifolia, Ambrosia trifida, Aster pilosus and Lactuca scariola.

Endophytic Bacteria Improve Root Traits, Biomass and Yield of Helianthus tuberosus L. under Normal and Deficit Water Conditions

  • Drought is more concerned to be a huge problem for agriculture as it affects plant growth and yield. Endophytic bacteria act as plant growth promoting bacteria that have roles for improving plant growth under stress conditions. The properties of four strains of endophytic bacteria were determined under water deficit medium with 20% polyethylene glycol. Bacillus aquimaris strain 3.13 showed high 1-aminocyclopropane-1-carboxylate (ACC) deaminase production; Micrococcus luteus strain 4.43 produced indole acetic acid (IAA). Exopolysaccharide production was high in Bacillus methylotrophicus strain 5.18 while Bacillus sp. strain 5.2 did not show major properties for drought response. Inoculation of endophytic bacteria into plants, strain 3.13 and 4.43 increased height, shoot and root weight, root length, root diameter, root volume, root area and root surface of Jerusalem artichoke grown under water limitation, clearly shown in water supply at 1/3 of available water. These increases were caused by bacteria ACC deaminase and IAA production; moreover, strain 4.43 boosted leaf area and chlorophyll levels, leading to increased photosynthesis under drought at 60 days of planting. The harvest index was high in the treatment with strain 4.43 and 3.13 under 1/3 of available water, promoting tuber numbers and tuber weight. Inulin content was unchanged in the control between well-watered and drought conditions. In comparison, inulin levels were higher in the endophytic bacteria treatment under both conditions, although yields dipped under drought. Thus, the endophytic bacteria promoted in plant growth and yield under drought; they had outstanding function in the enhancement of inulin content under well-watered condition.

녹비작물 재배가 토양화학성 및 인삼뿌리썩음병 발생에 미치는 영향 (Effect of Green Manure Crop Cultivation on Soil Chemical Properties and Root Rot Disease in Continuous Cropping Field of Ginseng)

  • Background: Some plants have harmful effects on fungi and bacteria as well as other plants. Incorporating such plant into soil as green manure is effective in reducing population densities of soil pathogens. Methods and Results: Twenty-three species of green manure crops were cultivated after the harvest of 6-year-old ginseng and then incorporated into the soil at the flowering stage. The following year, the root rot ratio of 2-year-old ginseng and soil chemical properties were investigated. In the absence of green manure addition, the $NO_3$ content, electric conductivity (EC), and K content decreased by 95%, 79% and 65%, respectively. In the presence of green manure addition, $P_2O_5$ and $NO_3$ contents reduced by 41% and 25%, respectively. The "survived root ratio" of 2-year-old ginseng significantly increased by 56.2%, 47.5%, and 47.3%, in the Sorghum sudanense, Ricinus communis and Helianthus tuberosus treatment, respectively. In addition, there was a significant increase in the "survived root ratio" in the Secale cereale, Chrysanthemum morifolium, Atractylodes macrocephala, and Smallanthus sonchifolius treatments. The "survived root ratio" of ginseng showed a significant positive correlation with the soil pH and a negative correlation with the $NO_3$ contents, and EC. Conclusions: Cultivation of plant form the Chrysanthemum family as green manure, using mainly the rhizomes was effective for the control of root rot disease of ginseng.

가야산국립공원의 식물상과 보전방안 (Flora and Conservation Plan of Gayasan National Park)

  • This study is to offer the raw data for conservation and restoration of national park by surveying and analysing the vascular plants in the Gayasan national park. The flora were summarized as 102 families, 328 genera, 469 species, 4 subspecies, 59 varieties and 9 forms. The endangered plants designated by Ministry of Environment were 2 taxa; Pedicularis hallaisanensis Hurus. and Gymnadenia conopsea for. albiflora Y.N.Lee. The rare plants designated by Korea Forest Service were 13 taxa; Crypsinus hastatus (Thunb.) Copel., Ligusticum tachiroei (Franch. & Sav.) M. Hiroe & Constance, Primula modesta var. fauriae (Franch.) Takeda, Lilium distichum Nakai ex Kamibay and so forth. The korean endemic plants were 15 taxa; Betula ermanii Cham., Carpinus laxiflora (Siebold & Zucc.) Blume, Stewartia pseudocamellia Maxim., Galium koreanum (Nakai) Nakai, Heloniopsis koreana Fuse, N.S. Lee & M.N. Tamura and so forth. The specific plants by floristic region were 66 taxa; Betula chinensis Maxim., Spiraea fritschiana Schneid., Vaccinium hirtum var. koreanum (Nakai) Kitam., Cimicifuga heracleifolia Kom., Adoxa moschatellina L. and so forth. The plants with approval for delivering oversea were 22 taxa; Salix hallaisanensis H.L$\acute{e}$v., Sanguisorba argutidens Nakai, Viola albida Palib., Weigela subsessilis (Nakai) L.H.Bailey and so forth. The naturalized plants were 30 taxa; Chenopodium album L., Lepidium apetalum Willd., Trifolium pratense L., Bidens frondosa L., Helianthus tuberosus L. and so forth.

밀양시 재약산 산들늪의 식물상과 복원방안 (Flora and Restoration Plan of Sandeul Wetland in Mt. Jaeyak, Miryang-si, Korea)

  • The objectives of this study were to offer the basic data for the restoration and conservation of forest wetland by surveying systematically the vascular plants of Sandeul wetland in Mt. Jaeyak, Miryang-si, Gyeongsangnam-do, Korea. The vascular plants in this wetland were recorded as 232 taxa; 74 families, 178 genera, 200 species, 27 varieties and 5 forma. The major communities were Quercus mongolica, Tripterygium regelii, Alnus japonica, Molinia japonica, Salix gracilistyla and Stephanandra incisa. The numbers of plant species by routes were 168 taxa in A-route, 126 taxa in B and 132 taxa in C. The ecological problems in Sandeul wetland were the afforestation of Pinus koraiensis and P. thunbergii, the appearance of naturalized plants, the invasion of species and the scour of valley. The Korean endemic plants were 5 taxa; Salix hallaisanensis, Hepatica insularia; Chrysosplenium barbatum, Ajuga spectabilis and Weigela subsessilis. The rare plants designated by Korea Forest Service were 3 taxa; Aristolochia manshuriensis, Chrysanthemum lineare and Iris ensata var, spontanea. The naturalized plants were 8 taxa; Rumex acetocella, R. crispus, Trifolium repens, Oenothera lamarckiana, Ambrosia artemisifolia var. elatior, Helianthus tuberosus, Erigeron annuus and Phleuum pratense.

Bio-control of Stem Rot in Jerusalem Artichoke (Helianthus tuberosus L.) in Field Conditions

  • Stem rot is a serious disease in Jerusalem artichoke (JA). To reduce the impact of this disease on yield and quality farmers often use fungicides, but this control method can be expensive and leave chemical residues. The objective of this study was to evaluate the efficacy of two biological control agents, Trichoderma harzianum T9 and Bacillus firmus BSR032 for control of Sclerotium rolfsii under field conditions. Four accessions of JA (HEL246, HEL65, JA47, and JA12) were treated or notreated with T. harzianum T9 and B. firmus BSR032 in a 4 × 2 × 2 factorial experiment in two fields (environments), one unfertilized and one fertilized. Plants were inoculated with S. rolfsii and disease was evaluated at 3-day intervals for 46 days. T. harzianum T9 and B. firmus BSR032 reduced disease incidence by 48% and 49%, respectively, whereas T. harzianum T9 + B. firmus BSR032 reduced disease incidence by 37%. The efficacy of T. harzianum T9 and B. firmus BSR032 for control of S. rolfsii was dependent on environments and genotypes. The expression of host plant resistance also depended on the environment. However, HEL246 showed consistently low disease incidence and severity index in both environments (fertilized and unfertilized). Individually, T. harzianum T9, B. firmus BSR032, or host plant resistance control stem rot caused by S. rolfsii in JA. However, no combination of these treatments provided more effective control than each alone.
