2단 흐름형 침출공정에 의한 돼지감자 줄기의 전처리 (Pretreatment of Helianthus tuberosus Residue by Two-Stage Flow Through Process)

  • 본 연구에서는 목질계 바이오매스인 돼지감자 줄기에 대한 전처리 공정을 수행하였다. 공정은 효소 당화 수율을 높이기 위하여 흐름형 침출 전처리 공정에 2단 전처리 공정으로 적용하였다. 전처리 용매로 암모니아수에 의한 탈 리그닌 효과와 황산 용액에 의한 헤미셀룰로오스의 분해가 효소당화 및 발효에 미치는 영향을 확인하였다. 암모니아수와 황산용액을 이용한 2단 전처리 공정을 수행하였다. 먼저 1단계 공정은 40분 동안 $163.2^{\circ}C$의 온도에서 암모니아수로 처리를 진행하였고 다음 2단계 공정은 $169.7^{\circ}C$에서 20분 동안 황산 용액으로 처리를 수행하여 물질수지를 구하였다. 그리고 앞의 공정과 반대의 순서로 황산 용액을 먼저 처리한 후 암모니아수를 처리한 2단 전처리 공정을 수행하였다. 이때 암모니아수를 먼저 처리한 공정에서 글루코오스 생산량은 30.7 g으로 72.4%의 수율이 나타났다. 반대로 황산 용액을 먼저 처리한 후 암모니아수를 처리한 2단 전처리 공정에서는 글루코오스 생산량이 20.9 g으로 49.3%의 수율을 보였다.

돼지감자분말 첨가가 쌀 스펀지케이크의 품질 특성과 항산화능에 미치는 효과 (Effects of Helianthus tuberosus Powder on the Quality Characteristics and Antioxidant Activity of Rice Sponge Cakes)

  • This study was conducted to evaluate the quality characteristics and antioxidant activities of rice flour sponge cakes containing 0, 20, 30, 40, and 50% Helianthus tuberosus powder (HTP). As HTP content of sponge cake increased, the pH of sponge cake batter significantly decreased, whereas specific gravity significantly increased (p<0.05). Meanwhile, volume and symmetry indices of sponge cake were inversely proportional to HTP amount. Further, with higher HTP content in sponge cake, L and b values of crumbs significantly decreased (p<0.05), whereas a value significantly increased (p<0.05). L, a, and b values of crust significantly increased with higher HTP content. For texture characteristics, hardness, cohesiveness, gumminess, and chewiness of sponge cake significantly increased (p<0.05),whereas cohesiveness did not significantly change. In the sensory evaluation test, rice sponge cake prepared with 10% HTP showed the highest sensory scores in terms of color, flavor, taste, softness properties, and overall preference. The levels of total polyphenol compounds and DPPH radical scavenging activities of rice sponge cakes significantly increased with higher HTP contents (p<0.05). The results of this study suggest that rice sponge cake containing 10% HTP is the most appropriate for quality characteristics and sensory evaluation. This study also provides a way to increase the quality, texture characteristics, and organoleptic properties of sponge cake while reducing HTP content to less than 10% in order to satisfy consumer tastes.

열처리에 따른 돼지감자 Methanol 추출물의 항산화 및 α-glucosidase 저해 효과 (Antioxidant Activity and α-Glucosidase Inhibitory Effect of Jerusalem Artichoke (Helianthus tuberosus) Methanol Extracts by Heat Treatment Conditions)

  • This study investigated the changes of antioxidant activity and ${\alpha}$-glucosidase inhibitory effect of Jerusalem artichoke (Helianthus tuberosus) 100% methanol extracts by various heat treatment. The contents of total phenolic and flavonoid compounds in methanol extract tended to increased gradually with the rise of temperature to 180$^{\circ}C$. The maximum yield of gallic acid (51.52 ${\pm}$ 2.17mg/g extract weight) and quercetin (13.39 ${\pm}$ 0.03mg/g extract weight) were obtained with extraction temperature of 180$^{\circ}C$ for 120min. In addition, the improving extraction efficiency resulted in the increased biological activities, such as electronic donation ability (EDA, 90.36${\pm}$ 0.57%), reducing power (Abs 1.14) and ${\alpha}$-glucosidase inhibitory effect (92.14 ${\pm}$ 1.14%). Overall, the results of this study indicate that the optimum conditions for the extraction process were an extraction temperature at 180$^{\circ}C$ for 120 min, and will provide the basis for future research on the improving extraction yield of phenolic and flavonoid compounds.

돼지감자(Helianthus tuberosus L.)로 부터의 알콜 생산을 위한 균주 선발 (Selection of Yeast Strains for Alcohol Production from Jerusalem Artichoke Tubers (Helianthus tuberosus L.))

  • 돼지감자 괴경에서의 알콜 생산을 위한 균주 선발에서, 먼저 11종의 효모를 YNB-inulin 배지에서 inulase 생성에 대한 실험 결과 K. fragilis와 K. marxianus가 각각 0.124unit/ml와 0.116unit/ml로 가장 양호하였으며, 다음으로 돼지감자 괴경착즙액에서 36시간 알콜 발효 실험을 행한 결과 알콜 발효능이 70% 이상인 균주는 K. fragilis, K. marxianus 및 T. colliculosa였으며, 이중 K. fragilis가 82.8%로 가장 좋은 결과를 나타내었다. 또한 위 세 균주 및 S. cerevisiae의 알콜 발효 kinetic parameter들을 비교 분석한 결과 K. fragilis가 가장 좋은 균주로 판명되었다. 즉, 균체 수득율$(Y_{x/s})$은 0.07g/g으로 다른 균주에 비하여 가장 적었으며, 알콜 수득율은 0.45(이론치의 88%)로 S. cerevisiae의 0.47(이론치의 92%)보다 낮았으나, 알콜 생성속도$(q_p)$, 알콜 발효 속도(Q) 및 알콜 발효능(F) 값은 각각 1.53g/g-hr, 2.35g/l-hr 및 77%로 다른 균에 비하여 가장 높았다.

HIT-T15 세포에서 돼지감자 추출물의 항당뇨 효과 (Helianthus tuberosus Extract Has Anti-Diabetes Effects in HIT-T15 Cells)

  • 본 실험에서는 hamster $\beta$-cell인 HIT-T15 cell을 이용하여 돼지감자추출물의 생리활성 및 기능을 검증하고자 하였다. 돼지감자추출물을 첨가한 NC(0 ${\muL/mL$), HT2(1.1 ${\muL/mL$), HT3(1.5 ${\muL/mL$)군과 inulin을 첨가한 NC(0 ${\muL/mL$), IN2(1.8 ${\muL/mL$), IN3(2.5 ${\muL/mL$)군으로 나누어 실험하였다. 세포 viability 측정한 결과 시료를 첨가하지 않은 군을 100%로 보았을 때 돼지감자추출물을 첨가한 HT3(1.5 ${\muL/mL$)군과 inulin을 첨가한 IN2(1.8 ${\muL/mL$), IN3(2.5 ${\muL/mL$) 군에서 세포생존율이 유의적으로 증가하였다(p<0.05). 시료처리 후 췌장 $\beta$-세포 파괴를 유도하지 않고 HIT-T15 cell의 cell culture supernatant를 이용하여 cytotoxicity를 측정한 결과 시료를 첨가하지 않은 NC(0${\muL/mL$)군에 비해 모든 군에서 cytotoxicity가 낮게 나타났다. Alloxan(4 mM)으로 $\beta$-세포 파괴를 유도하여 HIT-T15 cell에서 세포보호 효과를 측정한 결과 시료를 첨가하지 않은 NC(0 ${\muL/mL$)군에 비해 돼지감자추출물을 첨가한 HT2(1.1 ${\muL/mL$), HT3(1.5 ${\muL/mL$)군에서 세포생존율이 유의적으로 증가하였다(p<0.05). 또한 췌장 $\beta$-세포 파괴를 유도하여 HIT-T15 cell이 분비한 인슐린 분비능 및 세포 내 $NAD^+$/NADH 함량을 측정한 결과 시료를 첨가하지 않은 NC(0 ${\muL/mL$)군에 비해 돼지감자추출물을 첨가한 HT3(1.5 ${\muL/mL$)군에서 인슐린 분비량과 $NAD^+$/NADH 함량이 유의적으로 증가하였다(p<0.05). 이상의 연구 결과 돼지감자추출물은 HIT-T15 cell의 생존율을 높이고, 세포보호 효과를 가짐으로써 인슐린 분비능 정상화 및 $NAD^+$ 함량을 증가시켜 혈당 조절 및 당뇨에 긍정적 효과가 있을 것으로 사료된다.

Changes of Catbohydrate and Tuber Production on Red and White Skinned Tubers of Jerusalem Artichoke (Helianthus tuberosus L.)

  • This study was aimed to investigate the difference for carbohydrate accumulation in both the red skinned tuber and white skinned tuber of Jerusalem artichoke (Helianthus tuberosus L.), and to evaluate their tuber yield of seven lines collected from Korea. Jerusalem artichoke tubers were divided into two groups regarding to their skinned colors. Red skinned tuber collected from Euisung region showed the lowest tuber yield as 3,100 kg per 10a, otherwise white skinned tuber collected from Imdong region resulted in the highest tuber production as 6,300 kg per 10a among the six kinds of white skinned tubers. Yield of white skinned tuber was higher than that of red skinned tuber. It was inferred from the result that carbohydrate accumulation in white skinned tuber was highly increased compared to red skinned tuber since after early tuber enlargement.

금오산 분지의 삼종 초본식물개체군의 인의 순경 (Phosphorus Cycle in Three Herb Plant Populations in the Basin of Mt. Geumoh)

  • The seasonal pattern and the annual magnitude of the internal (or biochemical) and plant-soil cycles of phophorus in adjacent three different herb plant populations-Helianthus tuberosus L., Artemisia princeps Pampan and Phalaris anundinacea L.-in an old field of the basin in the Mt. Geumoh were investigated. A lot of the phosphorus demand for the three populations was supplied by the withdrawal from below-ground organ in early growing period, by the absorption from soil in mid-growing period and by the withdrawal from stem in later growing period. But in the A. princeps and P. arundianacea populations, a great deal of phosphorus seemed to be absorbed prior to the first withdrawal. The annual amount of the phosphorus flowing throughthe internal cycle was about 59% of the total hosphorus flow on theorgan level in the H. tuberosus population, 41% in the A. princeps population and 32% in the P. arundinacea population, indicating that the tuber plant had the most developed internal cycle system. The annual amount of phosphorus which took part in the plant-soil cycle in the stand of the three populations was in the range of 4.49-5.65 gPm-2, corresponding to only 3-8% of the extractable phosphorus contained in the soil of 0-20 cm depth. The fact that the H. tuberosus population is higher in the extent fo internal cycle but smaller in the magnitude of plant-soil cycle and lower in the turnover rate of phsophorus than the other two populations suggests that the growth of H. tuberosus population may be less dependent on soil phosphorus availability than those of the other two populations.

종피제거와 돼지감자종자의 발아 (Seedcoat removal and seed germination in Helianthus tuberosus L.)

  • Pericarp and seedcoat removal treatments were tested to determine their effectiveness in the hard-to-germinate botanical seeds of jerusalem artichoke. Fresh seed of five Helianthus tuberosus L.varieties were (A)untreated (B)water soaked overnight (C) soaked overnight followed by renoval of pericarp or (D) soaked overnight followed by renoval of pericarp and seedcoat. The results indicate that treatments which removed the pericarp and seedcoat were the very effective, giving germination of over 90%. A considerable increase in germination did not follow only the peri-carp removal treatments. The factors inhibitory to germination of fresh jerusalem artichoke seed is associated withthe seedcoat. The removal treatment of pericarp and seed-cost is recommended despite its complexity because it givesa high germination percentage and varies least from varietyto variety. Heliarthus tuberosus L., a biomass potential crop, is a member of the family compositae. The genus Helianthushas provided man with two food plants, the sunflower (H.annus) and the jerusalem artichoke or topinambour (H. tube-rosus) . (3) The jerusalem artichoke grown for its tubers, has always been an extremely minor crop, but it is stillgrown in many places as a food for man or livestock and forthe production of alcohoL. Though tubers are used for pro-pagation jerusalem artichoke also flower and produce seedin head-like in florescences. Flowers are developed acro-petaLly on flattened receptacles such that outermost flowers are oldest. Each of these epigynous flowers may develop an achene-type fruit in which outer layers of the overy wallpersist, while inner layers become disorganized. Insidethe ovary wall of mature fruit, there is a papery seedcoat, probably composed of compact cells from endosperm, integu-ments, and nucellus.In general, the efforts to improving this crop havebeen hampered by the hard-to-germinate botanical seed.Seeds did not germinate for at least IL months after harvest.Fresh seeds of some varieties require one year more to gar-minate. (5) Since the time factor between generations isof concern in a prospective breeding program of jerusalemartichoke , these observations led to investigation of thenature of delayed seed germination in jerusalem artichokeas a biomass potential crop.
