Tmp21, a novel MHC-I interacting protein, preferentially binds to β2-microglobulin-free MHC-I heavy chains

  • MHC-I molecules play a critical role in immune surveillance against viruses by presenting peptides to cytotoxic T lymphocytes. Although the mechanisms by which MHC-I molecules assemble and acquire peptides in the ER are well characterized, how MHC-I molecules traffic to the cell surface remains poorly understood. To identify novel proteins that regulate the intracellular transport of MHC-I molecules, MHC-I-interacting proteins were isolated by affinity purification, and their identity was determined by mass spectrometry. Among the identified MHC-I-associated proteins was Tmp21, the human ortholog of yeast Emp24p, which mediates the ER-Golgi trafficking of a subset of proteins. Here, we show that Tmp21 binds to human classical and non-classical MHC-I molecules. The Tmp21-MHC-I complex lacks ${\beta}_2$-microglobulin, and the number of the complexes is increased when free MHC-I heavy chains are more abundant. Taken together, these results suggest that Tmp21 is a novel protein that preferentially binds to ${\beta}_2$-microglobulin-free MHC-I heavy chains.

A Study on the Design and Strength Evaluation of the Pipe Support Structure and Hull Reinforcement (파이프 지지구조와 하부 보강의 설계와 강도 평가에 관한 연구)

  • In the case of gas carriers and oil tankers, pipes are installed on the upper deck as a moving passage to load LPG, LNG, crude oil, etc. Pipes used for loading or unloading liquid cargo in cargo holds are connected to the hull through support structures. However, many cases of hull damage have been reported where the various equipment and support structures are installed on the upper deck. It is assumed that not only the structural discontinuity where the hull and the pipe support structure meet, but also action due to the pipe loads and the hull girder bending moment are simultaneously affected. This paper deals with the design and strength evaluation of the support structure of pipes and cables installed on the upper deck of commercial ships and offshore structures. For these supporting structures, design conditions and working loads were defined. The design procedure was established through the structure analysis on the method of determining the member dimensions. A series of finite element analysis was performed on the factors to be considered in the design and the effects were discussed. The accuracy and design periods of the strength evaluation was improved and reduced by application of the automation program in the finite element analysis. It is also expected that the design reliability of the shipyard is improved.

Comparative Experimental Study on Sloshing Impact Loads of LNG Cargoes in Membrane Containment System of 160K LNGC (160K LNGC 멤브레인 화물창에 작용하는 슬로싱 충격 하중에 대한 비교 실험 연구)

  • A new state-of-the-art sloshing research equipment has developed to perform the model test of LNG tanks for the safer design of LNG cargo containment system in violent sloshing phenomena. This sloshing test system has developed by the Samsung Ship Model Basin (SSMB) and thoroughly verified. The accuracy of the motion of hexapods equipment for the excitation of a model tank has been verified. The maximum displacement in six degrees of freedom, harmonic motions of various frequencies, and irregular motions in wave conditions are measured and compared with input signals. In order to confirm the reliability of the post-processing program for measured impact pressure, the post-processed results were compared with those of the reference institute. A benchmarking sloshing test using 1/50 scale model of 160K LNGC tank was conducted for the verification of the whole testing system. The partial filing levels were considered. As a result of the experiment, it is confirmed that the results are in good agreement with those of the reference institute.

  • Consumption of Liquefied Natural Gas (LNG) has increased due to environmental pollution; therefore, the need for LNG carriers can efficiently transport large quantities of LNG, is increased. In various types of LNG Cargo Containment System (CCS), Membrane-type MARK-III composed of composite materials is generally employed in the construction of an LNG carrier. Among composite materials in a Mark-III system, glass-fiber composites act as a secondary barrier to prevent the inner hull structure from leakage of LNG when the primary barrier is damaged. Nevertheless, several cases of damage to the secondary barriers have been reported and if damage occurs, LNG can flow into the inner hull structure, causing a brittle fracture. To prevent those problems, this study conducted the applicability assessment of composite material manufactured by bonding glass-fiber and aluminum with epoxy resin and increasing layer from three-ply (triplex) to five-ply (pentaplex). Tensile tests were performed in five temperature points (25, -20, -70, -120, and -170℃) considering temperature gradient in CCS. Scanning Electron Microscopy (SEM) and Coefficient of Thermal Expansion (CTE) analyses were carried out to evaluate the microstructure and thermos-mechanical properties of the pentaplex. The results showed epoxy resin and increasing layer number contributed to improving the mechanical properties over the whole temperature range.

  • Maritime transportation consists of various situations such as navigation in the ocean, ship handling at harbor entrances, cargo handling and mooring in harbors. Generally, ships are built for the purpose of currying people and materials upon the seas. In order to accomplish the mission, a ship must be built to withstand the rigors of heavy weather and waves. In particular, the safety of ship motions at the entering/departing harbor and mooring under the effects of waves is very important for ship operation from the viewpoint of marine engineering. Therefore, safety and efficiency during entrance, departure, and mooring are extremely important aspects in the evaluation of ship operations from viewpoints of ship motions. However, the ship motions near a harbor entrance are not observed or studied as much. In this paper, to evaluate the difficulty of ship operations, field observations were performed using a new observation system with high accuracy in typhoon seasons, and grasp was done concerning about the time series characteristic that ship motions change rapidly within a harbor. Namely, such observations enable the quantitative safety evaluation under the effects of waves during ships entering and departing harbors in heavy weather.

  • When liquefied natural gas (LNG) is stored in a tank, it is necessary to maintain low temperature. It is very important that insulation techniques are applied to the LNG cargo because of this extreme environment. Hence, laminated wood, especially plywood, is widely used as the structural member and insulation material in LNG cargo containment systems (CCS). However, fracture of plywood has been reported recently, owing to sloshing effect. Therefore, it is necessary to increase the strength of the structural member for solving the problem. In this study, compressed wood, which is used as a support in LNG independent type B tanks, was considered as a substitute for plywood. Compression and bending tests were performed on compressed wood under ambient and cryogenic temperatures to estimate the mechanical behaviors and fracture characteristics. In addition, the direction normal to the laminates surface was considered as an experimental variable. Finally, the feasibility of using compressed wood for an LNG CCS was evaluated from the test results.

A Study on the Towing Force of Heavy Loaded Barge against Strong Current (강조류에서 중량물 운반 부선의 예인력에 관한 연구)

  • A marine collision accident occurred at the Jin-do water way. A barge loaded with heavy equipment of Power Generation Plant was being towed at its side by two tugs "A" and "B" from Byeg-pa harbour to the designated position to be used for power generation plant near Jin-do bridge. The Power Generation Plant was 40-meters high loaded atop the barge. When the "tugs and tow" were approaching the Jin-do the bridge, the two towing tugs lost maneuvering control of the barge and it went off-course, hit first the waterway embankment and finally struck the lower part of bridge's span and post musing the barge to capsize and sunk its cargo into the sea. The bridge also suffered damages where the top of the Plant struck At the time of the accident, there was a strong tidal current at the waterway. This paper calculates the safe towing force of barges with heavy loads when traversing in a strong tidal current situation in narrow waterway.

  • Recently, the size of Liquified Natural Gas (LNG) carriers has been increasing, in turn increasing the load generated during operation. To handle this load, the thickness of LNG Cargo Containment Systems (CCSs) should be increased. Despite increasing the thickness of LNG CCSs, a secondary barrier is still used in conventional thickness. Therefore, the mechanical performance of the existing secondary barrier should be verified. In this study, tensile test of the secondary barrier was performed to evaluate mechanical properties under several low- and cryogenic-temperature conditions considering LNG environment, and in each fiber direction considering that the secondary barrier is composed of anisotropic composite materials depending on the glass fibers. Additionally, the coefficient of thermal expansion was measured by considering the degradation of the mechanical properties of the secondary barrier caused by the generated thermal stress during periodical unloading. As a result, the mechanical performance of secondary barrier in the Machine Direction (MD) was generally found to be superior than that in the Transverse Direction (TD) owing to the warp interlock structure of the glass fibers.

  • Plywood is a laminated wood material where alternating layers are perpendicular to each other. It is used in a liquefied natural gas (LNG) carrier for an insulation system because it has excellent durability, a light weight, and high stiffness. An LNG cargo containment system (LNG CCS) is subjected to loads from gravity, sloshing impact, hydrostatic pressure, and thermal expansion. Shear forces are applied to an LNG CCS locally by these loads. For these reasons, the materials in an LNG CCS must have good mechanical performance. This study evaluated the shear behavior of plywood. This evaluation was conducted from room temperature ($25^{\circ}C$) to cryogenic temperature ($-163^{\circ}C$), which is the actual operating environment of an LNG storage tank. Based on the plywood used in an LNG storage tank, a shear test was conducted on specimens with thicknesses of 9 mm and 12 mm. Analyses were performed on how the temperature and thickness of the plywood affected the shear strength. Regardless of the thickness, the strength increased as the temperature decreased. The 9 mm thick plywood had greater strength than the 12 mm thick specimen, and this tendency became clearer as the temperature decreased.
