Beneficial Effect of Heat Fans on Quality and Yield of Korean Melon Cultivated in Greenhouses at Winter Season (히터팬 처리가 저온기 하우스 참외의 품질 및 수량에 미치는 긍정적 영향)

  • The purpose of this study was to investigate the changes of environmental conditions and the quality and yield of melon fruit by heat fan operation in greenhouses at winter season. The average daily temperature inside the tunnels during January 1 to 31, 2017 was $0.9^{\circ}C$ higher than that of the control $17.8^{\circ}C$. The air flow rate of heater fan treatment was 4.8 times higher than the control (untreated $0.05m{\cdot}s^{-1}$) at 20cm above the ground where the korean melon grew. The temperature of the heater pan was $5.6^{\circ}C$ higher than that of the untreated at $35.3^{\circ}C$ and the relative humidity was 8.1% lower than that of the untreated at 39.1%. The flowering rate of the heater fan treatment was 96%, 5% higher than the control. The number of first harvest days of heater fan treatment was shortened by 4 days than that of untreated treatment. Fruit quality and marketable fruit yield increased by 3.4% and 38% compared to untreated respectively, the heater fan treatment increased the temperature inside the greenhouse and air flow rete, which were beneficial for growing the korean melon in greenhouses at winter season.

Studies on the Immobilization of ${\beta}-Galactosidase$ from Bacillus subtilis (Bacillus subtilis ${\beta}-Galactosidase$의 고정화에 관한 연구)

  • The conditions for immobilization of the partially purified ${\beta}-galactosidase$ form Bacillus subtilis HP4 and the properties of the immobilized enzyme have been investigated. The crude enzyme precipitated with cold acetone was purified about 68-fold through DEAE-cellulose and sephadex G-100 chromatography and its recovery was 19.9% The optimal conditions for Immobilization of enzyme were obtained in 2%(w/v) sodium alginate, 15%(v/v) enzyme solution and 2%(w/v) calcium chloride, and also the optimal stirring thme was 2 hours on the above conditions. The optimum temperature and pH values for immobilized enzyme were $55^{\circ}C$ and 6.5, respectively. Its residual activity was show 25% after heat treatment for an hour at $65^{\circ}C$, and found its high stability in pH 6.0 to 8.0. The enzyme activity was not affected b)· EDTA, 2-mercaptoethanol, KCN, protective agents, and other methal ions except Hg ion and Cu ion. The $K_m\;and\;V_{max}$ values of the immobilized enzyme on ONPG were $1.82{\times}10^{-2}M\;and\;3.57{\times}10^{-8}mole/min$, whereas those on lactose were $2.94{\times}10^{-2}M\;and\;1.68{\times}10^{-7} mole/min$, respectively. The remained enzyme activity for the immobilized enzyme was 95%t of original activity after storage of 40 days at $4^{\circ}C$, and when reused for 5 times was 81%. When skim milk(4.8% lactose) and 5% lactose solution were reacted with the immobilized enzyme(250 units/g) of lactose were 51% and 43%, respectively.

Changes in Functional Constituents and Stability of Green Tea Beverage during Different Storing Conditions (저장조건에 따른 녹차음료의 기능성성분 변화 및 안정성)

  • Preservation stability of the extract from dried green tea produced in Jeju, extracted at $55^{\circ}C$ for 15 min and bottled in PET container after filtration and then settled at $4^{\circ}C$, indoor at room temperature and outdoor, respectively, was estimated. Color L-value was almost constant but color of the extract became darkened during storage, especially in indoor storage. Main catechins of green tea extract were (-)-epigallocatechin, (-)-epigallocatechin gallate, epicatechin, (+)-catechin, epicatechin gallate in order, and the content of epigallocatechin was 53.8%. Nevertheless total catechin content was not changed in cold storage, and $4.7\sim10%$ of total catechins were reduced in outdoor or indoor storage after 14 weeks. Caffeine content were increased but total polyphenols were not changed largely during storage. Election donating abilities (EDA) showed more than 60% without influence of storage conditions and periods. Nitrite scavenging abilities (NSA) showed more than 90% at pH 1.2, but those were decreased gradually as pH increased, and then disappeared at pH 6.0. Color, polyphenols, antioxidant activities of extract were not so changed for more than 3 months. Microbial growth was not shown during same periods, due to heat treatment of the extracts at $95^{\circ}C$ for 10 min.

α-Amylase Activity of Radish and Stability in Processing (무의 α-Amylase 활성 및 가공 안정성)

  • The effects of processing conditions on the ${\alpha}$-amylase activity of radish were investigated at various temperatures, pHs and drying conditions. The ${\alpha}$-amylase activity of radish root was 3.1-fold higher than that of radish trunk. As the freeze-dried radish was incubated at various temperatures and pHs, ${\alpha}$-amylase activity was stably maintained at pH range of $4{\sim}7$ and temperature of $25{\sim}40^{\circ}C$. When radish was processed to kakdugi and danmooji, the residual ${\alpha}$-amylase activity was 45.39% and 19.19%, respectively. Consequently, the ${\alpha}$-amylase activity was greatly affected by processing conditions such as heat treatment and pH. It is suggested that radish should be processed at below $60^{\circ}C$ and at neutral to acidic pH condition.

  • Various aspect of epidemics broke out continually from the middle of Joseon Dynasty due to the famine and drought caused by abnormal climate of the sixteenth century and the war. Thus the Dynasty performed sacrificial rites, isolated the patients and published plenty of medical books related epidemics in order to cure of the patients, and Heojun edited 『Byeokyeoksinbang』 as 'Dangdokyeok' broke out at Gwanbuk(關北) districts in 1613, Heojun explained the cause of Dangdokyeok as meteorology under the feudal conditions, and concluded Simhwa(心火) by fever toxin, Therefore he selected the method of puting out Simhwa by attack of fever toxin. In addition he presented emergency treatment that can maintain the airway by bleeding. To treat Dangdokyeok, Heojun presented lots of prescriptions so as Seungmagalgeuntang(升麻葛根湯), Cheongyeolhaedoksan(淸熱解毒散), Yeongyopaedok-san(連翹敗毒散), Bangpungtongsaongsan(防風通聖散), Jowiseunggitang(調胃升氣湯) and Hwangryeonhaedoktang(黃連解毒湯) etc. And he proposed Samdueum (三豆飮), Realgar(石雄黃) and so on to prevent infection from that. They presume from 120 to 150 years as the period of human adaptation to the first epidemics. Dangdokyeok put a large number of people to death at first, but it wasn't referred at the history any more after Byeokyeoksinbang. So we can say that the treatment of Heojun may be effective. Common cold and dyspeptic cold broke out in our country differently from 'Shanghan(傷寒)' in the China, so we had settled 'pestilence infectious epidemic disease(瘟疫)' while 'epidemic febrile disease(溫病)' of the China. Dangdokyeok of Heojun is similar to 'Scalet fever' belonging to 'virulent heat pathogen(溫毒)', 'newly epidemic febrile disease(新感溫病)'. As a cure of Dangdokyeok, the Korean medicine uses the treatment of removing fever state whereas the western medicine uses the antibiotics to kill the streptococcus. The symptoms of Dangdokyeok are remarkably similar to those of the Scarlatina, so this occupies a high position on the world history of medicine in aspects of the period and details of symptoms. These days we have the problems that the tolerance of antibiotics increases and disease of unknown cause is prevalent. It means the western medicine get to limits. So if we progress epidemiography based on Heojun's medicine, we may contribute to the world history of medicine.

Visual Color Deterioration of the Extract of Lithospermi radix (자초(紫草)추출물의 외관상 변색)

  • The effects of temperature on the visual color deterioration of Lithospermi radix were investigated under steady conditions of various pH and temperature. The changes of Hunter L, a, b, and ${\Delta}E$ values related to the color deterioration were sharply increased above $50^{\circ}C$ and the deteriorations were expressed linear relationships with the temperature above $60^{\circ}C$. Linear regression coefficients of Hunter L, a, b values decreased with an increase of heat treatment time, whereas the color difference was increased. The linear regression coefficients of Hunter L, a, b, ${\Delta}E$ values in 66.7% ethanol extract solution of Lithospermi radix for 1 hr were -0.3696, -0.4124, -0.2279, and 0.5983, respectively. Inear regression coefficients in color difference (${\Delta}E$) could be calculated from the coefficients in Hunter L, a, b values. The low pH treatment of extract from pH 6.07 to pH 1.35 led to decrease of Hunter a value and a little increase of Hunter L, b values, but the high pH treatment above pH 6.07 decreased all Hunter values. Particularly, the visual color of the extract of Lithospermiradix was appeared to be faborable at pH 4.0-6.5, which was a bright red color.

Development of Eco-friendly Combustion Process for Waste 2,4,6-trinitrotoluene (폐 2,4,6-trinitrotoluene의 환경 친화적 연소처리공정 개발)

  • In this study, an eco-friendly combustion process of waste 2,4,6-trinitrotoluene (TNT: 2,4,6-trinitrotoluene) was developed, and fundamental data for the quantity of the organic matter in the final combustion residues is presented. Because complete combustion of TNT is not possible theoretically, the combustion process was optimized to reduce organic matter content in the combustion residue by performing measures such as heating time changes, addition of propellant material, and after treatment using a high-temp electrical furnace. From the results, it was confirmed that the organic matter content in the residue could be decreased to 7 ~ 10% with each method. The quantity of the organic matter could be minimized by optimizing the combustion conditions of the process. With only a combustion time increase, the amount of organic matter in the combustion residues was measured at about 9 wt%. The environmental friendliness of the final exhaust gas was also confirmed by real time gas component analyses. In addition, the organic contents could be reduced by a further 2 wt% by applying an additional heat treatment using an external electric furnace after the first incineration treatment. In the combustion process of propellant added waste TNT, it was found that various TNT wastes could be treated using the same eco-friendly protocols because the organic content in the residue decreased in accordance with the amount of propellant. The amount of the organic matter content produced by all these methods fulfilled the requirements under the Waste Management Act.

Expression, Characterization and Regulation of a Saccharomyces cerevisiae Monothiol Glutaredoxin (Grx6) Gene in Schizosaccharomyces pombe

  • Glutaredoxins (Grxs), also known as thioltransferases (TTases), are thiol oxidoreductases that regulate cellular redox state in a variety of organisms. In the budding yeast Saccharomyces cerevisiae, Grx1 and 2 are cytosolic dithiol Grxs, while Grx3, 4 and 5 are monothiol Grxs. A gene encoding a new monothiol Grx, Grx6, was cloned from the genomic DNA of S. cerevisiae by PCR. Its DNA sequence contains 1,080 bp, and encodes a putative protein of 203 amino acid residues containing Cys-Phe-Tyr-Ser at the active site. Grx6 is similar to other monothiol Grxs in the same organism and to Grx3 in the fission yeast Schizosaccharomyces pombe. and its predicted three-dimensional structure resembles that of S. pombe Grx3. S. pombe cells harboring plasmid pFGRX6 containing the Grx6 gene had about 1.3-fold elevated Grx activity in the exponential phase, and grew better than the control cells under some stressful conditions. Synthesis of ${\beta}$-galactosidase from a Grx6-lacZ fusion gene in S. pombe was enhanced by potassium chloride, aluminum chloride and heat ($37^{\circ}C$) treatment. S. pombe cells harboring plasmid pFGRX6 had elevated ROS levels whereas S. pombe cells harboring extra copies of Grx3 had reduced ROS levels.

A Cyclophilin from Griffithsia japonica Has Thermoprotective Activity and Is Affected by CsA

  • Members of the multifunctional Cyp family have been isolated from a wide range of organisms. However, few functional studies have been performed on the role of these proteins as chaperones in red alga. For studying the function of cDNA GjCyp-1 isolated from the red alga (Griffithsia japonica), we expressed and purified a recombinant GjCyp-1 containing a hexahistidine tag at the amino-terminus in Escherichia coli. An expressed fusion protein, $H_6GjCyp-1$ maintained the stability of E. coli proteins up to $50^{\circ}C$. For a functional bioassay for recombinant $H_6GjCyp-1$, the viability of E. coli cells overexpressing $H_6GjCyp-1$ was compared with that of cells not expressing $H_6GjCyp-1$ at $50^{\circ}C$. After high temperature treatment for 1 h, E. coli overexpressing $H_6GjCyp-1$ survived about three times longer than E. coli lacking $H_6GjCyp-1$. Measurement of the light scattering of luciferase (luc) showed that GjCyp-1 prevents the aggregation of luc during mild heat stress and that the thermoprotective activity of GjCyp-1 is blocked by cyclosporin A (CsA), an inhibitor of Cyps. Furthermore, the Cyp-CsA complex inhibited the growth of E. coli under normal conditions. The results of the GjCyp-1 bioassays as well as in vitro studies strongly suggest that Cyp confers thermotolerance to E. coli.

Synthesis and magnetic properties of $Fe_3C$ fine particles ($Fe_3C$ 미립자의 제조와 자기적 특성)

  • It was investigated to obtain the relationship between magnetic properties and conditions of forming $Fe_{3}C$ single phase from acicular goethite by heat treatment under the atmosphere of CO and $N_{2}$ mixed gas. X-ray analyses, TEM and VSM measurements were imployed for the characteristics of the carbide. Acicular goethite was sinthesized under proper process parameters. $Fe_{3}C$ single phase was obtained above $550^{\circ}C$, 60min. and $Fe_{5}C_2$ was formed with $Fe_{3}C$ below that temperature. The soturation magnetization of $Fe_{3}C$ single phase was about 100emu/g regardless of the reaction temperature. The coersive force and the ratio of Mr/Ms decreased respectively from 780 to 400 Oe. and from 0.35 to 0.13 with increasing reaction temperature.
