The cellular responses of soil-borne bacterium, Pseudomonas sp. HK-6 to explosive 2,4,6-trinitrotoluene (TNT) were examined. Two stress shock proteins (SSPs), approximately 70-kDa DnaK and a 60-kDa GroEL were found in HK-6 cells in response to TNT. Analyses of SDS-PAGE and Western blot using anti-DnaK and GroEL revealed that SSPs were induced in HK-6 cells exposed to 0.5 M of TNT far 6-12 hrs. The maximum induction of proteins was achieved at 8-hr incubation point after HK-6 cells'exposure to TNT. Similar SSPs were found to be induced in HK-6 cells by heat shock (shift of temperature, from $30^{\circ}C$ to $42^{\circ}C$) or cold shock (shift of temperature,$30^{\circ}C$ to $4^{\circ}C$).2D-PAGE of soluble protein tractions from the culture of Pseudomonas sp. HX-6 exposed to TNT demonstrated that approximately 450 spots were observed on the silver stained gels ranging from pH 3 to pH 10. Among them, 12 spots significantly induced and expressed in response to TNT were selected and analyzed. Approximately 60-kDa protein, which was assumed highly expressed on the gel, was used for amino acid sequencing. N-terminal microsequencing with in-gel digestion showed that N-terminal sequence of the TNT-induced protein, <$^1XXAKDVKFGDSARKKML^17$, shared extensive similarity with $^1XXAKDVKFGDSARKKML^17$, N-terminal sequence of (P48216) GroEL of Pseudomonas putida.

Plans to mitigate overheating in urban areas requires the identification of the characteristics of the thermal environment of the city. The key information is the distribution of higher and lower temperatures (referred to as "hotspot" or "coldspot", respectively) in the city. This study aims to identify the areas within Jeonju City that are suffering from increasing land surface temperatures (LST) and the factors linked to such this phenomenon. To identify the hot and cold spots, Local Moran's I and Getis-Ord Gi* were calculated for the LST based on 2017 images taken using the thermal band of the Landsat 8 satellite. Hotspot analysis revealed that hotspot regions, (the areas with a high concentration of Land Surface Temperature) are located in the old town area and in industrial districts. To figure out the factors linked to the hotspots, a correlation analysis, and a regression analysis taking into account environmental covariates including Normalized Difference Vegetation Index (NDVI) and land cover. The values of NDVI showed that it had the strongest effect on the lowering LSTs. The results of this study are expected to provide directions for urban thermal environment designing and policy development to mitigate the urban heat island effect in the future.

The dehydration responsive element binding protein 2C (DREB2C) is a dehydration responsive element/C-repeat (DRE/CRT)-motif binding transcription factor that induced by mild heat stress. Previous experiments established that overexpression of DREB2C cDNA driven by the cauliflower mosaic virus 35S promoter (35S:DREB2C) resulted in increased heat tolerance in Arabidopsis. We first analyzed the proteomic profiles in wild-type and 35S:DREB2C plants at a normal temperature ($22^{\circ}C$), but could not detect any differences between the proteomes of wild-type and 35S: DREB2C plants. The transcript level of DREB2C in 35S: DREB2C plants after treatment with mild heat stress was increased more than two times compared with expression in 35S:DREB2C plants under unstressed condition. A proteomic approach was used to decipher the molecular mechanisms underlying thermotolerance in 35S:DREB2C Arabidopsis plants. Eleven protein spots were identified as being differentially regulated in 35S:DREB2C plants. Moreover, in silico motif analysis showed that peptidyl-prolyl isomerase ROC4, glutathione transferase 8, pyridoxal biosynthesis protein PDX1, and elongation factor Tu contained one or more DRE/CRT motifs. To our knowledge, this study is the first to identify possible targets of DREB2C transcription factors at the protein level. The proteomic results were in agreement with transcriptional data.

This study aims to propose management strategies of ventilation paths for improving urban thermal environments. For this purpose, Gimhae-si in Gyeongsangnamdo was selected as a study area. We analyzed hot spots and cool spots in Gimhae by using Landsat 8 satellite image data and spatial statistical analysis, and finally derived the vulnerable areas to thermal environment. In addition, the characteristics of ventilation paths including wind direction and wind speed were analyzed by using data of the wind resource map provided by Korea Meteorological Administration. As a result, it was found that a lot of hot spots were similar to those with weak wind such as Jinyoung-eup, Jillye-myeon, Juchon-myeon and the downtown area. Based on the analysis, management strategies of ventilation paths in Gimhye were presented as follows. Jinyoung-eup and Jillye-myeon with hot spot areas and week wind areas have a strong possibility that hot spot areas will be extended and strengthened, because industrial areas are being built. Hence, climate-friendly urban and architectural plans considering ventilation paths is required in these areas. In Juchon-myeon, where industrial complexes and agricultural complexes are located, climate-friendly plans are also required because high-rise apartment complexes and an urban development zone are planned, which may induce worse thermal environment in the future. It is expected that a planning of securing and enlarging ventilation paths will be established for climate-friendly urban management. and further the results will be utilized in urban renewal and environmental planning as well as urban basic plans. In addition, we expect that the results can be applied as basic data for climate change adaptation plan and the evaluation system for climate-friendly urban development of Gimhye.
Anatomically separate fat depots differ in size, function, and contribution to pathological states such as the metabolic syndrome. We isolated pre-adipocytes from different adipose depots, omental, subcutaneous and intramuscular, of beef cattle, and cultured in vitro to determine the basis for the variations and attribute these variations to the inherent properties of adipocyte progenitors. The proliferating cells from all depots before the confluence were harvested and the proteome was analyzed by a functional proteomic approach, involving 2-DE and MALDI-TOF/TOF. More than 252 protein spots were identified, selected and analyzed by Image Master (ver 7.0) and MALDI-TOF/TOF. Further, our analysis showed that there were specific differences in proteome expression patterns among proliferating precursor cells from the three depots. Sixteen proteins were found to be differentially expressed and these were identified as proteins involved in cellular processes, heat shock/chaperones, redox proteins, cytoskeletal proteins and metabolic enzymes. The results also enabled us to understand the basic roles of these proteins in different inherent properties exhibited by adipose tissue depots.
Objective : It has long been known about the osteogenic effect of CTF-HAS on bone tissues. However, it has not been determined the effect of CTF-HAS on cancer cells. The purpose of this study is to screen the CTF-HAS mediated differentially expressed genes in cancer cells such as HepG2 hepatoma cells lines. Methods : CTF-HAS was prepared by boiling and stored at $-70^{\circ}C$ until use. For proteomic analysis, total protein was analyzed by 2D gel electrophoresis and Q-TOF mass spectrometer. Results : In proteomic analysis, three spots were identified by 2D-gel electrophoresis and Q-TOF analysis. One down-regulated protein was heat shock 70kDa protein 5 and up-regulated proteins were chaperonin and 2-phospho -pyruvate-hydratase ${\alpha}-enolase$ by 1.5mg/ml of CTF-HAS. Discussion : Proteomic analysis approach were performed to screen the differential expression genes. The screened genes will be used for the better understanding in therapeutic effect of CTF-HAS on cancer field.

In our previous study, the expression of active H-ferritins in Saccharomyces cerevisiae was found to reduce cell growth and reactive oxygen species (ROS) generation upon exposure to oxidative stress; such expression enhanced that of high-affinity iron transport genes (FET3 and FTR1). The results suggested that the recombinant cells expressing H-ferritins induced cytosolic iron depletion. The present study analyzes metabolic changes under these circumstances via proteomic methods. The YGH2 yeast strain expressing A-ferritin, the YGH2-KG (E62K and H65G) mutant strain, and the YGT control strain were used. Comparative proteomic analysis showed that the synthesis of 34 proteins was at least stimulated in YGH2, whereas the other 37 proteins were repressed. Among these, the 31 major protein spots were analyzed via nano-LC/MS/MS. The increased proteins included major heat-shock proteins and proteins related to endoplasmic reticulum-associated degradation (ERAD). On the other hand, the proteins involved with folate metabolism, purine and methionine biosynthesis, and translation were reduced. In addition, we analyzed the insoluble protein fractions and identified the fragments of Idh1p and Pgk1p, as well as several ribosomal assembly-related proteins. This suggests that intracellular iron depletion induces imperfect translation of proteins. Although the proteins identified above result from changes in iron metabolism (i.e., iron deficiency), definitive evidence for iron-related proteins remains insufficient. Nevertheless, this study is the first to present a molecular model for iron deficiency, and the results may provide valuable information on the regulatory network of iron metabolism.

In lithium-ion batteries, separator membrane's, main role is to physically isolate a cathode and an anode while maintaining rapid transport of ionic charge carriers during the passage of electric current. As far as battery safety is concerned, the electrical isolation of electrodes is most crucial since unexpected short-circuits across the membrane induces hot spots where thermal runaway may break out. Internal short-circuits are generally believed to occur by protrusions on the electrode surface either by unavoidable deposits of metallic impurities or by dendritic lithium growth during battery operation. Another cause is shrinkage of the separator membrane when exposed to heat. If separator membrane can be engineered to prevent the internal short-circuit, it will not be difficult to improve lithium-ion batteries' safety. Commonly the separators employed in lithium-ion batteries are made of polyethylene (PE) and/or polypropylene (PP). These materials have terrible limitations in preventing the fore-mentioned internal short-circuit between electrodes due to their poor dimensional stability and mechanical strength. In this study we have developed a novel separator membrane that possesses very high thermal and mechanical stability. The cells employing this separator provided noticeable safety improvement in the various abuse tests.

During the last two decades the laser beam has progressed from a sophisticated laboratory apparatus to an adaptable and viable industrial tool. Especially, in its welding mode, the laser offers high travel speed, low distortion, and narrow fusion and heat-affected zones (HAZ). The principal obstacle to selection of a laser processing method in production is its relatively high equipment cost and the natural unwillingness of production supervision to try something new until it is thoroughly proven. The major objective of this work is focused on the combined features of gas tungsten arc and a low-power cold laser beam. Although high-power laser beams have been combined with the plasma from a gas tungsten arc (GTA) torch for use in welding as early as 1980, recent work at the Ohio State University has employed a low power laser beam to initiate, direct, and concentrate a gas tungsten arcs. In this work, the laser beam from a 7 watts carbon monoxide laser was combined with electrical discharges from a short-pulsed capacitive discharge GTA welding power supply. When the low power CO laser beam passes through a special composition shielding gas, the CO molecules in the gas absorbs the radiation, and ionizes through a process known as non-equilibrium, vibration-vibration pumping. The resulting laser-induced plasma (LIP) was positioned between various configurations of electrodes. The high-voltage impulse applied to the electrodes forced rapid electrical breakdown between the electrodes. Electrical discharges between tungsten electrodes and aluminum sheet specimens followed the ionized path provided by LIP. The result was well focused melted spots.

Four species of Trichoderma causing green mildew of Agaricus bisporus were isolated from 38spots of mushroom growing areas in Korea. These are T. koningi, T. lignorum, T. glaucum and an unidentified species, and their frequencies of occurrence are $50\%,\; 32\%,\; 13\%; and\; 5\%$ respectively. All of these species grew well in potato dextrose, Waksman's and Richard's solution, and preferred acid (pH. 4) to neutral. The temperature in mushroom trouse should be kept at $15^{\circ}C$ during cropping period, not only for the high yield of mushroom but also for the prevention of green mildew of mushroom caused by Trichoderma spp . T. lignorum was killed in soil on an exposure of 60 minutes at $70^{\circ}C$ and when exposed for 30minutes at $80^{\circ}C$. Peak heat procedure of compost eliminated T. lignorum and T koningi.
