Preservation Experiment of Hibernating Silkworm Eggs by 2 Year Cold Refrigeration (월년잠종의 2년간 냉장에 의한 보존 시험)

  • Long term preservation experiment through refrigeration was conducted for 2 year on 300 lines of silkworm races preserved, as one method for the development of long term safe preservation technique. Experiment with 6 treatments was conducted for 680 days from July 1 st 2000 to May 1st 2002. Embryonic development was conducted to each treatment. There are no differences among treatments and races in 400 days preservation, the stage of whole embryonic development was Eul B and condition of eggs was good. In 650 days preservation experiment, differences were reveled among races and treatment, the level of whole embryonic development was Byeong A and condition of eggs was good. The order of embryonic development is European races >Tropical, Korean races >Japanese, Chinese races, thus European races showed fast embryonic development. Control(treatment A) and treatment C showed faster development than other treatments. And treatment D and F showed stable individual stage among all treatments. The test of shape characteristics and embryo which were conducted in hatching period showed 61% of high line succession possibility in average of 6 treatments. But treatment A and B showed no hatching, 3 lines of treatment C, 48 lines of treatment D, 1 line of treatment E, and 29 lines of treatment F showed slow development. And treatments D and F which showed stable embryo condition had highest possibility. The two treatments D and F showed good result among six treatments, and the preservation period of treatment D and F are 235 days and 310 days, and exposure period in $-2.5^{\circ}C$...was longer than other treatments. Numbers of hatched lines of treatment D and F are 48 and 29, and occupied 15.6% and 9.4% of all tested lines, respectively. Average hatching ratio of treatment D and F were 54.% and 71.6%, and average dead egg ratio were 12.6% and 2.4%, respectively. These results show that average ratio of hatching dead eggs in treatment D and F are higher than general line. Thus reconsideration of hatching condition on treatment D and F is needed.

Breeding Status of the Black Woodpecker (Dryocopus martius) in Gayasan National Park (가야산국립공원 까막딱다구리의 번식 현황)

  • This study was conducted from April 2012 to June 2012 to analyze breeding status of Black woodpeckers (Dryocopus martius) in the Gayasan National Park. Nest height, tree diameter which a nest built and altitude at the nest site of Black woodpeckers were 17.4 m, 63.8 cm and 587 m, respectively. A pair of Black woodpeckers completed a clutch in 4 eggs on 7 May and 3 of eggs hatched on 24 May 2012. Both female and male incubated eggs and never left eggs without one of parents. A male more contributed to build a nest than a female. Incubation duration was higher in a male than in a female. Nestlings made a begging call in a nest during the growing period. Begging frequency of nestlings increased from 11 days up to 20 days after hatching. After then, nestlings reduced begging and fledged a nest 24 days after hatching. This study provided information of breeding ecology to protect and manage population of Black woodpeckers which are endangered species in South Korea.

Ontogenetic Development of the Digestive System in Chub Mackerel Scomber japonicus Larvae and Juveniles

  • Chub mackerel, Scomber japonicus, larvae and juveniles were reared from hatching to 35 days after hatching (DAH), and the development of their digestive systems was histologically investigated. The larvae were initially fed on rotifers and Artemia nauplii starting around 19 DAH, and thereafter on Artemia nauplii, fish eggs, and a formulated feed mixture. The primitive digestive system differentiated at 3 DAH; the digestive tract was distinctively divided into the buccopharyngeal cavity, esophagus, stomach, air bladder, intestines, and rectum. The gastric gland and pyloric caeca first appeared at 5 and 7 DAH, respectively. The stomach was divided into cardiac, fundic, and pyloric regions in the preflexion phase. The number of gastric glands and pyloric caeca, as well as the volume of the gastric blind sac increased markedly, with development continuing into the juvenile stage. The precocious development of the digestive system during the larval period might be related to the early appearance of piscivory, which is able to support high growth potential. The organogenesis results obtained for this precocial species represent a useful tool to aid our understanding of the physiological requirements of larvae and juveniles to ensure optimal welfare and growth under aquaculture conditions, which will improve current rearing practices of this scombrid species.

Quality Analysis of the Free Amino Acids during the Early Development Stages of Hynobius leechi (한국산 도롱뇽(Hynobius Leechi BOULENGER 의 초기발생단계에 있어서의 유리 아미노산의 정성분석)

  • Free amino acids at five different developemntal stages (Gastrulation-Hatching -out stage) of Hynobius leechi BOULENGER were analyzed qualitatively by the use of paper paitition chromatography. It was found that the number of free amino acids increased as the development proceeded. The free amino acids identified at each stages are as follows : Gastrulation stage : Alaninie, Aspartic acid, Glutamin acid, Histidine, Methionine. Neural plate formation stage : Alanine , Aspartic acid, Glutamic acid, Glycine, Histidine, MEthionine, Phenylalanine, Proline, Serine, Trypotophan. Middle tail-bud stage : Alanine, Arginine, Asparagine,Aspartic acid, Citrulline, Glutamic acid, Glycine, Histidie,Hydroxyproline, Proline, Leucine, Methionine, Ornithine, Phenylalanine, Serine, Threonine, Tryptophan. Late tail-bud stage : Alanine, Arginine, Asparagine, Aspartic acid, Citrulline, Glutamic acid. Glycine, Histidine, Hydroxyproline, Leucine, Methionine, Ornithine, Phenylalanine, Proline, Serine, Threonine, Tryptophan, Valine. Hatching -out stage : the same with the late tail-bud stage. It seems probable that the metabolic systems of amino acids before and after the middle tail-bud stage are quite different from each other. Before the middle tail=-bud stage, the reaction system of amino acids is thought not to be completed while after that stage the system has been completed , because in the former period of the development , the number of freeamino acids increased rapidly with the development , and after that stage, there is practically no change in the number of free amino acids.

Egg Development of the Ussurian Bullhead Fish, Leiocassis ussuriensis (Pisces: Bagridae) and Morphological Development of Its Larvae and Juveniles

  • This study was examined the ovogenesis of Ussurian bullhead, Leiocassis ussuriensis and the morphological development of its larvae and juveniles and to use the results as basic information for the preservation of species and resource enhancement. For artificial egg collection, human chorionic gonadotropin (HCG) was injected at a rate of 10 IU per gram of fish weight. During breeding period, water temperature maintained at $24.5{\sim}26.5^{\circ}C$ (mean $25.0{\pm}0.05^{\circ}C$). The process of ovogenesis reached the two-cell stage in 50 minutes after fertilization. In 73 hours of fertilization the movement of the embryoid body became active state and the larvae began to hatch from the tail through the oolemma. Length of prelarvae were 6.33~6.50 mm long (mean $6.40{\pm}0.06mm$) just after hatching having yolk with their mouth not opened. After thirty eight days of hatching, juveniles were 30.6~32.5 mm long (mean $31.5{\pm}0.65mm$). The color was dark yellowish brown throughout the entire body, and the number of caudal fin rays developed to thirty six perfectly.

Screening and Utilization of Antagonistic Plants to Control Northern root-knot Nematode in Ginseng Fields (인삼포장에 발생하는 당근뿌리혹선층의 방제를 위한 길항식물의 탐색)

  • This study was conducted to screen the antagonistic plants on northern root-knot nematode (Meloidogyne) and to utilize those in its control. Egg hatching of M. hapla was found to be inhibited by 17 plant methanol extracts, and 11 plant extracts among them were also toxic to M. hapla second stage juvenile. Egg hatching of iW. hapla was also found to be inhibited by squeezed extracts of Cassia tora and Zea mays, and they were also toxic to M. hapla second stage juvenile. Extracts of Achyranthes japonica, Melia axedrach and Acorus graminens were toxic to M. hapla second stage with a juvenile mortality above 70clc at the 10 folds diluted concent ration and A. graminens was toxic to tested juvenile mortality above 50% at the 100 folds diluted concentration. The toxicity was directly propotional to the diluted concentration of the plant extracts and to the exposure period. Punica granatum, Acorns graminens and Melia axedrach were effective in inhibiting root penetration of JW. hapla juveniles, among of them p. granatum is most effective Percent inhibition of penetration by second and third stage juveniles into tomato slants penetrating by it was 72.7 and 82.4%, respectively.

  • Studies were carried out to investigate physiological and biochemical analysis of recessive lethal mutant "nmn" and to observe the cuticle formation and dermal gland. All nmn homozygous larvae continued to eat a few mulberry leaves, and died without entering into molt. In case of artificial hatching, eggs had a higher nmn individual segregation ratio than in hibernating eggs. The percentage of nmn individuals with the hatching days was alike during 3days period. As a result of histological observation, nmn mutants dermal gland was different from normal dermal gland in form and size. Normal was formed new cuticle in the middle of the molting, but not in nmn mutant. Total protein content in haemolymph of the normal larva was more than that of nmn smutant and there was a difference between normal and nmn mutant protein components.omponents.

Distributional Data and Ecological Characteristics of Parnassius bremeri Bremer in Korea (붉은점모시나비의 국내 분포정보 및 생태적 특성 조사)

  • This study was carried out to survey and confirm the occurring sites of Parnassius bremeri in Korea, and to investigate ecological characteristics to develop a mass rearing technique. In the field survey, adults were found in the two previously known sites in Gyungnam Province and another site was newly found in Samcheok, Gangwon Province. Emergence period of adults was from middle of May to middle of June. Oviposition took place on various material, including hostplant, debris, dead leaves, etc. Eggs were laid singly, up to 126.7 eggs per female. The egg-period was 221.3${\pm}$2.3 days, eggs were hatched from 11th to 22nd of January in the natural condition, and started to feed for about 10 days after hatching. Survival rate of the 1st larvae was 67.6%. Developing period of each instar in the insectary (25$^{\circ}C$, 75% RH, 16L:8D) was 11.2 days for the 1st instar, 7.3 days for 2nd, 12.8 days for 3rd, 16.2 days for 4th, and 18.2 days for 5th, and the pupal period was 21.3 days. The average longevity of adults was 26.2 days. Oviposition rate was higher in the natural condition with enough space to fly for 3♀ : 1 ♂ coupled, at least 3 ${\times}$ 3${\times}$4 m-sized room, than in smaller cage. In a comparison of the preference for visiting on sugar source, black sugar and fructose were effective.

Study on the Hatching Characteristics and Diet of the Stick Insect, Baculum elongatum (Phasmida: Phasmatidae) for Artificial Mass Rearing

  • Baculum elongatum has several peculiarities such as parthenogenesis, unique external features, changing body color, and dropping oviposition, which makes it a potential economically useful insect. Oviposition was performed by females without fertilization by the sperm of males. The oviposition period was $42.2{\pm}22.7$ days and the number of eggs per female was $109.5{\pm}70.5$ eggs. The hatch rate was 73.3% at $25^{\circ}C$ and 66.7% after low temperature treatment ($8^{\circ}C$ for 60 days). In nature, B. elongatum overwinters as an egg, but it can be assumed from the results that cold temperatures were not required for hatching. The hatch rate was 98.2% in the treatment using floral foam, fermented sawdust, and leaves. The developmental period was $100.9{\pm}4.2$ days for eggs, $55.3{\pm}4.6$ days for nymphs, and $49.7{\pm}16.0$ days for adults. The length of the eggs was $0.33{\pm}0.0$ cm and the lengths of the nymphs were clearly distinguishable according to the instar stage. Clover(Trifolium repens) was an excellent diet as it was similar to the host plant and could be used as an alternative diet. The rate of reaching adulthood for the insects was 66.7% on an artificial diet containing 25% acacia leaves. For sustainable mass rearing of nymphs or adults of B. elongatum, a natural diet could be used such as acacia (Robinia pseudoacacia), white oak (Quercus aliena), chestnut (Castanea crenata var. dulcis), and bush clover (Lespedeza bicolor), or an alternative diet such as clover or artificial diet.

  • We investigated the effects of rearing water temperatures during sex differentiation period on sex ratios in olive flounder, Paralichthys olivaceus. Control and gynogenetic diploid juvenile flounder reared at water temperature of 18, 21, 24 and $27^{\circ}C$ for 65 days from 35 to 100 days after hatching. Fish were sampled to examine sex ratios at 195 or 260 days after hatching. Female ratios of control and gynogenetic diploid flounder declined rapidly as water temperature increased. Sex ratio of gynogenetic diploid reared at $27^{\circ}C$ was very close to 1 : 1 ratio (P<0.01), The survival rates of control and gynogenetic diploid reared at $27^{\circ}C$ were different from other water temperature groups. The growth of body weight of control and gynogenetic diploid reared at $18^{\circ}C$ were different from other water temperature groups.
