ICM - Trophectoderm Cell Numbers of Mouse IVF/IVC Blastocysts (체외생산된 생쥐 배반포기배의 ICM과 Trophectoderm 세포수에 관한 연구)

  • This work has been carried out to examine the number of Total, ICM and TE cells of F1 mouse blastcysts at day 4 after IVF by differential labelling of the nuclei with polynucleotide-specific fluorochromes and to obtain a fundamental information of preimplantation mouse embryo development. Blastocysts produced by superovulated B6CBA F1(C57BL/${\times}$CBA) eggs were inseminated with $1{\times}10^6$spermatozoa/ml and cultured in M16 medium at $37^{\circ}C$, 5% $CO_2$ incubator for 95hrs. Blastocysts were classified as early, middle, expanded and hatching stage according to the developmental morphology; blastocoel expansion and zona thickness. The results obtained in these experiments were summarized as follows; 1) The development rate of blastocysts at 95hrs after IVF was 86.7% and classified blastocysts to early, middle, expanded and hatching were 16.3%, 18.9%, 10.5% and 40.9%, respectively. 2) The numbers of total blastomere using bisbenzimide in the classified blastocysts to early, middle, expanded and hatching were 35.6${\pm}$1O.4, 49.4${\pm}$8.6, 60.8${\pm}$1O.7 and 62.7${\pm}$13.9, respectively. 3) In ICM and TE cell number by using differential labelling with polynucleotide-specific fluorochrome in the classified blastocysts to early, middle, expanded and hatching; ICM numbers were 9.6${\pm}$3.0, 13.6${\pm}$3.9, 16.0${\pm}$3.3 and 19.5${\pm}$4.6, respectively and TE cell numbers were 30.6${\pm}$5.1, 39.9${\pm}$5.8, 42.2${\pm}$8.1 and 43.7${\pm}$11.1, respectively. These results showed the same increase pattern according to development advance level. Also, when compared with the results of total count were obtained between bisbenzimide only and differential labelling, both of them showed the same increase pattern according to development level and at the same time their cell numbers were almost the same. So, rapid and simple cell count method using differential labelling can be used for the examination of later preimplantation development or as an indicator of embryo quality according to the variables of culture conditions.

Early Larval Growth of Flesh Prawn (Fenneropenaeus chinensis) (대하(Fenneropenaeus chinensis)의 초기 유생성장)

  • Early larval growth for total length (TL) and body weight (BW) in flesh prawn (Fenneropenaeus chinensis) was studied from hatching to 25 days after hatching. Mean total length at nauplius and zoea stages was 0.21-0.39 mm and 0.62-1.03 mm, respectively. Mean total length of mysis stage grew from 1.28 mm (9 days after hatching) to 5.98 mm (21 days after hatching). The increase of body weight during experimental period showed step by step growth pattern due to metamorphosis of this species. The relationship between BW and TL from hatching to 25 days after hatching was BW = 2E-5TL/sup 4.1707/ (r²= 0.9746).

  • Microstructure of otoliths and early growth in total length of laboratory-reared river puffer, Takrifugu obscurus were observed from 1 to 63 days after hatching. The total length of Takifugu obscurus was 3.09${\pm}$0.02 mm, 4.96${\pm}$0.24 mm, 5.39${\pm}$0.42 mm,9.13${\pm}$0.59 mm, 10.78${\pm}$0.96, 16.40${\pm}$2.31 mm, 30.49${\pm}$l.53 mm in 1, 10, 20, 30, 34, 43, 63 days respectively. The mean radius of otoliths was 14.20${\pm}$1.21 $\mu\textrm{m}$ in 1 day, 31.06${\pm}$2.31 $\mu\textrm{m}$ in 10 days,57.19${\pm}$6.19 $\mu\textrm{m}$ in 20 days,127.13${\pm}$13.13 $\mu\textrm{m}$ in 34 days,190.79${\pm}$17.08 $\mu\textrm{m}$ in 43 days and 281.85${\pm}$23.81 $\mu\textrm{m}$ in 63 days after hatching. Growth of otoliths were similar to the growth pattern in total length during 20-63 days after hatching. Hatch mark was distinguished with other growth increments and within a hatch mark two or three increments were observed. The number of growth increments was correlated with the days after hatching between 20 and 63 days after hatching.

Studies on Early Life History of the Korean Mandarin Fish, Siniperca scherzeri(Perciformes: Centropomidae) (쏘가리 Siniperca scherzeri 의 초기 생활사에 관한 연구)

  • The early history of Korean mandarin fish, Siniperca scherzeri was studied to obtain some information required in aquaculture and reinforcement of natural population. During the period from June 1996 to July 1997, the mature adults of Siniperca scherzeri were collected from the middle Soyang Lake at Puksan - myon, Chunchon - shi, Kangwon - do, Korea. The eggs from females were obtained by injecting HCG or/and GnRH - a and fertilized by dry method in the laboratory. The fertilized eggs, measuring 1.70~2.10 mm in diameter and expanded to 2.20~2.66mm after absorption of water, were globosity, light orange yellow, separative, submergence and had one large oil globules of 0.5~0.7 mm. The blastodisc was formed in 1 hour and cleavage started in 1 hour 30 min. after fertilization, and the intervals of each stage of cleavage was about 50 min. at the water temperature of $21{\sim}24^{\circ}C$. Hatching occurred 131 hours 30 min. after fertilization and newly hatched larvae were 5.86~6.85 mm in total length(TL) and numerous stellate melanophores were distributed on the yolk and abdomen of caudal peduncle. The yolk was almost absorbed and the teeth development. 3 days after hatching, at 6.98~7.60 mm TL. The head spines and the teeth were largely developed and all fins were completely formed and became postlarva stage 15 days after hatching, at 10.10~12.90 mm TL. The body shape and the color pattern were similar to adult, 25 days after hatching, at 15.3~23.8 mm TL. In 5 months after hatching were reached at 154.10~175.02 mm TL and 49.32~82.67 g in body weight.

Egg Development and Early Life History of Zacco koreanus (Pisces: Cyprinidae) (참갈겨니 Zacco koreanus (Pisces: Cyprinidae)의 난 발생과 초기생활사)

  • We investigated developmental stages of egg and early life history of the Korean indigenous fish, Korean Chub Zacco koreanus from the South Han River in 2011 for phylogenetic study and conservation of this species. Eggs of Zacco koreanus were artificially fertilized by the dry method in the laboratory. The fertilized eggs were demersal, almost spherical in shape, transparent and unpigmented, with a pale yellow yolk, and no oil globule and average $3.09{\pm}0.07$ mm (n=10) in diameter. The hatching of the embryo took place in about 68 hrs after fertilization under water temperature of $20.0{\sim}23.0^{\circ}C$ and the newly hatched larvae were average $10.30{\pm}0.40$ mm (n=10) in total length (TL). Six days after hatching, the larvae grew up to $16.12{\pm}0.42$ mm (n=8) in TL and york sac absorption, mouth and anus opening were shown postflexion larvae stage. 17 days after hatching, most of fin-rays appeared at $18.21{\pm}0.38$ mm (n=6) in TL and brown spot appeared on the abdomen. 27 days after hatching, the larvae were brought up to $20.01{\pm}1.12$ mm(n=5) in TL and all their fin-rays were formed. 120 days after hatching, the larvae (juvenile) were $23.29{\pm}3.12$ mm (n=10) in TL and their body shape and color pattern were similar to the adult fish.

Relationship Among Reproductive Traits and Brood Production Pattern of Caridean Shrimp, Palaemon gravieri (Decapoda: Caridea: Palaemonidae)

  • Reproductive traits of Palaemon gravieri such as embryo size, number of embryo (fecundity), incubation period, larval development mode, larval development period, larval survival and larval growth were described and compared to analyze the correlation among those traits. Embryo volume is a primary factor determining other ensuing reproductive features. Egg volume was $0.042mm^3$ in the first developmental stage. Embryo volume in P. gravieri was comparatively small which is indicative of great number of embryo (y = 3.0161x + 0.0185 $R^2$ = 0.74 positive isometric relationship) and relatively long incubation period. Larvae survived from zoea 1 to post-larvae and it took 45 days at $22^{\circ}C$. Survival rate of the larvae was rather great in the early stage and thereafter steadily decreased. Daily growth rate of larvae in P. gravieri at $22^{\circ}C$ was 0.0195 mm on average. They grew steadily as time went by. Incubation period was between 10-14 days at $22^{\circ}C$. Larval development mode was almost complete planktotrophic. PNR (point of no return) appeared to be the third day on average. Survival rate of larvae without feeding declined rapidly between 3 and 4 days. Larval development period and stage frequency were 23-30 days and 11 stages which imply prolonged larval period and high mortality. The pattern of brood production followed fast successive parturial pattern. Most ovigerous female had mature ovary when they performed parturial molt soon after hatching (larval release).

Reproductive Pattern of the Epifaunal Amphipod Pontogeneia rostrata (Crustacea) on Dolsando Sandy Shore in Korea

  • Reproductive patterns of an epifaunal amphipod, Pontogeneia rostrata, were studied on Dolsando sandy shore in Korea. The life history pattern was iteroparous, with recruitment mainly occurring from winter to spring. The sex ratio was malebiased, especially during breeding periods. The mean body length of females was significantly larger than that of males. Brood size and egg volume were positively related to the body length of ovigerous females. There was no significant difference in brood size between successive breeding periods, but egg volumes were significantly higher in early winter (December) than in late spring breeding (May and June), increasing the probability of survival to hatching. These traits contribute to more reproductive potential in early winter than in late spring breeding. The mean brood size of epifaunal P. rostrata was larger but the mean egg volume smaller than that of infaunal amphipods in this sampling area. We suggest that reproductive effort for epifaunal species may be proportionally greater than for infaunal species in risky environments.

  • The early life history of black bullhead, Pseudobagrus koreanus, endemic to Korea was investigated to get biological information needed in artificial production of seedlings and in recovering natural resources. The fertilized eggs showed some characteristics in having heavy sticky material and minute folds which is formed radical pattern on the egg membrane. The shape of egg was spherical and $2.59{\pm}0.08$(2.45~2.70, n=10)mm in diameter. The yolk had not oil globule. The first cleavage was observed 2 hrs after insemination at $21{\sim}23^{\circ}C$, and the progressive cleavage were done about 30 min. interval. The characteristic changing of the yolk surface started at morula stage and continued to the end of gastrula. Hatching was started 72 hrs and completed 90 hrs after fertilization. The size of the larvae were 5.41~5.81mm in total length and 2.76~2.94mm in preanal length, and the number of so mites was 15-16+33~34(48~50). The barbels and swimbladder were completed and all the fins except second dorsal were appeared 1 week after hatching. The larvae attained 9.67~10.52mm in total length and 5.20~5.65mm in preanal length. All the fin sets and color pattern were completed 2 weeks after hatching and body mucus was secreted at that stage. The juvenile attained 14.59~16.02mm in standard length, 3.31~4.16mm in head length and 8.07~9.31mm in prenal length.

Quality Assay of Human Fetal-Cord Serum for Human IVF-ET with Mouse 2-Cell Embryos (생쥐 2-세포배아에 의한 시험관아기 배양용 대아제대혈청의 절적평가에 관한 연구)

  • The purpose of this study was to examine the qualitative variation of human fetal-cord sera (HCS) and to accept the sera in human lVF-ET program. One hundred and sixteenth RCS were tested with 1772 2-cell embryos of F1 (C57BL x CBA) virgin mice, Ten to sixteenth embryos were cultured in m-KRB medium with a aliquot of each serum (10%, v/v) or with bovine serum albumin(O.4%, w/v) as a control medium. Embryonic development were recorded at every 24hr for 4 days by such events as cellular compaction, cavitation, and hatching. In the control groups of eight assays, 98.1%(106/ 108) of 2-ce1l embryos developed above expanded blastocyst and the embryonic development was unified through the tests. But the developmental pattern in medium with each serum was various. Namely, the sera that supported development of 100% 2-cell embryos to above morula, early blastocyst, expanded blastocyst and hatching blastocyst was 45,7%(53/116) , 35.3%(41/116), 15.5%08/116.) and 6.9-%(8/116), respectively. And the sera that supported development of above 80% 2-cell embryos to the each embryonic stage was 92.2% (107/116), 83.6%(97/116), 63.8%(74/116) and 36.2%(42/116), respectively. Meanwhile two kinds of toxic pattern to the embryonic development were observed in some sera. The first pattern is that some sera arrested development of most embryos in pre- or post-stage of morula or blastocyst. The second pattern is that some sera promoted or arrested a part of embryos in the same dish. The ability of serum was depended on the batch of serum. Finally we could accept 69%(80/116) of the tested sera for human IVF-ET program. The base line for acceptance was the ability that supported above 80% 2-ce1l embryos to blastocyst. But some deterious sera were contained in this range. We cut off about 10% of the sera (83.6% , 97/116) that passed the baseline. This final percent of sera was similar to that of grade N of this study.

Spatio-temparal Pattern Formation of Abdominal Muscle in Xenopus Iaevis

  • The final pattern of the skeletal muscle of a vertebrate depends on the position-specific behavior of the muscle precursor cells during early developmental process and the abdominal muscle is made of cells which migrate a relatively long distance from their original tissue, myotome of dorsal mesoderm. We report the spatia-temporal migration pattern of abdominal muscle in Xenopus laevis by in situ hybridization and immunohistological studies. Shortly after hatching tadpole stage (stage 31/32), a group of myotomal cells detaches from the lower tip of the second somite and migrates ventrally to the lower position of abdomen. At stage 34/35, a second cell group migrates away from the third somite. Total 7 myotomal cell groups migrate ventrally one by one from the second to eighth myotome along their own pathways through the cell free space located between epidermis and subepidermal layer of the abdomen. During migration, the sizes of the cell groups (abdominal muscle anlagens) are increased to several tens fold. Around stage 40 all the abdominal muscle anlagens reaches their final positions and are interconnected side by side rostrocaudally. They are also connected to other types of muscles, forming a large multisegmented abdominal muscle. Heat shock study suggests that the disruption of segmentation of somites does not block the detachment of abdominal muscle anlagen, though the treatment gave stage- and dosagedependent effects on the migration speed.
