Occurrence of viral nervous necrosis(VNN) in red drum(Sciaenops ocellatus) larvae (홍민어 Sciaenops ocellatus에서의 바이러스성 신경괴사증 viral nervous necrosis)

  • Mass mortalities occurred among red drum larvae, 20 to 30 days old, culturing at hatcheries on southern costal area. No specific external signs were observed except abnormal swimming and spinal deformity. It was, however, suspected as a viral etiology due to high mass mortalities so that histopathological and molecular biological study was performed to evaluate the agent. Both vacuoles and necrosis were observed on nerve cells of brain and eye by H-E staining, and viral particles were observed on electronmicroscopic examination. On the other hand, DNA fragment, approximately 426 bps, was amplified with RT-PCR. The above results were able to diagnosis the etiological agent of mass moralities in red drum larvae as VNN(viral nervous necrosis)virus.

Energy Budget for Larval Development of Pandalus hypsinotus BRANDT (도화새우, Pandalus hypsinotus의 유생발생)

  • Zoeal stage's larvae of pandalid shrimp Pandalus hypsinotus, is distributed off the East sea and esteemed as a valuable shrimp resource in Korea, were reared in $10^{\circ}C$ temperature-controlled chambers and inverstigated the energy budget. The total energy intake per larva of zoea I to VI stages fed on Artemia nauplii was 140.88 J. The energy loss by respiration, molting, and excretion were 16.22 J, 1.19 J, and 106,40 J, respectively. The amount of energy used by growth was 17.07 J. Pandalid larvae assimilated $24.47\%$ of the ingested food. The gross efficiency ($K_1$) calculated by the equation of (growth+exuviae)/ingestion $rate{\times}100$ was $12.96\%$, and the net growth rate ($K_2$) calculated by the equation of (growth rate + exuviae)/(growth rate+ exuviae+ respiration rate)${\times}100$ was $52.96\%$. The percentage used for somatic growth and maintenance among the assimilated energy were $49.51\%$ and $47.04\%$ respectively.

Genetic Similarity and Variation in the Cultured and Wild Crucian Carp (Carassius carassius) Estimated with Random Amplified Polymorphic DNA

  • Random amplified polymorphic DNA (RAPD) analysis based on numerous polymorphic bands have been used to investigate genetic similarity and diversity among and within two cultured and wild populations represented by the species crucian carp (Carassius carassius). From RAPD analysis using five primers, a total of 442 polymorphic bands were obtained in the two populations and 273 were found to be specific to a wild population. 169 polymorphic bands were also produced in wild and cultured population. According to RAPD-based estimates, the average number of polymorphic bands in the wild population was approximately 1.5 times as diverse as that in cultured. The average number of polymorphic bands in each population was found to be different and was higher in the wild than in the cultured population. Comparison of banding patterns in the cultured and wild populations revealed substantial differences supporting a previous assessment that the populations may have been subjected to a long period of geographical isolation from each other. The values in wild population altered from 0.21 to 0.51 as calculated by bandsharing analysis. Also, the average level of bandsharing values was $0.40{\pm}0.05 $ in the wild population, compared to $0.69{\pm}0.08$ in the cultured. With reference to bandsharing values and banding patterns, the wild population was considerably more diverse than the cultured. Knowledge of the genetic diversity of crucian carp could help in formulating more effective strategies for managing this aquacultural fish species and also in evaluating the potential genetic effects induced by hatchery operations.

Induction of Changes in Morphology, Reactive Nitrogen/Oxygen Intermediates and Apoptosis of Duck Macrophages by Aflatoxin B1

  • The purpose of this study was to investigate the effects of aflatoxin $B_1$ ($AFB_1$) on the ultracellular morphology alteration, apoptosis induction and reactive nitrogen and oxygen intermediates production of peritoneal macrophages (DPM) from mule ducks. The ducklings were purchased from a commercial hatchery, and were fed a corn-soybean based diet. As the ducklings were grown up to 3 wk of age, the Sephadex-elicited peritoneal exudative cells (PEC) were used as the source for duck peritoneal macrophages. The ultracellular morphology study showed that significant number of cells shifted from category I (normal cell with ruffled membrane) and II (cell membrane blebbing) to category III (cell membrane blebbing and even rupture) after DPM were incubated with $AFB_1$ ($20{\mu}g/ml$) for 12 to 48 h. When DPM were exposed to $AFB_1$ in vitro, the production of NO, $H_2O_2$ and $O_2{^-}$ in macrophages was reduced after 12-48 h incubation with previous LPS stimulation. There was a DNA laddering pattern observed in DPM incubated with $AFB_1$ 5, 10, 20, 50 or $100{\mu}g/ml$ for 12 h. Evidence also revealed that the percentage of apoptotic cells was increased along with the elevation of $AFB_1$ concentration. The results suggest that $AFB_1$ exposure causes duck macrophages going on apoptotic pathway through evidence of ultracellular morphology alteration and DNA laddering in agarose electrophoresis. The production of reactive nitrogen and oxygen intermediates of duck macrophages also depressed after $AFB_1$ exposure, and this implied that $AFB_1$ could cause deteriorated functions of bacteriocidal and tumoricidal activity in duck macrophages.

Effects of 3,5,3'-triiodo-L-thyronine ($T^3$) on Growth of GR-transgenic Coho Salmon, Oncorhynchus Kitsutch

  • GH-transgenic coho salmon (Oncorhynchus kitsutch) juveniles in tGH*T$_3$and tGH*PTU were fed with the diets containing 1 ug/g fish of 3,5,3'-triiodo-L-thyronine (T$_3$) and 30 ug/g fish of 6-n-propyl-2- thiouracil (PTU), respectively, to assess the effect of these drugs on the change of physiological activity, growth and survival rate in comparison with normal transgenic (tGH*C) and nontransgenic coho salmon (Wild) for 90 days. Although the daily food intakes of all transgenic (tGH)-groups were higher than Wild, the amount was reduced by exogenous PTU supply. The fred efficiencies of tGH-groups were lower than Wild, but the efficiency was reduced both by T$_3$and PTU. The survival rate of tGH-group was significantly higher than that of Wild, but there was no significant difference among tGH-groups. Although the growth of tGH-coho salmon was faster than Wild. the growth rate of transgenic salmon was increased by exogenous T$_3$, but was reduced by PTU Plasma TT$_4$levels of tGH-groups was approximately 2-fold higher relative to Wild, but there were no difference of plasma TT$_4$levels among tGH-groups. plasma TT$_3$level or tGH-coho salmon was increased by exogenous T$_3$administration, but was reduced by exogenous PTU. In addition, although plasma GH levels of all tGH-groups were higher than that of Wild, the GH level in plasma of transgenic coho salmon was increased by exogenous T$_3$and reduced by exogenous PTU. In the meantime, the transgenic fishes also displayed head, jaw and opercular abnormalities typical of the offsets of this gene construct in coho salmon, indicating that some imbalance in growth processes has been induced. However, the abnormalities of transgenic coho salmon was reduced following exogenous PTU administration.

Comparison of Performances between Brown and White Egg Layers (백색산란계와 갈색산란계의 생산성 비교)

  • On account of the recent improvement in performance of brown layers, the market share taken by brown layers has increased to about 50% in the world and to almost 100% in Korea. There are several other reasons why the industry has moved from white to brown, such as : brown layers are used to be more robust, more docile and easier to manage ; e brown layers are easier to sex at the hatchery ; brown layers lay less second grade eggs, due to a better shell Quality ; brown eggs seem to be more attractive than white ; and a clear consumer preference, thus a better price per egg. More recently, however, the trend towards brown eggs has been slowing down. The main reasons for this lie in that white layers can still produce an egg at a lower cost and that white eggs have better de-shelling properties, easier candling and higher yolk and solid content of the liquid egg which are benefits for egg processing industry. Although the performance of the brown layers is still improving, there are increasing opinions in the poultry industry that the market portion of white layers should be increased based on the following reasons, such as : shell color has no effect on the nutritive value of eggs ; . brown layers consume more feed ; the percentage of meat spots is significantly higher in brown eggs than in white eggs ; . brown layers are less efficient in the second cycle of production than in the first ; white layers are more resistant to the disease of fowl typhoid. In order to increase the market share of white layers in Korea, it may be needed to enlighten the consumers not to prefer the brown and large eggs and to inform the excellencies of white eggs widely.

Induced Ovulation by using Human Chorionic Gonadotropin and Gonadotropin-Releasing Hormone Analogue plus Pimozide in Yellow Puffer, Takifugu obscurus (인간의 태반성 성선자극호르몬 또는 성선자극호르몬-방출호르몬 유도체와 Pimozide에 의한 황복의 배란유도)

  • Ovulation of maturing female yellow puffer, Takifugu obscrus, was induced by using single injection of human chorionic gonadotropin (HCG) or gonadotropin releasing hormone-analogue (GnRH-A) $des-Gly^{10}[D-Ala^6]$ GnRH-ethylamide plus pimozide. The response was evaluated using the fertilization and embryo-formation rate after insemination and the gonadotropin (GTH) level in blood plasma using radioimmunoassay. In the fertilization and embryo-formation, maximal effects were recorded by using 1,000 IU/kg HCG or $10\;{mu}g/kg$ GnRH-A plus 5 mg/kr pimozide. Pimozide (1, 5 mg/kg) or GnRH-A treatment alone was not effective in elevation of GTH level, however combinations of these treatments were particularly effective. Injection of dopamine blocked the rapid elevation of plasma GTH levels of blood. These data suggest that yellow puffer secrete GnRH and gonadotropin-releasing-inhibiting factor during the spawning or the other period.

Changes of Antioxidant Enzyme Activity in Bagrid Catfish, Pseudobagrus fulvidraco Exposed to Diethylhexyl Phthalate (Diethylhexyl Phthalate에 노출된 동자개, Pseudobagrus fulvidraco의 항산화 효소활성의 변동)

  • The effects of diethylhexyl phthalate (DEHP) on various oxidative stress responses in liver, kidney and gill tissues of freshwater bagrid catfish Pseudobagrus fulvidraco were investigated under laboratory conditions. Bagrid catfish were intraperitoneally injected with sunflower seed oil containing nominal concentrations of 0, 300 or 900mg DEHP per kilogram of body weight for 3 days and the effects after last injection were assessed in liver, kidney and gill tissues of the exposed organisms. The oxidative stress responses of fish were evaluated by analyzing the level of glutathione (GSH), as well as the activities of antioxidant enzymes such as glutathione S-transferase (GST), glutathione peroxidase (GPx) and glutathione reductase (GR). After exposure to the DEHP, there were significant decrease in GR, GPx activity and GSH content in liver of fish exposed to 900 mg DEHP per kilogram of body weight compared to the control group. Compared with the control group, significant decreases in renal GPx and GR activity were observed in the DEHP treatment groups (900 mg $kg^{-1}$ bw). However, no significant difference was observed in any oxidative stress responses in gills between the DEHP-treated and the untreated group of fish. The findings of the present investigation show that DEHP induce oxidative stress and the liver was the most affected organ followed by the kidney and gills. Furthermore, the changes of GPx and GR activities may be important indicators of oxidative stress responses but additional study is required to confirm the oxidative stress of DEHP.

Differences in Egg Quality and Larval Development among Four Populations of Sea Squirt Halocynthia roretzi Adults (난질과 유생발생을 이용한 4개 멍게(Halocynthia roretzi) 어미계군 특성 비교)

  • To compare four populations of sea squirt Halocynthia roretzi adults, their egg quality and larval development were investigated in the hatchery. The populations were: south sea wild (SSW), south sea cultured (SSC), east sea wild (ESW) and Iwate Japan cultured (IJC). Egg quality and larval development were compared using 13 factors (fertilization rate and diameter, proximate composition, amino acid and fatty acid contents of eggs, hatching rate and various sizes of tadpole larvae) which were obtained from each population. Fertilized egg diameter, hatching rate and size of tadpole larvae were significantly different among the four populations (P<0.05). Eggs were produced with higher crude protein ($5.20{\pm}0.00%$ SSW, $4.71{\pm}0.01%$ ESW, $4.66{\pm}0.01%$ SSC and $3.96{\pm}0.01%$ IJC) and lipid ($1.22{\pm}0.01%$ ESW, $1.01{\pm}0.00%$ SSW, $0.77{\pm}0.01%$ SSC and $0.69{\pm}0.00%$ IJC,) contents from domestic wild populations than from Japanese or cultured populations. Also amino acid and fatty acid contents were different. The extent of similarity between domestic and Japanese populations (30.5% IJC:SSW, 34.3% IJC:SSC and 40.7% IJC:ESW) was relatively low but was very high between SSW and SSC (73.9%). These results may have been due to differences in the abundance of food types and environmental conditions in the four localities and consequent differences in the diets of the sea squirts.

Selection of Copepods as Live Food for Marine Fish Larvae Based on Their Size, Fecundity, and Nutritional Value

  • Copepods are a major food source for marine fish larvae in nature. Many studies on copepods culture have been conducted to develop a new live food for the seedling production of marine fish larvae. But fish farmers still depend on rotifer and Artemia nauplii. This study was carried out to find suitable copepods as live food for the larvae in hatchery. Eight species of copepods (1 calanoid, 2 cyclpoid, 5 harpacticoid) that were fed Isochrysis galbana were examined in terms of the size of nauplii, fecundity, amino acids, and fatty acids contents. These species were divided into small (nauplii length 46-86 ${\mu}m$) and large (nauplii length 120-188 ${\mu}m$) size group. Nitokra spinipes in the small group and Tigriopus japonicus in the large group showed the highest fecundity with 151.1 and 139.6 nauplii production per gravid female, respectively. With regard to nutrients, essential amino acids were the highest with 21.2% in cyclopoid Paracyclopina nana in the small group and n-3 HUFA were the highest in calanoid Pseudodiaptomus inopinus (8.5 ${\mu}g/mg$) in the large group and P. nana (8.8 ${\mu}g/mg$). In terms of the size, fecundity, and nutritional value of copepods examined in this study, N. spinipes and P. nana seem to be suitable copepod species to develop as a new live food for small mouth fish larvae.
