In vitro and in vivo evaluation of erdosteine capsule

  • Purpose. The purpose of this study is to compare in vitro dissolution characteristics and bioavailability in beagle dog of a hard gelatine capsule containing erdosteine (Yuhan Erdosteine capsule$\^$TM/) with those of commercial product (Erdos capsule$\^$TM/). Methods. Yuhan Erdosteine capsule$\^$TM/ was prepared using erdosteine 300 mg, lactose, magnesium stearate, and others by powder filling method. The dissolution characteristics of Yuhan Erdosteine capsule$\^$TM/ and Erdos capsule$\^$TM/ were determined by USP dissolution apparatus 2. (omitted)

  • Purpose. To develop a hard gelatine capsule containing meloxicam (Yuhan Meloxam capsule$\^$TM/), in vitro dissolution characteristics and bioavailability in beagle dog were compared with commercial product (Mobic capsule$\^$TM/). Methods. Meloxicam capsule$\^$TM/ was prepared by powder filling method using meloxicam, lactose, magnesium stearate, and others. The release of Meloxicam capsule$\^$TM/ and Mobic capsule$\^$Tm/ were monitored by USP dissolution method under various dissolution donditions - dissolution medium (pH 1.2, 4.0, 6.8 and water).   • Water vapor sorption of corn starch in various protective film coated capsules in 100% RH chamber for 14 days were as follows: The percent of water vapor sorption were 28.63% in uncoated capsule, 25.16% in hydroxy propylcellulose(HPC) coated capsule, 15.59% in 2-methyl-5-vinyl pyridine-methyl acrylated-methacrylic acid (MPM) coated capsule and 15.50% in polyvinyl acetal diethyl amino acetate(AEA) coated capsule. 2. Water vapor sorotion of magnesium trisilicate in various protective film coated capsules in 100% RH chamber for 14 days were as follows. The percent of water vapor sorption were 13.91% in uncoated capsule, 13.30% in HPC-coated capsule, 10.87% in MPM-caated capsule and 9.9% in AEA-coated capsule. 3. Water vapor sorption of magnesium carbonate in various protective film coated capsules in 100% RH chamber for 14 days were as follows: The percent of water vapor sorption were 12.48% in uncoated capsule, 10.72% in HPC-coated capsule, 8.10% in MPM coated capsule and 7.8% in AEA-coated capsule. 4. MPM-coated capsules and AEA-canted capsules were mere effective to protect water vapor sorption than HPC-coated capsules.

컴퓨터 시각장치를 이용한 자동 캡슐 검사장치 (Automate Capsule Inspection System using Computer Vision)

  • In this study, we have developed a prototype of the automatic defects detection system for capsule inspection using the computer vision techniques. The subjects for inspection are empty hard capsules of various sizes which are made of gelatine. To inspect both sides of a capsule, 2-stage recognition is performed. Features we have used are various lengths of a capsule, area, linearity, symmetricity, head curvature and so on. Decision making is performed based on average value which is computed from 20 good capsules in training and permission bounds in factories. Most of time-consuming process for feature extraction is computed by hardware to meet the inspection speed of more than 20 capsules/sec. The main logic for control and arithmetic computation is implemented using EPLD for the sake of easy change of design and reduction in time for developement. As a result of experiment, defects on size or contour of binary images are detected over 95%. Because of dead zone in imaging system, detection ratio of defects on surface, such as bad joint, chip, speck, etc, is lower than the former case. In this case, detection ratio is 50-85%. Defects such as collet pinch and mashed cap/body seldom appear in binary image, and detection ratio is very low. So we have to process the gray-level image directly in partial region.

  • Stability is impaired in capsules in which there is a transfer of water vapor from capsule to the contents of the capsules. The water vapor transfer between AEA-coated capsules and corn starch, magnesium trisilicate in the closed container was examined. Experimental eguilibrium relative humidity after water vapor transfer and estimated values calculated from a formula of these substances were in close agreement with each other.

사이클로스포린 A 경질캅셀제에 대한 생물학적 동등성 평가 (Bioequivalence of Cyclosporin A Hard Capsules)

  • The bioequivalence of two cyclosporin A products was evaluated in 26 normal male volunteers (age 25 ~33 yr, body weight 56~84 kg) following single oral administration. Test product was a hard capsule containing the granule of cyclosporin A (Chong Kun Dang Corp., Korea) and reference product, Sandimmun", was a soft capsule containing surfactant, oil, alcohol and cyclosporin A (Sandoz, Swiss). Both products contain 100 mg of cyclosporin A. Four capsules of the test and the reference product were administered to the volunteers, respectively, by randomized two period cross-over study (2$\times$2 Latin square method). Average drug concentrations at each sampling time and pharmacokinetic parameters were not significantly different between two products (p>0.05); the area under the concentration-time curve to last sampling time (24 hr) (AU $Co_{24}$) (5034.8$\pm$ 1760.6 vs 4635.4$\pm$ 1158.9 ng . h/ml), maximum plasma concentration ( $C_{max}$) (1002.7$\pm$353.1 vs 980. 4$\pm$ 171.7 ng/71), and mean residence time (MRT) (6.16$\pm$0.81 vs 5.64$\pm$0.50 h). The differences of mean AUC 7-,4,7~, T_ and MRT between the two products (7.93,2.22,16 and 8.39%, respectively) were less than 20% given as a guideline. The power (1-$\beta$) and treatment difference ($\Delta$) for AU $Co_{24}$, $C_{max}$ and MRT were more than 0.8 and less than 0.2, respectively. Although $T_{max}$ of the two products was significantly different each other (p<0.05), $T_{max}$ might be an insignificant parameter because cyclosporin A generally requires long-term administration. From these results, the two products are bioequivalent.alent.t.

병솔꽃나무(Callistemon citrinus (Curt.) Skeels)의 개화결실 및 증식특성 (Characteristics of flowering, fruit setting and propagation of Callistemon citrinus (Curt.) Skeels)

  • 본 연구는 외래도입 병솔꽃나무(Callistemon citrinus)의 개화결실 습성과 효율적인 증식방법을 구명 하고자 수행하였다. 병솔꽃나무의 삭과는 개화후 4년간 가지에 존속하며, 종자의 수와 크기는 3년생 삭과와 2년생 삭과가 비슷한 반면 1년생 삭과는 크게 차이가 나는 것으로 나타났다. 삭과로부터 종자를 탈리시키기 위해 건조시킨 결과 삭과의 상대 수분함량이 약 75%일 때 삭과열개율이 100%에 달했다. 발아율은 3$0^{\circ}C$에서 건조시킨 종자가 가장 높았으며, 그 중 2년생 삭과에서 탈리된 종자의 발아율이 높았다. 반면 50, 7$0^{\circ}C$에서 건조되어 탈리된 종자의 발아율은 낮았다. 가을철 2차 개화를 유도하기 위하여 봄철 개화 후 삭과가 달린 가지를 전정한 개체에서는 모든 개체에서 가을에 개화하였으며 , 개화량은 평균 14송이였다. 반숙지를 IBA 100mgι$^{-1}$ 에 24시간 침적처리한 결과 86.4%가 발근되었으며, 평균 12.6개의 1차근을 가지며 생장이 가장 양호하였다.

하이드록시프로필화 녹두, 고구마, 물밤 전분의 이화학적 특성과 하드캡슐 제조 (Physicochemical Properties of the Hydroxypropylated Mung Bean, Sweet Potato and Water Chestnut Starches for Hard Capsules Formation)

  • 녹두, 고구마, 물밤 전분에 propylene oxide를 0~18% 첨가하여 하이드록시프로필화 전분을 제조하여 하드캡슐 제조시 젤라틴 대체제로서의 가능성을 검토하였다. RVA 시험결과 propylene oxide 첨가량이 증가할수록 호화온도와 setback은 감소했고 최고 점도와 breakdown은 증가하였다. 팽윤력은 녹두의 경우 propylene oxide 양이 12% 이상 첨가되었을 때 급격히 증가하였고 고구마와 물밤은 9% 첨가했을때부터 증가하기 시작하였다. 투명도는 녹두가 다른 전분보다 가장 높았으며 propylene oxide 첨가량에 유의적으로 증가했고 고구마와 물밤의 경우 하이드록시프로필화 전분이 일반 전분보다는 증가했으나 propylene oxide의 양에 따른 경향성은 보이지 않았다. 붕해도 시험에서는 물밤의 경우 propylene oxide 6% 이상 첨가하였을 때 완전히 용해되었고 녹두와 고구마는 12% 이상 첨가되었을 때 완전히 용해되었다. 본 실험에서는 하이드록시프로필화 전분이 젤라틴 또는 동물성단백질을 기본으로 제조된 하드캡슐의 대체제로써 의약 산업에서 활용될 수 있는 가능성을 보여주었다. 본 연구결과를 토대로 하이드록시프로필화 전분 필름의 물성, 온도와 pH에 따른 붕해도 실험등이 추가 연구된다면 전분 하드캡슐로 상용화도 가능할 것이라 생각된다.

Preparation of Micro-/Macroporous Carbons and Their Gas Sorption Properties

  • Micro-/macroporous carbons (MMCs) were prepared using a hollow mesoporous silica capsule (HMSC) as a sacrificial hard template. The carbonization process after the infiltration of furfuryl alcohol into the template-free HMSC material afforded MMC materials in high yield. The hard template HMSC could be removed by HF etching without deteriorating the structure of MMC. The MMC materials were fully characterized by SEM, TEM, PXRD, XPS, and Raman spectroscopy. The replication processes were so successful that MMCs exhibited a hollow capsular structure with multimodal microporosity. Detailed textural properties of MMC materials were investigated by volumetric $N_2$ adsorption-desorption analysis at 77 K. To explore the gas sorption abilities of MMCs for other gases, $H_2$ and $CO_2$ sorption analyses were also performed at various temperatures. The multimodal MMC materials were found to be good sorbents for both $H_2$ and $CO_2$ at low pressure.

난용성 항진균제 이트라코나졸의 액상제제화에 의한 생체이용율 개선 (Enhanced Bioavailability of Itraconazole in Liquid Preparation)

  • This study was to develop an effective itraconazole liquid preparation which exhibits an enhanced bioavailability. The solubility of itraconazole was increased (72-fold) in itraconazole liquid preparation as compared with itraconazole powder. The dissolution rate of itraconazole was higher for itraconazole liquid preparation filled into a hard gelatin capsule with 90% release within 20 min as compared to 55% for $Sporanox^{\circledR}$capsules. The oral absorption of itraconazole liquid preparation and $Sporanox^{\circledR}$tablets were studied in the rat. The area under the concentration-time curve $(AUC_{0-24hr})$ of itraconazole liquid preparation ($90.25\;{\pm}\;8.36\;{\mu}g{\cdot}hr/ml$) increased by 6.2 times compared to that of Sporanox tablets ($14.58\;{\pm}\;1.26\;{\mu}g{\cdot}hr/ml$) after oral administration of itraconazole 15 mg/rat each. $C_{max}$ also increased to $6.87\;{\pm}\;1.15\;{\mu}g/ml$ after administration of liquid preparation $1.58\;{\pm}\;0.16\;{\mu}g/ml$ of $Sporanox^{\circledR}$tablets. These results indicate that in vivo bioavailability of itraconazole liquid preparation was significantly enhanced as compared with $Sporanox^{\circledR}$tablets.
