Evaluation of Sperm Sex-Sorting Method using Flow Cytometry in Hanwoo (Korean Native Cattle)

  • This study evaluated a method of sorting X and Y chromosomes based on size using the forward angle light scatter related refractive index (FSC) of a flow cytometer. Hanwoo bulls sperm were separated to X and Y chromosomes by the parameters of FSC or Hoechst 33342 intensity. As a result, using monitor program linked flow cytometry during sorting processing, the purities were $97{\pm}0.57$ or $96{\pm}0.67%$ for the X-fraction and $96{\pm}0.33$ or $97{\pm}1.33%$ for the Y-fraction in the two sperm sorting methods. There were no differences in the X and Y ratios (X and Y %) between the sperm sorting methods based on FSC or DNA content. The proportions of female and male embryos used for in vitro fertilization and development were $66.03{\pm}3.31$ or $69.37{\pm}1.41%$, and $70.56{\pm}2.42$ or $56.11{\pm}3.09%$ when sperm were processed using the sex sorting method by FSC or Hoechst 33342. In conclusion, further study is needed to determine the optimum procedure and improve the nozzle to enhancing sorting accuracy or efficiency. Also, the findings of this study do not negate the possibility that the difference method of sperm sorting cannot use a UV laser beam.

The Expression of Plasma Progesterone and Estradiol-17$\beta$ Level before Parturition in the Recipients Pregnant by Hanwoo SCNT Embryos (체세포 복제란 이식 한우의 분만 전 혈장 Progesterone과 Estradiol-l7$\beta$ 농도 변화)

  • This study was performed to analyze the characterization of plasma hormonal levels during pregnancy in the Hanwoo recipients pregnant by artificial insemination (AI) or somatic cell nuclear transfer (SCNT) embryos. The synchronized recipients pregnant by SCNT embryos produced by Hanwoo fetal fibroblast cells (n=8) and by AI (control, n=5) were used. The plasma hormonal levels were measured by RIA (P4 and E2) and ELISA (cortisol), respectively. In control, the increase of E2 and the decrease of P4 were occurred immediately before the initiation of parturition. The expression pattern of plasma P4 was similar in both groups from 50 to 10 days before parturition, however, it did not decrease even at the expected date of labor in the SCNT recipients. The plasma cortisol was expressed a lower level during pregnancy in the SCNT recipients. But, the cortisol was increased in the cow aborted around 100 days of pregnancy (n=1). Based on these results, it can be postulated that the failure of the hormonal changes immediately before parturition in the SCNT recipients may be one of the most important reasons for a delayed parturition in clone calving.

Superovulation Response by FSH Treatments in Hanwoo (FSH 투여 용량과 방법Ul 따른 한우의 과배란 처리 효율)

  • The present study was carried out to assess the effect of superovulation response and efficiency of embryo production induced with injection of FSH dissolved in polyethylene glycol (PEG) in Hanwoo. Eighty-eight cows were divided into four groups. In group 1, cattle were intramuscularly treated with twice-daily administration of 50mg FSH for 4 days. Group 2 and 3 were subcutaneously single injection of 400mg and 200mg FSH dissolved in 30% PEG, respectively. Group 4 were subcutaneously single injection of 200 mg FSH dissolved in 30% PEG at 7 day after CIDR insertion. The number of corpus luteum (CL) in group 2 resulted in significantly (p<0.05) higher compared to group 1, 3 and 4 (18.5 vs. 11.2, 13.1 and 13.9, respectively). However, the number of total ova $(7.9{\sim}10.4)$, transferable embryos $(3.7{\sim}4.7)$, degenerate embryos $(1.9{\sim}3.5)$ and unfertilized ova $(1.8{\sim}2.7)$ did not differ among treatment groups. No difference was observed in pregnancy rate after transferring the recovered embryos among groups $(36.0{\sim}50.0%)$. In addition, blood progesterone concentrations at embryo recovery did not differ among all groups. In conclusion, although no differences were observed in the number of total ova, transferable embryos and pregnancy rate after transfer, a single injection of reduced dose of FSH (200mg FSH) at 7 day after CIDR insertion is more practical for superovulation treatments than frequent injection because of reduction of stress in Hanwoo and decreases of cost and laber.

Potential functional roles of follistatin on bovine somatic cell nuclear transfer embryos

  • To demonstrate that follistatin treatment enhances the efficiency of nuclear transfer (SCNT), cell allocation and preimplantational development were determined in bovine SCNT embryos in the present study. Treatment of activated SCNT embryos with 10 ng/ml follistatin significantly increased the proportion of blastocyst development compared to untreated SCNT embryos. In addition, an increase in trophectoderm (TE) cell numbers and relatively higher proportion of TE cells to total cells were observed, but the number of inner cell mass (ICM) cell and total cell numbers were not changed (P < 0.05). No significant effect of other doses of follistatin was observed for the above endpoints. However, treatment with 1 and 10 ng/ml follistatin reduced the proportion of nuclear transfer blastocysts with an ICM ratio of > 60% relative to untreated nuclear transfer blastocysts at Day 7. No significant effect of follistatin treatment on proportions of nuclear transfer blastocysts with ICM ratio of 20-40% or 40-60% was observed. Taken together, these results suggested that follistatin can be used to increase developmental competence of SCNT embryos in terms of cell allocation, particularly TE cells, during preimplantation stages, subsequently enhancing placentation and birth of live offspring.

Effects of biological Factors on In Vitro Production of Hanwoo Embryos (한우 난포란 유래 배반포의 체외생산을 위한 생물학적 요인들의 영향)

  • This study was carried out to investigate the effect of biological factors on the in vitro production(IVP) of bovine oocytes for development of simple culture methods and medium. Oocytes from the slaughterhouse ovaries were matured and fertilized using general protocol and this study was examined if there were necessary to co-culture, media change, media type and embryo density. This results were as follows: 1. The development rate according to co-culture with cumulus cells and non co-culture as drop culture was not significantly different in cleavage (68.9 vs 71.7%), 8-cell stage (41.2 vs 44.1%) and blastocyst stage (12.2 vs 13.8%), respectively (p<0.05) 2. The blastocyst development rates in YS and CRIaa were higher than that in TCM199 (12.4, 10.4$ vs 3.7%), but the cleavage (69.0, 77.8 and 61.0%) and 8-cell stage (31.7, 37.0 and 35.7%) development accoring to YS, TCM199 and CRIaa ws not significantly different, respectively (p<0.05). 3. There was no significantly different in cleavage (62.6, 59.5 and 61.2%), 8-cell(34.7, 37.9 and 34.0%) and blastocyst (9.5, 11.6 and 12.8%) development among medium change time as control, Group I and Group II, respectively (p<0.05). 4. Blastocyst formation of 8-cell stage according to embryo density was not significantly different in 1, 10 and 25 embryos (27.3, 22.5 and 34.0%), respectively (p<0.05). These results indicated that simple culture system could reduce bovine IVP embryos as drop culture as non co-culture system, high density embryo (25 embryos/50 $\mu$1 drop). YS defined medium and no medium change for whole culture period, although other biological factors need to examine in order to produce efficient IVP bovine embryos.

Studies on the Embryo Transfer of In Vitro Matured/ In Vitro Fertilized Embryo in Hanwoo (한우 체외성숙.체외수정란의 수정란이식에 관한 연구)

  • These studies were carried out to improve the reproductive efficiency through embryos transfer of Hanwoo IVM/IVF embryos. Following routine IVM/IVF procedure, oocytes and zygotes were cultured far 40 to 44 h in CRlaa medium with BSA. Then 2 to 8-cell embryos were removed the cumulus cell and were cultured in CRlaa medium containing 10% fatal bovine serum and 2.5 mM taurine in 5% $O_2$ and 5% $CO_2$ at 38.5$^{\circ}C$. The fresh embryos of the morulae and blastocysts cultured for 6 to 9 days in vitro or the frozen-thawed embryos were transferred into recipients. The pregnancy rates of the blastocyst produced for 6, 7, 8, and 9 days in vitro culture were 59.4, 68.2, 66.0 and 100%, respectively. In the developmental stage, pregnacy rates of early blastocysts (61.1%), blastocysts(64.7%) and expanded blastocysts(69.5%) were higher than that of morulae stage(20.0%). The pregnancy rates according to the corpus luteum grades of A, B and C in recipients were 73.6, 62.9 and 50.0%, respectively. Effects of donor-recipients synchrony of after day 2, 1 and 0, before day 1 and 2 on the pregnancy rates were 35.7, 65.5, 72.6, 67.9 and 60.0%, respectively. Pregnancy rates of the body condition score of recipients $\leq$2(71.3%) were higher than those of $\geq$3.0 score(40.0%). The pregnancy rates according to the parity of recipients when embryo was transferred to cow(70.6%) was higher than in heifer(59.1%). The pregnancy rates according to hormone treatment before embryo transfer were 69.9% in hCG + GnRH administration group and 63.0% in control group. Fresh and frozen-thawed embryos on the pregnancy rates were 70.6 and 36.4%, respectively. Pregnancy rates in single and AI+single was 90.0% and 64.8%. Pregnancy rates in twin induction was better than in single. These results indicate that pregnancy rates after transfer were affected on the embryo ages, donor-recipient synchrony, body condition score of recipients, corpus luteum status, parity and hormone treatment to recipients.

The Studies on In Vivo Embryo Production and Transfer in Hanwoo - II. Factors Influencing Pregnancy Rate after Embryo Transfer (한우에 있어서 체내수정란의 생산과 이식에 관한 연구 - II 한우 수정란 이식이 수태율에 미치는 요인)

  • These studies were carried out to establish an effective in vivo embryo transfer methods in Hanwoo by analyzing several factors that influence this process. In an embryo transfer, recipients with grade A corpus luteum of the right ovary and that of grade B of the left one showed a higher pregnancy rate(p<0.1) than others. The pregnancy rates of frozen embryos were significantly lower(P<0.01) than those of fresh ones; the former resulting in 35% and the latter resulting in 56.2%. Transfer of embryos according to the estrus cycle(6.0 ∼ 9.0 days) did not show a significant difference in pregnancy rate with fresh embryos recording 45.4 ∼65.7% and frozen ones recording 22.0 ∼ 50.0%. According to the status of corpus luteum and embryo freezing or not, the pregnancy rate was higher on grade A corpus luteum with 40.8 ∼67.9% than B and C which ranged from 25.0∼56.0%. The results of embryo transfer according to the development stage and grade of embryos showed that regardless of the embryo's grade. transfer of morula recorded an average pregnancy rate of 46.3%. This results higher than the transfer of blastocyst which was 34.1%.

Production of Superior Cows by Sexed Embryo Transfer Using In Vivo Embryos in Hanwoo (한우 체내수정란의 성판별 후 이식으로 우수 암송아지 생산)

  • The objective of this study was to produce superior cows by sexual distinction of embryos collected from the donor with pedigree information. Collected embryos distinguished by biopsy using punching or bisection methods and Loop-mediated isothermal amplification of sex-determined DNA for sexing. Six embryos predicted as female were transplanted to recipients and then 2 pregnant cows were normally delivered of calves at 278 and 285 days after embryo transfer. Birth weights of calves named Barani and Borani were 18kg and 25kg, respectively. Adjusted body weights for 90 days were 61.1kg and 88.8kg, respectively. Average daily gains were 0.48kg and 0.71kg, respectively.

Effects of hCG Treatment on the Pregnancy Rates and Progesterone Concentrations in Hanwoo Recipients with SCNT Embryos (hCG 투여가 복제란 이식 한우 대리모의 임신과 Progesterone 농도에 미치는 영향)

  • This study was performed to investigate the effects of hCG treatment on pregnancy and delivery rates in the Hanwoo recipients. There were significantly higher pregnancy and delivery rates in the recipients treated with hCG at 7 days after artificial insemination (p<0.05), respectively. The SCNT embryos from bovine fetal fibroblast cells were transferred into the synchronized recipients. The recipients were administered saline (n=89) or hCG (1,500 IU) (n=48) at 7 days after heat, respectively. The pregnancy rate was significantly higher in the recipients treated with hCG compared to that of saline treated group (p<0.01), however, the delivery rate was not different in both treated groups. The concentration of plasma progesterone (P4) was not different in both groups before hCG treatment, but the P4 level was increased significantly in hCG treated group after hCG injection (p<0.05). Although the pregnancy rate was very high in early stage of pregnancy, it was decreased dramatically after 50 days of pregnancy and maintained basal level. Taken together, the treatment of hCG in the SCNT recipients after day 7 of heat was effective method to increase the P4 concentration and to increase the pregnancy rate. But it did not affect directly to delivery.
