Estimation of genetic parameters using real-time ultrasound measurements in Hanwoo (한우 암소의 생체 초음파 성적을 이용한 유전모수 추정)

  • This study was conducted to estimate genetic effects on economically important traits for genetic improvement in Hanwoo by using the real-time ultrasound measurements for longissimus dorsi muscle area (LMA), backfat thickness (BFT), and marbling score (Marb). The phenotypic data were obtained from 1,648 pedigreed cows, and general linear models were applied to test the effects of age, region, and body condition socre. The cows between 50 and 60 months of age had the greatest scores for LMA and BFT, and Marb (P<0.05). The cows in region C had the greatest scores for body condition socre, LMA and BFT, while in region J Marb was the lowest (P<0.05). There was positive relation with LMA, BFT, and Marb according to increase body condition socre. Heritabilities for LMA, BFT, and Marb were estimated as 0.136, 0.351, and 0.236, respectively. These results would provide primary information for the efficient implementation of genetic improvement schemes in Hanwoo.

Analysis of the influence of sex, slaughter season, and feeding system on carcass traits in Hanwoo

  • This study was conducted to investigate the effects of sex, environmental factors, and feeding system on the carcass traits of Hanwoo (Korean Native Cattle). Data were collected from 7,866 heads slaughtered in Chungnam province during one-year period. Using the collected carcass traits data which greatly influence a Hanwoo's carcass grade, the effects of sex class, slaughter season, and feeding system were estimated. Backfat thickness of steers was significantly higher than that of bulls (p < 0.05). Marbling score was also the highest in steers when compared with cows and bulls (p < 0.05). Live weight and carcass weight were significantly higher in winter than in summer (p < 0.05). However, backfat thickness was significantly lower in summer than in other seasons (p < 0.05). Marbling score was higher in spring and fall than in summer and winter (p < 0.05). In terms of feeding systems, TMR (Total mixed ration) and TMF (Total mixed fermentation feed) fed groups showed the highest carcass grade (p < 0.05). However, the group fed TMR and formula feed at the final fattening period showed the lowest performance (p < 0.05) and it is assumed that some stress was associated to the feed change. The results reconfirm that castration may be recommended in order to improve meat quality and marbling scores in bulls. There was no consistent trend of seasonal effects of slaughter on carcass traits although some traits were significantly affected. Regarding the feeding system, either TMR or TMF can be supplied to achieve high feed efficiency and good carcass characteristics in Hanwoo.

Comparison on genomic prediction using pedigree BLUP and single step GBLUP through the Hanwoo full-sib family

  • Objective: When evaluating individuals with the same parent and no phenotype by pedigree best linear unbiased prediction (BLUP), it is difficult to explain carcass grade difference and select individuals because they have the same value in pedigree BLUP (PBLUP). However, single step GBLUP (ssGBLUP), which can estimate the breeding value suitable for the individual by adding genotype, is more accurate than the existing method. Methods: The breeding value and accuracy were estimated with pedigree BLUP and ssGBLUP using pedigree and genotype of 408 Hanwoo cattle from 16 families with the same parent among siblings produced by fertilized egg transplantation. A total of 14,225 Hanwoo cattle with pedigree, genotype and phenotype were used as the reference population. PBLUP obtained estimated breeding value (EBV) using the pedigree of the test and reference populations, and ssGBLUP obtained genomic EBV (GEBV) after constructing and H-matrix by integrating the pedigree and genotype of the test and reference populations. Results: For all traits, the accuracy of GEBV using ssGBLUP is 0.18 to 0.20 higher than the accuracy of EBV obtained with PBLUP. Comparison of EBV and GEBV of individuals without phenotype, since the value of EBV is estimated based on expected values of alleles passed down from common ancestors. It does not take Mendelian sampling into consideration, so the EBV of all individuals within the same family is estimated to be the same value. However, GEBV makes estimating true kinship coefficient based on different genotypes of individuals possible, so GEBV that corresponds to each individual is estimated rather than a uniform GEBV for each individual. Conclusion: Since Hanwoo cows bred through embryo transfer have a high possibility of having the same parent, if ssGBLUP after adding genotype is used, estimating true kinship coefficient corresponding to each individual becomes possible, allowing for more accurate estimation of breeding value.

Protein Patterns on a Corpus Luteum during Pregnancy in Korean Native Cows

  • Luteal cells produce progesterone that supports pregnancy. Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism. In the present study, the corpus luteum (CL) in early pregnancy established from luteal phase and pregnant phase was analyzed. The first study determined progesterone changes in the bovine CL at day 19 (early maternal recognition period) and day 90 in mid-pregnancy and compared them to the CL from day 12 of the estrous cycle. CL alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF). Comparing CL from luteal phase to those from pregnant phase counterparts, significant changes in expression level were found in 23 proteins. Of these proteins 17 were not expressed in pregnant phase CL but expressed in luteal phase counterpart, whereas, the expression of the other 6 proteins was limited only in pregnant phase CL. Among these proteins, vimentin is considered to be involved in regulation of post-implantation development. In particular, vimentin may be used as marker for CL development during pregnancy because the expression level changed considerably in pregnant phase CL tissue compared with its luteal phase counterpart. Data from 2-DE suggest that protein expression was disorientated in mid pregnancy from luteal phase, but these changes was regulated with progression of pregnancy. These findings demonstrate CL development during mid-pregnancy from luteal phase and suggest that alternations of specific CL protein expression may be involved in maintenance of pregnancy.

Physicochemical Meat Quality, Fatty Acid and Free Amino Acid Composition of Strip loin, Chuck Tender, and Eye of Round Produced by Different Age Groups of Hanwoo Cow

  • This study was performed to investigate the influence of age on the physicochemical properties of strip loin (m. longissimus lumborum), chuck tender (m. supraspinatus), and eye of round (m. semitendinosus) of Hanwoo cows. Hanwoo cows (n=126; 24-194 mon; live weight, 270-500 kg) were slaughtered and three muscles were obtained according to 3 age groups (G1, < 5 years old; G2, 6-8 years old; G3, > 9 years old). The chuck tender had significantly higher protein contents in G3 than in G1 or G2 (p<0.05). For strip loin and chuck tender, G1 had significantly higher intramuscular fat contents than G3 (p<0.05). The chuck tender had significantly higher $b^*$ (yellowness) values for G2 than for G1 (p<0.05). The three muscles had significantly higher cooking loss (%) and lower WHC (%) in G3 than in G1 or G2 (p<0.05). WBS values of strip loin were significantly higher in G3 than in the other groups (p<0.05). The three muscles had higher C18:1n9 in G1 than that in the other groups. The total content of saturated fatty acids was significantly higher in G3 than in G1 for all 3 muscles (p<0.05). Regarding free amino acid contents, G1 had significantly higher contents of threonine, alanine, valine, methionine, phenylalanine, leucine, and lysine in the strip loin than G2 or G3 (p<0.05). In conclusion, young cow beef were higher in the WHC, intramuscular fat and free amino acids contents, whereas old cow beef had higher cooking loss and WBS (p<0.05).

  • The objective of this study was to investigate response of corpus luteum, recovery rate of embryos and production of transferable embryos according to superovulation by PEG 30% FSH in Hanwoo. Cows were selected as recipient, subsequent were superovulated with a total of 400 mg NIH-FSH-P1(Folltropin-V, Canada) given by one shot subcutaneously. At the time of five days after Folltropin injection, 25mg of a PGF$_2$a was injected and cows were inseminated 12 and 24h after the onset of estrus. Seven days after insemination, embryos were collected non-surgically and were cryopreserved by direct transfer methods. The results are summarized as follows : 1. Response of corpus luteum following the superovulation in Hanwoo, right ovary were 52.8%(271/513) and left ovary were 47.1%(242/513), respectively (P<0.05). 2. Recovery rate of embryos following the number of corpus luteum were 83.0%(426/513). 3. Mean number of embryos recovered and transferable embryos were 7.74 and 6.43, respectively (P<0.05). 4. In the total number of transferable embryos per flush were collected 6.4 and all saved transferable embryos were 355.

Effect of Recombinant Bovine Somatotropin (rbST) Treatment on Concentration of Progesterone, Volume of Luteal Tissue and Pregnancy Rate in Hanwoo (한우에서 rbST 처리가 Progesterone 농도와 황체의 크기 및 수태율에 미치는 영향)

  • The purpose of this study was to determine the effect of rbST treatment on progesterone concentration, volume of luteal tissue and pregnancy rate following embryo transfer. Recipient cows were assigned to control and rbST group, of which was given a single injection of rbST (500 mg. sc) at estrus detection. The concentration of progesterone was not significantly different between control and at 0, 3, 6 days after rbST treatment. However, the concentration of progesterone at 9, 12 days was significantly higher than in control group (4.6 and 6.8 vs. 3.9 and 4.5 ng/ml P4). The pregnancy rate after embryo transfer in rbST treatment was significantly higher than in control group (64.0 vs. 47.1 %; p<0.05). The results indicated that rbST treatment in recipient cows could be improved the efficiency of pregnancy rate after embryo transfer.

  • The purpose of this experiment is to compare the pregnancy rate (PR) according to the state of the ovaries and uterus, according to the number of embryos transferred from cows and heifers and to investigate the method of artificial twin induction with Hanwoo $in$ $vitro$ fertilized (IVF) embryos by embryo transfer (ET). Looking at the PR according to the condition of the ovaries and uterus, the result was not influenced by the condition of the ovaries, but was significantly influenced by the state of the uterus. The PR according to the number of embryos transferred from cows was 36.8%, 53.0%, 50.5% for 1, 2, and 3 embryos respectively, and although there was a higher frequency of twin calves with 3 embryos than with 2, the calving rate was the highest with 2 embryos. In case of heifers, the transfer of 1 embryo showed the best pregnancy and calving rate, and although the PR was similar with 2 embryos (67.7 versus 66.4), in case of 2 embryos transferred there was high frequency of embryonic loss( 6.1%) occurred when a cow was diagnosed at 28 and 53 d after ET, total loss (21.3%; sum of fetal death, abortion and stillbirth after pregnant diagnosis at 60 day).
