Current situation and future prospects for beef production in South Korea - A review

  • Hanwoo cattle are an important food source in Korea and their supply can have a major impact on meat availability for Korean consumers. The Hanwoo population was 1.8 million head in 2005 and gradually increased to 2.6 million in 2015. Per capita beef consumption has also increased, to 11.6 kg per year in 2015, and is expected to continue to increase. Because intramuscular fat percentage is a critical contributor to meat quality, Hanwoo cattle are fed a high-energy corn-based diet for long fattening periods. Long fed diet causes significant alterations in fat percentage in the loin muscle and other areas of the carcass. However, these long feeding periods increase feeding costs and beef prices. Recently, there has been increased Korean consumer demand for lean beef which has less fat, but is tender and priced more reasonably. These consumer demands on the Korean beef industry are driving differing beef production systems and also changes to the beef grading methodology. Korean government has made a significant investment to select bulls with favorable production traits using progeny testing. Progeny tested bull semen has been disseminated to all Hanwoo farmers. A beef traceability system has been employed for all cattle breeds in Korea since 2009. Hanwoo cattle are ear-marked with a 12-digit identification number from birth to slaughter. This number allows traceability of the management history of individual cattle, and also provides information to consumers. Traceability including management information such as herd, farm, year of birth, and carcass data can determine estimated breeding values of Hanwoo. For a sustainable Hanwoo industry, research scientists in Korea have attempted to develop feeds for efficient fattening periods and precision feeding systems based on genetic information for Hanwoo cattle. These initiatives aim to Korean consumer demands for beef and provide more precision management in beef production in Korea.

Analysis of Myosin Heavy Chain Isoforms from Longissimus Thoracis Muscle of Hanwoo Steer by Electrophoresis and LC-MS/MS

  • The purpose of this study was to analyze myosin heavy chain (MHC) isoforms in bovine longissimus thoracis (LT) muscle by liquid chromatography (LC) and mass spectrometry (MS). LT muscles taken from Hanwoo (Korean native cattle) steer (n=3) used to separate myosin bands by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The peptide queries were obtained from the myosin bands by LC-MS/MS analysis following in-gel digestion with trypsin. A total of 33 and 43 queries were identified as common and unique peptides, respectively, of MHC isoforms (individual ions scores >43 indicate identity or extensive homology, p<0.05). MHC-1 (IIx), -2 (IIa), -4 (IIb), and -7 (slow/I) were identified based on the Mowse score (5118, 3951, 2526, and 2541 for MHC-1, -2, -4, and -7, respectively). However, more analysis is needed to confirm the expression of MHC-4 in bovine LT muscle because any query identified as a unique peptide of MHC-4 was not found. The queries that were identified as unique peptides could be used as peptide markers to confirm MHC-1 (14 queries), -2 (8 queries), and -7 (21 queries) in bovine LT muscle; no query identified as a unique peptide of MHC-4 was found. LC-MS/MS analysis is a useful approach to study MHC isoforms at the protein level.

Detection of Quantitative Trait Loci for Growth and Carcass Traits on BTA6 in a Hanwoo Population

  • The purpose of this study was to detect quantitative trait loci (QTL) for growth and carcass quality traits on BTA6 in a population of Hanwoo cattle. Three hundred and sixty one steers were produced from 39 sires that were sired by 17 grandsires in the two Hanwoo farming branches of the National Livestock Research Institute of Korea, between Spring 2000 and Fall 2002. DNA samples were collected for all of the steers, sires and grandsires, and the phenotypes for six growth and carcass quality traits were measured at 24 months of age. Twelve microsatellite markers were chosen on BTA6 and a linkage map was constructed by using seven of the twelve markers. Then, a chromosome-wide QTL scan was performed by applying an Animal Model, in which effects of QTL alleles within the grand sires were fitted as a random term. Three QTL were detected at the 5% chromosome-wise level for backfat thickness, average daily gain, and final weight. The most likely positions for the QTL were in the proximal region, i.e. 0 cM, 35 cM, and 63 cM, respectively. Also, another QTL for longissimus dorsi muscle area was detected at the 10% chromosome-wise level at 67 cM. These results were, in general, consistent with our previous report, in which candidate gene analyses showed that a SNP near ILSTS035 flanked by BM4621 (62.5 cM) and BMS2460 (81.3 cM) was associated with final weight, carcass weight, average daily gain, and longissimus dorsi muscle area in the same Hanwoo population.

Identification of Candidate SNP (Single Nucleotide Polymorphism) for Growth and Carcass Traits Related to QTL on Chromosome 6 in Hanwoo (Korean Cattle)

  • As genetic markers, single nucleotide polymorphisms (SNP) are very appropriate for the development of genetic tests for economic traits in livestock. Several microsatellite markers have been identified as useful markers for the genetic improvement of Hanwoo. Among those markers, ILSTS035 was recently mapped at a similar position with four SNPs (AH1_11, AH1_9, 31465_446, and 12273_165) in a linkage map of EST-based SNP in BAT6. Among the four SNPs, two SNPs (31465_446 and 12273_165) were analyzed using BLAST at the NCBI web site. The sequences including the 12273_165 SNP were identified at the intron region within the LOC534614 gene on the gene sequence map (Bos taurus NCBI Map view, build 3.1). The LOC534614 gene represents a protein similar to myosin heavy chain, fat skeletal muscle, embryonic isoform 1 in the dog, and myosin_1 (Myosin heavy chain D) in Macaca mulatta. In cattle, the myosin heavy chain was associated with muscle development. The phenotypic data for growth and carcass traits in the 415 animals were analyzed by the mixed ANCOVA (analysis of covariance) linear model using PROC GLM module in SAS v9.1. By the genotyping of Hanwoo individuals (n = 415) to evaluate the association of SNP with growth and carcass traits, it was shown that the 12273_165 SNP region within LOC534614 may be a candidate marker for growth. The results of the statistical analyses suggested that the genotype of the 12273_165 SNP significantly affected birth weight, weight of the cattle at 24 months of age, average daily gain and carcass cold weight (p<0.05). Consequently, the 12273_165 SNP polymorphisms at the LOC534614 gene may be associated with growth in Hanwoo, and functional validation of polymorphisms in LOC534614 should be performed in the future.

한우의 도체 형질이 경제성에 미치는 영향 (The Effect of Carcass Traits on Economic Values in Hanwoo)

  • 본 연구는 한우 농가의 소득에 직접적인 영향을 미치는 경매단가(AP) 및 도체판매가(CP)에 영향을 미치는 도체 특성(근내지방도(MS), 배최장근단면적(LMA), 등지방 두께(BFT), 도체중(CW) 및 육량 지수(MYI))와의 상관관계를 조사하였다. AP의 경우는 근내지방도(MS)가 가장 높은 상관(r=0.73)을 나타내었으며, CP는 도체중이 0.71로 가장 높고, 다음으로 근내지방도가 영향력이 높게 나타났다. 하지만, LMA, MYI는 비교적 낮은 상관을 나타내었다. 이상의 결과 농가의 소득의 향상과 한우의 MS 및 CW 향상이 유의적인 상관도가 있었다.

Investigation of blood biomarkers related to meat quality and quantity in Hanwoo steers

  • Objective: This study was conducted to investigate the correlation between blood components and carcass traits, and to find the biomarkers related to meat quality and quantity in Hanwoo steers. Methods: One hundred twenty-six Hanwoo steers (8 to 9 months of age, body weight of $185.6{\pm}24.38kg$) were used to find the correlation between blood compositions and carcass traits. The steers were fed concentrate and rice straw (30 steers) or total mixed rations (96 steers) during the whole experimental period. Blood samples were collected from the jugular vein at the growing (8 to 12 months), fattening (13 to 23 months) and finishing phases (24 to 30 months). Steers were slaughtered at 30 to 31 months of age (body weight of $701.6{\pm}60.45kg$) and the carcass traits were evaluated. Blood metabolites and hormones were analyzed and the correlation coefficients and regression equations with carcass traits were determined. Results: Average concentrations of retinol, leptin and insulin were 1.10 IU, 30.34 ng, and 35.35 ng per mL of blood plasma, respectively. Retinol has negative correlations (p<0.01) with insulin and leptin. Blood insulin and total protein decreased with the age of steers, but retinol, aspartic acid transaminase (AST), glucose, cholesterol and triglyceride increased. In the finishing phase, significant (p<0.01) negative correlations occurred between blood retinol content and marbling score, and between blood AST content and longissimus muscle area of 13th rib, and the following regression equations were obtained: Marbling score $(1-9)=-0.009{\times}retinol(IU/100mL)+9.125$ ($R^2=0.643$), Longissimus muscle area $(cm^2)=-0.250{\times}AST(U/L)+112.498$ ($R^2=0.450$). Conclusion: It is possible to make highly marbled beef by controlling the blood retinol content during the fattening and finishing phases of Hanwoo steers. Accordingly, blood retinol and AST could be biomarkers for determining beef quality and quantity, respectively, prior to slaughter.

Correlation Analysis between the Breeding Value of Carcass Traits in Hanwoo (Korean Brown Cattle), Bos Taurus, L. and Spot Intensity on Two-dimensional Gel Electrophoresis

  • In order to investigate the genetic marker associated with economic performance in Hanwoo (Korean Brown Cattle), proteomic approach was used. Breeding values were estimated from performance tested steers. The top 20 and bottom 19 steers based on carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS) evaluation for one progeny testing period was used. Meat samples dissected from longissimus dorsi muscles were taken from the slaughter house and analyzed for two-dimensional gel electrophoresis. A total of 102 significant spots out of total 146 on each gel were detected and compared with the reference gel (synthetic gel) to be evaluated. Four candidate spots for marbling score were identified: 205, 84, 204 and 198. The study confirmed the relationship between breeding values of economic traits of Hanwoo cattle and spot intensity.

국내 한우산업 현황과 정밀축산을 활용한 한우고기 생산전망 (Current situation of Hanwoo industry and future prospects for precision livestock farming)

  • 한국 육우의 거의 90%를 차지하는 한우의 역사, 산업 특성 및 소고기 생산을 중심으로 한우의 현안과 미래를 논의했다. 한우산업에 많은 변화가 있었지만 여전히 한우는 한국 소비자에게 최고의 쇠고기 공급원으로서 프리미엄의 위치를 가지고 있다. 다른 농업 기반경제와 비교할 때, 한우 농민의 수는 최근 수십 년 동안 급격히 감소했으며 농장의 규모가 커지고 시스템이 보편화되었다. 수입 소고기와의 차별화, 사료비 증가, 송아지 가격의 증가, 토지 비용의 증가 등 한우산업이 직면 한 어려움을 해쳐나가기 위해서는 보다 젊고 똑똑한 농부들이 필요하다. 기존의 축적된 노하우와 새로운 기술을 잘 접목시켜 한우를 잘 기르는 사람들이 많이 생겨나야 하고 소득을 창출하여야 한다. 우리나라의 발전된 ICT기술과 농민들의 근면함이 잘 융합되어 고급데이터 들이 꾸준히 축적된다면 여러 가지 어려운 상황에서도 새로운 고급육 생산 프로그램을 개발할 수 있고 이를 활용한 원천기술 개발로 지속가능한 한우산업을 유지할 수 있을 것이다. 아울러 기존의 방식과 다르게 변화하고 있는 4차 산업혁명시대에 한국형 정밀축산을 개발하여 축산선진국으로 나아갈 수 있을 것이라 기대한다.

한우 성장단계 특이발현 유전자의 발현양상 분석 (Expression Patterns of the Differentially Expressed Genes During Growth Stages of Hanwoo(Korean Cattle))

  • 한우 성장단계 특이발현 유전자 탐색에 관한 연구(장 등, 2002)에서 선정된 후보유전자 중, EPV 20, TCTP 및 aldolase A를 비롯하여 24개월령 cDNA probe에 대하여 강한 signal을 나타낸 ADRP 유전자의 발현양상을 분석하기 위하여 성장단계별 한우 등심조직 시료를 사용하여 semiquantitative RT-PCR 및 northern blot 분석을 실시하였다. EPV 20 유전자는 한우 등심조직에서 성장단계에 따른 발현량 차이가 거의 없는 결과를 근거로, 근육조직에서 항상 발현되는 유전자로 판단하였다. 또한 발달단계에 따라 발현량 차이가 있는 것으로 보고된 aldolase A 유전자 역시 뚜렷한 발현량 차이는 없었으며, aldolase A의 muscle specific promoter와 근육분화 관련 transcription factor에 관한 연구를 통하여 근육분화 및 근육수축에 있어 aldolase A의 역할 및 조절작용을 밝힐 수 있을 것으로 사료된다. 성장관련 단백질로 알려져 있는 TCTP의 발현양상을 분석한 결과, 한우 등심조직에서 성장함에 따라 발현량이 약간 증가하는 양상을 나타내었다. 이와 같은 발현양상 분석결과로 TCTP 유전자는 성장과 관련된 기능이 있지만 동물의 성장에 있어 직접적으로 관여하지는 않을 것으로 판단된다. 지방세포 분화의 초기단계에서 발현량이 급증하고 lipid droplet 형성에 관여하며 지방축적과도 관련이 있는 것으로 보고된 ADRP 유전자의 transcript의 크기는 약 1.82 kb 이었고, 24개월령 한우 등심조직에서 발현량이 급격히 증가하였으며, 30개월령 조직에서는 감소하는 양상을 나타내었다. 이와 같은 결과로부터 ADRP 유전자를 한우 성장단계 특이발현 유전자로 선정하였으며, ADRP 유전자의 발현양상은 근육내 지방축적과 관련이 있을 것으로 추정하였다. 이후에는 발현조절 기작의 해명 및 근육내 지방축적과의 관련성 분석을 위하여 ADRP 유전자의 전체적인 구조 및 전사조절 인자 분석을 비롯하여 다른 지방대사 및 지방축적 관련 유전자와의 상호작용 및 다형성 탐색분석에 관한 연구가 필요 할 것으로 판단하였다.

한우(Bos taurus coreanae)의 CAST 유전자 내 변이지역 탐색 및 경제형질과의 연관성 분석 (Identification of Polymorphisms in CAST Gene Associated with Economic Traits in Hanwoo (Bos taurus coreanae))

  • Calpastatin (CAST)은 calpain의 억제제 역할을 하는 유전자이며 BTA7에 위치하고 있다. Calpain을 억제하는 CAST 유전자 내 다형성이 육질의 연도에 영향을 미치고 있는 것이 알려져 있다. 또한 여러 연구를 통해 calpain 시스템은 일반 골격근의 성장에 중요한 역할을 하는 것으로 보고되었다. 따라서 본 연구는 한우 집단의 CAST 유전자 내 변이 지역을 탐색하여 경제형질과의 연관성을 분석하기 위하여 실시하였다. CAST 유전자(Accession no. NC_007305) 단일염기변이지역을 탐색 결과 총 5개의 변이(109749T/C, 116151G/A, 109737G/A, 109823T/C, 109926G/A) 지역이 탐색 NCBI에 등록되어 있지 않은 새로운 변이로 본 연구를 통해 탐색되었다. 한우집단 내에서 탐색된 총 5개의 SNP를 대상으로 연관불균형(LD, Linkage disequilibrium) 분석을 수행하였다. 변이들 간의 연관불균형(LD) 정도가 가장 낮은 수준에 있는 2개의 변이지역(109926G/A와 116151G/A, $r^2$=0.038)을 대상으로 경제형질과의 연관성 분석을 실시한 결과 등심단면적(109926G/A, p<0.05)과 18개월령 체중(116151G/A, p<0.05)에서 유의적인 연관성이 검출 되었다. CAST유전자는 도축 후 숙성과정에서 고기의 연도를 증가시키는데 영향을 미칠 뿐만 아니라 가축의 성장에도 깊은 연관성이 있으며, 본 연구를 통해 CAST 유전자 내 변이는 성장 관련 형질과의 연관성이 유의적인 것으로 확인되었다.
