Adipose tissue deposited within muscle fibers, known as intramuscular fat (IMF or marbling), is a major determinant of meat quality and thereby affects its economic value. The biological mechanisms that determine IMF content are therefore of interest. In this study, 48 genes involved in the bovine peroxisome proliferator-activated receptor signaling pathway, which is involved in lipid metabolism, were investigated to identify candidate genes associated with IMF in the longissimus dorsi of Hanwoo (Korean cattle). Ten genes, retinoid X receptor alpha, peroxisome proliferator-activated receptor gamma (PPARG), phospholipid transfer protein, stearoyl-CoA desaturase, nuclear receptor subfamily 1 group H member 3, fatty acid binding protein 3 (FABP3), carnitine palmitoyltransferase II, acyl-Coenzyme A dehydrogenase long chain (ACADL), acyl-Coenzyme A oxidase 2 branched chain, and fatty acid binding protein 4, showed significant effects with regard to IMF and were differentially expressed between the low- and high-marbled groups (p<0.05). Analysis of the gene co-expression network based on Pearson's correlation coefficients identified 10 up-regulated genes in the high-marbled group that formed a major cluster. Among these genes, the PPARG-FABP4 gene pair exhibited the strongest correlation in the network. Glycerol kinase was found to play a role in mediating activation of the differentially expressed genes. We categorized the 10 significantly differentially expressed genes into the corresponding downstream pathways and investigated the direct interactive relationships among these genes. We suggest that fatty acid oxidation is the major downstream pathway affecting IMF content. The PPARG/RXRA complex triggers activation of target genes involved in fatty acid oxidation resulting in increased triglyceride formation by ATP production. Our findings highlight candidate genes associated with the IMF content of the loin muscle of Korean cattle and provide insight into the biological mechanisms that determine adipose deposition within muscle.
This experiment was prosecuted to estimate the carcass traits and their relationships in Hanwoo steers. The mean carcass weight(CWT), back fat thickness (BFT), meat, bone, intra- muscular fat (IMF) and eye muscle area (EMA) of steers were 300.27 kg, 9.23 mm, 58.99%, 12.92%, 28.08% and 74.00 $cm^2$, respectively. The pHB(before aging), pHA(after aging), cook loss before aging($CL_B$) and cook loss after aging ($CL_A$) were 5.32, 5.60, 16.43% and 19.21%, respectively. It was also found that the MS and FC were 3.67 and 3.17, respectively. The EMA and quantity index (QIX) were positively correlated with CWT, meat% and bone% but negatively correlated with IMF%. The BFT was positively correlated with CWT and meat% but negatively correlated with bone% and IMF%. The pH (both before and after aging) was negatively correlated with CL (both before and after aging), water loss during freezing (WLF) and FC. The muscle score(MS) was negatively correlated with CL but positively correlated with WLF and FC. The ARM, HRD, CHW and CTM increased with the increasing of fat% and sharply decreased with the increasing of meat% in the carcass.
Genetic parameters of carcass weight (CWT), dressing percent (DP), cook loss (CL), eye muscle area (EMA), back fat thickness (BFT), and meat tenderness in terms of mastication (MAS), shear force (SFR) and penetration (PEN) in Korean native cattle were estimated in this study. Effects of sire, location and their interaction on these traits were also evaluated. Sire effects were found to be significant on all the traits studied except for PEN. The CWT and DP were also significantly affected both by location (p<0.01) and by interaction effect between sire${\times}$location (p<0.05). The EMA was significantly (p<0.05) affected by location but not by interaction effect between sire${\times}$location. All the traits were positively correlated ($r_g$ and $r_p$) with each other except between CL and meat tenderness (negatively correlated). Moderate to high genetic correlations between CWT and other important traits were obtained; indicating that selection for CWT would lead to improve carcass quality. Heritability estimates were 0.64, 0.52, 0.37, 0.25, 0.19 and 0.18 for MAS, SFR, CWT, PEN, DP and EMA, respectively.
The objective of this research was to investigate the effect of immersion in Rhus verniciflua Stokes extract (RVSE) on the total reducing ability (TRA), protein oxidation and myoglobin oxidation of beef steak (Hanwoo longissimus muscle) stored with high oxygen-modified-atmosphere packaging (MAP) (HOMAP, 75% $O_2$+20% $CO_2$+5% $N_2$) and low oxygen-MAP (LOMAP, 0% $O_2$+20% $CO_2$+80% $N_2$) at $4^{\circ}C$ for nine days. RVSE induced TRA (p<0.05), metmyoglobin (MetMb) formation, and the CIE $H^{\circ}$ value but reduced the carbonyl content and R630-R580, as an index of the intensity of redness by oxymyoglobin, and the CIE $L^*$,$a^*$, and $C^*$ values. HOMAP maintained a lower TRA, MetMb concentration, and CIE $H^{\circ}$ value but had higher R630-R580 and CIE $L^*$, $a^*$, and $C^*$ values compared to LOMAP. Therefore, RVSE induced TRA and protein oxidation stability but reduced myoglobin stability in Hanwoo beef steak. In addition, the effects of HOMAP were opposite those of RVSE.
This study was conducted to determine effects of different selenium (Se) sources on performance, carcass characteristics, blood measures (whole blood Se concentration and plasma glutathione peroxidase (GSH-Px) activity), and Se concentrations in tissues of finishing Hanwoo steers (Korean native steers). Twenty finishing Hanwoo steers (average body weight=536${\pm}$23.4 kg, average age=approximately 20 months) were allotted to treatments in four groups of five steers per pen for 16 weeks preceding slaughter. Treatments were control (CON), spent mushroom composts from Se-enriched mushrooms (Se-SMC), selenized yeast (Se-Y), and sodium selenite (SS). Dietary Se levels of all treatments except CON were 0.9 mg Se/kg on the dry matter basis. Body weight was measured at the first and final day of trial, and blood samples were collected to analyze whole blood Se concentration and plasma GSH-Px activity at 2, 4, 8, and 16 weeks. At the end of trial, steers were slaughtered to collect muscle and liver samples for their Se analyses, and carcass data were recorded. In terms of dry matter intake, body weight gain and carcass characteristics, no significant differences among treatments were observed. Whole blood Se concentrations were significantly higher (p<0.05) for Se-SMC and Se-Y treatments than for CON at each collection period, with no significant difference between SS and CON. For weeks 2 and 8, there was no significant difference for whole blood Se concentration between Se-SMC and Se-Y, but for weeks 4 and 16, Se-Y treatments were significantly higher (p<0.05) than Se-SMC. No differences were observed for plasma GSH-Px activity between Se-SMC and Se-Y. The Se concentrations in hind leg and liver were significantly different among treatments (p<0.05) and those in both tissues ranked the greatest in Se-Y, followed by Se-SMC, SS, and CON treatments. However, tissue Se concentration for SS was not different from that for CON. These results showed that feeding organic Se sources such as Se-SMC and Se-Y enhanced Se concentration in tissues, while SS, the most common supplement of inorganic Se, was inefficient in Se deposition. Even though Se-Y had a higher Se concentration in tissues than Se-SMC, replacing Se-Y with Se-SMC in diets of beef steers would be an inexpensive way to increase Se concentration in beef.

This study was conducted to evaluate the effect of medicinal plant by-products (MPB) supplementation to a total mixed ration (TMR) on growth, carcass characteristics and economic efficacy in the late fattening period of Hanwoo steers. Twenty seven steers (body weight [BW], $573{\pm}57kg$) were assigned to 3 treatment groups so that each treatment based on BW contained 9 animals. All groups received ad libitum TMR throughout the feeding trial until slaughter (from 24 to 30 months of age) and treatments were as follows: control, 1,000 g/kg TMR; treatment 1 (T1), 970 g/kg TMR and 30 g/kg MPB; treatment 2 (T2), 950 g/kg TMR and 50 g/kg MPB. Initial and final BW were not different among treatments. Resultant data were analyzed using general linear models of SAS. Average daily gain and feed efficiency were higher (p<0.05) for T1 than control, but there was no difference between control and T2. Plasma albumin showed low-, intermediate- and high-level (p<0.05) for control, T1 and T2, whereas non-esterified fatty acid was high-, intermediate- and high-level (p<0.05) for control, T1 and T2, respectively. Carcass weight, carcass rate, backfat thickness and rib eye muscle area were not affected by MPB supplementation, whereas quality and yield grades were highest (p<0.05) for T1 and T2, respectively. Daily feed costs were decreased by 0.5% and 0.8% and carcass prices were increased by 18.1% and 7.6% for T1 and T2 compared to control, resulting from substituting TMR with 30 and 50 g/kg MPB, respectively. In conclusion, the substituting TMR by 30 g/kg MPB may be a potential feed supplement approach to improve economic efficacy in the late fattening period of Hanwoo steers.
The present study was conducted to examine the effect of a stair-stepped feed intake pattern on growth, feed efficiency, and meat quality of Hanwoo steers. Twenty-seven 11-month-old Hanwoo steers were randomly divided into three groups. The control group was fed according to the Korean steer feeding program, and the other two groups were fed according to an alternated feeding schedule of 3-2-4-2 months. During the first three months of the experiment, treatment group 1 (T1) and treatment group 2 (T2) were fed 20% and 30% less than the control group, respectively. For the following two months, the T1 group was fed 20% more than the control group while the T2 group was fed 20% less than the control group. In the third step, T1 and T2 groups were fed 20% and 10% less, respectively, than the control group for four months. In the last two months, T1 and T2 groups were fed 20% more than the control group. After the stair-step feeding trial, steers were fed concentrated feed ad libitum for five months. The altered feed intake pattern did not affect daily body weight gain. However, daily feed intake tended to decrease and growth efficiency tended to increase in the two treatment groups compared to the control group. Altered feed intake also affected blood metabolite levels. The serum glucose and BUN level of the T1 group increased in the first re-fed period compared to the T2 and control groups. The serum cholesterol level of the T2 group decreased in the first restricted-re-fed growth period compared to the T1 and control groups. The serum NEFA levels of the two treatment groups increased from the first restricted period compared to the controls. The serum insulin level of the T2 group increased in the last period compared to the T1 and control groups. Regarding meat yield index, the control group was significantly higher than the T2 group (p<0.05). Regarding meat yield grade, the carcass back fat thickness of the T2 group was significantly higher than the control group (p<0.05). In marbling score, the T1 group was the highest (4.9), followed by the control group (4.1) and the T2 group (4.0). These results indicate that using a stair-stepped growth pattern (T1) can contribute to improvements in growth efficiency and muscle marbling.

The present study was conducted with twenty-four Korean native (Hanwoo) steers to observe the effect of mixed oil and monensin supplementation and duration of feeding on c9,t11-CLA content in plasma and fat tissues. The steers were randomly assigned to three groups of eight animals each according to body weight. Hanwoo steers in the control group were fed the commercial concentrate for the late fattening stage. The other groups of steers were fed the same diet as control steers, but the concentrate was supplemented with high-$C_{18:2}$ oil mixture (soybean oil, sunflower oil, safflower oil) and fish oil at 6% level of concentrate (DM basis), and monensin (20 ppm). The second and third group of steers was fed the oil mixture supplemented diet with monensin for the last 10 weeks and 20 weeks, respectively, prior to being slaughtered. The oil mixture consisted of 45% soybean oil, 20% sunflower oil, 20% safflower oil and 15% fish oil. Average daily gain (p<0.049) and feed efficiency (p<0.018) of the steers fed the diet supplemented with oil mixture and monensin (OM-M) for 20 weeks were higher than those of the other groups of steers. Dressing percent, fat thickness and longissimus muscle area were not affected by the OM-M supplementation and duration of its feeding. The OM-M supplementation increased the content of total-cholesterol (p<0.0001-0.0007) and HDL-cholesterol (p<0.0001) in the plasma of steers compared to the control diet. The steers fed the OM-M diet had a higher proportion of c9,t11-CLA in plasma (p<0.048-0.044) than the control steers. Feeding the OM-M diet for 20 weeks increased the proportion of CLA in intramuscular (p<0.015), intermuscular (p<0.039) and subcutaneous (p<0.001) fat tissues compared with both steers fed the control diet and the OM-M diet for 10 weeks. Increased (p<0.007) proportion of total unsaturated fatty acids in steers fed the OM-M diet for 20 weeks compared to those in control steers was related to the increased (p<0.001) $C_{18:2}$ and decreased (p<0.001) $C_{18:0}$ proportions in subcutaneous tissue.
Effects of spent composts of selenium-enriched mushroom (Se-SMC) on plasma glutathione peroxidase (GSH-Px) activity and selenium (Se) deposition in finishing Hanwoo (Bos taurus coreanae) steers were investigated. Twenty-five Hanwoo steers (average body weight = 613 kg, average age = 22 months) were allotted to treatments in five groups of five steers per pen for 12 weeks preceding slaughter. Treatments were SMC alone (CON; 0.1 ppm Se), 0.3 ppm (0.3 Se-SMC), 0.6 ppm (0.6 Se-SMC), 0.9 ppm (0.9 Se-SMC), and 0.9 ppm (sodium selenite; SENI) Se. During the experimental period, blood samples were taken to analyze Se concentrations and GSH-Px activities. Muscle and liver samples were collected for analyses of Se contents after slaughter. Dry matter intake and body weight gain were not affected by Se-SMC or sodium selenite supplementation. Selenium concentration in the whole blood and GSH-Px activity in plasma were linearly increased (p<0.01) with increasing levels of Se-SMC. The whole blood Se concentration of SENI treatment was significantly higher (p<0.05) than that of CON treatment from 4 weeks, whereas there was no significant difference in GSH-Px activities between both treatments at 8 and 12 weeks. Selenium content in the hind leg and liver increased linearly (p<0.05) with increasing levels of Se-SMC, but those of SENI treatments were not significantly different from CON treatments. These results suggested that Se in the Se-SMC was highly bioavailable to blood and tissues of ruminants, especially compared with Se in the sodium selenite. Therefore, Se-SMC might be used not only as an inexpensive way of providing Se for ruminants but also as another way of producing Se-fortified beef.
The objective of this study was to investigate the effects of environments (farms born, testing groups, age at the tests, date at slaughter, ages at slaughter) on body weights at 6, 12, 18 and 24 months of ages, body type measurements at 18 months of age and carcass characteristics in Hanwoo steer populations that were collected from commercial farms and reared in a progeny testing station. Performances of a total of 1,838 steer calves set for tests from 2004 to 2008 were recorded. Carcass characteristics were the carcass grading results evaluated and data collected slaughter scores at 24 months of age. For growth traits of all age classes and body type traits measured at 18 months of age, farms born, test group and linear covariate of age at test were fit in the models. For carcass traits, date at slaughter and linear covariate of ages at slaughter were fit in the models. Effect of farm at birth was not significant for body weight at 24 months of age. Carcass weight, eye muscle area, yield score and back fat thickness were affected by dates at slaughter but not by the ages at slaughter. Marbling score, however, was affected by these two effects. Farms at birth did not seem to affect body type measures greatly. This study will be utilized for Hanwoo Steers genetic evaluation.
