The fat mass and obesity associated (FTO) gene plays an important role in the regulation of energy homeostasis, fat deposition and obesity. For this reason, the FTO gene is a physiological and functional candidate gene for carcass and meat quality traits in beef cattle. The objectives of this study were to identify SNPs in the exonic regions of FTO gene and to evaluate the association of these SNPs with carcass traits in Hanwoo (Korean cattle). In this study, we newly identified two exonic SNPs in Hanwoo population. The g.125550A > T SNP was located in exon 3 and the g.175675C > T SNP was located in exon 6. Genotyping of the two SNP markers was carried out using PCR-RFLP analysis in Hanwoo steers to evaluate their association with carcass traits. As a result, g.125550A > T SNP genotype was significantly associated with effects on marbling score. Animals with the AA and TT homozygous genotypes had a significantly higher marbling score (p < 0.001) than those with AT heterozygous genotype, and this was significant after Bonferroni correction of the significance threshold (p = 0.003). Dominance effect was also observed for the marbling score (P < 0.05) with higher marbling score of homozygous animals. However, no significant associations with meat quality traits were observed for the g.175675C > T SNP. Our results suggest that the exonic SNP g.125550A > T in the FTO gene may be used as a DNA marker for the selection of Hanwoo with higher marbling.
The purpose of this study is to demonstrate antimicrobial activities of the lactoferrin and its peptic hydrolysates obtained from the colostrums of Hanwoo(Korean native cattle) and Holstein cattle. In the measurement of antimicrobial activity to E. coli O111 and other microorganisms, bovine lactoferrin showed a higher antimicrobial activity than that of Hanwoo cow's lactoferrin . The minimal inhibitory concentration (MIC) of lactoferrin against E. coli O111 was exhibited 1.5mg/ml(Holstein) and 2.75mg/ml (Hanwoo). The same result was also observed between bovine lactoferrin hydrolysate (0.12mg/ml) and Hanwoo cow's lactoferrin hydrolysate (0.25mg/ml). In addition of lysozyme, antimicrobial activity of lactoferrin was increased.
The effect of isoprothiolane(di-isopropyl-1,3-dithiolan-2-ylidenemalonate) aganist fat necrosis in Hanwoo(Korean Cattle) sire was evaluated. The 10 heads of Hanwoo sire suffering from fat necrosis were given 50mg/kg body weight of isoprothiolane(0.2g/kg of Fujix, Japan) orally once a day for 8 weeks. In 30% of these, the size of the necrotic fat masses had decreased significantly 7 months after the adminstration. Isoprothiolane did not affect on live body weight and semen characteristics. However the sire affected with fat necrosis had higher MCHC(Mean corpuscular hemoglobin concentration) than normal sire in hematologic values 10 weeks after administration. Number of RBC(red blood cell) and PCV(packed cell volume) 10 weeks after administration had been increased than those before administration(p < 0.05). The serum concentrations of creatinine, triglyceride, and total cholesterol 10 weeks after administration were higher than those before administration while the concentration of glucose was vice versa. The isoprothiolane may reduce the oxidation of glucose, increase the glucose transfer to lipids, and increase blood supply to necrotic masses. These results indicate that isoprothiolane may be useful as the therapeutic agent aganist fat necrosis.
Recently, summer temperatures have frequently been abnormal in Korea owing to global warming. In summer, a decrease in feed intake rate and biological activity were observed in Hanwoo (Korean Native Cattle), leading to lower production rates in the industry. However, the precise scale of damage was not reported as with other animals of economic value. This study was conducted to investigate the effects of birth season on birth weight in Hanwoo. Data were collected from 100 local breeding farms from 2016 to 2019. A total of 41,081 Hanwoo calves were classified and analyzed by sex, year, month, and season (March-May, spring; June-August, summer; September-November, fall; and December-February, winter) of birth. The birth weight of Hanwoo calves differed according to birth month. The average birth weight of male calves was 30.47 kg and that of female calves was 28.16 kg. Hanwoo birth weight was the highest in March-born calves and the lowest in July-born calves. The birth weights of calves born in February, March, April, November, and December were significantly larger than those of calves born in July. In addition, the birth weight of Hanwoo calves from the summer was significantly lower than that of calves born in other seasons. Furthermore, Hanwoo steer slaughter age showed a negative correlation, whereas carcass weight had a positive correlation with birth weight. In the beef cattle industry, birth weight is a very important economic characteristic that is related to growth rate. These data will contribute toward planning the reproduction of Hanwoo and analysis of changes in characteristics of economic value owing to high temperatures.
As the number of scaled-up ranches increased and agile responses to market changes became possible, decision-making by Hanwoo cattle farms also began to affect short-term shipments. Considering the changing environment of the Hanwoo supply market and the response speed of producers, it is necessary quickly to grasp the forecast ahead of time and to respond accordingly in an effort to stabilize supply and demand in the Hanwoo market. In this study, short-term forecasting model centered on the supply of Hanwoo was established. The analysis conducted here indicates that the slaughter of Hanwoo males increases by 0.248 as the number of beef cattle raised over 29 months of age in the previous month increases by one, and 0.764 Hanwoo females were slaughtered under average conditions for every Hanwoo male slaughtered. With regard to time, the slaughtering of Hanwoo was higher in January and August, which are months known for holiday food preparation activities for the New Year and Chuseok in Korea, respectively. Simulations indicated that errors were within 10% in all simulations performed through the Hanwoo supply model. Accordingly, it is considered that the estimation results from the supply model devised in this study are reliable and that the model has good structural stability.
Despite the importance of mutation rate, some difficulties exist in estimating it. Next-generation sequencing (NGS) data yields large numbers of single-nucleotide polymorphisms, which can make it feasible to estimate substitution rates. The genetic substitution rates of Hanwoo and Holstein cattle were estimated using NGS data. Our main findings was to calculate the gene's substitution rates. Through estimation of genetic substitution rates, we found: diving region of altered substitution density exists. This region may indicate a boundary between protected and unprotected genes. The protected region is mainly associated with the gene ontology terms of regulatory genes. The genes that distinguish Hanwoo from Holstein in terms of substitution rate predominantly have gene ontology terms related to blood and circulatory system. This might imply that Hanwoo and Holstein evolved with dissimilar mutation rates and processes after domestication. The difference in meat quality between Hanwoo and Holstein could originate from differential evolution of the genes related to these blood and circulatory system ontology terms.
Hanwoo cattle have been genetically selected for enhancing meat quality and meat quantity since the last few decades. Beef consumption in South Korea has also increased to 11.6 kg per year in 2016 and is expected to continue to increase. For improving meat quality, Hanwoo cattle has been raised a high-energy corn-based diet for long fattening periods, which causes not increment of fat percentage in the loin muscle but increment of feeding costs. Consumer demands on the Korean beef industry are driving differing beef production systems and also changes to the beef grading system. For a sustainable Hanwoo industry, researcher in Korea have attempted to develop feeds for efficient fattening periods and precision feeding systems based on genetic information for Hanwoo cattle. Therefore, representatives of government, academics, and market association need to provide efficient collaboration for the future prospective or develop a precise management system for Hanwoo industry in Korea.

The objective of this study was to compare the effects of the levels of inbreeding on body weight traits between two breed populations, Hanwoo and Korea Brindle cattle. Birth weight (BW), weaning weight (WW), body weight at 6 months of age (W6) and yearling weight (YW). Records of 1,745 calves (1,513 from Hanwoo, and 232 from Korea Brindle calves) were collected from Livestock Research Institutes in Kangwon, Gyeongbuk and Chungbuk provinces. The least squares means (LSM) and their standard errors for BW, WW, W6 and YW were $25.4{\pm}0.1$ kg, $81.0{\pm}1.8$ kg, $146.1{\pm}3.7$ kg and $291.5{\pm}2.4$ kg, respectively in Hanwoo calves and $22.6{\pm}0.3$ kg, $79.9{\pm}2.3$ kg, $137.6{\pm}4.6$ kg and $249.3{\pm}6.6$ kg, respectively in Korea Brindle calves. Pedigree data showed that 14.8% (316 out of 2131) of Hanwoo was inbred and the average inbreeding coefficient was 0.0209 (2.09%). Inbreeding coefficients of ten calves out of 316 total inbred Hanwoo calves were 12.5% or higher, whereas those of the other 306 calves were less than 12.5%. Genotypes of the nucleotide substitution g.23655332G>C of SNP marker rs385360448 at Lysophospholipase I (LYPLA1) gene intron 7, were tested for their effect on the carcass traits of Hanwoo and Jeju Crossbred cattle ($F_1$ progeny produced between Hanwoo ${\times}$ Jeju Black cattle) populations on Jeju Island. In the Hanwoo steer population, the meats containing LYPLA1 rs385360448 G/- genotypes showed significantly higher marbling scores and greater texture indices, compared to those of rs385360448 C/C homozygous animals (p<0.05). However, the LYPLA1 genotypes were not associated with the levels of carcass weight, backfat thickness, eye muscle area (EMA), meat color, and fat color (p>0.05). On the other hand, in the JCC steer population, the LYPLA1 G/- harboring meats showed significantly greater EMA levels, compared to those of C/C homozygotes (p<0.05). The results of the present study indicate that the LYPLA1 genotypes could alter the levels of intramuscular fat deposition, texture index, and eye muscle area via phospholipid metabolism in the Longissimus dorsi muscle of the cattle. These findings suggested that LYPLA1 genotypes may effect molecular genetic markers in the improvement of carcass traits of Hanwoo and Jeju Black industrial cattle populations on Jeju Island.
This study was conducted to establish an individual identification system comprising of 19 microsatellite markers located on different bovine autosomes. The markers were typed on 257 animals from five cattle breeds. In total, 112 alleles were detected from the genotyping of 19 microsatellite markers. The average heterozygosities ranged from 0.292 to 0.824 and the polymorphic information content (PIC) ranged from 0.274 to 0.817 in Hanwoo. We found that there were differences in allele frequencies in Hanwoo when compared with other cattle breeds. The calculated cumulative power of discrimination (CPD) was 99.999% when nine microsatellite loci were used for analysis in the individual identification system. Also the matching probability, the probability that two unrelated animals would show the same genotypes, was estimated to be $0.44{\times}10^{-9}$. Therefore, the nine markers used in this study will be used for individual identification in two million Hanwoo individuals.
