Identification of the SNP (Single Necleotide Polymorphism) of the Stearoyl-CoA Desaturase (SCD) Associated with Unsaturated Fatty Acid in Hanwoo (Korean Cattle)

  • Fatty acid composition of beef intramuscular tissue is an important trait because high proportions of mono-unsaturated fatty acid are related to favorable beef flavor. In this study, we investigated the effects of genetic factors, such as stearoyl-CoA desaturase (SCD), on beef carcass traits, including fatty acid composition, in the Hanwoo. Analysis of fatty acids in Hanwoo was performed using a breed raised in Gyeonbuk province (n = 395). Compared to the homozygote, the GA, CT, and CT genotypes of exon 5 in the SCD polymorphism showed a higher content of oleic acid (p<0.05) and higher contents of mono-unsaturated fatty acid (p<0.05) and marbling scores (p<0.05) in intramuscular fat. Results of haplotype analysis showed a significant presence of unsaturated fatty acids and marbling score in the $ht1^*ht2$ and $ht2^*ht2$ groups (p<0.05). Furthermore, haplotype effects more powerful than a single gene were also observed. These ht1 and ht2 types also showed a significant difference in unsaturated fatty acids and marbling score, affecting beef flavor in the Hanwoo groups. Therefore, it can be inferred that the ht1 and ht2 types might be valuable new markers for use in improvement of Hanwoo.

Identification of Hanwoo and Holstein meat using MGB probe based real-time PCR associated with single nucleotide polymorphism (SNP) in Melanocortin 1 receptor (MC1R) gene (소 모색관련 MC1R 유전자의 SNP와 관련한 MGB probe에 기초한 real-time PCR을 이용한 한우육과 Holstein육의 판별)

  • The melanocortin 1 receptor (MC1R) plays an important role in regulation of melanin pigment synthesis within mammalian melanocytes. Mutations within the gene encoding MC1R have been shown to explain coat color variations within several mammalian species including cattle. To develope a rapid and accurate method for the identification of Hanwoo meat, we performed a single nucleotide polymorphism (SNP) analysis in Melanocortin 1 receptor (MC1R) gene using TaqMan$^{(R)}$ MGB probe-based real-time PCR. Two specific probes (one for Hanwoo and the other for Holstein and Black angus) were designed. At the 5' end of 2 TaqMan$^{(R)}$ MGB probes, 6-carboxyfluorescein (FAM) was labeled for Hanwoo, and VIC for Holstein and Black angus. As a result, Hanwoo samples showed FAM-positive signal only, whereas other samples showed VIC-positive. This result suggests that the TaqMan$^{(R)}$ MGB probe based real-time PCR technique would be very accurate, easy and reproducible method to discriminate between Hanwoo meat and Holstein/Black angus meat.

Production of In-Vivo Embryos by Superovulation and Result of Transfer with Fresh or Frozen Embryos for Hanwoo and Holstein Cattle (한우 및 젖소에서 과배란 처리를 이용한 체내수정란 생산과 신선 및 동결 수정란 이식 결과)

  • To investigate the feasibility of embryo transfer technology to promote productivity of cattle, 36 cows(18 Holstein, 18 Hanwoo) were superovulated. Fresh embryos were transferred to 25 recipients(14 Holstein, 11 Hanwoo), whereas frozen embryos were transferred to 17 recipients(10 Holstein, 7 Hanwoo). Two embryos were transferred at a time to 13 recipients(9 Holstein, 4 Hanwoo) to produce twin calves. 1. 75.0% of donor cattle were reacted to hormonal treatment far superovulation. 2. The rate of embryo recovery by non-surgical method for Holstein and Hanwoo was 90.4 and 95.8% in comparison with numbers of corpus luteum. 3. Of all the ova collected non-surgically, the rate of viable blastocyst was 66.4% and the rate of transferrable blastocysts was 48.6%. 4. The rate of embryo collection by one-catheter method was 75.0%. 5. The rate of pregnancy/delivery following embryo transfer with fresh embryos was 60.0%. 6. The rate of pregnancy/delivery following embryo transfer with frozen embryos was 35.3%. 7. In embryo transfer to produce twin calves, the rate of pregnancy/delivery was 28.6% with fresh embryos and 16.7% with frozen embryos.

Establishment of Genetic Characteristics and Individual Identification System Using Microsatellite loci in Domestic Beef Cattle (초위성체 DNA표지인자를 이용한 국내 육우집단의 품종특성 및 개체식별 체계설정)

  • DNA marker information is used to identify or distinguish cattle breeds or individual animal. The purpose of this study was to apply Bovine Genotypes Kit Version 1.1/2.1 to bovine DNA samples (National Institute of Animal Science) taken from Australian / American beef (n=148), Holstein beef (n=170) and Hanwoo cattle (n=177) bred in Jeongeub, Jeonbuk, Korea, so that it could distinguish Hanwoo breed. The Bovine Genotype Kits consist of 16 ISAG MS markers, which were used to build a database of genotypes in each group. Genotyping results were analyzed using MS Tool kit and Phylip program to create phylogenetic tree. The GeneClass 2.0 was used to estimate breed identification. These analyses found that this kit had 100% capacity to distinguish Hanwoo beef, 95.3% capacity to differentiate Australian / American beef and 90% capacity to identify Korean Holstein steer beef. Hence, it is expected that 16 commercial microsatellite markers is useful to categorizegenetic characteristics of Hanwoo breed and also identify Hanwoo individuals and the origin of beef. In particular, it is expected that these markers will be advantageous in discriminating domestic Holstein beef from Australian / Americanbeef.

Association of Microsatellite Marker in FABP4 Gene with Marbling Score and Live Weight in Hanwoo

  • The bovine fatty acid binding protein 4 (FABP4) plays an important role to uptake intracellular fatty acid. It has been previously reported as a positional candidate gene for marbling score in livestock. The re-sequencing of FABP4 gene detected a polymorphic AT repeated sequence in intron II of FABP4 gene. Allelic distribution for this microsatellite marker was examined in other cattle breeds. A total of 8 alleles were detected with diverse repeat units (14 to 21 AT repeat) in Hanwoo and 7 breeds. Of the 8 alleles, the predominant alleles were $[AT]_{16}$, $[AT]_{18}$ and $[AT]_{19}$ in the Hanwoo and 7 cattle breeds. The linear mixed model for genotypic effect (3237AT) on carcass traits showed a significant effect on marbling score (MAR P=0.025) and live weight (LWT; P=0.04) in the 583 Hanwoo cattle population. Live weight (LW) was highest in the homozygous $(AT)_{17}$ genotype ($557.5{\pm}6.94$) and lowest in the heterozygous $(AT)_{16/17}$ genotype ($521.7{\pm}7.70$). On the other hand, the homozygous $(AT)_{17}$ genotype ($3.0{\pm}0.15$) has the highest effect on marbling score and the lowest effect was in homozygous (AT)$_{18}$ genotype ($2.2{\pm}0.15$). The marbling score difference between both groups was 0.8 which is around two times higher than SNP genotype effect on marbling score in Limousin $\times$ Wagyu crosses.

PCR-based Detection of Bovine Papillomavirus DNA from the Cutaneous Papillomas and Surrounding Environments in the Korean Native Cattle, Hanwoo

  • Two 1-year old calves of Korean Native cattle (Hanwoo) presented cutaneous papillomas on the face and neck. Type 2 bovine papillomavirus (BPV-2) was identified in the cutaneous papillomas based on BPV-specific PCR and subsequent DNA sequencing analysis results. Using DNA samples extracted from two affected calves and unaffected animals reared in the same stable, BPV-2 was not only detected in the cutaneous papillomas of affected animals based on BPV-specific PCR analysis, but also detected in normal skins, hairs, and their environments based on nested PCR analysis. BPV-2 was also detected in DNA samples isolated from animals and environments of that distinct stable with affected calves. However, no BPV-2 was detected in the drinking water of both stables (infected and unaffected). These findings concluded that BPV-2 was transmitted by direct or indirect contact, not by drinking water. This is the first report to show molecular evidence of BPV-2 infection. Rapid and precise molecular identification can be used to screen BPV-2 in cattle farms to understand the biological roles of BPV in animal diseases.

An Outbreak of Blindness Caused by Hypovitaminosis A in Korean Native Cattle (한우에서 vitamin A 결핍증에 기인한 맹안의 집단발생)

  • This report described an outbreak of blindness caused by hypovitaminosis A in Hanwoo (Korean native cattle) in Gyeongsangbukdo province, Korea. The diagnosis was based on clinical findings, analysis of diet, and concentration of vitamin A and carotene in serum and/or feedstuff. Thirty eight of 48 cows were found to be affected. The causative diet contained higher levels of crude protein, net energy and crude fat and lower level of total fiber and crude fiber. Surprisingly, vitamin A was not detected in the causative diet. Treatment of vitamin A to Hanwoo that had blindness did not induce further progress of the symptom. Furthermore, there were no additional diseases detected. Based on the observations described above, this case was diagnosed as hypovitiminosis A.

Treatment of acute bovine theileriosis in grazing Korean native cattle (방목중인 한우에서 발생한 급성 타일레리아증 치료)

  • Bovine theileriosis caused by Theileria sergenti is a tick-borne hematoprotozoan disease that is characterized by chronic anemia and fever in cattle. In this study, results of microscopic examination and PCR detection confirmed 17 Korean native cattle with emaciation and fever as acute bovine theileriosis caused by T. sergenti. Buparvaquone was injected as treatment, but was proved to be an inappropriate measure according to our study. After 6 months of injection, clinical signs and hematological values were recovered, but T. sergenti was still identified in blood sample as a result of microscopic exam and PCR. These results suggest that continuous management is necessary to control bovine theileriosis. Therefore, findings of this study may provide significant guideline on the control of bovine theileriosis.

Characteristics of Livestock Manure in Hanwoo Farms Centered in Gyeongsangbuk-do Province (경북지역을 중심으로 한우농가의 축분 특성 조사)

  • This study focused on the characteristics of manure in five beef cattle farms and two breeding farms in Hanwoo centered in Gyeongsangbuk-do province. The results of analysis and evaluation are as follows; First, the pH of manure in all the farms in this study was within the appropriate range of 8.81 to 9.45. Second, the dry matter content varied from 44.7% to 70.5% for all the farms. Third, the total nitrogen content in all the farms was 2.05-3.04%, which is higher than the reported range; however, it could play a role as a soil improvement agent when used as compost. Fourth, although the exact range of ammonia-N (VBN) content was not reported, results analyzed in all the farms showed that the level of impact on the environment was insignificant. In conclusion, soil and environment improvement effects are expected to be effective if the manure at Hanwoo farms in the Gyeongbuk area are used as compost through appropriate management.

Effect of Mixed-Sowing of Legume and Applying of Cattle Manure on the Productivity, Feed Values and Organic Hanwoo Carrying Capacity of Rye (Secale cereale L.) (콩과 사료작물의 혼파와 우분의 시용이 호밀의 생산성, 사료가치 및 단위면적당 유기한우 사육능력에 미치는 영향)

  • This study was conducted to investigate the effect of mixed-sowing of legume forage and applying of cattle manure on the productivity of rye in order to produce the organic roughage to be effectively reducing the environmental contamination and to provide Hanwoo rearing farmer with safe organic animal products. The present study also aimed to evaluate optimal applying level of cattle manure and carrying capacity of Korean native cattle (Hanwoo) per unit area. The annual amount of crude protein (CP) of rye in Gyeongju were higher than those of in Gyeongsan and Yeongju, and the mixed-sowing of forage pea was the highest (p<0.05) compared with the single-sowing. The amount of CP were increased proportionately corresponded to the applying level of fermented cattle manure, and found significantly (p<0.05) high at the level of 100 and 150 kg/ha groups. With feed value of forage, CP was tended to be higher in the mixed-sowing of legume than the single-sowing of rye at the wintering experimental sites of legume, and in particular the forage peas was higher (p<0.05) than other crops. Carrying capacity of organic Hanwoo (head/ha) was higher (p<0.05) in Gyeongju (3.28 head) than that in Gyeongsan (1.94) and Yeongju (1.50). The carrying capacity in the single-sowing of rye also was lower than that in the mixed-sowing of legume, and the forage peas was the highest among the mixed-sowing groups (p<0.05). Overall, the present results showed that it would be recommended to take into account the wintering for mix-sowing of legume. The fermented cattle manure may be applied for legume in the wintering unavailable regions. In addition, the applying the fermented cattle manure at the level of 100~150kg/ha and the mix-sowing of legume with forage peas may increase the productivity per unit area and feed value including CP for improving carrying capacity of organic Hanwoo.
