Semen Properties of a Hanwoo bull persistently infected by BVDV (한우에서 BVDV 지속감염우의 정액 성상에 관한 연구)

  • BVDV causes significant infections in ruminants, resulting in reproductive disorders, diarrhea, reduced milk production and enormous damage to farms. In particular, identification and culling of persistent infectious calf is an important task to eliminate infectious nidus in cattle households. However, studies on physiological characteristics of PI bull are still insufficient to understand reproductive effects of BVDV. In this study, one PI bull was confirmed in herd and complete blood analysis was performed. The lymphocyte count of PI at age 4 was below the normal range and the number of WBCs was also in the lower level of normal range in blood. The sperm number produced by PI male becomes lower and the viability of fresh sperm comes to poor with ages (P<0.05). The sperm abnormality was also increased, especially in nuclear vacuoles of head and droplets of midpeace (P<0.05). The PI male becomes infertile due to poor semen quality at age 4. With these results, we concluded that BVDV in PI bull cause decreased sperm cell and abnormality in semen so causes infertility. However, it appears that BVDV could not be transmitted by indirect contact of PI bull, because there was no evidence of BVDV infection in the herd, when regular vaccination program was applied.

Effect of bovine recombinant somatotropin protein on milk yield in Hanwoo (소 재조합 somatotropin 단백질을 이용한 한우 유량 증진 효과)

  • This study was carried out to increase the breeding rate using sustained recombinant bovine somatotropin (bST) and examine the reproductive disorder and disease prevalencies of cow and calf, respectively. Sixty cows were allotted randomly to three groups in randomized complete block design; (1) No treatment, (2) 500 mg bST two times + 250 mg bST SC injection five times from 3 weeks before to 9 weeks after parturition every two weeks, (3) 500 mg bST two times before 3 weeks parturition + 250 mg bST SC injection three times from five to 9 weeks after parturition every two weeks. Developmental data of birth weight, weaning weight, total gain and average daily gain in control was $22.9{\mu}0.45$ kg, $88.0{\mu}1.13$ kg, $65.1{\mu}0.74$ kg and $0.54{\mu}0.08$ kg, respectively. There is no significant difference between Group 1 and 3 in the examined factors. However, there is significant difference between Group 1 and 2 in the birth weight ($24.4{\mu}0.88$ kg), weaning weight ($101.0{\mu}1.77$ kg), total gain ($76.7{\mu}0.9$ kg) and average daily gain ($76.7{\mu}0.9$ kg). Total gain was showed higher in Group 2 and 3 than Group 1 in the >2nd parity, and in Group 2 than Group 1 and 3 in the 1st parity. There is no significant difference in the BCS, no return days and disease occurrence of cow and calf by bST treatment. Number of service per conception was slightly increased in the Group 3 compared to Group 1 and 2. To increase the breeding ability of Hanwoo, it is recommended that 500 mg bST two times + 250 mg bST injection five times from 3 weeks before to 9 weeks after parturition every two weeks.

Comparison of Fatty Acid Composition of Hanwoo Beef by Different Quality Grades and Cuts (한우육의 육질등급 및 부위 별 지방산 조성 비교)

  • The fatty acid composition of Hanwoo beef was investigated by different quality grades and cuts. Five cuts (strip loin, loin, chuck roll, top round, brisket) were obtained from 15 Hanwoo beef cattle [bulls and steers, 24-30 mon old]. For each quality grade of $1^{++}$, $1^+$, 1, 2, 3, three animals were selected. The contents of C18:0 in chuck roll, strip loin, brisket, top round and loin were significantly higher in 3 quality grades than those of the other grades respectively (p<0.05). The C18:1n9 content in the chuck roll was not significantly different among the different grades, but those of strip loin, top round, brisket, and loin were significantly higher in $1^{++}$ quality grade than those of 3 quality grade (p<0.05). The C18:3n3 content was significantly higher in brisket of $1^{++}$ quality grade and loin of 3 quality grade than the other grades for the same cut (p<0.05). However, those of strip loin, top round and chuck roll were not significantly different (p>0.05) among the different grades. The C18:3n6 content in chuck roll and loin was significantly higher for 2 quality grade than in those of the other grades (p<0.05). For five cuts, total SFA contents for 3 quality grade were significantly higher than in other cuts. The total MUFA content of $1^{++}$ quality grade was significantly higher than in 3 quality grade (p<0.05). The total n-3 PUFA content was not significantly different in chuck roll, strip loin and top round; however, those of brisket were significantly higher in $1^{++}$ quality grade (0.15%) and those of loin were significantly higher in 3 quality grade than in the other grades (p<0.05). The total n-6 PUFA content was significantly higher in 3 quality grade than in those of the other grades (p<0.05).

Effects of Total Mixed Rations on Ruminal Characteristics, Digestibility and Beef Production of Hanwoo Steers (섬유질배합사료 급여가 비육후기 거세한우의 반추위 발효성상, 소화율 및 산육성에 미치는 영향)

  • This experiment was carried out to compare the effects of feeding commercial formula feed and rice straw separately (control) versus a total mixed ration (TMR) on productivity of Hanwoo steers in late stage of fattening and on ruminal fermentation characteristics and digestibilities. Ruminal digesta from the cannulated cattle were sampled at 0, 1, 2, 3, 5, 8 hour after feeding. The steers fed TMR consumed 7.4kg per day and there was no difference between feeding systems. Daily weight gain was not significantly (P>0.05) between feeding systems, however, TMR group showed lower daily gain than control group. The amount of feed consumption per kg weight gain was higher in TMR group than control group (10.5kg and 9.7kg, respectively), resulting in a greater efficiency of feed utilization for gain. In the result of appearance rates of quality grade A were 33% higher for TMR group than those in control group. Appearance rates of grade 1 showed 56% and 75% when fed the control and TMR, respectively. Digestibilities of dry matter, crude protein, crude fiber and gross energy for TMR treatment were significantly higher (P<0.01) than those of control. Prior to feeding (0 h) and each subsequent hour, the TMR resulted in higher rumen pH (P<0.05) when compared with control ration. The concentration of NH3-N for TMR treatment maintained at higher level up to 8hr after feeding, especially increased up to 28.2mg/$d\ell$ during 1-2 hour which was two times (P<0.05) more than control. The amount of total VFA showed same trends between feeding systems. However, the ratios of branched chained fatty acid such as iso-butyric acid and iso-valeric acid for TMR treatment were significantly(P<0.01) higher than control for 3-5hr. Results showed that TMR in these trials is effective feeding system for fattening Hanwoo steers in the respect of ruminal characteristics, total tract digestibility and productivities.

Superovulation Response by FSH Treatments in Hanwoo (FSH 투여 용량과 방법Ul 따른 한우의 과배란 처리 효율)

  • The present study was carried out to assess the effect of superovulation response and efficiency of embryo production induced with injection of FSH dissolved in polyethylene glycol (PEG) in Hanwoo. Eighty-eight cows were divided into four groups. In group 1, cattle were intramuscularly treated with twice-daily administration of 50mg FSH for 4 days. Group 2 and 3 were subcutaneously single injection of 400mg and 200mg FSH dissolved in 30% PEG, respectively. Group 4 were subcutaneously single injection of 200 mg FSH dissolved in 30% PEG at 7 day after CIDR insertion. The number of corpus luteum (CL) in group 2 resulted in significantly (p<0.05) higher compared to group 1, 3 and 4 (18.5 vs. 11.2, 13.1 and 13.9, respectively). However, the number of total ova $(7.9{\sim}10.4)$, transferable embryos $(3.7{\sim}4.7)$, degenerate embryos $(1.9{\sim}3.5)$ and unfertilized ova $(1.8{\sim}2.7)$ did not differ among treatment groups. No difference was observed in pregnancy rate after transferring the recovered embryos among groups $(36.0{\sim}50.0%)$. In addition, blood progesterone concentrations at embryo recovery did not differ among all groups. In conclusion, although no differences were observed in the number of total ova, transferable embryos and pregnancy rate after transfer, a single injection of reduced dose of FSH (200mg FSH) at 7 day after CIDR insertion is more practical for superovulation treatments than frequent injection because of reduction of stress in Hanwoo and decreases of cost and laber.

Possible Application of Artificial Insemination Buffer for Increasing Production Efficiency of Female Cow Offspring

  • The present research was carried out to evaluate the possibility of increasing female offspring production ratios using artificial insemination buffer (AIB) before artificial insemination (AI). In this experiment, we optimized AIB composition, made an AIB gun and analyze factors affecting AI non-return rate after AIB treatment. The AIB was made with the base of Tris-buffer supplemented with L-arginine and several other chemicals that might reduce the motility of male sperm compared to the female counterpart, therefore, increasing the possibility of fertilization by female sperm. AIB must be deposited into $2^{nd}$ to $4^{th}$ cervix by AIB gun. After 15 min of AIB deposition, frozen semen was deposited into the same place. A total of 348 cattle were inseminated with AIB insemination, and there were no significant differences between AIB and traditional AI non-return rates (56.8% vs. 55.7%). The AI non-return rate in AIB group, however, differed significantly among 7 Hanwoo farms. The parturition numbers ($1^{st}$ to $7^{th}$) of cows did not affect AIB AI rate. The proportion of AIB AI success rates was significantly higher in Hanwoo cows than in dairy cows (61.0% vs. 48.7%), but the average AI success rate did not differ significantly between AIB and conventional AI (56.8% vs. 55.7%). The female offspring production rate in $2^{nd}$ to $4^{th}$ cervix deposition place was significantly higher than that in the uterus body (77.7% vs. 59.6%, p<0.05). The injection volume of AIB in 5 and 10 ml was significantly higher than that in 2 ml (77.7%, 78.7% vs. 51.8%, p<0.05), but there were no differences in AIB injection volume between 5 and 10 ml. The best exposure time of AIB in the cervix was 10 to 15 min rather than 5 min (79.2%, 77.2% vs. 52.6%, p<0.05). AIB therefore needs to have an exposure time of at least over 10 min for a higher production rate of female offspring. In conclusion, AIB could be used in AI industry to increase the female offspring ratio and AIB AI can increase the AI success rate.

The Effects of Consecutive Superovulation in Hanwoo and Holstein Cattle (한우와 젖소에서 연속적인 다배란 처리의 효과)

  • 본 연구는 한우와 홀스타인 젖소에서 연속적인 다배란 처리 시 효율성 및 부작용에 대한 문제점을 연구하고자 다른 종간 다배란 처리효과를 비교, 조사하였다. 연속적인 다배란 처리를 위하여 estradiol benzoate(EB)와 CIDR plus, FSH 호르몬을 이용하여 한우에서는 17두, 젖소에서는 9두를 2회 이상 반복 이용하였다. 다난포 발생을 위해서는 FSH 호르몬을 이용하고 호르몬 처리 동기화를 위해서는 CER plus 기구를 질 내 삽입하였으며, 주요 조사 항목으로는 번식기관의 문제점, 배란 효율, 회수된 수정란 수, 우수 수정란 수 등을 분류하고, 종합적으로 종간 연속적인 다배란 효과를 비교, 조사하였다. 한우에서는 17두 중 4두(23.5%)에서 자궁경관의 협착이 발생하였고, 자궁 내에 손상이 발생하여 채란이 되지 않는 경우도 4두(23.5%)의 발생이 조사되었으나, 상대적으로 젖소에서는 9두 중 4두에서 일시적으로 배란이 전혀 일어나지 않는 경우가 있었다. 연속 처리 후 채란된 수정란의 수를 종합해 보면, 젖소에서는 평균 $4.3{\pm}3.4$개, 한우에서는 $2.7{\pm}2.5$개였으며, 종간 호르몬 효율과 연관된 배란율 조사에서는 한우에서는 89.7%, 젖소에서는 80.4%의 성적을 보였다. 또한, 3회까지 연속 다배란 처리를 한 결과, 한우나 젖소에서 채란된 수정란의 수는 비슷한 수준으로 채란되었다. 이와 같은 성적을 종합해 볼 때 EB제나 CIDR plus, FSH를 이용하여 1개월 주기로 연속적인 수정란 채란을 위한 다배란 처리가 소의 종에 무관하게 가능하다고 사료되지만 일시적으로 배란이 일어나지 않는 소에 대한 문제나 번식기관의 손상, 채란되는 수정란의 수가 적다는 것에 더 많은 연구가 필요하다고 사료된다.

Optimization of In Vivo Embryo Production and Pregnancy following Embryo Transfer in Hanwoo Cattle

  • Embryos formed in vivo were collected from 171 donors housed in Chung Cheong Buk-Do Institute of Livestock and Veterinary Research of the Chungbuk community during the years 2009~2012. We evaluated annual embryo collection, effect of follicle stimulating hormone (FSH), controlled internal drug release (CIDR) and prostaglandin (PG) administration to the donor for superovulation and controlling the estrus cycle, seasonal effects of embryo collection and compared the number of embryos recovered as per the collection days and pregnancy rate. In all, 1,243 embryos were collected from 118 donors with an average of $7.31{\pm}5.35$ embryos per donor, out of which 69.4% were transferable. Dosages of FSH required for inducing superovulation in various donors were compared. Average number of embryos collected from donors administered with 30 AU of FSH ($7.13{\pm}5.74$ per donor) was not significantly different from that of donors who were given an injection of 24 AU of FSH ($7.53{\pm}4.91$ per donor). However, the percentage of transferable embryos in the 30AU FSH-administered group (63.2 %, 449 of 711) was higher than that in the 24AU FSH-administered group (77.8%, 414 of 532). In the group of donors under a natural estrus cycle, the FSH dose administered did not influence the number of transferable embryos produced ($7.49{\pm}6.25$ per donor for 30 AU of FSH vs $7.49{\pm}4.92$ per donor for 24 AU of FSH). However, in donors administered with CIDR and PG for controlling the estrus cycle, the FSH dose affected the average number of transferable embryos collected ($4.25{\pm}2.87$ per donor for 30 AU of FSH vs $8.50{\pm}6.36$ per donor for 24 AU of FSH). We collected embryos from donors 6, 7 or 8 days after artificial insemination (AI). Results showed that the percentage of transferable embryos among those collected 8 days after AI was significantly higher than that among embryos collected 6 or 7 days after AI. Seasonal variations did not affect number of recovered embryos and pregnancy rates in natural estrus cycle and CIDR treatment groups (48.28% and 42.55%) but higher than pregnancy rate of frozen embryos (19.63%). These results indicated that administration of FSH beyond a threshold dose (at least 24 AU) has no beneficial effect on the production embryos and that collection of embryos 7~8 days after AI is optimal for embryo recovery. CIDR treatment induced superovulation in short term and had no influence on the natural estrus cycle. Finally, although good-quality embryos were transferred, freezing significantly reduced the pregnancy rates after transfer.

Characteristics of the Absorption of Cherry Tomato by the Application of Chelated Calcium and Germanium (킬레이트화 칼슘 및 게르마늄의 방울토마토 시용에 따른 흡수 특성)

  • Bloods from slaughtered Korean native cattle (Hanwoo : Bos taurus coreanae) can be used in agriculture with various beneficial effects on the environment. This study was performed to find out the effect of the application of calcium and germanium, which are chelated with peptides made of cattle bloods on the absorption of them by cherry tomato (Lycopersicon esculentum Mill.). Peptide was purified by enzymatic hydrolysis and ultrafiltration from serum protein, which is composed of Asp, Glu, Lue etc. Chelated Ca and Ge were produced by reacting the peptide with $CaCl_2$ and Ge-$132^{(R)}$. Then, they were applied to cherry tomato during cultivation period at the rate of $300mL{\times}3times$ (10 days interval) for each tomato tree. Application of chelated-Ca increased the Ca contents in leaves and fruits of cherry tomato showing as 19.9% and 23.4% in newerly prepared chelated-Ca-200 ($200mg\;L^{-1}$), 8.1% and 6.8% in commecrial Calciolid Ca-300 ($300mg\;L^{-1}$) compared to 3152.6 and $63.2mg\;kg^{-1}$ in control, respectively. Application of chelated-Ge showed the increase in the germanium contents in both leaves and fruits of cherry tomato by over 6 times than those of control and over 4 times than Ge $132^{(R)}$-20 application. As a result of this study, it seemed that the application of chelated-Ca and chelated-Ge would be more effective than the existing commertial one in the absorption of calcium and germanium by cherry tomato.

Effect of Embryo Factors on Pregnancy and Abortion Rate after Transfer of In Vitro Produced Korean Native Cattle Embryos (한우 체외수정란이 이식된 수란우의 임신과 유산에 영향을 미치는 수정란측 요인)

  • In this study, we investigated the effects of number, grade, stage and day of embryos on pregnancy and abortion rates of Holstein heifer recipients after transfer of in-vitro produced Korean Native Cattle (Hanwoo) embryos. When one, two or three blastocysts were transferred to recipients, pregnancy (32.0, 44.0 and $32.0\%$) and abortion (12.5, 13.6 and $6.3\%$) rates did not differ among treatments. The pregnancy and abortion rates of different grades of blastocysts were $40.8\~44.2\%\;and\;9.4\~16.3\%$, respectively. However, they were not significantly different. According to the stage of blastocyst, the pregnancy rates were similar between $39.1\%\;and\;45.3\%$. However, the abortion rate in hatched blastocyst stage $(20.8\%)$ was significantly higher than that in late blastocyst stage $(9.0\%)$. The pregnancy rates in day $7\cdot7$ and day $7\cdot8$ blastocyst (47.1 and $53.9\%$, respectively) was significantly higher than that in the day $9\cdot9\;and\;8\cdot9$ blastocyst (29.8 and $27.7\%$, respectively). The abortion rate was the highest in the day $9\cdot9$ blastocyst $(30.8\%)$ among treatments, and it was significantly different compared to that in the day $7\cdot7$ blastocyst $(10.1\%)$. Therefore, this study suggested that the pregnancy rate was affected by only day of embryos, while the abortion rate was affected by stage and day of embryos.
