Analysis of Bovine Seminal Plasma Proteins from Korean Native Cattle, Hanwoo, and Korean Native Brindle Cattle

  • After spermatogenesis, spermatozoa come in contact with fluids in the epididymis where they mature. During ejaculation, spermatozoa are mixed with secretions from prostate gland, vesicular glands, and bulbourethral glands. During natural mating, seminal plasma is deposited in the female reproductive tract eliciting various physiological and immunological responses. With the advances in proteomics, the components of seminal plasma have been identified and the information may be valuable in identifying markers for fertility. Components of seminal plasma that affect fertility have been discovered and the mechanism of action of these factors has been determined. The objective of this study was to determine the specific seminal plasma proteins from Korean native cattle, Hanwoo, and Korean native brindle cattle (KNBC) with the long term goal of improving fertilization rate. After SDS-PAGE and 2-dimensional gel electrophoresis, proteins were identified by Q-ToF analysis. They include plasma serine protease inhibitor precursor and platelet-activating factor acetylhydrolase after SDS-PAGE. Number and density of the spots in 2-dimensional gels were higher in KNBC than Hanwoo. Proteins identified from the paired spots of both breeds include chain A, bull seminal plasma PDC-109 Fibronectin Type II module, BSP-30 kDa precursor, and Spermadhesin Z13 or its precursor. Interestingly, some proteins were identified from multiple spots. The functional differences of these diverse forms of the proteins may require further studies. With their previously reported roles in sperm capacitation by these proteins, the studies on the mechanism of action, ligand interaction and the variation in the genome may help improving fertility in cattle.

Characteristics and Health Benefit of Highly Marbled Wagyu and Hanwoo Beef

  • This review addresses the characteristics and health benefit of highly marbled Wagyu and Hanwoo beef. Marbling of Wagyu and Hanwoo beef has been increased in Japan and Korea to meet domestic consumer preferences. Wagyu and Hanwoo cattle have high potential of accumulating intramuscular fat (IMF) and producing highly marbled beef. The IMF content varies depending on the feeding of time, finishing diet, and breed type. IMF increases when feeding time is increased. The rate of IMF increase in grain-fed cattle is faster than that in pasture-fed cattle. Fatty acid composition are also different depending on breeds. Highly marbled Wagyu and Hanwoo beef have higher proportions of monounsaturated fatty acid (MUFA) due to higher concentrations of oleic acid. MUFAs have little effect on total cholesterol. They are heart-healthy dietary fat because they can lower low-density lipoprotein (LDL)-cholesterol while increasing high-density lipoprotein (HDL)-cholesterol. Clinical trials have indicated that highly marbled beef does not increase LDL-cholesterol. This review also emphasizes that high oleic acid beef such as Wagyu and Hanwoo beef might be able to reduce risk factors for cardiovascular disease.

한우 종모우의 지방괴사증에 대한 Isoprothiolane 투여효과 (Effects of Isoprothiolane administration on fat necrosis of Hanwoo(Korean Cattle) sire)

  • The effect of isoprothiolane(di-isopropyl-1,3-dithiolan-2-ylidenemalonate) aganist fat necrosis in Hanwoo(Korean Cattle) sire was evaluated. The 10 heads of Hanwoo sire suffering from fat necrosis were given 50mg/kg body weight of isoprothiolane(0.2g/kg of Fujix, Japan) orally once a day for 8 weeks. In 30% of these, the size of the necrotic fat masses had decreased significantly 7 months after the adminstration. Isoprothiolane did not affect on live body weight and semen characteristics. However the sire affected with fat necrosis had higher MCHC(Mean corpuscular hemoglobin concentration) than normal sire in hematologic values 10 weeks after administration. Number of RBC(red blood cell) and PCV(packed cell volume) 10 weeks after administration had been increased than those before administration(p < 0.05). The serum concentrations of creatinine, triglyceride, and total cholesterol 10 weeks after administration were higher than those before administration while the concentration of glucose was vice versa. The isoprothiolane may reduce the oxidation of glucose, increase the glucose transfer to lipids, and increase blood supply to necrotic masses. These results indicate that isoprothiolane may be useful as the therapeutic agent aganist fat necrosis.

Correlation Analysis between the Breeding Value of Carcass Traits in Hanwoo (Korean Brown Cattle), Bos Taurus, L. and Spot Intensity on Two-dimensional Gel Electrophoresis

  • In order to investigate the genetic marker associated with economic performance in Hanwoo (Korean Brown Cattle), proteomic approach was used. Breeding values were estimated from performance tested steers. The top 20 and bottom 19 steers based on carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS) evaluation for one progeny testing period was used. Meat samples dissected from longissimus dorsi muscles were taken from the slaughter house and analyzed for two-dimensional gel electrophoresis. A total of 102 significant spots out of total 146 on each gel were detected and compared with the reference gel (synthetic gel) to be evaluated. Four candidate spots for marbling score were identified: 205, 84, 204 and 198. The study confirmed the relationship between breeding values of economic traits of Hanwoo cattle and spot intensity.

Comparison of the Microsatellite and Single Nucleotide Polymorphism Methods for Discriminating among Hanwoo (Korean Native Cattle), Imported, and Crossbred Beef in Korea

  • The identity of 45 Hanwo and 47 imported beef (non-Hanwoo) samples from USA and Australia were verified using the microsatellite (MS) marker and single nucleotide polymorphism (SNP) methods. Samples were collected from 19 supermarkets located in the city of Seoul and Gyeonggi province, South Korea, from 2009 to 2011. As a result, we obtained a 100% concordance rate between the MS and SNP methods for identifying Hanwoo and non-Hanwoo beef. The MS method presented a 95% higher individual discriminating value for Hanwoo (97.8%) than for non-Hanwoo (61.7%) beef. For further comparison of the MS and SNP methods, blood samples were collected and tested from 54 Hanwoo ${\times}$ Holstein crossbred cattle (first, second, and third generations). By using the SNP and MS methods, we correctly identified all of the first-generation crossbred cattle as non-Hanwoo; in addition, among the second and third generation crossbreds, the ratio identified as Hanwoo was 20% and 10%, respectively. The MS method used in our study provides more information, but requires sophisticated techniques during each experimental process. By contrast, the SNP method is simple and has a lower error rate. Our results suggest that the MS and SNP methods are useful for discriminating Hanwoo from non-Hanwoo breeds.

Determination of Genetic Diversity among Korean Hanwoo Cattle Based on Physical Characteristics

  • This study was conducted to establish genetic criteria for phenotypic characteristics of Hanwoo cattle based on allele frequencies and genetic variance analysis using microsatellite markers. Analysis of the genetic diversity among 399 Hanwoo cattle classified according to nose pigmentation and coat color was carried out using 22 microsatellite markers. The results revealed that the INRA035 locus was associated with the highest $F_{is}$ (0.536). Given that the $F_{is}$ value for the Hanwoo INRA035 population ranged from 0.533 (white) to 1.000 (white spotted), this finding was consistent with the loci being fixed in Hanwoo cattle. Expected heterozygosities of the Hanwoo groups classified by coat colors and degree of nose pigmentation ranged from $0.689{\pm}0.023$ (Holstein) to $0.743{\pm}0.021$ (nose pigmentation level of d). Normal Hanwoo and animals with a mixed white coat showed the closest relationship because the lowest $D_A$ value was observed between these groups. However, a pair-wise differentiation test of $F_{st}$ showed no significant difference among the Hanwoo groups classified by coat color and degree of nose pigmentation (p<0.01). Moreover, results of the neighbor-joining tree based on a $D_A$ genetic distance matrix within 399 Hanwoo individuals and principal component analyses confirmed that different groups of cattle with mixed coat color and nose pigmentation formed other specific groups representing Hanwoo genetic and phenotypic characteristics. The results of this study support a relaxation of policies regulating bull selection or animal registration in an effort to minimize financial loss, and could provide basic information that can be used for establishing criteria to classify Hanwoo phenotypes.

Linkage Mapping and QTL on Chromosome 6 in Hanwoo (Korean Cattle)

  • The QTL(quantitative traits loci) linkage mapping of Hanwoo (Korean Cattle) chromosome 6 for daily gain and marbling score was performed using 378 individuals from 18 paternal half-sib families in Hanwoo. Hanwoo chromosome 6 were mapped to total length of 394.2 cM between 28 microsatellite loci using 36 microsatellite primers of BTA 6 linkage group. The QTL analysis for daily gain in Hanwoo showed 8 microsatellite loci (BM3026-5.66, EL03-5.58, BM4311-5.29, ILSTS035-4.50, BMS1242-4.37, BM1329-3.67, BM415-3.11, BMS2460-3.03) in larger than LOD score 3.0. Based on the QTL analysis for marbling score, LOD scores of 12 microsatellite loci (BM415-8.88, BM3026-7.15, ILSTS093-5.45, ILSTS035-4.91, EL03-4.69, BMS690-4.52, BM1329-4.43, BMS511-3.74, BMS1242-3.66, BMS518-3.65, BM4311-3.41, BMC4203-3.36) were found larger than 3.0.

Novel SNP in the coding region of the FTO gene is associated with marbling score in Hanwoo (Korean cattle)

  • The fat mass and obesity associated (FTO) gene plays an important role in the regulation of energy homeostasis, fat deposition and obesity. For this reason, the FTO gene is a physiological and functional candidate gene for carcass and meat quality traits in beef cattle. The objectives of this study were to identify SNPs in the exonic regions of FTO gene and to evaluate the association of these SNPs with carcass traits in Hanwoo (Korean cattle). In this study, we newly identified two exonic SNPs in Hanwoo population. The g.125550A > T SNP was located in exon 3 and the g.175675C > T SNP was located in exon 6. Genotyping of the two SNP markers was carried out using PCR-RFLP analysis in Hanwoo steers to evaluate their association with carcass traits. As a result, g.125550A > T SNP genotype was significantly associated with effects on marbling score. Animals with the AA and TT homozygous genotypes had a significantly higher marbling score (p < 0.001) than those with AT heterozygous genotype, and this was significant after Bonferroni correction of the significance threshold (p = 0.003). Dominance effect was also observed for the marbling score (P < 0.05) with higher marbling score of homozygous animals. However, no significant associations with meat quality traits were observed for the g.175675C > T SNP. Our results suggest that the exonic SNP g.125550A > T in the FTO gene may be used as a DNA marker for the selection of Hanwoo with higher marbling.

Analysis of seasonal effect on Korean native cattle (Hanwoo) birth weight

  • Recently, summer temperatures have frequently been abnormal in Korea owing to global warming. In summer, a decrease in feed intake rate and biological activity were observed in Hanwoo (Korean Native Cattle), leading to lower production rates in the industry. However, the precise scale of damage was not reported as with other animals of economic value. This study was conducted to investigate the effects of birth season on birth weight in Hanwoo. Data were collected from 100 local breeding farms from 2016 to 2019. A total of 41,081 Hanwoo calves were classified and analyzed by sex, year, month, and season (March-May, spring; June-August, summer; September-November, fall; and December-February, winter) of birth. The birth weight of Hanwoo calves differed according to birth month. The average birth weight of male calves was 30.47 kg and that of female calves was 28.16 kg. Hanwoo birth weight was the highest in March-born calves and the lowest in July-born calves. The birth weights of calves born in February, March, April, November, and December were significantly larger than those of calves born in July. In addition, the birth weight of Hanwoo calves from the summer was significantly lower than that of calves born in other seasons. Furthermore, Hanwoo steer slaughter age showed a negative correlation, whereas carcass weight had a positive correlation with birth weight. In the beef cattle industry, birth weight is a very important economic characteristic that is related to growth rate. These data will contribute toward planning the reproduction of Hanwoo and analysis of changes in characteristics of economic value owing to high temperatures.

한우와 젖소 초유로부터 분리한 Lactoferrin과 가수분해물의 항균활성 (Antimicrobial Activities of Lactoferrin and its Hydrolysate Obtained from the Colostrum of Hanwoo and Holstein Cattle)

  • 본 연구는 우리나라 재래종인 한우로부터 초유를 얻어 Lf을 분리정제한 후 한우 Lf와 젖소 Lf의 항균활성을 확인하였다. E. coli O111및 기타 미생물에 대한 항균성은 젖소 Lf가 한우 Lf 보다 높았으며, 젖소 Lf 가수분해물도 한우 Lf 가수분해물보다 마찬가지로 높았다. MIC에서는 E. coli O111 경우 젖소 Lf가 1.5mg/ml, 한우 Lf은 2.75mg/ml이며 젖소 Lf 가수분해물은 0.12mg/ml, 한우 Lf 가수분해물은 0.25mg/ml로 항균성 실험과 동일하게 젖소 Lf 가수분해물의 항균활성이 더 높은 것으로 나타났다. 한편 Lf과 lysozyme의 첨가는 항균활성을 상승시키는 효과를 나타내었다.
