Assessment of intestinal permeability of EGCG by piperine using Caco-2 cell monolayer system

  • (-)-epigallocatechin-3-gallate (EGGG), a flavonoid found in green tea, is known to have low bioavailability. In this study, we determine whether piperine, a natural bioenhancer, can increase the absorption rate of EGCG. Using a Caco-2 cell monolayer, permeability experiments were performed in Hanks' balanced salt solution (HBSS) and EGCG stability was adjusted to pH 6.5 and pH 5.5 by ascorbic acid treatment. When HBSS was adjusted to pH 6.5, EGCG remained at 94.78% for up to 2 h and remained at 86.04% after 4 h and the net efflux decreased compared to the control. As a result, uptake was significantly increased in the piperine co-administered group compared to the EGCG-alone group, showing that piperine increased the permeability of EGCG in the Caco-2 cell monolayer. These results suggest that piperine inhibits EGCG glucuronidation and efflux, allowing for greater absorption of EGCG.

  • Ti-Zr-Ni coatings deposited by low vacuum plasma spray technique consisted of nanometer-sized $W-Ti_{50}Zr_{35}Ni_{15}$ 1/1 cubic approximant and TiZrNi Laves phases as well as a low volume fraction of $ZrO_2$ phase. The shift of composition during deposition of the quasicrystalline powders and the presence of $ZrO_2$ phases are believed to be responsible for the reduced corrosion performances evaluated by means of electrochemical tests in a Hanks' Balance Salt Solution at $37^{\circ}C$.

  • 이온이 첨가된 BCP(biphasic calcium phosphate) 분말을 제조하기 위하여 $Ca(NO_3)_2{\cdot}4H_2O$, $(NH_4)_2HPO_4$를 출발 물질로 공침법(co-precipitation process)을 이용하여 합성하였다. Si-BCP 합성을 위하여 TEOS를 Mg-BCP 합성을 위해서는 $Mg(NO_3)_2{\cdot}6H_2O$를 사용하였다. 합성된 분말의 열처리 후 결정상을 분석한 결과 HAp와 ${\beta}$-TCP가 혼재된 상을 관찰할 수 있었다. 분광학적 분석에서도 인산칼슘의 $PO^{3-}_4$$OH^-$ 그룹에 해당하는 밴드도 관찰되었다. 이온이 첨가된 BCP 분말의 생체활성 거동을 평가하기 위하여 Hanks' Balanced Salt Solution(HBSS)에 침적시켜 시간에 따라 형상의 변화 및 결정상을 분석한 결과, 두 샘플 모두에서 빠른 표면 활성을 나타내었다. 세포 성장률 평가에서도 대조군에 비하여 세포성장률이 우수함을 관찰하였다.

  • 본 연구의 목적은 다공성의 티타늄 산화막에 불소를 처리한 결과를 평가하는 것이다. 디스크에 양극산화법을 통하여 다공성의 티타늄산화막표면을 얻은 후 불소를 처리하고 일정시간동안 Hank액에 침전을 시켰다. 양극산화를 통해 일정거칠기의 표면을 얻었으며 SEM과 XRD를 통하여 표면의 형상과 성분을 분석하였다. 생성된 표면은 빠른 표면 활성도를 보였으며, 적절한 거칠기와 좋은 골반응으로 골유착에 기여할 수 있을 것으로 여겨진다.

Ti-6Al-4V 합금 표면에 생성된 $TiO_2$ 나노튜브의 전석회화 처리 (Precalcification Treatment of $TiO_2$ Nanotube on Ti-6Al-4V Alloy)

  • 연구목적: 최근 치과용 임플란트의 임상 경향이 전체 치료기간을 줄일 수 있는 방법에 관심이 집중됨에 따라 불활성의 티타늄 임플란트 표면에 활성을 부여하기 위한 다양한 표면처리 방법이 검토되고 있다. 본 연구에서는 높은 강도가 요구되는 부위의 임플란트 재료로서 사용되고 있지만 표면 특성이 순 티타늄에 비해 떨어지는 Ti-6Al-4V 합금의 골전도성을 개선할 목적으로 시행되었다. 연구 재료 및 방법: $20{\times}10{\times}2\;mm$의 Ti-6Al-4V 합금판을 준비한 다음 $TiO_2$ 나노튜브를 형성하기 위해 DC 정전원 장치의 양극과 음극에 각각 시편과 백금판을 결선하고 0.5 M $Na_2SO_4$와 1.0 wt% NaF를 함유하는 전해액을 사용하여 전압 20 V와 전류밀도 $30\;㎃/cm^2$ 조건에서 2시간 동안 양극산화 처리하였다. $TiO_2$ 나노튜브 형성 후 산화 피막층의 결정화를 유도하기 위해 $600^{\circ}C$에서 2시간 동안 열처리하였고, 표면활성도를 개선하기 위해 0.5 M $Na_2HPO_4$ 수용액 24시간 침적과 $Ca(OH)_2$ 포화 수용액에 5시간 침적을 시행하였다. 준비한 시편의 표면 반응성을 조사하기 위해 pH와 무기이온의 농도를 사람의 혈장과 유사하게 조절한 Hanks 용액 (H2387, Sigma Chemical Co., USA)에 2주간 침적하였다. 결과: 20 V에서의 양극산화처리로 직경 48.0 - 65.0 ㎚ 범위의 무정형의 $TiO_2$ 나노튜브가 전체 표면에 걸쳐서 균일하게 생성되는 양상을 보였다. $TiO_2$ 나노튜브는 $600^{\circ}C$에서 2시간 열처리 후 상대적으로 강한 anatase 피크와 함께 rutile 피크가 관찰되었다. $TiO_2$ 나노튜브의 표면활성도는 0.5 M $Na_2HPO_4$ 수용액 24시간 침적과 $Ca(OH)_2$ 포화수용액에 5시간 침적으로 개선되었다. 열처리와 전석회화 처리 후 SBF에 침적한 결과, $TiO_2$ rutile 피크의 상대적 강도는 크게 증가되었지만 HA의 석출은 저하되는 경향을 보였다. 결론: 이상의 결과로 미루어 볼 때, 양극산화 처리한 $TiO_2$ 나노튜브는 $600^{\circ}C$에서의 열처리에 의해 피막층이 안정화되고, 0.5 M $Na_2HPO_4$ 수용액 24시간 침적과 $Ca(OH)_2$ 포화수용액에 5시간 침적으로 표면에 인산칼슘층을 형성하는 것이 표면활성도를 개선하는데 유효함을 알 수 있었다.

Fe-31Cr-27Ni-1.6Mo-1.5W-0.26N계 초내식성 스테인리스강의 생체적합성 및 부식특성에 미치는 Co함량의 영향 (Effect of Cobalt Contents on the Biocompatibility and Corrosion Properties of Fe-31Cr-27Ni-1.6Mo-1.5W-0.26N Alloy)

  • Super austenitic stainless steels shows the high PRE (Pitting Resistance Equivalent) number and the good corrosion resistance. This work controlled the Co contents in Fe-31Cr-1.7Mo-27Ni-0.25N alloys to elucidate the effect of cobalt contents on the biocompatibility and corrosion resistance. Increasing Co contents, the hardness of the annealed alloys tends to be reduced. In aged alloys, cobalt decreased the increments of hardness by aging treatment. Cobalt decreased the critical pitting temperature (CPT) in 6% $FeCl_3$ + 1% HCl solution, but improved the anodic polarization behavior in Hanks' balanced salt solution and artificial saliva solution. Repassivation rate in artificial body solutions was improved by increasing cobalt contents, but didn't show the linear relationship to PRE number of the alloys. The experimental alloys showed the non-cytotoxicity because of its high corrosion resistance.

음극환원법에 의한 Pure Ti의 전기화학적 열수처리 (Electrochemical hydrothermal treatment on Pure Titanium by the method of Cathodic reduction)

  • The purpose of this study was to examine the optimum condition of impulse during the anodic spark oxidation applying pulse current as well as to find the excellent condition for HA precipitation the after electrochemical hydrothermal treatment by cathode reduction method. After anodic spark oxidation, the anodized specimen and the Pt plate connected cathode and anode, respectively. Hydrothermal treatment performed at 90, 120, $150^{\circ}C$ for 2 hours in the electrolyte containing $K_2HPO_4$, $CaCl_2{\cdot}2H_2O$, Tris(Hydroxymethyl)-$(CH_2OH)_3\;CNH_2$(Aminomethane), and NaCl. The optimum impulse voltage for anodic spark oxidation was 350V. The optimum pulse cycle measured at 10 mS. The HA crystals precipitated excellently by cathode reduction at $150^{\circ}C$ for 2 hours. The phases of anatase, rutile, and HA coating on the surface of modified titanium surface immersed in Hanks' solution for 3weeks were detected by XRD measurement and the intensity of HA crystal phase has increased by temperature and time of hydrothermal treatment. According to the our experiments, we found that Pure Ti will be good materials of bioactivity and biocompatibility.

Effect of Ti Intermediate Layer on Properties of HAp Plasma Sprayed Biocompatible Coatings

  • The objective of this study was to improve properties of plasma sprayed HAp layer to titanium substrate by introducing an intermediate layer with two different methods. Before applying Zn doped HAp coating on titanium substrate, an intermediate layer was introduced by titanium plasma spray or titanium anodization. Heat treatments were conducted for some samples after titanium intermediate layer was formed. Zn doped HAp top layer was applied by plasma spraying. Three-point bending test and pull-off adhesion test were performed to determine the adhesion of Zn doped HAp coatings to substrates. Long-term credibility of Zn doped HAp plasma sprayed coatings on titanium was assessed by electrochemical impedance measurements in Hanks' solution. It was found that both titanium plasma sprayed and titanium anodized intermediate layer had excellent credibility. Strong adhesion to the titanium substrate was confirmed after 12 weeks of immersion for coating samples with titanium plasma sprayed intermediate layer. Samples with titanium anodized intermediate layer showed good bending strength. However, they showed relatively poor resistance against pulling off. The thickness of titanium anodized intermediate layer can be controlled much more precisely than that of plasma sprayed one, which is important for practical application.

Optimization of Experimental Conditions for In vitro P-glycoprotein Assay Using LLC-GA5 Cells

  • Identification of compounds that function as P-glycoprotein (P-gp) substrates or inhibitors can facilitate the selection and optimization of new drug candidates. The purpose of this study is to optimize the experimental conditions for in vitro P-gp assay using LLC-GA5 cells, which is a well-known transformant cell line derived by transfecting LLC-PK1 with human MDR1. The amount of rhodamine123 transported by the LLC-GA5 and LLC-PK1 cells was evaluated under the following experimental conditions: 3 different types of transport media, colchicine pretreatment or nontreatment of the cells in the culture media, and with and without poly-L-lysine coating of the culture plates. The assay sensitivity was found to considerably differ depending on the diluents used in the transport media. P-gp-mediated transport in LLC-GA5 cells was most clearly characterized in the Hanks' balanced salt solution based transport media. The sensitivity of P-gp-mediated transport was not changed by colchicine pretreatment or poly-L-lysine coating of the culture plates.

양극산화와 열수처리한 니오비움 금속의 표면특성 (Surface Characterization of Anodized and Hydrothermal Treated Niobium Metal)

  • This study was performed to investigate the surface properties of electrochemically oxidized pure niobium by anodic oxide and hydrothermal treatment technique. Niobium specimens of $10mm\times10mm\times1.0mm$ in dimension were polished sequentially from $\#600,\;\#800,\;\#1000$ emery paper. The surface of pure niobium sperimens was anodized in an electrolytic solution that was dissolved calcium and phosphate in water. The electrolytic voltage was set in the range of 250 V and the current density was $10mA/cm^2$. The specimen was hydrothermal treated in high-pressure steam at $300^{\circ}C$ for 2 hours using an autoclave. And all specimens were immersed in the in the Hanks' solution nth pH 7.4 at $37^{\circ}C$ for 30 days. The surface of specimen was characterized by surface roughness, scanning electron microscope(SEM), energy dispersion X-ray analysis(EDX), X-ray photoemission spectroscopy(XPS) test. The value of surface roughness was the highest in the anodized sample and $0.41{\pm}0.04\;{\mu}m$. The results of the SEM observation show that oxide layers of the multi porosity in the anodized sample were piled up on another, and hydroxyapatite crystal was precipitate from the surface of the hydrothermal treated sample. In the XPS analysis, O, Nb, C peak and small amounts of N peak were found in the polished specimens while Ca and P peak in addition to O, Nb, C and peak were observed in the hydrothermal treated sample.
